1.Correlation Study between Soothing Liver-qi Stagnation bySi-Ni-San and Effect of Improving Sleep

Pharmacological evaluation on the improvement of sleeping effect usingSi-Ni-San(SNS) in order to conduct further study on scientific connotation of soothing liver-qistagnation. Rats, mice and fruit flies were used as animal model. Pharmacological assessment was conducted and done on the improvement of sleep effect by SNS with the cortical EEG scanning and analysis, biochemistry, molecular pharmacological methodology and technology. SNS can prolong the sleeping time of mice which were induced by pentobarbital sodium. Its effect showed the dose-dependent and time-dependent manner. It can prolong the slow-wave sleep time in normal and sleep deprivation rats. It influenced several neurotransmitters and endogenous sleep factors in brain of normal and sleep deprivation rats. SNS can improve sleeping. The effect was closely related the effect of soothing liver-qi stagnation. The function was realized through multiple pathways and targets.
2.Impact of conflict psychological stress on rats'sleep time and sleep phrase

ObjectiveTo discuss the impact of conflict psychological stress on rats'sleep time and sleep phrase.Methods20 Wistar male rats were randomly divided into vehicle group and model group,10 in each group.The improved experiment of Vogel drinking water conflict was used to establish the model of conflict psychological stress in rats and monitoring the changes of rats'weight and concentration of corticosterone (CORT) in serum after a week.After one week,the 12 h EEG and EMG of rats were traced by the polygraph in order to clarify the effect of conflict psychological stress on rats′ sleep time and sleep phrase.ResultsAfter one week,compared with vehicle group the weight of model group decreased significantly ( t =2.87,P < 0.05 ),the concentration of CORT in serum raised significantly( t=5.19,P < 0.05 ).A week later,compared the vehicle group,total sleep time( TST),light sleep,slow wave sleep(SWS) and rapid eye movement sleep (REM) time in model group rats ( (337.8 ±84.2)min,( 169 ±49.2) min,( 136.9 ±40.8)min,(33.1 ± 10.1 ) min for vehicle group; ( 155.4 ±16.7 ) min,( 111.6 ± 16.4 ) min,( 30.6 ± 4.9 ) min,( 12.6 ± 3.3 ) min for model group) were significantly reduced ( t =6.75,2.43,5.51,5.78,P < 0.05 ),and the proportion of lightsleep increased ( (49.6 ± 3.4 ) %,( 71.7 ±5.4)% ) ( t =6.90,P<0.05),the proportion of SWS decreased( (40.3 ± 10.9)%,( 19.8 ±3.2)% ) ( t=6.74,P < 0.05 ) ;but no significant change in the proportion of REM ( t =1.57,P> 0.05 ).Rats,sleep-wake rhythm figure indicated that the rats'sleep duration reduced and the number of awakening increased ; sleep tended to be fragmentation.ConclusionsConflict psychological stress lead to hypothalamus-pituitary-adrenal axis hyperactivity,weight loss,and the rhythm of sleep and sleep phase change in rats.
4.Decoction of Fructus Schisandrae,s constituents absorbed into blood of rats after oral administration

Objective:For the further studies on the physical foundation of medicinal effi cacy of decoction of Fructus Schisandrae,to study its constituents absorbed into blood of rats after oral administration.Methods:Serum pharmacochemistry of Chinese medicine was used,based on the established high performance liquid chromatography analytical method of plasma effective constituents,to identify the constituents originated from the preparation and their metabolites in the serum by comparing fi ngerprint in vitro with that in vivo.Results:Five constituents have been detected in the blood,they were compound 1,2,3,4,5.Conclusion:Compound ⅣandⅤexist originally in Fructus Schisandrae decoction(including schisandrin),compound 1,2and 3 might be the metabolites of original constituents.
5.HPLC fingerprinting similarity analysis of Radix Paeoniae Alba

AIM: To establish a sensitive and specific HPLC method for controlling the quality of Radix Paeoniae Alba. METHODS: HPLC method was applied for quality assessment of Radix Paeoniae Alba.Column: Hypersil C_(18) column(4.61 mm?200 mm,51 ?m);flow-rate: 1.0 mL/min;column temperature: 25?C.mobile phase: acetonitrile(A) and H_2O(acidified to 0. 05% with phosphoric acid)(B).The concentrations of solvent A were 95%,86%,86% and 65% at 0,25,35,60 min,respectively. RESULTS: The HPLC chromatographic fingerprinting of Radix Paeoniae Alba,showing 14 characteristic peaks,was established from 21 batches of Radix Paeoniae Alba products.The results of similarity analysis was the same as that of herarchical cluster analysis. CONCLUSION: The chromatographic fingerprinting of Radix Paeoniae Alba with high specificity can be used to control its quality and ensure consistency between batches.
6.Studies on chemical components of lyophilized powder of Sinisan

Objective The chemical components of lyophilized powder of Sinisan were analyzed by the method of RP-HPLC.Methods HPLC Method was applied for quality assessment on lyophilized powder of Sinisan.Column: Kromasil C18 column(250 mm ? 4.6 mm,5 ?m);wavelength: 210 and 240 nm;flow-rate: 1.0 mL/min;column temperature: 28 ℃.Mobile phase: acetonitrile(A) and H2O(acidified to pH=3 with phosphoric acid)(B).Concentration of lyophilized powder of Sinisan was resolved into different parts and analyzed by the same method,the variation of the chemical components was analyzed.Results At 240 nm,21 peaks were detected and two of them were confirmed to be from Bupleurum chinense,three from Paeonla lactiflora,ten from Citrus aurantium,and five from Glycyrrhiza uralensis.At 210 nm,24 peaks were detected and one of them was confirmed to be from B.chinense,four from P.lactiflora,fourteen from C.aurantium,and five form G.uralensis.Conclusion This method can be used to establish the fingerprint chromatograph of lyophilized powder of Sinisan and elucidate the variation of chemical components completely.
7.The effects of olmesartan on ambulatory blood pressures and blood pressure variability in patients with mild to moderate essential hypertension

Objective To evaluate the effect of olmesartan medoxomil tablets (olmesartan) in comparison with Olmetec on 24 h ambulatory blood pressure (ABPM) and blood pressure variability (BPV)in patients with mild to moderate hypertension.Methods A randomized,double-blind,double-mimic controlled trial was performed.Forty-eight patients with mild to moderate essential hypertension were randomly into treatment group (olmesartan) and control group (Olmetec) for eight weeks.The ABPM was taken before and at the end of the trial.Results After eight weeks,treatment with olmesartan induced a significant reduction in ABPM in patients [(9 ± 3)/(11 ± 3) mmHg (1 mmHg =0.133 kPa)],which is similar with the reduction by Olmetec [(9 ± 4) / (9 ± 5) mmHg],P > 0.05.This situation holds for BPV with the standard deviations of 24 h,systolic blood pressure/diastolic blood pressure of pre-treatment and pro-treatment were (10 ± 2)/(11 ± 3) mmHg vs (10 ± 3)/(12 ± 2) mmHg in olmesartan group,and (10 ± 3)/(11 ±3) mmHg vs (12 ±3)/(12 ±4) mmHg in Olmetec group.(3) There is no difference in the rate of adverse event between olmesartan (10.42%) and Olmetec (8.33%) treatment(P > 0.05).Conclusion Similar to Olmetec,treatment with olmesartan once daily can significantly reduce ABPM in patients with mild to moderate essential hypertension.
8.The effect of different illumination condition on drosophila circadian rhythm and 24 h sleep time

Objective To investigate the influence of illumination changes on the drosophila circadian rhythm and 24 h sleep time.Methods We collect the unmated fruit flies (a total of 128,single-sex) under CO2 anesthesia within 8 hours of eclosion.And put them into the group of normal light group,day and night reversed group,24 h light group and 24 h dark group with the simple random sampling method,male and female with 4 single-sex groups(n=32).Change the illumination conditions of the living environment of the fruit flies and use Drosophila Activity Monitoring System (DAMS) and Data Acquisition System (DAS),to observe and compare the changes of drosophila circadian rhythm and 24 h sleep time between the different illumination conditions.Results Compared with the normal light group,drosophila circadian rhythm of the day and night reversed group was significantly reversed,the number of fruit flies' activities of 24 h light group was increased,and circadian rhythm was decline,drosophila circadian rhythm of the 24 h dark group is still there,but frequent activity appears during the day,and the 19 : 00 activity peak disappears.Conclusion The drosophila circadian rhythm and 24 h sleep time changes with illumination conditions change.
9.Effect of Salidroside on the sleep phase of insomnia rats

Objective:To observe the effect of the salidroside on the sleep phase of insomnia rats. Methods: Under the conditions of constant temperature,constant humidity,light automatically and electromagnetic shielding,the effect of the salidroside on the sleep phase of insomnia rats was observed with the method of the cortical EEG.Results:The results showed that salidroside can obviously decrease the duration of W and increase TST,primarily prolong SWS2 and REMS.Conclusion:Salidroside can prolong SWS2 and REMS of insomnian rats and regulate the sleep phase of insomnia rats near the normal rats.
10.Clinical study and analysis of pathogenic bacteria of pulmonary infection in patients with terminal cancer

Objective To analyze of pathogenic bacteria of pulmonary infection in patients with terminal cancer.Methods The clinical data were retrospectively analyzed including the general cultivation of pathogenic bac-teria in sputum and the results of drug sensitivity of hospitalization of patients with lung cancer from July 2013 to May 2014.Results 132 strains of pathogenic bacteria were cultured from120 cases of patients with pulmonary infection, including gram-negative bacilli accounted for the vast majority of 27.3%,accounted for 59.8% of Gram-positive coc-ci,Ranked in the top 5 pathogenic bacteria were Streptococcus pneumoniae,Staphylococcus aureus,Candida albicans, Klebsiella pneumoniae,there arevarying degrees of resistance to commonly used antibiotics in bacteria.Conclusion Selecting and using the antimicrobial agents according to the results of pathogen detection in patients,could improve the effectiveness of antibiotic treatment and reduce bacterial resistance.
