1.Research progress of bacterial flagellin as a vaccine adjuvant

Vaccines play an important role in the prevention and control of infectious diseases. Bacterial flagellin can activate Toll like receptor 5(TLR5) and NOD like receptor C4(NLRC4) in host cells,and has immune adjuvant effect. As a new immune adjuvant,flagellin can not only share with antigen protein,but also fuse with antigen protein,which can significantly improve the immune effect. At the same time,it has achieved good application effect in mucosal immunity and antitumor immunity,becoming a hot spot in the research and development of vaccine adjuvants. This review mainly discussed the research progress on application of bacterial flagellin as vaccine adjuvant,so as to provide new ideas for the development of flagellin adjuvant vaccine.
2.Clinical efficacy of esomeprazole, mosapride, magnesium plus aluminum in reflux esophagitis

Objective To investigate clinical effect of esomeprazole, mosapride, magnesium plus aluminum in patients with reflux esophagitis. Methods 98 patients with reflux esophagitis who were treated in our hospital from October 2014 to October 2016 were selected and randomly divided into control group and observation group, with 49 cases in each group.In the control group, patients were treated with esomeprazole and mosapride.In the observation group, patients were given esomeprazole, mosapride and magnesium plus aluminum.The levels of serum gastrin, serum motilin and 24-hour pH in the esophagus were compared before and after treatment .Results After treatment,The levels of serum gastrin and serum motilin in the observation group were significantly higher than those in the control group, the difference was statistically significant (P<0.05), while the total time percentage of pH<4 at standing position, total time percentage of pH <4 at supine position, total reflux time percentage of pH <4, and the duration of continuous reflux≥five mins in the observation group were significantly lower than those in the control group, the difference was statistically significant (P<0.05). Conclusion Esomeprazole, mosapride and magnesium plus aluminum have significant clinical effect in patients with reflux esophagitis, and can effectively reduce 24-hour pH in the esophagus, decrease the number of reflux, increase serum gastrin and serum motilin levels.
3.Effect of omeprazole combined with Kangfuxin liquid on immune function, gastrin family and NO levels in gastric ulcer

Objective To observe the effect of omeprazole combined with Kangfuxin liquid on immune function,gastrin family and NO levels in patients with gastric ulcer.Methods 120 cases of patients with gastric ulcer from October 2014 to September 2016 in our hospital were selected and randomly divided into the observation group and the control group with 60 cases in each group.The control group was given omeprazole based on conventional treatment, and the observation group was given Kangfuxin liquid based on the control group.For four weeks, to observe the differences of immune function (CD3 +,CD4 +,CD8 +,CD4 +/CD8 + and CD4 +CD25 +FoxP3 +Treg), gastric gastrin family (GAS, CCK, MTL) and NO levels between the two groups.Results After treatment, the CD3 +, CD4 +, CD8 +, CD4 +/CD8 +and CD4 +CD25 +FoxP3 +Treg levels of the two groups were compared, the difference was statistically significant (P<0.05), CD3 +, CD4 +, CD4 +/CD8 + of the groups showed a gradually increasing, the difference was statistically significant (P<0.05) and CD8 +, CD4 +CD25 +FoxP3 +Treg showed a gradually decreasing, the difference was statistically significant (P<0.05), and the change range of the observation group was more than that of the control group, the difference was statistically significant (P<0.05), the GAS,CCK,MTL levels of the two groups were compared, the difference was statistically significant (P<0.05), the GAS, CCK of the two groups showed a gradually decreasing, the difference was statistically significant (P<0.05), and MTL showed a gradually increasing, the difference was statistically significant ( P<0.05 ) , and the change range of the observation group was more than that of the control group , the difference was statistically significant (P<0.05), the NO levels of the two groups were compared, the difference was statistically significant (P<0.05), NO of the groups showed a gradually increasing, the difference was statistically significant (P<0.05), and the change range of the observation group was more than that of the control group, the difference was statistically significant (P<0.05).Conclusion Omeprazole combined with Kangfuxin liquid can get better regulating results of immune function and gastrin family, NO in patients with GU, and have more effective than omeprazole alone.
4.Effect of itraconazole on pharmacokinetics of viaminate in healthy subjects

Aim To investigate the effect of oral itraconazole on pharmacokinetics of viaminate and under-stand the metabolism pathway of viaminate in healthy humans,as well as guide the reasonable medication for viaminate and itraconazole coadministration.Methods The study was conducted in a double-blind randomized and crossover manner with two phases of treatment with placebo-viaminate or itraconazole-viaminate.Eight healthy male subjects received an oral dose of 200 mg itraconazole(QD) or matched placebo for five consecutive days,and took an oral 100 mg dose of viaminate on day 4 after each treatment phase.The liquid chromatography-mass spectrometry(LC-MS) was established to measure the blood drug concentration of viaminate.Results Our results showed that the mean peak concentration of viaminate was increased with itraconazole [(21.21?11.65) ?g?L~(-1) vs(27.12?13.83) ?g?L~(-1),P0.05].Additionally,the mean elimination half life was prolonged [(2.71?0.38) h vs(4.43?0.93) h,P
5.Advanced oxidation protein products modulate autophagy of renal tubule epithelial cells

Objective To investigate the effect of advanced oxidation protein products (AOPPs) on human renal tubular epithelial cells(HK-2) autophagy.Methods HK-2 cells were stimulated with AOPPs.RT-qPCR and Western blot were used to determine the expression of autophagy related protein LC3-Ⅱ/LC3-Ⅰ,Beclin1 and p62;Western blot was utilized to examine the activation of p38 MAPK pathway.Then p38 MAPK inhibitor (SB203580) was added and co-processed with AOPPs.The change of autophagy was observed Also,autophagy inducer rapamycin was added and co-processed with AOPPs.RT-qPCR and Western blot were used to detect the expression of cell cycle inhibitory protein p27.The cell total protein level was detected by the bicinchoninic acid (BCA) method.The hypertrophy change was observed.Results AOPPs down-regulated the expression of LC3-Ⅱ/LC3-Ⅰ and Beclin1,up-regulated expression of p62 and activated p38 MAPK pathway;in comparison with the AOPPs alone treatment group,the expression of LC3-Ⅱ/LC3-Ⅰ and Beclin in the SB203580 co-processing group was increased,while p62 was decreased;the p27 expression and cells total protein in the sirolimus co-processing group were down-regulated.Conclusion AOPPs inhibits the autophagy of HK-2 cells by activating p38 MAPK pathway and the autophagy inhibition participates in HK-2 cell hypertrophy.
6.Advances in research on VEGF-related signaling pathways in angiogenesis

Tissues and organs generate angiogenesis under the stimulation of angiogenic factors in physiological or pathological conditions.Multiple signal pathways including VEGF, Notch, Wnt/β-catenin, Ang1(2)/tie2 and PIK-Akt etc.have effects on various stages of angiogenesis.VEGF exerts irreplaceable effects on the whole process of angiogenesis through multiple signal pathways.Over the past few years, new progress has been made in the researches of mechanisms regulating angiogenesis through VEGF-related signal pathways both at home and abroad.These findings provide us new theoretical basis for clarification of the pathogenesis of many diseases and clinical drug development.In this article we will summarize the recent research progress in this field, hoping to provide new possibilities for the treatment of angiogenesis-related diseases.
7.Determination of bulleyacinitine A in mouse plasma by liquid chromatography tandem mass spectrometry

AIM:To develop the method for analyzing bulleyacinitine A in plasm from mice through the treatment of subcutaneous injection of bulleyacinitine A by liquid chromatography tandem mass spectrometry(LC-MS/MS).METHODS:50 ?L mouse plasma sample added zolpidem as internal standard and pH 9.2 buffer solution was extracted with ethylether,followed by liquid chromatography and mass spectrometric detection.The mobile phase was consisted of methanol-buffer solution(85∶15)(buffer solution consisted of 20 mmol/L ammonium acetate and 0.1% formic solution).The flow rate was 300 ?L per minute.The separation column was C_ 18 column.The electrospray ionization tandem mass spectrometry and the multiple reaction monitoring mode were applied to detecting the bulleyacinitine A in plasma.RESULTS:The assay was linear from 0.1 to 1 000 ng/mL.The recovery rate was more than 80%.CONCLUSION:The method could be used to detect bulleyacinitine A level in mouse plasma,which offers advantages of sensitiveness,specificity and simpleness.
8.Expression, purification of nattokinase in Pichia pastoris and preparation of its polyclonal antibody

Purpose To indicate the expression of nattokinase (NK) in Pichia pastoris , an emulsion was prepared with the purified NK to prepare polyclonal antibody. In order to establish sandwich enzyme-linked immunosorbent assay (ELISA) to assay NK in organism, furthermore to lay the foundation for researching in vivo metabolism and function of NK. Methods The NK gene was cloned into a Pichia pastoris expression vector pHBM905A to construct the recombinant plasmid pPRONK.The recipient cell of Pichia pastoris GS115 was transformed with pPRONK which had been cut by restriction enzyme Sal I , under the induction of methanol. The expressed production is purified by salting out and ultrafiltration membrane. An emulsion was prepared with the purified NK and injected into rabbits to prepare polyclonal antibody. Results NK was expressed and identified by SDS-PAGE.The molecular mass of expressed production is about 27 kD.The fibrin plate assay indicated that the NK protein can cleavage fibrin effectively. ELISA analysis indicated that the polyclonal antibody titer is about 1:8 000. Western blot demonstrated that there was a special strap nearby 27 kD. Conclusion NK was successfully expressed in Pichia pastoris , the production can cleavage fibrin effectively and it had great immunogenicity.

In 2020, the World Allergy Organization released a position paper on the immunoglobulin E (IgE) allergy diagnostics and tests, which provided a thorough and updated appraisal of the most frequently used diagnostic tests, both in vivo and in vitro.This article focused on the interpretation of in vitro tests, mainly introduced three test methods of serum total IgE, allergen specific IgE detection and basophil activation test, aiming to help medical workers better understand the position paper, and further understand the diagnosis methods, so as to make accurate diagnosis and formulate the optimal treatment.
10.Prevalence of orthostatic hypotension in patients aged 80 or over and associated risk factors

Objective To investigate the prevalence of orthostatic hypotension in hypertensive patients aged 80 or over and associated risk factors at level A tertiary hospitals.Methods A multicenter cross-sectional study was conducted among very old hypertensive patients (≥ 80 years) at 28level A tertiary hospitals by using questionnaire-based surveys.A total of 1298 hypertensive subjects aged between 80 and 101 years were enrolled in this study.Blood pressures was measured in the supine position and the upright position within three minutes of standing,and 316 patients were assigned to the orthostatic hypotension(OH) group and the other 982 patients to the non-orthostatic hypotension(NOH) group.Additional information was acquired through patients' medical records and the questionnaire.Results The prevalence of OH was 24.3% (316 cases) There were no significan different betueen OH patienl with NOHones in gender and age.OH patients had higher supine systolic and diastolic blood pressure(P=0.003 and 0.000,respectively),lower standing systolic and diastolic blood pressure(P=0.000 and 0.000,respectively),and higher rates of past coronary heart disease,stroke and renal abnormalities (P =0.037,0.001 and 0.014,respectively) than NOH patients.Logistic regression analysis showed that the prevalence of OH in the patients was positively correlated with supine systolic blood pressure(OR=1.196,CI:1.153-1.242,P=0.000) and supine diastolic blood pressure (OR =1.740,CI:1.602-1.885,P =0.000) and was negatively correlated with standing systolic blood pressure(OR =0.824,CI:0.795-0.855,P=0.000) and standing diastolic blood pressure (OR =0.584,CI:0.539-0.634,P =0.000).Conclusions The prevalence of orthostatic hypotension is high in hypertensive patients aged 80 years or over at level A tertiary hospitals.Poor blood pressure control may increase the risk of orthostatic hypotension.
