Depression and anxiety is one of common mental illness that are being seriously harm to the physical and mental health of human being in recent years.Depression and anxiety are difficult to be well treated with a single and targeted drug, owing to its biological-psychological-social interaction pathogenic feature.Due to its own multicomponent characteristics, traditional Chinese medicine (TCM) compound Xiao-Yao (XY) powder can exert the antidepression and anxiety effects from multi-level,multi-link and multi-target.Based on previous studies on effect and mechanism of XY powder on anti-depression and anxiety of our group,we proposed that XY powder had two main characteristics when treating depression and anxiety, which were "two-way adjustment" and "multi-target,multisystem,multi-level".Additionally,to further clarify the pharmacodynamic material basis of XY powder, more importance is that we should depend on the variety of disciplinary research methods, focused on the TCM syndrome and the combination of disease and TCM syndrome, brought both disease evaluation and syndrome evaluation method, took the importance of gut-brain axis (GBA),and combined with the correlation between the formula and the syndrome.

Objective:To develop and examine the reliability and validity of the adolescent health literacy evaluation scale under public health emergencies (AHLES-PHE) in junior middle school students. Methods:The initial version of AHLES-PHE was formed by combining the characteristics of public health emergencies and adolescent health literacy. The expert version of AHLES-PHE was formed by two rounds of Delphi expert consultation.Totally 1 729 adolescent students from three junior middle schools in Changzhou City were tested. The formal version of AHLES-PHE was formed through item analysis and factor analysis (structural validity). The content validity of the formal version of AHLES-PHE was evaluated by specialist analysis and sensibility analysis, and its correlation validity with the everyday health information literacy (EHIL) was tested. The internal consistency reliability, parity split-half coefficient and test-retest reliability were calculated.Results:(1)Item analysis: the correlation coefficient between the scores of each item and the total score of AHLES-PHE expert version was 0.420 to 0.722 ( P<0.01), and the CR of decision value was 10.140 to 66.980 ( P<0.01). (2)Validity analysis: the formal version of AHLES-PHE with 45 items and 8 factors was obtained by exploratory factor analysis. The 8 factors could explain 61.30% of the total variation. The fitting indexes of the confirmatory factor analysis model were χ2/ df=3.325, RMSEA=0.052, GFI=0.853, CFI=0.912, TLI=0.904, NFI=0.880.Sensibility analysis of the formal version of AHLES-PHE showed the Cronbach's α ranged from 0.957 to 0.958.The correlation coefficient between the total score of AHLES-PHE formal version and the total score of EHIL was 0.340 ( P<0.01). (3)Reliability analysis: the Cronbach's α coefficient and parity split-half coefficient of the formal version of AHLES-PHE were 0.958 and 0.975, respectively. The test-retest reliability of the formal version of AHLES-PHE was 0.753 ( P<0.01). Conclusion:The formal version of AHLES-PHE developed in this study has good reliability, structural validity and content validity, and the correlation validity with EHIL is common. This scale has a certain degree of applicability to the assessment of health literacy for junior middle school students under public health emergencies.

Journal of Zhejiang University. Medical sciences 2024;53(4):472-480
Objective:To investigate the association of R-loop binding proteins with prognosis and chemotherapy efficacy in lung adenocarcinoma. Methods:The data related to R-loop regulatory genes were obtained from literature of R-loop proteomics and relevant databases. We used 403 cases of lung adenocarcinoma in the Cancer Genome Atlas as training set,and two datasets GSE14814 and GSE31210 in Gene Expression Omnibus as validation sets. The weighted gene co-expression network analysis(WGCNA)was employed to identify R-loop genes with a significant impact on the clinical phenotype of lung adenocarcinoma.Least absolute shrinkage and selection operator(LASSO)regression analysis was utilized to eliminate genes exhibiting multicollinearity.A multivariate Cox regression analysis was employed to scrutinize clinical variables and R-loop characteristic genes that exert independent prognostic effects on patient survival.Subsequently,a risk score model was constructed. The predictive capacity of this model for the prognosis of patients was analyzed and validated.Additionally, the performance of risk model on the anti-tumor drug sensitivity was assessed. The mutations of R-loop genes were analyzed by maftools. The effect of PLEC expression on anti-tumor drug sensitivity was tested on non-small cell lung adenocarcinoma H1299 and A549 cells in vitro. Results:A collection of 1551 R-loop genes were obtained, and 78 genes exhibited significant effects on the clinical phenotype shown on WGCNA.The LASSO regression analysis retained fourteen R-loop genes.A multivariate Cox regression analysis further identified three R-loop genes(HEXIM1,GLI2,PLEC)and a clinical variable(tumor grading)that were associated with patient prognosis.Risk prediction model was established according to the regression coefficients of each parameter.Kaplan-Meier survival analysis showed that the prognosis of high-risk group was significantly worse than that of low-risk group(P<0.01).The time-dependent ROC curve showed that the risk model had good predictive ability in both training and validation sets.Predictive analyses of anti-neoplastic drug sensitivity indicated a diminished responsiveness to both chemotherapy and targeted treatment drugs among high-risk patients. The expression of PLEC was strongly correlated with sensitivity to gefitinib,a classical EGFR inhibitor.Conclusion:R-loop binding proteins have been identified as significant determinants in the prognosis and therapeutic strategies for lung adenocarcinoma, which indicates that therapeutic interventions targeting these specific R-loop binding proteins might contribute to a better survival of the patients.
