1.Effect of dietary vitamin A intake on plasma vitamin A concentration in preschool children of Banan district, Chongqing, China

Objective:To study the relationship between dietary vitamin A intake and plasma vitamin A concentration , and establish the theoretical basis for dietary intake predicting vitamin A nutritional status . Methods:By using cluster sampling , 492 children aged 2-7 years in kindergartens in Banan district of Chongqing were selected .A cross-sectional nutrition and health survey was conducted , including the clinical examination , anthropometry , laboratory test and dietary survey .Results: Among the children surveyed, 229 were boys, and 263 girls, the mean age was (4.54 ±0.87) years, height (107.50 ± 7.20) cm, and weight (18.42 ±3.41) kg, the mean value of plasma vitamin A was (1.04 ±0.30)μmol/L.The prevalence of marginal vitamin A deficiency ( MVAD) was 43.5%.No cases of severe clinical vitamin A deficiency were found (plasma vitamin A≤0.35μmol/L).Clinical examination found no conjunctiva, corneaor skin abnormalities, and no Bitot’s spots.Prevalence of the last two weeks colds were 27.4% (135/492), no diarrhea and other gastrointestinal or digestive diseases were found .The proportion of insufficient dietary vitamin A intake ( <600 μg RE/d) was as high as 50.0%.By using correlation analysis, plasma retinol concentrations were related to dietary vitamin A intake (r=0.162, P<0.001), and to dietary energy intake (r=0.107, P=0.017).After adjustment for the effects of other non-dietary factors on vitamin A deficiency , the multivariate logistic regression showed that vitamin A-rich foods of liver intake=0 g/d (OR=1.95, 95% CI:1.05-3.61, P=0.034), vitamin A-rich fruits intake=0 g/d (OR =1.55, 95% CI: 1.03 -2.33, P =0.034), vitamin A-rich vegetables intake<200 g/d (OR=3.47, 95%CI:1.37-8.75, P=0.009) were important risk factors of vitamin A deficiency .But we had not found the correlation between the intake of meat , eggs and milk and vita-min A deficiency .Conclusion:Dietary factors may be the major risk factor of vitamin A deficiency in the three kindergartens .The dietary vitamin A intakes are significantly related to plasma retinol concentra-tions, and the vitamin A-rich foods intakes can predict the body ’ s vitamin A nutritional status .
2.Effect of self-practice-oriented teaching method on negative emotions of first-time pen insulin injection patients

Objective To observe the effect of self-practice-oriented teaching method on injection skills, psychological status and blood glucose control of type 2 diabetes mellitus patients receiving pen insulin injection for the first time. Methods A total of 105 patients with type 2 diabetes mellitus who received insulin injection for the first time were divided into the control group and the observation group according to admission sequence. The control group adopted the method of one-to-one bedside health education of primary nurses. Insulin injection demonstration and disease-related knowledge education were conducted on the day of the medical order to start insulin treatment. Health education was conducted once a day for 3-5 minutes each time for a total of 5 days. The observation group adopted centralized health education, which insulin injection teaching desk, video and other teaching aids, combined with the guidance and correction of primary nurses were used, focusing on the use of teaching aids and self-injection as early as possible during hospitalization. The levels of fasting and postprandial blood glucose, insulin injection skills and anxiety and depression scale (HADS) were compared between the two groups. Results There was no statistically significant difference in blood glucose level, anxiety scores and depression scores between the two groups before the intervention (P>0.05). Two weeks after the hospital discharge, the anxiety and depression scores of the observation group was 7.31 ± 1.78, 7.00 ± 1.73, significantly lower than the anxiety and depression points of the control group 9.33 ± 2.21, 8.61 ± 1.79 (t=2.492, 3.097, P<0.05);The insulin injection skills assessment score of patients in the observation group was (90.90 ± 4.15) points, significantly higher than the points of the control group (83.74 ± 6.22) (t=-6.593,P<0.01);In the observation group, fasting blood glucose and postprandial 2-hour blood glucose were (7.56 ± 1.86) mmol/L and (10.61 ± 2.25) mmol/L, respectively Compared with the control group (8.55 ± 1.96) mmol/L ,(12.91 ± 2.95) mmol/L, there was statistically significant difference between the two groups(t=2.542, 4.301, P<0.05). Conclusion The self-practice-oriented teaching method can effectively alleviate the negative emotions of the patients with the first insulin injection, improve their insulin injection skills to better control blood glucose.
3.Exosomes and skin wound healing

BACKGROUND:Exosomes play a role in all stages of wound repair,and there is currently a large body of research on exosomes in skin wound repair,which has been shown to have great potential for clinical applications. OBJECTIVE:To summarize and discuss the main mechanisms and clinical applications of exosomes in the treatment of skin wounds,in order to promote the clinical translation of exosomes. METHODS:PubMed,clinicaltrials.gov,China National Knowledge Infrastructure,Food and Drug Administration database,and Chinese Clinical Trial Register were searched from inception to March 2023.The English search terms were"exosomes,wound healing,stem cells,chronic wound,immunoregulation,inflammation,skin,therapeutic use,isolation,characterization,infections".The Chinese search terms were"exosomes,wound healing,stem cells,immunomodulation,clinical applications".A total of 79 articles were included for the summary. RESULTS AND CONCLUSION:(1)Exosomes can improve and accelerate wound healing through inflammation regulation,immune protection,angiogenesis,cell proliferation and migration,and collagen remodeling.(2)Exosomes derived from stem cells have mature preparation techniques and related mechanism research,which is currently the mainstream research direction.Non-stem cell-derived exosomes have the advantages of convenience,economy,and easy production,and can be used as a supplement for clinical applications.(3)The clinical application of exosomes is still in its infancy,but has great potential for application.Various exosome modification techniques have laid the foundation for the future development of clinically personalized services and require further research.(4)The clinical translation of exosomes faces many challenges,such as low yield,high heterogeneity,lack of unified standards for isolation,purification,and quality control,and difficulties in storage.
4.Pharmacological inhibition of BAP1 recruits HERC2 to competitively dissociate BRCA1-BARD1, suppresses DNA repair and sensitizes CRC to radiotherapy.

Radiotherapy is widely used in the management of advanced colorectal cancer (CRC). However, the clinical efficacy is limited by the safe irradiated dose. Sensitizing tumor cells to radiotherapy via interrupting DNA repair is a promising approach to conquering the limitation. The BRCA1-BARD1 complex has been demonstrated to play a critical role in homologous recombination (HR) DSB repair, and its functions may be affected by HERC2 or BAP1. Accumulated evidence illustrates that the ubiquitination-deubiquitination balance is involved in these processes; however, the precise mechanism for the cross-talk among these proteins in HR repair following radiation hasn't been defined. Through activity-based profiling, we identified PT33 as an active entity for HR repair suppression. Subsequently, we revealed that BAP1 serves as a novel molecular target of PT33 via a CRISPR-based deubiquitinase screen. Mechanistically, pharmacological covalent inhibition of BAP1 with PT33 recruits HERC2 to compete with BARD1 for BRCA1 interaction, interrupting HR repair. Consequently, PT33 treatment can substantially enhance the sensitivity of CRC cells to radiotherapy in vitro and in vivo. Overall, these findings provide a mechanistic basis for PT33-induced HR suppression and may guide an effective strategy to improve therapeutic gain.
