1.The change of high mobility group box-1 protein expression in the moose model with acute hepatic failure

Objective To study the expression-mode and dynamic transmutation of high mobility group box-1 (HMGB1) in hepatocytes of the mouse model with acute hepatic failure and to study the interaction beween HMGB1 and tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β). Methods The mouse model of acute hepatic failure was established by injecting D-galactosamine (D-GalN) and lipopolysaccharide(LPS). Immunohistochemistry SABC method was used to detect the HMGB1 expression at 6 time points. The enzyme linked immunosorbent assay (ELISA) was used to determine serum TNF-α. IL-1β levels. Paired t test was used for statistical analysis. Results The HMGB1 expression was detectable at 2 hours after injection, which dramatically increased over time and peaked at 24 hours after injection. The serum TNF-a level and IL-1β level increased right after injection. The TNF-a level peaked at 8 hours after injection with a maximum value of (473.42±22. 99) pg/mL. The IL-1β level peaked at 2 hours after injection with a maximum value of (724. 49±34. 24) pg/mL. Both cytokine levels slowly decreased after peaking. IL-1β level returned normal with (51. 49±36. 87) pg/mL. Conclusions HMGB1 is one of the most important factors during the development of acute hepatic failure, which can promote the secretion of TNF-α and IL-1β at early stage and be abundantly expressed under the effect of these cytokines at middle and late stages with the result of liver damage. This process is directly correlated with the development and severity of hepatic failure.
2.Influencing factors of retinal nerve fiber layer thickness in myopia

Conclusion RNFLT in myopia measured by OCT may have its own rule. Avg-RNFLT may have positive correlation with diopter in severe myopia group.
3.Feasibility analysis of Triptolide's role in treating filtering bleb fibrosis after the filtration surgery of glaucoma

At present, filtration surgery remains an important treatment of glaucoma, and filtering bleb fibrosis is the main cause for treatment failure. Filtering bleb fibrosis is a common fiber hyperplastic disease, and it relates to the activation and proliferation of fibroblasts and the excessive production of extracellular matrix ( ECM) such as collagen protein. The most frequently-used drugs for filtering bleb fibrosis in clinic are 5-fluoro-2,4 (1h, 3h) pyrimidinedione ( 5 - Fu ) and mitomycin ( MMC ) . Although they are effective in some degree, they also have some serious side effects which restrict their clinical use. Triptolide ( TPL) is a major active component of the medicinal plant, tripterygium wilfordii hook. f. ( TWHF) . TPL has multiple pharmacological activities including immunosuppressive, anti- inflammatory, anti- cancer and anti-fertility activity. Reviewing related literatures published in recent ten years, we confirmed that TPL seemed to possess a pharmacological activity in treating filtering bleb fibrosis. Since it has three major functions:1. inhibit the activation and proliferation of fibroblasts and the excessive production of collagen protein; 2. alleviate the inflammatory reaction after surgical wound to suppress fibrous scar formation; 3. TPL has a protective effect on retinal ganglion cells ( RGCs ) . We further find that TPL's anti-fibrosis activity mainly results from that it inhibits TGF-β/Smad,NF-κB and PI3K/AKT signal transduction pathway. This comprehensive analysis about the feasibility of Triptolide's role in treating filtering bleb fibrosis after the filtration surgery of glaucoma can help us develop new drugs for filtering bleb fibrosis and exploit TPL's clinical value on some level.
4.Methods for Components in Chlorine Dioxide Disinfectant

Objective In order to evaluate the quality and performance of chlorine dioxide disinfectant,it is necessary to develop a set of methods to determine the components in chlorine dioxide disinfectant.Methods Iodometry,malonic acid iodometry,improved malonic acid iodometry and five-step iodometry were employed to determine the components including ClO_2, Cl_2,ClO_2~-,and ClO_3~-in chlorine dioxide disinfectant,the advantages and disadvantages of the test methods were compared.Results Using iodometry,the liquid stabilized chlorine dioxide products activated by hydrochloric acid,the content of ClO_2 was 20.23 mg/ml that was accord with the content which was marked in the product label.Using malonic acid iodometriy,the liquid stabilized chlorine dioxide contents of ClO_2 was 19.99 mg/ml,Cl_2 was 0.35 mg/ml,in which a little chlorine was detected.Using improved malonate iodometry and five-step iodometry,the activation rates of the liquid stabilized chlorine dioxide products were 88.0% and 75.6% respectively,ClO_2 and ClO_2~-in the liquid stabilized chlorine dioxide products could be distinguished.Conclusion Active chlorine dioxide and residual ClO_2~-can be detected by using iodometriy and improved malonic acid iodometry,but iodometriy and malonic acid iodometry can not.
5.Strategies for automatic extraction of key words from medical news

The strategies for automatic extraction of key words from medical news were put forward by adding the MeSH terms into the general classification table in combination with the length of MeSH terms and location-weigh-ted MeSH terms.The key words randomly selected from 100 papers reporting medical news on 10 Websites were in-dexed and verified by machine indexing.The extraction accuracy was 0.34 and the recall rate was 0.30, showing that the strategies can be used for automatic extraction of key words from medical news.
6.Effect of minimally invasive percutaneous nephrolithotomy titanium laser lithotripsy for upper ureteral calculi

Objective To analyze the clinical effect of minimally invasive percutaneous nephrolithotomy titanium laser lithotripsy(MPCNL)on upper ureteral calculi.Methods According to digital table,68 patients with upper ureteral calculi were randomly divided into the study group and control group,cach group in 34 cases.The study group received the MPCNL treatment,the control group received extracorporeal shock wave lithotripsy(ESWL)therapy. The therapeutic effects were compared.Results The success rate of gravel,stone clearance rates of the study group were 91 .1 8% and 91 .1 8%,which were higher than 70.59%,67.65% of the control group,there were significant differences between the two groups(χ2 =4.660,5.757,P =0.031 ,0.0164).Conclusion Percutaneous nerve titanium laser lithotripsy for upper ureteral stones can achieve good therapeutic effect.
7.Progress of immunotherapy aiming at preserving residual beta cells of islet

As patients with type 1 diabetes,both the function and number of their beta cells decreased.Despite treating with insulin in early stage,it can't avoid the occurrence of serious complications or death.However,Immunotherapy,different from the previous replacement therapy,emphasizes re-balance between affected T cells and immune regulative effect,suppresses autoimmune reaction,and strengthens the tolerance of beta cells of islet to it.At present,a variety of immune medicines have already been approved for the treatment of type 1 diabetes,such as anti-CD3 monoclonal antibody,GAD-alum,etc.
8.Anti-inflammatory action of recombinant mouse HMGB1-A box in RAW264.7 cells

Objective: To study the influence of HMGB1-Abox on the expression of HMGB-1 in RAW264.7 cells stimulated by lipopolysaccharide(LPS)and its inhibition on the secretion of inflammatory factor.Methods: Use cloning vector to make up prokaryotic plasmid PET28a-HMGB1-Abox, which included the gene fragment of the A box of HMGB1, and recombinant vector was further transformed into Escherichia coil strain BL21(DE3),the recombinant plasmid was induced by isopropylthiogalactoside(IPTG)to express target protein.The protein was purified by Ni-column.We tested the effect of the different concentration of rHMGB1-Abox on the viabihty of RAW264.7 cells stimulated by LPS using CCK-8.Experimental group(EG)was treated with rHMGB41-Abox and control group(CG)was deh with PBS.TNF-α and IL-1β levels were detected by the enzyme linked immunosorbent assay(ELISA).Western blot and immunofluorescence staining method were used to compare the expression of HMGB1 in RAW264.7 cells with the experiment group and control group.Reverse transcription polymerase chain reaction(RT-PCR)was used to detect the variation tendency of HMGB1-mRNA.All procedures were manipulated at 2 \ 6 \ 12 \ 24 \ 48 h after treatment.Results: The recombinant prokaryotic expression vector PET28a-HMGB1-Abox was successfully constructed and mouse HMGB1-Abox protein about 14 kD was successfully expressed.There was a positive correlation between the viability of RAW264.7 cells and the concentration of protein and acting time.Compared to CG, TNF-α and IL-1β levels in EG declined significantly.In EG, Western blot and Immunofluorescence staining showed that the expression of HMGB1 protein was decreased and the expression of HMGB1-mRNA was reduced markedly and delayed.Conclusion: The rHMGB1-Abox could reduce expression and secretion of HMGB1 in RAW264.7 cells stimulated by LPS significantly,thereby to prevented the promotion of HMGB1 on pro-inflammatory mediator and inhibit the expression and secretion of inflammatory factors,which has some anti-inflammatory action.
9.Inhibitory effect of RNA interference targeting vascular epithelial growth factor-C on proliferation of human colon cancer cell line LOVO in vitro and In vivo

Objective This study was to explore the inhibitory effect of shRNA-VEGF - C on growth of human colon cancer cell line Lovo in vitro and vivo. Methods Recombinant VEGF-C short hairpin RNA (shRNA) plasmid was constructed and transfected into Lovo cells. The expression of VEGF-C was detected at mRNA and protein levels by real-time reverse transcription-polymerase chain reaction (RT-PCR). In vivo study, xenograft tumors were established by injecting LOVO cells into nude mice, then shR-NA-VEGF-C were injected into the tumors, the tumor volume and weight and the incidences of lymph node metastasis were detected. Immunohistochemical staining was used to detect the lymphatic microvessel density of colon cancer tissues. Results After transfection of shRNA-VEGF-C, the mRNA of VEGF-C in Lovo cells were down-regulated. Four weeks after injection, the tumor volume and tumor weight in VEGF-C-shR-NA group were significantly smaller than that in empty plasmid group and NS group [(324. 9 ± 64. 8 ) mm3 vs. (553.5±90. 1)mm3 and (570. 1±85.4)mm3; (3.01 ±0.55)g vs (4.65 ±0.65)g and (4.75 ±0. 75)g]. The incidences of lymph node metastasis (30. 1% ) were significantly inhibited compared with empty plasmid group (50. 2% ) and normal saline group (53. 1% ). In shRNA-VEGF-C group, and microlymphatic density (15.5 ± 6. 90) was also decreased compared with empty plasmid group (24. 18 ±6. 45 ), and normal saline group (29. 59 ± 8. 21 ) ( all P <0. 01 ). Conclusion shRNA-VEGF-C can inhibit the growth of LOVO cells in vitro and vivo. VEGF-C may inhibit the lymph node metastasis of colon cancer by suppressing lymphangiogenesis.
