1.Induction of apoptosis of ovarian carcinoma cell strains with As_2O_3 via G_1 phase cell cycle arrest

Objective To study the apoptosis of ovarian carcinoma cell strain COC1 induced by arsenic trioxide (As_2O_3). Methods The effect of arsenic trioxide on the proliferation of ovarian carcinoma cell strains were examined, using Methyl thiazolyl tetrazolium(MTT) assay. Flow cytometry(FCM) was used to detect apoptosis percentage and phase distribution of cell cycles. After being exposed to As_2O_3 solution of different concentration, apoptosis morphologic features of COC1 were observed by TdT-mediated dUTP nick end labeling (TUNEL). Results Inhibition of As_2O_3 on the growth of COC1 was increasing with passing of time and increasing of concentration. After 48h of exposure to 1.5 ?mol/L As_2O_3, COC1 cell strains presented apoptosis morphologic change, and the number of apoptosis cells increased with the passage of time. After treatment with 3.0 ?mol/L or 1.5 ?mol/L As_2O_3, the percentage of COC1 cells in G_2/M phase declined, and the percentage of cells in G_1 phase increased with the passage of time. These results suggested that As_2O_3 inhibited the cellular proliferation of COC1 cells via arrest of cell cycle. Conclusion Arsenic trioxide can induce apoptosis of ovarian carcinoma cell strain COC1. It can block the cell cycle at the G_1 phase, which is one of the possible mechanisms of apoptosis induced by As_2O_3.
2.The exploration and practice on the self-designed experiments of extract preparations in phar-macy teaching

The self-designed experiments of extract preparations is practiced in pharmacy teach-ing. Students work in groups, first consulting literature material, selecting the prescription and devis-ing a plan. Then, if the plan has carried on the feasible proof, students begin to purchase raw materi-als, complete the extraction and separation of effective components, the preparation of molding and the analysis of experimental results and finally form their self-evaluation. This experiment can strengthen students' experiment skill and promote their comprehensive application ability of all the subjects in the pharmacy field. At the same time it can also play an important role in the improvement of the students' social practice ability and the cultivation of students' creative thinking.
3.Establishment of blood purification system for rats

Objective To establish blood purification system for rats and provide a safe and reliable experimental platform for further research of blood purification. Methods The right carotid artery and the contralateral jugular vein of adult male Sprague?Dawley rats were cannulated to creat vascular access for blood purification, by which continuous arteriovenous hemofiltration blood purification system was established. Blood flow, substitution fluid flow and ultrafiltration rate were regulated by rotary mini?pumps. Blood purification therapy continued for 4 hours on the basis of maintained anesthesia and effective anticoagulation. The safety of continuous arteriovenous hemofiltration blood purification systems was evaluated by comparing the arterial blood gas, electrolyte indexes and blood glucose during the blood purification therapy. Closely monitoring the vital signs of rats, such as blood pressure and heart rate, and observing whether there were any side effects, such as massive haemorrhage, thrombogenesis and gas embolism in the therapeutic process. Results There were no obvious changes of arterial blood gas, electrolyte indexes and blood glucose during the blood purification therapy (P>0.05). The vital signs did not fluctuate acutely before and after the blood purification therapy (P>0.05). The incidence rate of side effects was very low. Conclusions
Continuous arteriovenous hemofiltration blood purification system had no obvious adverse effects on healthy rats. Our blood purification system for rats appears to be safe and reliable.
4.The clinical significance of predicting the contrast-induced nephropathy after PCI by the ratio of contrast ;medium volume and glomerular filtration rate

Objective To evaluate the significance of contrast medium (CM) volume and estimated glomerular filtration rate (CM/eGFR) in predicting contrast-induced nephropathy (CIN) after PCI. Methods A total of 307 patients after PCI were enrolled from Nanfang Hospital from May 2014 to October 2015. The patients were divided into the CIN group(n = 29) and the non-CIN group(n = 278) according to whether CIN within 72 hours after PCI. The baseline renal function was assessed by the sCr and CyC, respectively. Results Twenty-nine patients (9.4%, 29/307) developed CIN. There were significant differences in Age, CM、NTpro-BNP、IABP、 Periprocedural Hypotension、Preprocedural sCr/CyC between two groups (P < 0.05, respectively). The result of multivariate logistic regression analysis showed that Age, Cardiac function ≥Ⅲ level, IABP, use CCB, CM/eGFRMDRD, CM/eGFRCyC were independent risk predictors for CIN, respectively. Receiver Operating Characteristic (ROC) curve analysis showed that the area under the curve of CM/eGFRMDRD(AUC = 0.838) was superior to CM/eGFRCyC (AUC = 0.805) without significant difference. The sensitivity and specificity were 79.3%and 76.3%(Cut-off Point = 2.094), respectively. Conclusion Both the CM/eGFRMDRD and CM/eGFRCyC may be good methods to determine maximum CM before PCI and to predict CIN after PCI currently, without significant differences between these two predictors.
5.Differential analysis of gene expression profiles for lymphonode metastasis of colon cancer

Objective To investigate the differences in the gene expression profiles between SW480 and SW620 cell lines.Methods A dataset of GDS756 containing the gene expression profiles of SW480 and SW620 was downloaded from the GEO database in NCBI.The differential expression genes between SW480 and SW620 were analyzed with gene set enrichment analysis (GSEA) and leading edge subset analysis.The genes in leading edge subset were re-annotated by FunRich software.The core genes of leading edge subset closely relating to SW480 or SW620 were analyzed with the STRING on-line analytical system.The functional core genes closely relating to SW480 or SW620 were obtained by the combined analysis of the core genes and high frequency genes from leading edge subset.Results GSEA identified 12 significantly enriched gene sets,491 leading edge genes and 7 highly overlapping genes from SW480 and 80 significantly enriched gene sets,870 leading edge genes and 6 highly overlapping genes from SW620.The STRING system identified 5 core genes from SW480 and 8 from SW620.The combined analysis of GSEA and bionetwork obtained 2 functional core genes,TOP2A and CDK1,from SW620.Conclusion The SW480 and SW620 cells with identical genetic background have different functional gene expression profiles,and the functional core genes TOP2A and CDK1 in SW620 cells may be related to the signal pathways of colon cancer metastasis.
6.Near-total laryngectomy and voice prosthesis reconstructive surgery

OBJECTIVE To investigate the application of near-total laryngectomy on the treatment of advanced laryngeal and hypopharyngeal carcinoma. METHODS Thirty patients with laryngeal carcinoma(T3N0~T3N1 17cases,T4N0~T4N1 13cases) and nine with hypopharyngeal carcinoma(T3N0~T3N1 6cases,T4N0~T4N1 3cases) were treated with near-total laryngectomy from May 1995 to March 2003 according to Peaeson modifying method of Tangpingzhang. Concurrent neck dissection were performed in sixteen patients. RESULTS The speech function was obtained in 36 cases (92.3 %) and good swallowing function was obtained in 38 cases(97.4%). Three year survival rates were 73.3% (22/30) in laryngeal carcinoma and 55.6 %(5/9)in hypopharyngeal carcinoma and five year survival rate in laryngeal carcinoma was 61.1%(11/18) . Local recurrence rate was only 2.56 %. CONCLUSION Near-total laryngectomy and speech rehabilitation can be used in most cases that might be treated with total laryngectomy in the past. It can provide satisfying survival rate, good swallowing and speech function.
7.Expression and significance of signal transducer and activator of transcription 3 and c-myc in laryngeal squamous cell carcinoma.

OBJECTIVE:
To investigate the expression and clinical significance of signal transducer and activator of transcription 3(STAT3) and its target gene c-myc in laryngeal squamous cell carcinoma (LSCC), and the relationship between the two genes.
METHOD:
Formalin-fixed and paraffin-embedded tissues, which came from 56 cases of LSCC and 30 samples of normal mucosas from 30 patients with total or subtotal laryngectomy over 2.0 cm away from tumor margin, were detected for the expression of STATS, c-myc by in situ hybridization and SP immunohistochemistry. Micro-image analysis system was used to determine the optical density, and the result was analyzed statistically.
RESULT:
There is overexpression of STAT3, c-myc mRNA and protein in LSCC. The expression of STAT3 and c-myc mRNA in LSCC was associated with clinical stage, differentiation grade and lymph nodal metastases (P < 0.05 or 0.01). The expression of STAT3 and c-myc protein in LSCC was associated with clinical stage and lymph nodal metastases (P < 0.05 or 0.01). There was a positive correlation between the expression of STAT3 and c-myc genes. mRNA and protein, The correlation coefficient (r) was 0.6224 (P < 0.01) for the mRNA expression and 0.7012 (P < 0.01 )for the protein expression.
CONCLUSION
The expression of STAT3 and c-myc may play an important role in the tumorigenesis, metastases and poor prognosis of LSCC. There was a positive correlation between the overexpression of STAT3 and c-myc genes in LSCC.

8.Effects of cucurbitacin B on cell proliferation and apoptosis in Hep-2 cells.

OBJECTIVE:
To investigate the mechanism underlying the anticancer activity of cucurbitacin B on human laryngeal cancer.
METHOD:
Hep-2 cells were treated with different concentrations of cucurbitacin B for different time. MTT assay was used to evaluate cell proliferation. Flow cytometry with PI staining and fluorescent microscopy with Hoechst 33258 staining were used to estimate cell cycle distribution and cell apoptosis. Expression of p-STAT3, cyclin B1 and Bcl-2 proteins was evaluated by Western blot assay. In vivo inhibitory effects of cucurbitacin B on tumor growth was evaluated in a nude mouse xenograft model.
RESULT:
Cucurbitacin B inhibited cellular proliferation in a dose and time dependent manner (P <0.05 or 0.01). Flow cytometry analysis showed that treatment with cucurbitacin B resulted in accumulation of cells at the G2/M phase of the cell cycle and cell apoptosis in a dose and time dependent manner (P <0.05 or P <0.01). Marked morphological changes of cell apoptosis including condensation of chromatin, nuclear fragmentation and apoptotic bodies were observed clearly by Hoechst 33258 staining. Western blot analysis demonstrated that the expression of p-STAT3, cyclin B1 and Bcl-2 proteins was suppressed significantly. In vivo studies showed that the inhibitory rates on laryngeal squamous carcinoma xenograft model were 32.43%, 43.24% and 70.27% for lower, moderate and higher dosage group, respectively.
CONCLUSION
Cucurbitacin B inhibited cell proliferation and induced apoptosis of Hep-2 cells by suppressing STAT3 signal pathway, down regulating the expression of cyclin B1 and Bcl-2 proteins.

9.Status survey on pediatric emergency in secondary and tertiary public hospitals in Shanghai city

Objective:To understand the current situations and existing problems of pediatric emergency in Shanghai city and provide a basis for the construction and management of pediatric emergency.Methods:The questionnaire survey was used to investigate the current situations of pediatric emergency department in secondary and tertiary public hospitals in Shanghai city.Results:A total of 28 questionnaires were delivered, and 28 were responded.Six of the 28 hospitals had no administratively independent pediatric emergency or no separate pediatric emergency area.Of the 22 hospitals, each had an average of 9.7 professional emergency pediatricians, with the exception of one secondary hospital that lacked professional emergency pediatricians.Professional emergency pediatricians accounted for an average of 70.0% of all pediatricians.Of the 22 hospitals, 18(81.8%) were open 24 hours a day.Fourteen(63.6%) had independent triage.Eight children′s emergency rooms(36.4%) were shared with adults.Among the 22 hospitals, 20 hospitals filled in the annual total number of pediatric emergency visits, and the total number of pediatric emergency visits ranged from 2 791 to 467 428, with an average of 93 966.65.Conclusion:There are still some problems in the development of pediatric emergency department in secondary and tertiary public hospitals in Shanghai city, such as insufficient human resources, substandard personnel training, incomplete equipment and lack of critical first-aid techniques.Therefore, the construction and management of pediatric emergency department still need to be improved.
10.The value of synthetic MRI combined with field of view optimized and constrained undistorted single shot diffusion weighted imaging in differential diagnosis of BI-RADS 4 breast lesions

Objective To evaluate the differential diagnostic value of synthetic magnetic resonance imaging(syMRI)and field of view optimized and constrained undistorted single shot diffusion weighted imaging(FOCUS DWI)in patient with breast imaging reporting and data system(BI-RADS)4 breast lesions.Methods A total of 68 patients who underwent breast MRI and were classified as BI-RADS category 4 were prospectively enrolled.Among them,there were 31 cases of benign lesions(benign group)and 37 cases of malignant lesions(malignant group).All patients underwent T1WI,T2WI,FOCUS DWI,dynamic contrast-enhanced magnetic resonance ima-ging(DCE-MRI)and syMRI.Two radiologists delineated the region of interest(ROI)on quantitative parameters images of syMRI and the apparent diffusion coefficient(ADC)images generated from FOCUS DWI and calculated T1,T2,proton density(PD)and ADC values,respectively.The thresholds and diagnostic efficacy of syMRI and FOCUS DWI parameters were evaluated by the receiver op-erating characteristic(ROC)curve.Logistic regression analysis was used to combine the parameters of the two sequences and evalu-ate the diagnostic efficacy.Results The T2,PD,and ADC values in the malignant group were significantly lower than those in the benign group(P<0.05),but there were no statistically significant difference in T1 values between the groups(P>0.05).T2 and ADC values could be used as important diagnostic indexes in patient with malignant lesions of BI-RADS 4 breast lesions,and the area under the curve(AUC)of T2 and ADC was 0.833 and 0.867,respec-tively.There was no significant difference in AUC of T2 and ADC between benign and malignant groups(Z=0.485,P=0.627).The AUC of T2 value combined with ADC value was 0.910,and the sensitivity and specificity was 90.3%and 89.2%,respectively.Conclusion T2 value of syMRI and ADC value of FOCUS DWI can be used as quantitative diagnostic indicators in patients with BI-RADS 4 breast lesions.The combination of T2 and ADC values can further improve the differential diagnosis efficiency.
