3.Cytocompatibility of oxygen plasma-treated polylactic acid and bioactive glass guided bone regeneration membrane

BACKGROUND:Currently, bioactive glass and polylactic acid have been used in clinical dentistry and plastic surgery; however, their therapeutic outcomes are not satisfactory, because the material properties have some limitations. OBJECTIVE:To explore the cytocompatibility of oxygen plasma-treated polylactic acid and bioactive glass guided bone regeneration membrane. METHODS:Bioactive glass and polylactic acid were used as the basic materials to prepare polylactic acid membrane, polylactic acid and bioactive glass composite membrane and oxygen plasme-treated polylactic acid and bioactive glass composite membrane, al of which were used to culture MG63 cels. Cel adhesion rate, cel proliferation rate and alkaline phosphatase activity of MG63 cels on these three kinds of membranes were observed. RESULTS AND CONCLUSION: With the growth of time, in these three groups of membranes, the cel adhesion rate and cel proliferation rate were al significantly increased. Alkaline phosphatase activity showed a decreasing trend after the first increase, and reached its peak at the 7thday of culture. The cel adhesion rate and cel proliferation rate in oxygen plasma-treated polylactic acid and bioactive glass group were significantly higher than those in the other two groups, while the cel adhesion and proliferation rates in polylactic acid and polylactic acid and bioactive glass groups were similar. At the 3rd day of culture, the alkaline phosphatase activity in the polylactic acid and bioactive glass group and oxygen plasma-treated polylactic acid and bioactive glass group was significantly higher than that in the polylactic acid group. At the 7th and 14th days, there was no significant difference in the alkaline phosphatase activity among these three groups. These results show that oxygen plasma-treated polylactic acid and bioactive glass composite membrane has good biocompatibility, which can better promote cel adhesion, proliferation and matrix secretion from osteogenic cels.

6.Experimental study on regeneration of articular cartilage defects with bone marrow stromal cells

[Objective]To retrieval a best cell factor which can induce bone marrow stromal cells (bMSCs) into chondrocyte in vitro and to explore an effective plan to repair rabbit's chondrocyte defect.[Method]Isolated bMSCs were cultured in vitro. rh Fibroblast Growth Factor 1(rhFGF-1)、rh Transforming Growth Factor-?1(rhTGF-?1)、rh Insulinlike Growth Factor-Ⅰ(rhTGF-Ⅰ) were utilizated. The proliferation of cells was detected by MTT assay, and the macroscopic histology , HE staining and immunohistochemical examinations were performed to seek the best cell factor. In vivo , to investigate the repair of the articular cartilage, bMSCs combined with fibrin glue and rhTGF-?1、rhIGF-I was compared with control group.[Result]The cells induced by rhTGF-?1 and rhIGF-I were similar to chondrocytes in morphology, and immunohistochemical examinations showed the cells possessed phenotype of chondrocytes. RhTGF-?1 and rhIGF-I could differentiate bone marrow stromal cells into cartilage cells in vivo, and repair the articular cartilage defect. The control group could not repair the articular cartilage defect efficiently.[Conclusion]rhTGF-?1 and rhIGF-I are best group which stimulate bMSCs to differenate into cartilage cells. BMSCs combined with fibrin glue and rhTGF-?1、rhIGF-I can repair the articular cartilage defect.
7.Selectivity treatment of patellar fractures with Cable-Pin system

[Objective]To study the effects of Cable-Pin system used as a treatment alternative for patellar fractures.[Method]Thirty-five cases of patellar fracture were operated with the Cable-Pin system from October 2005 to May 2007.Their diagnosis were 19 transverse close ractures and 16 comminuted of patella.There were 21 males and 14 females.Their ages ranged from 21 to 75 years old.After open reduction,the author used a drill bit vertically through the patella in a superior direction,drilled the threaded portion of the pin across the fracture site,inserted a second Cable Pin through the second hole in a similar manner.A horizontally directed drill hole anterior to the pins was created in the superior pole of the patella.The two cables were crossed in a fighre eight over the an-terior surface of the patella.The Crimper Tensioning Handle was,the cable carefully,the handles of the crimper were squeezed until the crimp was fully crimped.[Result]The 35 patients were followed up for 12 to 18 months(mean 13.2 m).Fractures healed in all the cases without such complications as loosening of screw and cable loop,loss of reduction or irritation to soft tissue.Restoration of the knee joint motion was excellent in 32 cases and good in 3 cases according to Bostman score system.[Conclusion]Cable-Pin system as a new option for the treatment of patella fracture,can result in reliable fixation and low rate of complications.

9.Morphological changes of left and right atrial myocytes in rheumatic heart disease patients with atrial fibrillation

6 months; Control group (n=8) without AF. Atrial muscle samples of the left and right atrial appendage were cut off during heart valve replacement. Morphological changes were observed by light microscopy and electron microscopy. Results ① The incidence rates of Aschoff body were 37.5% (3/8), 46% (5/11) , and 50% (6/12) in Group Ⅰ, Group Ⅱ, and the control group. There was no difference in Aschoff body between the left and right atrial myocytes. ② There was no difference in myocytic degeneration, interstitial hyperplasia, and lymphocytes infiltration between the left and right atrial myocytes in each group. ③ Myolysis, accumulation of glycogen, and fibrosis were detected by electron microscopy in the 3 groups. Changes of myolysis and fibrosis were more obvious in patients with AF than patients without AF. Conclusion There are similar pathological changes in the left and right atria in rheumatic heart disease patients with or without AF. Fibrosis of atrial myocytes may be involved in the occurrence and maintenance of AF.
