1.Effects of DNMT1-siRNA on the pain behavior and the expression of SOCS1,p-ERK,p-CERB in CCI rats

Objective To investigate the effects of DNMT1 on neuropathic pain behavior and neuropathic pain modulation.Methods Thirty-two male SD rats, weighing 200-250 g, were randomly assigned into 4 groups (n =8 each):sham operation group (group S),chronic constrictive injury group (group CCI),CCI+ DNMT1-siRNA group (group CDS),CCI+ control-siRNA group (group CCS).Group CDS were intrathcally injected of DNMT1-siRNA (2 μg/10 μl),and group CCS were intrathcally injected of control-siRNA 7,8,9 days after operation.Mechanical withdrawal threshold (MWT)and thermal withdrawal latency (TWL)were measured before operation and on day 3,5,7,9,12,14 after operation.The rats were then sacrificed and L4-L6 segments of the spinal cord were removed for determination of SOCS1,p-ERK,p-CREB expression using Western blot on day 14.Results Compared with group S,MWT and TWL in group CCI and CCS were significantly decreased on day 3,5,7,9,12,14 after operation (P <0.05).Compared with group CCS,MWT and TWL in group CDS were significantly increased on day 9,12,14 after operation (P < 0.05 ). Compared with group S and CDS,SOCS1 was significantly downregulated,p-ERK and p-CREB were significantly upregulated in group CCI and CCS (P <0.05 ).Conclusion Intrathcal injection of DN-MT1-siRNA significantly relieves neuropathic pain by upregulating SOCS1,downregulating p-ERK and p-CREB in rats spinal cords.
2.Proliferation and apoptosis in primary gastrointestinal B-cell non-Hodgkin’s lymphoma and its association with Helicobacter pylori

Objective: To investigate primary gastrointestinal B cell non Hodgkin’s lymphoma for clinicopathological features, proliferation, apoptosis and its association with Helicobacter pylori (HP). Methods: Classification of tumors, expression of HP, proliferation and apoptosis related gene products were studied by immunohistochemistry. Apoptosis was studied by TUNEL (TdT [terminal deoxynucleotidyl transferase] dUTP nick end labeling). Results: There were 15 cases of mucosa associated lymphoid tissue (MALT) lymphoma (9 in stomach, 6 in intestine) and 42 cases of diffuse large B cell lymphoma (DLBCL) (28 in stomach, 14 in intestine) in all the 57 cases of primary gastrointestinal BCL. The average apoptotic indexes (AI) were 0.16%, 2.54% in MALT lymphoma and DLBCL respectively. The average proliferation indexes (PI) were 2.22% and 8.71%,respectively. The p53 positive rates were 6.7% and 35.7%. PI ( P =0.026) and p53 ( P =0.044) had significant differences; But Bcl 2 positive rates were 60.0% and 35.7%,and HP positive rates were 66.7% and 23.8%, respectively. HP had significant differences ( P =0.005). Conclusion: HP and gastrointestinal MALT lymphoma had significant relation. Apoptosis and proliferative activity were higher in DLBCL than in MALT. The higher Bcl 2 expression in MALT may be related to lower apoptosis. p53 promotes apoptosis and p53 gene mutation may play a role in the progression from MALT to DLBCL. HP antibody may be used in HP detection in gastrointestinal BCL.
3.The effect of Feixian Fang treating the idiopathic pulmonary fibrosis patients in MMPs and TIMPs

Objective:To research the therapeutic effect dnd pulmonary ground substance rebuilding in the treatment on IPF patients by Feixian-Fang and.Methods:Treatment 20 cases IPF by Feixian Fang RCT prednisone three months.obsive The therapeutic efficacy and the levels of matrix met alloproteinase 1,2,tissue inhibtor of met alloproteinase(TIMP-1,2).Results: The total effective rate of Feixian-Fang group is 84.2%.The total effective rate of prednisone group is 81.3%.Feixian-Fang can reduce the level of MMP-1,MMP-2,TIMP-1,TIMP-2 in the serum.Concluds:Feixian-Fang can treat IPF probably through inhibiting pulmonary ground substance rebuilding.
4.Correlation between health value and health-promoting lifestyle in elderly people at nursing home

Objective To explore the correlation between health value and health-promoting lifestyle in elderly people at nursing home. Methods Totally 130 elderly people from nursing home were investigated with health value scale and health-promoting lifestyle profile II to understand the status of their health value and health-promoting lifestyle and the correlation between them. Results The mean score of health value was 7.78±1.58, which was in the middle level;The mean score of health-promoting lifestyle was 132.78±14.99. Among all dimentions. nutrition scored highest, while health responsibility and physical activity scored lowest. The health value was positively correlated with the health-promoting lifestyle among nursing home elderly people (P<0.01). Their own health status and health value were influencing factors of health promoting-lifestyle, which could be explained at 43.5%of the variance. Conclusions The nursing home for elderly people were good in health value and average in health-promoting lifestyle. The elderly with a higher health value had better health-promoting lifestyle. Health care workers should improve elderly people′s level of health value, and give more help and guidance to the elderly people for physical activity and health responsibility, making them establish healthy lifestyle.
5.Pilot-scale purification of rF1-V fusion protein of Yersinia pestis and characterization of its immunogenicity.

Recombinant Fl-V (rFl-V) fusion protein is the main ingredient of the current candidate vaccine against Yersinia pestis infection, which has been under investigation in clinical trial in USA. We investigated the soluble expression conditions of rF1-V in Escherichia coli BL21 (DE3) that we constructed before. After scale-up and optimization of fermentation processes, we got the optimized fermentation process parameters: the culture was induced at the middle exponential phase with 50 µmol/L of IPTG at 25 °C for 5 h. Soluble rFl-V protein was isolated to 99% purity by ammonium sulfate precipitation, ion exchange chromatography, hydrophobic chromatography and gel filter chromatography. The protein recovery was above 20%. Protein identity and primary structure were verified by mass spectrometry and Edman sequencing. Results of purity, quality and western blotting analysis indicated that the target protein is a consistent and properly folded product. Furthermore, the immunogenicity of various antigens formulated with aluminum hydroxide adjuvant was evaluated in mice. Serum antibody titers of 4 groups including 20 µg rFl, rV and rFl-V and 10 µg rFl+10 µg rV, were assayed by ELISA after 2 doses. The antibody titers of anti-Fl with 20 µg rFl-V were obviously higher than titers with other groups; meanwhile there were no significant difference of anti-V antibody titers among them. These findings confirm that rFl-V would be the active pharmaceutical ingredient of the plague subunit vaccine.

6.Expression of Osteopontin and molecules in EMT of rats with chronic allograft nephropathy

Objective To investigate the expression of OPN, α-SMA, E-cadherin and their correlation in the chronic allograft nephropathy (CAN) rat model, and to explore the possible role of OPN in CAN.Methods Orthotopic renal-transplantation using Fisher rats as donors and Lewis rats as recipients was done to establish CAN group, and Lewis to Lewis rats as control group. Rats in each group were sacrificed 12 weeks after the surgery. Blood and urine were collected for further test. Allograft samples were collected and sectioned for HE, Sirus-red staining, immunohistochemistry and Western blot.Results There were CAN morphological changes of the allograft in CAN group. As compared with control group, immunohistochemistry and Western blot revealed that the expression of OPN and α-SMA in CAN group was significantly increased, and that of E-Cadherin reduced. Its trend was correlated with the inflammatory response and the EMT of tubule epithelial cells.Conclusions OPN expression in rat CAN model is significantly up-regulated. OPN may play a role in CAN. OPN might affect the CAN by promoting EMT of tubule epithelial cells.
7.Influence of fluoride on the expression TM9SF1 mRNA and Ras mRNA of human osteoblasts

Objective To detect the influence of fluoride on the expression TM9SF1 mRNA and Ras mRNA of osteoblasts. Methods The third generation of primary cultured osteoblasts were exposed to a series concentrations of 0,2.5,5.0, 10.0,20.0 mg/L fluoride for 10 days. The influence of different doses of fluorine on the expression of TM9SF1 mRNA and Ras mRNA of osteoblasts cultured in vitro was investigated by SYBR Green I methods. Results The osteoblasts of the control group and the 2.5 mg/L group were in the shape of long spindle, triangle or irregular polygon and had processes, and the cytoplasm was translucent, adjacent cells affixed to each other under light microscope. Those of the 20.0 mg/L group shaped as long spindle or irregular polygon, and some vacuolization and granular materials appeared in cytoplasm. The number of the cells decreased and the volume increased significantly. After exposed to fluoride for 10 days, osteoblasts of 2.5 mg/L group morphologically proliferated. There were statistical siguificances between each groups of TM9SF1 mRNA in human osteoblasts(F = 322.82, P < 0.01). The highest in the 2.5 mg/L group(9326.0 ± 115.97), the expression of TM9SF1 mRNA decreased along with the increasing dose of fluorine. There were statistical significances between 5.0, 10.0,20.0 mg/L groups(6495.0 ± 323.9, 4387.5 ± 545.2, 5962.5 ± 536.7) and control group(9221.0 ± 107.5, all P< 0.01). There was a statistical significance between each groups of Ras mRNA in human osteoblasts(F = 703.28, P < 0.01). The highest in the control group, the expression of Ras mRNA decreased along with the increasing of dose of fluorine. There were statistical significance between 2.5, 5.0, 10.0, 20.0 mg/L groups(6144.5 ± 270.82,5603.5 ± 88.39,3181.0 ± 159.81,4067.5 ± 37.4) and control group(6571.0 ± 196.58). Conclusion The influence on TM9SFI mRNA and Ras mRNA expression in osteoblasts correlates with the dose of fluorine.

The practical experiences with subject service in Medical Library of Chinese PLA were summarized in terms of the establishment of user information records, accumulation and summation of information, and construc-tion of information platform. Suggestions were put forward for the improvement of subject service in Medical Library of Chinese PLA, such as holding the big information concept in the big data era, using the biomedical data as the target of study, providing various kinds of service, integrating seamless, dynamic and interactive library service into user service. The fact that focus on artificial intelligence, computer, information and coomunication technologies is one of the future directions was pointed out.
9.Effect of high-flux hemodialysis on immune state in maintenance hemodialysis patients

ObjectiveTo study the changes of serum immune state in maintenance hemodialysis (MHD) patients with high-flux hemodialysis. MethodsSixty MHD patients were divided into high-flux hemodialysis group (group T, 30 cases, receiving high-flux hemodialysis, 3 times per week and routine drug therapy) and conventional dialysis group (group C, 30 cases, taking conventional dialysis, 3 times per week and routine drug therapy) by random number table. They were treated for 12 weeks. Serum IgG,IgA, IgM,C3, C4, T lymphocyte subsets and albumin, prealbumin, hemoglobin were detected at the experiment onset and 12 weeks later, and compared with those of 20 normal medical examination adults (group N ), meanwhile the ratio of infection was calculated. ResultsThe levels of IgG,IgA,IgM,C3,C4,CD3+,CD4+,CD4+/CD/8+ in group C had no significant changes before and after treatment (P > 0.05 ), but there were significant changes in group T[(12.20 ± 3.98) g/L vs.(6.18± 1.45) g/L, (1.89 ±0.58) g/L vs. (0.63 ±0.15) g/L, (1.29 ± 0.47)g/L vs. (0.51 ± 0.13) g/L, (0.94 ± 0.36) g/L vs.(0.58 ± 0.20) g/L, (0.28 ± 0.06) g/L vs. (0.11 ± 0.04) g/L,(63.11 ± 9.43 )% vs. (53.26 ± 9.08 )%, ( 38.21 ± 6.15 )% vs. ( 31.56± 6.02 )%, 1.48 ± 0.37 vs. 1.25 ± 0.43](P< 0.05), and there were significant differences between group T and group C after treatment (P< 0.05).The ratio of infection in group T was decreased obviously [46.7% (14/30) vs. 133.3% (40/30), P < 0.05], but there was no significant difference in group C [126.7%(38/30) vs. 136.7%(41/30),P> 0.05]. Conclusion High-flux hemodialysis can improve the immune state and the nuuitional state in MHD patients, and it could decrease the infection rate.
10.Effects of Celastrol on HepG2 Cells Migration

Objectives To explore a better inhibitory effect of concentration and time of Celastrol on migration of HepG2 cells. Methods HepG2 cells were planted and cultured in 6-well plates. When the adherent cell density reached 70%-80%, cell migration was manufactured by scratch experiment model. Then, cell morphology and cell migration were observed under microscope with different concentrations of Celastrol 5, 1, 0.5, 0.1, 0.01μmol/L and DMSO at 0, 6, 12, 24 h. They were pictured and rates of cell migration and inhibition ratios of all groups were calculated. Results Celastrol inhibited HepG2 cell migration, and its inhibitory effect on the migration velocity was concentration-dependent in a certain range. The higher the concentration of Celastrol, the stronger effect is. Celastrol of the same concentration at different times had different inhibitory effect on cell migrationof HepG2 cells (P<0.05). Celastrol of different concentrations at the same time had different inhibitory effects on cell migration of HepG2 cells (P<0.05);Celastrol of high concentration cause dsevere changes in the cell morphology. Conclusion Celastrol of high concentration causes changes in the cell morphology and cell apoptosis of HepG2 cells. Celastrol inhibits HepG2 cell migration, which is dependent on the concentration and action time. The inhibitory effect of Celastrol on HepG2 cell migration is most obvious under final concentration of 5μmol/L at 6 h.
