1.The value of blood lipid and CA125 in identification diagnosis between lung cancer and pulmonary tuberculosis

Objective To explore the value of blood lipid and CA125 in identification diagnosis between lung cancer and pulmo‐nary tuberculosis .Methods The blood lipid and serum CA125 levels in 131 patients with lung cancer( Ⅰ - Ⅳ stage) ,53 patients with pulmonary tuberculosis and 60 normal controls were determined by biochemstry ,immunoturbidimetry and electrochemilumi‐nescence immunoassay and was done compared study .Results The serum TC and HDL‐C level in 131 patients with lung cancer were decreased with severity and were lower than in those of in 53 patients with pulmonary tuberculosis(TC :P< 0 .05 ,< 0 .05 ,<0 .01 and < 0 .001 respectively ;HDL‐C :P< 0 .05 ,< 0 .05 ,< 0 .01 and < 0 .01 respectively) .The serum LDL‐C level was no differ‐ences between lung cancer( Ⅰ - Ⅲ stage) and in 53 patients with pulmonary tuberculosis(P> 0 .05) only the lung cancer( Ⅳ stage) was a little increased(P< 0 .05) .The serum TG and ApoB/ApoA1 ratio levels in 131 patients with lung cancer( Ⅰ - Ⅳ stage) were increased with severity and were significantly higher than in those of 53 patients with pulmonary tuberculosis (TG :P < 0 .05 ,< 0 .05 ,< 0 .01 and < 0 .01 respectively ;ApoB/ApoA1 :P< 0 .05 ,< 0 .05 ,< 0 .01 and < 0 .01 respectively) .The level of CA125 in 131 patients with lung cancer was significantly higher than in those 53 patients with pulmonary tuberculosis(P< 0 .001) .Conclusion The serum TC and HDL‐C ,TG ,ApoB/ApoA1 ratio and CA125 levels may be the indexs of identification diagnosis between lung cancer and pulmonary tuberculosis .
2.Establishment of a gemcitabine-resistant human nasopharyngeal carcinoma cell line and its biological characteristics

Objective To establish a gemcitabine-resistant human nasopharyngeal carcinoma(NPC)cell line and study its characteristics.Methods The drug-resistant cell line CNE2/Gem was established by a procedure of treating the human NPC cells CNE2 with gemcitabine by pulse drug selection and continuous stepwise selection.The IC50 and resistance index(RI)to several commonly used anti-tumor drugs were tested by MTT assay.Fluorescence activated cell analysis(FACS)was employed for determining the cell cycle and concentration of fluorescence dye rhodamine 123 within the cells.Cell growth curve,doubling time and cell morphology were measured and observed.Results Duplication time of CNE2 and CNE2/Gem was 17.13h and 23.13h,respectively,as evaluated by the growth curve.The IC50 to gemcitabine increased from 2.84?1.63?g/ml in CNE2 to 42.95?7.53?g/ml in CNE2/Gem as tested by MTT assay at 48~72h exposure,the RI was 23.61(P
3.Effect of bevacizumab on apoptosis of drug-resistant nasopharyngeal carcinoma cells

Objective To investigate the effect of bevacizumab on apoptosis of drug resistant nasopharyngeal carcinoma(NPC) cells in vitro.Methods The human nasopharyngeal carcinoma(NPC) cell line CNE2 and its cisplatin-resistant CNE2/DDP cells were treated with bevacizumab and DDP in various concentrations.The lethal effects of the both drugs and the apoptosis rates on CNE2/DDP cells were then measured separately with MTT assay and flow cytometry,the mRNA expressions of Bcl-2 and Bax in CNE2,CNE2/DDP and CNE2/DDP/Bev cells were detected with semi-quantitative RT-PCR.Results The killing rate in 10?g/ml bevacizumab group was similar to that of blank group(0.0?4.1% vs.5.2?4.3%,P=0.180),while the rates of combination of 10?g/ml bevacizumab and DDP(0.1?g/ml and 0.2?g/ml,separately) were higher than that of using DDP only(42.3?6.5% vs.34.4?5.4%,P=0.041;62.6?5.5% vs.50.0?5.9%,P=0.009).The apoptotic rate of CNE2/DDP,which was caused by combining 0.1?g/ml DDP and 10?g/ml bevacizumab,was much higher than what was produced by DDP alone(87.29?3.38% vs.50.58?8.83%,P=0.049).By means of semi-quantitative RT-PCR assay,the expressions of Bcl-2 mRNA in CNE2,CNE2/DDP and CNE2/DDP/Bev cells were 0.613,0.952 and 0.135,respectively.Meanwhile,the expressions of Bax mRNA were 0.665,0.387 and 1.751,separately.Conclusion Bevacizumab can greatly increase the drug sensitivity of CNE2/DDP cells to DDP;apoptosis is inhibited in MDR cells,but it can be promoted by bevacizumab.
4.Studies of Peripheral Nerve Electrophysiology in Uremic Patients

Objective To analyze electrophysiology of peripheral nerve in uremic patients. Methods 24 uremic patients and 19 normalvolunteers were included, surface electrodes were used to observe the electrophysiology performance, including tibial nerve, peroneal nerve,sural nerve and skin sympathetic response. Results The motor conduction, sensory conduction and skin sympathetic response were significantlydifferent between uremia patients and normal volunteers (P<0.05). The electrophysiology performance showed that F wave latencyprolonged, nerve conduction slowed, distal latencies prolonged, sensory fibers damaged more serious than motor fibers. Conclusion Peripheralneuropathy is one of the most common complications in uremia patients, neural electrophysiological abnormalities are significantly earlierthan clinical symptoms, nerve electrophysiological examination is of great significance in diagnosis and evaluation uremic peripheralneuropathy.
5.The value of serum intestinal fatty acid binding protein measurement in discriminating intestinal ischemia in patients with acute abdomen

ObjectiveTo assess the differential diagnostic value of serum intestinal fatty acid binding protein (I-FABP)in distinguishing intestinal ischemia patients from acute abdomen patients.MethodsA total of 151 patients with acute abdomen and 17 healthy controls from the PLA General Hospital were enrolled from November,2009 to August,2011. Serum I-FABP levels were measured by ELISA.According to the ROC curve,the cut-off value,sensitivity,specificity,positive likelihood ratio (PLR),negative likelihood ratio ( NLR),positive predietive value (PPV) and negative predictive value (NPV) were calculated. ResultsOf the 151 acute abdomen patients,there were 24 intestinal ischemia patients and 127 without intestinal ischemia.Serum I-FABP level in intestinal ischemia group [( 109.67 ±48.82) μg/L]was significantly higher than those in patients without intestinal ischemia [(36.78 ± 11.25) μg/L]and healthy controls[(8.33 ±6.25) μg/L]( all P values <0.01 ).The serum I-FABP cut-off value for the diagnosis of intestinal ischemia was 87.52 μg/L.Serum I-FABP was efficient in terms of sensitivity (0.762),NPV(0.963),PLR(3.05) and NLR (0.24) in the diagnosis of intestinal ischemia.ConclusionI-FABP is potentially useful for discriminating intestinal ischemia from acute abdomen.
6.Effects of mechanical ventilation with different tidal volumes on right ventricular hemodynamics in acute respiratory distress syndrome rats

Objective:To explore the effect of different tidal volumes (VT) on the hemodynamics of right heart in acute respiratory distress syndrome (ARDS) rats induced by oleic acid (OA).Methods:Sixty adult male Sprague-Dawley (SD) rats were divided into control group ( n = 20), ARDS model group ( n = 20), low VT (LVT) group ( n = 10) and high VT (HVT) group ( n = 10) by random number table. ARDS model was reproduced by injecting OA 0.15 mL/kg through a jugular vein. The control group was given the same amount of normal saline. The success of modeling was judged by the oxygenation index (PaO 2/FiO 2) 2 hours after modeling, at the same time, the lung tissues were collected, the wet/dry weight (W/D) ratio was determined, and the lung histopathological changes were measured by lung injury score. The rats in the LVT group and HVT group were given mechanical ventilation with VT of 6 mL/kg or 20 mL/kg for 4 hours, respectively at 2 hours after modeling. The rats in the control group and the ARDS model group maintained spontaneous breathing. After mechanical ventilation for 4 hours, the heart rate (HR), right ventricular systolic pressure (RVSP), the maximum rate of rising of right ventricular pressure (dp/dt max), and the blood pressure (BP) were measured. Meanwhile, arterial blood samples were collected for blood gas analysis, including pH value, arterial partial pressure of oxygen (PaO 2), arterial partial pressure of carbon dioxide (PaCO 2) and PaO 2/FiO 2. Results:The rats in the ARDS model group showed symptoms of respiratory distress 1 hour after modeling, and the lung tissue samples showed obvious patchy bleeding 2 hours after modeling, while the control group showed no such changes. The PaO 2/FiO 2 in the ARDS model group was significantly lower than that in the control group [mmHg (1 mmHg = 0.133 kPa): 294.3±5.9 vs. 459.0±4.4, P < 0.01], and the lung W/D ratio and lung injury score were significantly higher (lung W/D ratio: 8.24±0.25 vs. 4.48±0.13, lung injury score: 0.60±0.03 vs. 0.12±0.02, both P < 0.01). It indicated that ARDS model was successfully reproduced. The arterial blood gas analysis and hemodynamic parameters of the ARDS model group were significantly worse than those of the control group. After 4-hour mechanical ventilation, the blood gas parameters of the LVT group were better than those of the ARDS model group and the HVT group [pH value: 7.36±0.02 vs. 7.24±0.02, 7.13±0.01; PaO 2 (mmHg): 92.4±2.1 vs. 61.8±2.3, 76.6±2.2; PaCO 2 (mmHg): 49.6±1.7 vs. 61.8±1.8, 33.6±1.3; PaO 2/FiO 2 (mmHg): 440.0±10.2 vs. 274.3±21.4, 364.7±10.5; all P < 0.05]. HR, BP and dp/dt max in the LVT group were significantly higher than those in the ARDS model group and the HVT group [HR (bpm): 346.9±5.4 vs. 302.3±10.1, 265.5±12.2; BP (mmHg): 125.4±2.2 vs. 110.0±2.5, 89.2±2.8; dp/dt max (mmHg/s): 1 393.3±30.3 vs. 1 236.4±20.5, 896.1±19.5; all P < 0.05], and RVSP was significantly lower than that in the ARDS model group and the HVT group (mmHg: 31.3±0.4 vs. 34.0±1.0, 38.8±0.9, both P < 0.05). Conclusion:Mechanical ventilation with low VT can improve the hemodynamic parameters of the right ventricle and protect the function of the right heart in ARDS rats.
7.Advance in memory NK cells study

Natural killer ( NK) cells have historically been considered short-lived cytolytic cells that can rapidly respond against microbial pathogens and tumors in an antigen-independent manner.Recently,NK cells have been shown to possess features of a-daptive immunity with immunological memory in a manner similar to that of T and B cells.Three major viewpoints of NK cell memory initially arose from the studies of NK cell memory to recall to mouse cytomegalovirus ( MCMV ) , cytokine and skin-contact hypersensitive chemical antigens.Currently,NK cell memory has been reported in acute infection of mouse HSV ,influenza virus,HCMV and simian immunodeficiency virus ( SIV).Here,we review the latest discoveries and unsolved questions regarding NK cell memory in these models.Studies to reveal the mechanisms for NK cell memory may provide opportunities for the therapeutic use of NK cells in in -fectious diseases and cancer.
8.Protective effect and mechanism of astragalus injection in cerebral ischemia reperfusion injury in rats

Objective To investigate the neuroprotective effect and mechanism of astragalus injection in cerebral ischemia reperfusion injury in rats .Methods A total of 70 healthy adult male Wistar rats were subjected to establish middle cerebral artery occlusion reperfusion models by inserting a monofilament thread from the external -internal carotid artery and treated by injecting 1g/L intraperitoneally astragalus injection (3ml/kg).The neurobehavioral function of rats was evaluated by Longa ’ s test and the cerebral infarction volume was calculated by tetrazolium chloride staining .The shape and ultrastructure of neurons in parietal cortex were observed by HE stain TEM .The early apoptotic ratio of neurons was detected by flow cytometry .The expressions of c-Jun N-terminal kinase 3 ( JNK3) mRNA and protein were determined by RT-PCR and Western blotting , respectively.Results After treatment with astragalus injection , the expressions of JNK3 mRNA and protein reduced significantly , the number of neuronal apoptosis in parietal cortexminus , the cerebral infarction volume shrink, the neuronal shape and ultrastructure and animal neurobehavioral function were improved significantly than those in model group rats .Conclusion The results suggest that astragalus injection may inhibit neuronal apoptosis , reduce the infarction volume and improve the animal neurobehavioral function by down -regulated the expression of JNK 3 gene following cerebral ischemia in rats .
9.Response of glucagon to the changes of glucose and blood pressure in elderly patients with type 2 diabetes

The response of glueagon to the change of glucose and its relation to blood pressure in 71 eidely patients with type 2 diabetes was investigated. The results showed that the postprandial increment of glucagon in the group of patients with postprandial 2h plasma glucose increment<2.5 mmol/L was significantly higher than that in the group with plasma glucose inerement≥ 2.5 mmoi/L (P<0. 05). The postprandial increment of glucagon in patients with normal blood pressure was significantly higher than that in patients with hypertension (P<0.05). The results suggest that the decreased response of glucagon to the change in plasma glucose in elderly patients with type 2 diabetes is related to increased blood glucose and high blood pressure.
10.Effects of FXR gene deletion on neurobehaviors and neurotransmission systems in female mice

Aim To investigate how Farnesoid X re-ceptor (FXR),a nuclear hormone receptor,acts on neurological behaviors such as emotion,social behav-ior,memory and so on.Methods FXR’s function in central nervous system was evaluated by conducting a battery of behavioral tests including elevated plus maze test (EPMT),forced-swimming test (FST),social in-teraction test (SIT ), and passive avoidance test (PAT),and the contents of neurotransmitters were de-termined by the LC-MS /MS method in FXR knockout (KO)female mice and their wild-type controls.Re-sults FXR KO mice showed significantly increased immobility time in FST (P <0.01 ),and it showed in-creased tendency to enter the open arms in EMPT (P<0.01 ).The number of probing the open arms by FXR KO mice was more than that of the controls.Mo-reover,in SIT,FXR KO mice had remarkably in-creased sniffing interactions with the stranger mouse in the same cage (P <0.01 ).But in PAT,the latency for FXR KO mice to enter the dark chamber on the test day and the number of FXR KO mice to enter the dark chamber didn’t differ from those wild-type mice.In hippocampus,the contents of GABA,Glu,and NE were decreased prominently in FXR KO mice (P <0.05,P <0.05 and P <0.01 ,respectively)as well as the ratio of GABA to Glu (P <0.05).But in pre-frontal cortex,none of the neurotransmitters examined showed any difference between FXR KO mice and their controls.Conclusion FXR may be involved in main-tenance of the homeostasis of neural transmission in the central nervous system,thereby influences the emotion-al and social behavior in animals.
