1.Related substances in orlistat detected with UPLC-MS/MS.

The study aims to identify related substances in orlistat pharmaceutical substance from different production technologies. Advanced technologies including UPLC-QTOF-MS and UPLC-QTrap-MS were used and the structures or molecules of 10 kinds of related substances in orlistat were identified. The experimental data were valuable to the quality control and development of orlistat.
2.Effect of dexmedetomidine combined with sufentanil on postoperative analgesia and the vascular crisis in patients undergoing free flap transplantation

Objective To investigate the effect of dexmedetomidine combined with sufentanil on postoperative analgesia and the vascular crisis in patients undergoing free flap transplantation. Methods Forty patients with ASA physical statusⅠ-Ⅱhaving underwent free flap transplantation were divided into groupⅠand groupⅡaccording to the random digits table method with 20 cases each. The drugs of patient controlled intravenous analgesia (PCIA) in groupⅠwere sufentanil 1.0μg/(kg·d)+tropisetron 5 mg/100 ml, and the drugs of PCIA in group Ⅱ were sufentanil 1.0 μg/(kg · d) +dexmedetomidine 1.0 μg/(kg · d) + tropisetron 5 mg/100 ml. The PCIA pump was set in 2 groups as follows: continuous perfusion rate 2 ml/h, single bolus 2 ml, lockout time 15 min. The visual analogue score (VAS), Ramsay sedation score, press number of PCIA pump within 48 h after operation, satisfaction score after operation, untoward reaction and vascular crisis were recorded. Results The VAS at 6, 12, 24 and 48 h after operation in group Ⅱ were significantly lower than those in group Ⅰ: (2.18 ± 0.41) scores vs. (3.00 ± 0.63) scores, (2.64 ± 0.51) scores vs. (3.82 ± 0.60) scores, (2.55 ± 0.52) scores vs. (3.36 ± 0.51) scores and (2.27 ± 0.47) scores vs. (3.09 ± 0.70) scores, the Ramsay sedation scores were significantly higher than those in group Ⅰ: (2.42 ± 0.51) scores vs. (2.00 ± 0.58) scores, (2.74 ± 0.45) scores vs. (2.11 ± 0.57) scores, (2.58 ± 0.51) scores vs. (2.05 ± 0.52) scores and (2.53 ± 0.51) scores vs. (2.00 ± 0.47) scores, and there were statistical differences (P<0.05). The press number of PCIA pump within 48 h after operation in groupⅡwas significantly lower than that in groupⅠ:(3.75 ± 1.45) times vs. (8.25 ± 2.61) times, the satisfaction score after operation was significantly higher than that in groupⅠ:(2.47 ± 0.51) scores vs. (1.60 ± 0.50) scores, and there were statistical differences (P<0.05). There were no statistical differences in the incidences of untoward reaction and vascular crisis (P>0.05). Conclusions Dexmedetomidine can relieve anxiety and nervousness, reduce postoperative consumption of sufentanil, and decrease the incidence of nausea and vomiting. In addition, dexmedetomidine does not affect the incidence of vascular crisis.
3.Effect of epidermal growth factor receptor inhibitor AG1478 induces cell death in HeLa cells

Objective To investigate the biological features of epidermal growth factor receptor inhibitor AG1478 on cervical carcinoma cell. Methods The proliferation of HeLa cells under AG1478 stimulation was determined by CCK8 assay. The expression of EGFR downstream signaling protein and apoptotic relative protein were examined by Western blot and transcription of apotosis?related genes were measured by RT?qPCR in AG 1478 treated HeLa cells. Nuclear transport of phosphorylated ERK were measured through ICC assay. TUNEL assay was used to determine early stage of apoptosis. Results CCK8 assay showed that AG1478 inhibit the proliferation of HeLa cells and also block phosphorylated level of EGFR ,ERK and AKT. Furthermore ,nuclear transport of phosphorylated ERK upon EGF stimulation were blocked and pro?apoptotic proteins were up?regluated with activat ed cleaved Caspases. Conclusion AG1478 inhibited the proliferation and induced apoptosis in HeLa cells and it could be a potential therapeutic drug for cervical carcinoma.
4.Pathogenic bacteria causing lower respiratory tract infections in the elderly orthopedics patients after general anesthesia and analysis of the risk factors

Objective To explore the incidence rates of lower respiratory tract infections after general anes-thesia in the elderly orthopedics patients and to analyze the related risk factors.Methods A total of 45 patients with lower respiratory tract infections in 479 elderly orthope -dics patients who received general anesthesia were enrolled in the study.The bacterial identification and drug susceptibility testing were preformed after the collection of sputum specimens.The univariate factor analysis and multivariate logistic regression analysis were performed to identify the risk factors for the lower respiratory tract infections after general anesthesia,including the age,gender,smoking histo-ry,chronic respiratory diseases history,basic cardiovascular disease,diabetes,mental disease,and endotracheal tube type,anesthesia ventilation time,postoperative extubation time,and postoperative analgesia.Results Smoking history (Wald =5.058,OR =4.618),chronic respiratory diseases history(Wald =37.950,OR =20.507),mental disease (Wald =26.303,OR =75.644),endotrac heal tube type(Wald =9.035,OR =11.758),anesthesia(Wald =15.035, OR =20.163)and postoperative extubation time(Wald =14.243,OR =12.613)were identified as the risk factors for lower respiratory tract infections after general anesthesia in the elderly orthopedics patients (all P <0.05 ). Conclusion Risk factors should be intervened in elderly orthopedics patients who receive general anesthesia,and the antibiotics should be chosen on the basia of the drug susceptibility testing.
5.Three-dimensional finite element stress distribution of condyle during the different vertical dimensions of edentulous jaw

Objective To find the characteristics of stress distribution on condyle during different vertical distances of edentulous jaw.Methods Three dimensional finite element models of the mandible and temporomandibular joint were established with the complete dentures in different heights by helix-CT scanned transverse sections.The stress distribution of the condyle was analyzed under load.Results The compressive stress and tensile stress coexisted on the whole condyle with the different vertical distances.The trend of distribution was consistent practically,but the stress presented a large difference on different locum.With the decrease of the vertical distance(from H-2 to H-6),the stress presented a decreasing trend but an increasing trend at H-8.Conclusion Changes in the vertical distance produce different effects of stress on edentulous jaw under the same load.
6.Improvement in Determination of Diethylene Glycol and Ethylene Glycol in Glycerol

Objective:To establish an improved method for the determination of diethylene glycol and ethylene glycol in glycerol on the basis of the method in Chinese pharmacopoeia (2010 edition). Methods: Glycerol samples were extracted by methanol, and detected by gas chromatography with a flame ionization detector. The column was a DM-624 capillary column(30 m × 0. 53 mm,3μm), and the initial temperature was 80℃ in the first 6min and then risen to 100℃ at the speed of 50℃·min-1, kept for 8min,fi-nally risen to 220℃ at the speed of 10℃·min-1 , kept for 10min. The split injection was used with the split ratio of 100∶1. The in-jection temperature was 250℃ and the detector temperature was 280℃. The flow rate was 6. 0ml·min-1 . Results:The RSD(%) of the method was lower than 5%, and the resolution of all chromatographic peaks met the requirements of the pharmacopoeia. The meth-od was used to analyze the commercial product of glycerol, and the contents of diethylene glycol and ethylene glycol were both less than the specified limits. Conclusion:The method is simple and rapid, and suitable for glycerol impurity inspection.
7.Analysis of selection of acupoints and medication rules in treating angina pectoris of coronary heart diseases with acupoint sticking therapy based on data mining

Objective:To explore the rules of acupoint sticking therapy based on data mining technology for the treatment of angina pectoris of coronary heart diseases, and to provide a reference for guiding acupoint selection and rational use of Chinese medicine.Methods:To search for the clinical research literature of acupoints for the treatment of angina pectoris of coronary heart diseases from China Academic Journal Database (Wanfang Data), China National Knowledge Resources Database (CNKI), China biomedical literature service system (SinoMed) and Chinese science and technology journal database (Chongqing VIP) dated from January 31, 2020. Using Microsoft Excel 2016, SPSS Statistics 21.0, and SPSS Modeler 14.2 statistical software as tools to conduct association analysis and cluster analysis on those acupoints and Chinese medicine.Results:A total of 102 acupoints application prescriptions were included, involving 38 acupoints. The top 5 frequently used acupoints were Danzhong (RN17), Neiguan (PC6), Xinshu (BL15), Zusanli (ST36), and Zhiyang (DU9). The top 5 acupuncture were Neiguan (PC6)- Sanyinjiao (SP6), Xinshu (BL15)- Geshu (BL17), Danzhong (RN17)- Geshu (BL17), Danzhong (RN17)- Jueyinshu (BL14), Danzhong (RN17)- Geshu (BL17) and Xinshu (BL15). There are 102 prescriptions of applied medicines with 88 kinds of Chinese medicine. The top 5 frequently used Chinese medicines were Chuanxiong Rhizoma, Borneolum Syntheticum, Salviae Miltiorrhizae Radix et Rhizoma, Olibanum, Santali Albi Lignum, the top three drug pairs were Chuanxiong Rhizoma- Cinnamomi Cortex, Allii Macrostemonis Bulbus- Trichosanthis Fructus, Olibanum- Myrrha. Conclusion:The commonly used single points to treat angina pectoris of coronary heart disease are Danzhong, and usually combined with Neiguan and Xinshu, and the acupoints along the Urinary Bladder Meridian of Foot- Taiyang are used frequently; the Chinese medicines are mainly used for activating blood circulation, genenrally reflects the therapeutic principle of removing blood stasis, dredging collaterals and relieving pain.
8.The Role of PP1 in the Cochlea of Mimetic Aging Mice Induced by D -galactose

Objective This study was designed to investigate the expression and potential role of the protein phosphatase 1 (PP1) impairment in D -galactose-induced inner ear aging mouse model .Methods Forty Kunming mice were randomly divided into two groups :the control group and D -galactose group ,20 mice for each .The D-galactose group mice were treated with a daily subcutaneous injection of the D -galactose solutions (800 mg · kg -1 · d-1 ) or an equal volume of normal saline(for the control group) in the nape back for 8 weeks .Eight weeks after D-galactose administration ,the effects of were measured by total Superoxide Dismutase (SOD) activity and Malon-dialdehyde (MDA) level in plasma .Immunofluoresence was performed to detect the location of the PP1 expression in the cochlea .Real-time PCR was performed to detect the level of PP1 mRNA in cochlea .A Western blot analysis was performed to analyze the protein levels of protein phosphates 1 nuclear targeting subunit (PNUTS) ,PP1 and caspase-3 in the inner ear .Results The MDA level was more significantly increased in the D -galactose group than in the control group ;however ,the total SOD activity was significantly decreased in the plasma of D -galactose- induced aging mice(P<0 .01) .The results showed that PP1 was predominantly localized in the nucleus and cyto-plasm of the hair cell ,spiral ganglion cell and stria vascularis cell .And the protein levels of PP1 and caspase-3 sig-nificantly increased ,and the level of PNUTS was decreased in the cochlea of the D -galactose group when compared to the control group .Conclusion PP1 contributes to the development of D -galactose-induced aging mice .
9.Characteristics of acupoint application for the sub-healthy condition treated with ancient and modern acupuncture based on data mining exploration.

The acupoint selection was retrieved from the ancient and modern literature on the treatment of sub-healthy condition with acupuncture. The law of acupoint application was analyzed so as to provide a certain reference to the determination of acupoint prescription in clinical acupuncture. The ancient literature was retrieved from Chinese basic ancient literature database. The modern literature was retrieved from Cochrane Library, Medline, PubMed, Ovid evidence-based medicine database, Chinese biomedical literature database, China journal full-text database, VIP journal full-text database and Wanfang database. The database mining software was adopted to explore the law of acupoint application in treatment of sub-healthy conditions with ancient and modern acupuncture. The acupoint use frequency, compatibility association rule, law for meridian use and the use regularity of specific points were analyzed. In the ancient treatment for sub-healthy condition, the top five commonly used acupoints are Shenmen (HT 7), Zhaohai (KI 6), Taibai (SP 3), Daling (PC 7) and Taixi (KI 3). The most commonly combined points are Zhangmen (LR 13), Taibai (SP 3) and Zhaohai (KI 6). The most commonly used meridians are the bladder meridian of foot-taiyang, kidney meridian of foot-shaoyin and liver meridian of foot-jueyin. The most commonly used specific points are the five-shu points. The most commonly used acupoints are located in the lower limbs. In the modern treatment, the top five commonly used acupoints are Zusanli (ST 36), Sanyinjiao (SP 6), Baihui (GV 20), Shenshu (BL 23) and Guanyuan (CV 4). The most commonly supplemented points are Hegu (LI 4) and Taichong (LR 3). The most commonly used meridians are the bladder meridian of foot-taiyang, the conception vessel and the governor vessel. The most commonly used specific points are the back-shu points. The most commonly used acupoints are located in the lower limbs. After the systematic comprehension of the relevant ancient and modern literature, the most commonly used acupoints are selected along the bladder meridian of foot-taiyang, and the most commonly used specific points are the back-shu points, the five-shu points and the front-mu-points. the acupoints are mostly located in the lower limbs.

10.BRICS AIDS medicines and antiretroviral therapy protection strategy and implications for China

BRICS has been dealing with the problem of increase in the number of the patients who require an-tiretroviral therapy and this therapy’s price-rise by promoting medicine to domestic production and reducing the impor-ted drug price. This paper reviewed the situation of BRICS HIV epidemic and prevention, anti-retroviral therapy drugs production and supply, drug security policy and strategy, and the following seven recommendations are straight forwarded to China based on the BRICS AIDS antiretroviral treatment coverage strategic comparison:(1) The estab-lishment of an ARV drugs co-ordination mechanism;(2) The reduction of the drug patent licenses while increasing the domestic generic drugs possibility;(3)Negotiations with the original research process of domestic pharmaceutical enterprises to obtain a voluntary license or speed up the technology transfer;(4) The use of antitrust laws to promote access to medicines for a voluntary license pharmacy localization; ( 5 ) If necessary, starting the compulsory medi-cines licensing to achieve localization;(6) Reducing the drug prices by bargain and negotiation;and (7) Strengthe-ning NGO built-in capacity.
