Chinese Journal of Radiological Medicine and Protection 2009;29(1):80-82
Objective To observe the efficacy and acute toxicity in non-myeloablative low-dose tatal body irradiation. Methods From January 2006 to January 2008, 27 cases of hematopoietic stem cell transplantation patients received non-myeloablative pretreatment program involving low-dose whole body irradiation. Pretreatment program: the total dose of TBI was 2 Gy, once completely. According to primary disease, physical condition, age, organ function and HAL coincide situation, the patients received different chemotherapy, including FUL, CTX, Ara-C, melphalan and so on. To prevent the graft-versus-host disease (GVHD), CSA/MMF was used. Results Hematopeietic reconstruction: the peripheral blood WBC in all 27 cases reduced to (0.05-0.9) × 109/L in 4 to 8 days, with the neutrophil count > 0.5 × 109/L in 8 to 22 days (median + 10.5 days), the platelet count > 30 × 109/L in 11 to 28 days (median + 14.5 days). The transplant rate was 85.2% (23/27). The fever rate was 65% and the infection rate was 37% (10/27). Hemorrhagic cystitis, hepatic veno-occlusive disease (HVOD), and other complications did not occurred. 4 cases developed aGVHD (15 %) and 5 cases developed cGVHD (19 %) out of 27 cases. Follow-up 3 to 22 months, 21 eases are still alive (77.8%). Conclusions Pretreatment with low-dose whole body irradiation of non-myeloablative hematopeietic stem cell transplantation proved to be simple and safe with fewer complications and broad indications. Program of TBI with total dose of 2 Gy (once completely) is a safe, less toxic and effective method. It could achieve the effectiveness of immune suppression.

Tianjin Medical Journal 2010;38(3):186-188
Objective:To detect the effects of porcelain-fused-to-metal(PFM)crown on the periodontal tissue of the restored teeth, and the relationship between the levels of intedeukin(IL)-1β,aspartate transaminase(AST)and alkaline phosphatase(ALP)in gingival crevicular fluid(GCF)and the health condition of periodontal tissue. Methods:Among 28 patients,34 maxillary incisors with PFM crowns were selected for the study. There were 19 Ni-Cr PFM crowns and 15 Au-Pt PFM crowns. The contralateral homonymy natural teeth were healthy and treated as control teeth. The patients were followed up 6 to 8 months after restoring crowns. The values of IL-1β,AST and ALP in GCF were analysed. Results:The concentrations of IL1β and ALP were significantly higher in GCF of Ni-Cr PFM compared with those of control, whereas no significant difference in Au-Pt PFM group compared with that of control. The level of GCF-AST was higher in Ni-Cr PFM and Au-Pt PFM group than that of healthy control.Conclusion:Ni-Cr PFM crowns have negative influence on periodontal tissue, but Au-Pt PFM have little influence on periodontal tissue of the restored teeth. The activity of IL-1β,AST and ALP of GCF are sensitive to reflect the effect of PFM crowns of different materials on periodontal tissue.
3.Differentiation of marrow stromal cells into neurons: current progress
Chinese Pharmacological Bulletin 2003;0(07):-
Bone marrow stromal cells(MSCs) have been shown to be extremely versatile.Recently,it is demonstrated that MSCs differentiate into neurons. Moreover, under the appropriate conditions,MSCs differentiate into special phynotype of neurons, such as 5-hydroxytryptamine-sensitive neurons, and dopaminergic neurons. The mechanism remains to be elucidated, and an overexpression of neuronal specific genes(such as Sox,Pax,Notch gene) may play an important role, especially RB/PBO gene. However,RB triggers cholinergic differentiation.Extrinsic factor such as concentration of cAMP plays certain role in differentiation.MSCs have also been shown to have certain therapeutic effect on neuronal degeneration disease. These cells have great potential because it is easy to isolate, culture, amplify, and no ethics neither immunologic rejection problem was concerned.

BACKGROUND: Differentiation of neural stem calls (NSCs) was mediated by many environmental factors. Several factors can induce NSCs to differentiate into neurons in varying degrees and it is now a focus on the control of NSCs differentiation.OBJECTIVE: To study the effects of combination of basic fibroblast growth factor (bFGF) and brain-derived neurotrophic growth factor (BDNF) on the differentiation of NSCs into neurons.DESIGN, TIME AND SETTING: The in vitro cytology observation was performed at the Neurotomia Laboratory of China Medical University in May 2008.MATERIALS: Three adult male SD rats were provided by Experimental Animal Center of China Medical University.METHODS: Dispositions to the rats were consistent with ethical standards of animals. The rat brain hippocampus was removed sterilely. After trypsin digestion, NSCs were cultured in serum-free medium. Cell suspension was prepared and diluted when the diameter of the fourth passage of clone sphere was 200 μm by mixture of DMEM/F12 containing 2% B27, 20 μg/L of epidermal growth factor (EGF), and 20 μg/L bFGF. Monoclonal calls were passagad. NSCs were divided into blank control, bFGF, BDNF and bFGF+BDNF groups by different growth factors added into the media. Fetal bovine serum of 0.1 volume fraction was added in blank control group. The media in the other three groups were added bFGF, BDNF and bFGF+BDNF respectively for 1 week. The concentration of bFGF was 10 μg/L and the concentration of BDNF was 200 μg/L.MAIN OUTCOME MEASURES: Immunocytochemistry staining was used to identify NSCs as well as to detect the differentiation of NSCs into neurons.RESULTS: The monoclonal calls expressed nestin and the differentiated call expressed neuron specific enolase and glial fibrillary acidic protein. Compared to blank control group, the proportion NSCs into neurons in the bFGF group, BDNF group and bFGF+BDNF group were much higher (t=3.409-7.558, P < 0.05), with the highest in bFGF+BDNF group (t =7.558, P < 0.05).CONCLUSION: Combination of bFGF and BDNF can promote the differentiation of adult hippocampus NSCs into neurons.
