1.Whether nano-hydroxyapatite particles can influence apoptosis of mononuclear macrophage in rat abdominal cavity?

BACKGROUND: Because of their size effect, nanometer particles (NPs) can combine molecular within cells, which can result in cell necrosis or apoptosis. But there are no systematic mechanisms of apoptosis induced by NPs about biological safety of NPs.OBJECTIVE: To investigate the effect of nano-hydroxyapatite particles on mononuclear macrophage in rat abdominal cavity at celluar and molecular level.DESIGN, TIME AND SETTING: A materials-cytology observation was performed at Shanghai Biomaterials Research & Testing Center from January 2001 to December 2008.MATERIALS: SD rats of clean grade were provided by SINO-BRITISH SIPPR/BK LAB. ANIMAL Co., Ltd.; NPs were provided by Shanghai Institute of Ceramics.METHODS: Peritoneal fluid was extracted under a sterility environment to in vitro separate and culture mononuclear macrophage using adherence method. The concentration of cell was adjusted at 2×10~9/L. At 300 W/40 kHz ultrasound, cell suspension containing 20,100 and 200 mg/L nano-hydroxyapatite particles was prepared to induce mononuclear macrophage for 24 hours, respectively. A normal control group was established.MAIN OUTCOME MEASURES: Ultrastructural phenotype was detected using transmission electron microscope; apoptotic rate was measured using AnnexinV-EGFP/PI staining; variation of apoptosis-related P53 gene expression was detected using Western Blot.RESULTS: Pseudopodia of mononuclear macrophage in the normal control group were intact, nuclear membrane was normal, and nucleoplasm was uniformed. After inducing by 20,100, 200 mg/L nano-hydroxyapatite particles for 24 hours, apoptotic morphological characteristics were typical in mononuclear macrophage. Compared with normal control group, apoptotic rate was significantly increased following the induction of 20 and 100 mg/L nano-hydroxyapatite (P< 0.01), and the increasing in the 100 mg/L nano-hydroxyapatite particle group was greater than 20 mg/L nano-hydroxyapatite particle group (P < 0.05). P53 protein was not observed in mononuclear macrophage in the normal control group. Following induction of varying concentrations of nano-hydroxyapatite particles, P53 protein expression was increased in the mononuclear macrophage, and the expression was positively related to concentration of nano-hydroxyapatite particles.CONCLUSION: Nano-hydroxyapatite particles can induce p53 expression through phosphorylation, which promotes downstream genes and finally results in cell apoptosis.
2.The Impact of the New Rural Cooperative Medical Scheme on Rural Medical Institutions

Objective: Analyze the impact of NCMS(new cooperative medical scheme) on rural medical institutions,and give suggestions for the sustainable development of medical institutions and the NCMS.Methods: Collect and analyze the efficiency and the account materials of the county-level healthcare departments and the town-level health centers of Zhen'an County in 2003 and 2006.Results: The implementation of NCMS produced an effect on the increase of the outpatient medical service;the service quality of the county-level medical institutions and the town-level health centers needs to be enhanced in spite of the increase of in-patient medical service;the medical institutions' income increased,and the medical service fees increased rapidly;the expenditure per person and time rose sharply.Conclusions: Management mechanism of NCMS should be further developed,construction and supervision of the county-level medical sectors and the town-level health centers,especially supervision of the quality of medical sectors be enhanced;the payment system be changed to control the growth rate of medical expenses.
3.Analysis the cupric ion release characteristics of different copper raw materials in intrauterine device in vitro using ICP method.

OBJECTIVETo study the Cupric ion release characteristics of different copper raw materials in intrauterine device in vitro by ICP. Reveal the relationship between purity and shape of Cu-IUD copper and copper ion release.
METHODSAccording to a certain proportion, the copper raw materials were 100 times diluted into the simulated uterine solution at 37 +/- 0.5 degrees C. Replaced medium at certain time points and collected soaking liquid. Using ICP analyzed the concentration of copper ion released.
RESULTSThe largest daily release of copper ions was in the first 7 days. There was no statistically significant difference between the copper ion release amount of 99.99% and 99.95% purity copper wire (P > 0.05). The release of copper ion of the copper wire was far greater than that of the copper pipe in early stage (P < 0.01). The release amount decreased and stabilized at 56 day.
CONCLUSIONRelease characteristics of copper ion could effectively analysis by ICP. And in the same area, the release amount of copper ions of copper wire was greater than that of copper pipe.

4.Effect of Social Support, Coping Styles and Self-efficacy on Depression of Adults with Epilepsy

ObjectiveTo explore the effect of the social support, coping styles and self-efficacy on depression of adults with epilepsy. Methods84 patients with epilepsy were evaluated with Zung's self rating depression scale (SDS), general conditions questionnaire, Social Support Scale, Coping Styles Questionnaire, General Self-efficacy Scale. Based on the SDS, they were divided into the depression group and the epileptic group.Results52 patients suffered from depression (61.2%). There were significant differences in general self-efficacy, coping styles and social support between the depression group and the epileptic group (P<0.05). The score of SDS was positively correlated with the frequency of seizure, duration of epilepsy, fantasy, self-accusation and avoiding (P<0.05), and negatively correlated with the economic status, total support, objective support, subjective support, utilization of support, problem solving, help seeking, rationalizing and self-efficacy (P<0.05). Multiple regression analysis showed that the score of SDS was correlated with the self-efficacy, help seeking, self-accusation, economic status and frequency of seizure.ConclusionThe depression of adults with epilepsy are affected by the social support,coping styles and self-efficacy.
5.Effect of the ioversol on early renal function in patients undergoing cerebral vascular intervention

Objective To investigate the effect of different doses of ioversol on renal function,and to explore early renal injury biomarkers on contrast induced kidney injury and safe ioversol dosage.Methods A total of 158 cases (98 males and 60 females) undergoing cerebral vascular intervention (CVI) in our department was selected with age ranging from 23 to 81 years old (average age 59.70 ± 12.02).Based on ioversol dosage in surgery,patients were divided into three groups:low dose group (≤ 150 ml,n =49),middle dose group (151-200 ml,n =74),and high dose group (>200 ml,n =35).U-κ,U-λ,urinary transferrin (UTRF),urine microalbumin (UMA),urinary immunoglobulin IgG (UIgG),urine beta2-microglobulin (Uβ2-MG),Uα1-MG,urinary N-acetyl-beta-D-glucosaminidase (UNAG),plasma cystatin C (CysC) and Scr were detected by scattering turbidimetry,immune turbidimetry and fully automatic biochemical analysis pre-surgery 24 h and post-surgery 72 h.Contrast-induced acute kidney injury (CI-AKI) was defined as laboratory increase of Scr value≥44.2 μmol/L or ≥25% from baseline measurement at 48 hours after surgery.The relationship in ioversol dosage and various factors was assessed by Single and multiple factors binary logistic regression analysis.Results According to the criterion that Scr increase value were ≥44.2 μmol/L,of 158 cases,3 cases occurred CI-AKI,the AKI incidence was 1.90%.Based on the criterion that Scr increase value was ≥25%,33 cases occurred CI-AKI,the incidence was 20.89%.The concentration of U-κ,UTRF,Uα1-MG,UNAG and plasma CysC were significantly different in high dose group compared to low ioversol dose group (P < 0.05),while the other biomarkers had no significant difference (P > 0.05).Conclusions The contrast media-ioversol could lead to CI-AKI;when the dosage of ioversol was more than 200 ml one-time,the concentration of U-κ,UTRF,Uα1-MG,UNAG and plasma CysC increased significantly.U-κ,UTRF,Uα1-MG,UNAG and plasma CysC could predict the early renal injury in patients who undergoing CVI.The rise of U-κ,UTRF,Uα1-MG,UNAG and plasma CysC are related to the dosage of ioversol.Furthermore,possibility of kidney injury is significantly high when ioversol dosage is more than 200 ml one-time.
6.Correlation of sole prenatal indication to fetal chromosomal karyotype abnormality

Objective To analysis the clinical high risk factors for fetal chromosomal abnormalities.Methods Amniocentesis,chromosomal karyotype analysis and other related methods were performed on 4829 pregnant women,who presented sole indication of prenatal diagnosis such as advanced age,high risk factors and fetal ultrasound abnormalities,for analyzing the correlations of those women to the incidence of fetal chromosomal abnormalities.Results The detection rates of abnormal karyotype were 5.0% (57/1143),1.7% (40/2367) and 4.3% (57/1319) in the older women group (age>35),abnormal maternal serological screening group and abnormal fetal ultrasound finding group,respectively.The detection rats of karyotype abnormality were 6.9% (23/333) in women with fetal congenital heart diseases,8.5% (20/234) in those with abnormal amniotic fluid,1.1% (1/89) in those with fetal ventriculomegaly,1.1% (10/898) in those with fetal intracardiac hyperechogenicity,5.9% (2/34) in those with fetal choroid cyst and 5.6% (1/18) in those with fetal renal pelvis broadening.Conclusion The pregnant women with age>35,fetal sonographic structural anomalies or two or more soft marker abnormalities should be prenatally diagnosed and doing the genetic counseling combined with the family history.
7.Effect of Acupoint Magnetic Therapy on Mild Cognitive Impairment

Objective To observe the effects of mind-regulating brain-developing acupoint magnetic therapy in patients with mild cogni-tive impairment (MCI). Methods From May to October, 2015, 60 patients with MCI were randomly divided into control group (n=30) and observation group (n=30). The control group received magnetic therapy on Yangming meridians, and the observation group received magnet-ic therapy on mind-regulating brain-developing acupoints, for two months. They were assessed with Montreal Cognitive Assessment (Mo-CA), digit-span and digit symbol of Wechsler Adult Intelligence Scale (WAIS). Results After treatment, the scores of MoCA, digit-span and digit symbol of WAIS significantly increased in the observation group (t>4.858, P<0.001), and were higher than that in the control group (t>2.188, P<0.05), however, only the scores of MoCA and digit symbol of WAIS significantly increased in the control group (t>5.527, P<0.001). The scores of visual space and execution, and delayed recall of MoCA increased in the observation group (t>2.324, P<0.05), and were higher than that in the control group (t>2.262, P<0.05) after treatment, and no significant difference was found in other domains (P>0.05). Conclusion Magnetic therapy on acupoints related with cognition could improve the cognitive function in patients with MCI.
8.Effect of hydrogen on endoplasmic reticulum stress during hypoxia-reoxygenation in PC12 cells

Objective To investigate the effect of hydrogen on endoplasmic reticulum stress during hypoxiareoxygeuation(H/R)in PC12 cells.Methods PC12 cells were randomly divided into 4 groups:normal control group(group NC),positive control group(group PC),H/R group and hydrogen group(group H).In group NC.the cells were cuhnred routinely for 25 h.In group PC,the cells were cultured routinely for 1 h and then in RPM1-1640 culture medium saturated with hydrogen for 24 h.In H/R group,the cells were exposed to 1 h of hypoxia followed by 24 h of reoxygenation.In group H,the cells were exposed to 1 h of hypoxia and then reoxygenated in RPMI-1640 culture medium saturated with hydrogen for 24 h.Hypoxia-reperfusion was produced by 1 h exposure of cells to 5% CO2 in an incubator at 37 ℃ in RPMI-1640 culture medium containing Na2S2O4 with the final concentration of 5.0 mmoI/L,followed by 24 h reoxygenation in the normal RPMI-1640 culture medium.The relative rate of cell proliferation was detected by WST-1,The concentration of MDA was determined by thiobarbituric acid method.The expression of caspase-3 was determined by immuno-histochemistry.The expression of activating transcription factor-4(ATF4)mRNA and C/EBP homologous protein(CHOP)mRNA was determined by RTPCR.Results Compared with groups NC and PC,the relative rate of cell proliferation was significantly decreased,MDA concentration was significantly increased,and the expression of caspase-3,ATF4 mRNA and CHOP mRNA was up-regulated in group H/R,and the expression of ATF4 mRNA and CHOP mRNA was up-regulated in group H(P < 0.05).There was no significant difference in the relative rate of cell proliferation,MDA concentration,and the expression of caspase-3,ATF4 mRNA and CHOP mRNA between groups NC and PC(P > 0.05).Compared with group H/R,the relative rate of cell proliferation was significantly increased,MDA concentration was significantly decreased,and the expression of caspase-3,ATF4 mRNA and CHOP mRNA was down-regulated in group H(P < 0.05).Conclusion Hydrogen can decrease cell apoptosis and attenuate H/R injury to PC12 cells through inhibiting endoplasmic reticulum stress.
9.Coating technique of pH-time lag colon-specific matrine delivery mini-pill

AIM:To study the coating technique of pH-time lap colon-specific matrine delivery mini-pill consisted of the time lag release coating (inner layer) and enteric coating (out layer). METHODS:To filter the coating composition based on the index of dissolution of matrine and oxymatrine in vitro and the appearance rating of miui-pill. RESULTS:4 The coating composition of inner layer was the alcoholic solution,consisted of 2% EC,0.4% DEP and 2% talc powder. Then the coating composition of out layer was the alcoholic solution consisted of 5% Eudragit S100,3% talc powder and 0.5% TEC. The dissolution tests in vitro indicated that matrine and oxymatrine were not dissolved in the simulated gastric juice in 2 h. The accumulative amount of matrine and oxymatrine were less than 15% in the simulated intestinal fluid in 4 h. The amount of matrine and oxymatrine were 80.7% and 83.5% in the simulated colon juice in 2 h. CONCLUSION:The mini-pill could achieve the goal of delivering in the specific colon.
10.Effects of a fusion protein LTβR-Fc on ovalbumin-induced dermatitis in a mouse model

Objective To evaluate effects of a fusion protein LTβR-Fc, which can block the herpesvirus entry mediator ligand (LIGHT-HVEM)signaling pathway, on ovalbumin-induced dermatitis in a mouse model. Methods Thirty BALB/c mice were randomly and equally divided into 3 groups: blank control group treated with 100 μl of sodium chloride physiological solution, model group sensitized with 100 μl of sodium chloride physiological solution containing 100 μg ovalbumin, blocker group firstly blocked with 100 μl of sodium chloride physiological solution containing 100 μg LTβR-Fc followed by sensitization with 100 μl of sodium chloride physiological solution containing 100 μg ovalbumin at 24 hours after the blocking. Disease severity was evaluated by eczema area and severity index (EASI)score, and lesional size was measured on day 0, 4, 8, 12, 15, 20, 23, 27, 31 and 34 after the first sensitization. A total of three sessions of sensitization were carried out. At the end of treatment, all the mice were sacrificed after serum was obtained from their orbital cavities. Thereafter, tissue specimens were obtained from skin lesions, and single cell suspensions of the spleen were prepared. RT-PCR was performed to detect mRNA expressions of interferon γ (IFN-γ), interleukin 4 (IL-4)and IL-5 in murine lesions, ELISA to measure IFN-γ, IL-4 and IL-5 levels in culture supernatants of murine splenocytes, as well as ovalbumin-specific and total IgE and IgG1 levels in murine sera. Results LTβR-Fc significantly suppressed inflammatory response in the mouse model of dermatitis induced by ovalbumin. Compared with the model group, the blocker group showed significantly decreased lesion area and EASI score (both P < 0.05). In addition, a significant decrease was observed in the mRNA expressions of IL-4 (0.88 ± 0.25 vs. 1.81 ± 0.25, P < 0.05), IL-5 (0.75 ± 0.15 vs. 1.24 ± 0.26, P < 0.05)and IFN-γ (0.62 ± 0.09 vs. 1.11 ± 0.19, P < 0.05)in murine lesions, and in supernatant levels of IL-4 (9.58 ± 1.44 ng/L vs. 20.12 ± 5.39 ng/L, P < 0.05), IL-5 (11.37 ± 2.02 ng/L vs. 22.77 ± 4.07 ng/L, P < 0.05)and IFN-γ (16 167 ± 950.40 ng/L vs. 23 930 ± 44.20 ng/L, P < 0.05)in the blocker group compared with the model group. The serum levels of both total IgE and ovalbumin-specific IgE were significantly lower in the blocker group than in the model group(total IgE: 27 466.67 ± 2 052.64 μg/L vs. 32 277 ± 407.53 μg/L, P < 0.05; ovalbumin-specific IgE: 1 296.33 ± 32.72 μg/L vs. 2 323.33 ± 502.43 μg/L, P < 0.05), so were those of total IgG1 (0.46 ± 0.11 μg/L vs. 0.84 ± 0.11 μg/L, P < 0.05)and ovalbumin-specific IgG1 (0.62 ± 0.11 μg/L vs. 0.86 ± 0.07 μg/L, P < 0.05). Conclusion The fusion protein LTβR-Fc can alleviate symptoms of ovalbumin-induced dermatitis in the mouse model likely by suppressing the LIGHT-HVEM signaling pathway, suggesting that this signaling pathway may serve as a target for the treatment of dermatitis(such as atopic dermatitis).
