1.Problems in medicinal materials research of new traditional Chinese medicine.

Medicinal materials research and development of new drug of traditional Chinese medicine (TCM) research is the premise and foundation of new drug research and development, it throughout the whole process of new drug research. Medicinal materials research is one of the main content of the pharmaceutical research of new drug of TCM, and it is also the focus of the new medicine pharmaceutical evaluation content. This article through the analysis of the present problems existing in the development of TCM research of new drug of TCM, from medicine research concept, quality stability, quality standard, etc are expounded, including medicine research idea value medicine study should focus on the important role and from the purpose for the top-level design of new drug research problem. Medicinal materials quality stability should pay attention to the original, medicinal part, origin, processing, storage, planting (breeding), and other aspects. Aspect of quality standard of medicinal materials should pay attention to establish the quality standards of conform to the characteristics of new drug of TCM. As the instruction of TCM new drug research and development and the scientific nature of the review, and provide the basis for medicinal material standards.

2.Effect of CPD_4 photodynamic therapy on endothelial cell in vitro

Objective:To study the PDT effects on culturing endothelial cell in vitro under the differens content of CPD 4. Methods: According to the existing techniques obtaining prime umbilical vein endothelial cell in 24 well culturing plate,10 5/well.Add 1640 medium mixed with different final concentration of CPD 4 individuallly and incubate for 1h. Then shine them with 630~640 nm red light. Observe the morphological changes of EC. After PDT 6 h,measure the contents of Prostaglandin F 1?(PGF 1?) and and LDH in the culturing medium and the percentage of the residual cell. Results:With the concentration of CPD 4 increasing, the damage degree of EC aggravates. There was emergence of EC contracting ,falling off , cytoplasmic swelling,floating and dying. The contents of PGF 1? and LDH were increasing and the percentage of residual cell decreases simultaneously. There was statistical significance between the five groups. Conclusion:The effect of PDT is direct.The effectiveness has two characteristic: ①CPD 4 induced PDT has direct photosensitised damage to EC. There exists CPD 4 dose dependence.②The PDT target of EC is membrane and cytoplasma organelle. The damage is lethal and irreversible.
3.The experience for reconstructing calcaneus partial defect with deep iliac circumflex artery ilium lamella and sural nerve flap:7 cases report

Objective To explore the surgical treatment effect reconstructing calcaneus partial defect with deep iliac circumflex artery illium lamella and sural nerve slap. Methods Sevens of calcaneus fracture with posterior part defect from July 2005 to July 2010 were rehabilitated with deep iliac circumflex artery ilium lamella whose artery were anastomosed to posterior tibial artery and whose skin were repaired with sural nerve slap. Results All patients were obtained follow-up,which ranged from 9 to 30 months with a mean of 19.5 months. Its Bohler angle maintain a range from 35 to 45 degree and foot bridge maintain its figure,as well as perfect skin.According to Maryland grades system,the result were excellent in 3 cases with score 90,good in 2 with score 85 and fair in 2 with score 75. Conclusion It is a perfect method for calcaneus trauma which lose its foot bridge to rehab its figure and function with deep iliac circumflex artery ilium lamella and sural nerve slap.
4.Investigation of sources of work stress and influence factors of neurology nurses in grade Ⅲ class A hospital

Objective To investigate the stress status of neurology nurses and related factors in grade Ⅲclass A hospital.Methods 185 neurology nurses selected by the convenience sampling method were surveyed with a questionnaire for basic information and work stressor scale and analysed by SPSS 11.5.Results The major source of work stress of neurology nurses was shown to be nursing profession and work(3.00±0.56)score,the overall score of the work stress was (2.52 ±0.46).Factors shown signigicant affect on the work stress included the marital status single(2.43 ±0.42) and married(2.60±0.48)(t =6.382,P <0.05) ; professional ranking between entry rank and junior professional rank;entry rank and medium rank (t=25.258,4.185,respectively;P <0.05),health condition:between very good and bad;good and average;good and bad (t =4.058,10.734,8.215,respectively;P <0.01) and sleep quality:between no sleep disorders and suspicious of insomnia;no sleep disorders and insomnia (t =4.061,9.195,respectively;P<0.05).Related factors of work stress of neurology nurse by linear regression analysis were educational level,whether the individual has siblings or not,professional ranking,marital and family status,work schedule,sleep quality,self-evaluation of work performance and health.Conclusions Administrative staft of hospital should value the related factors affecting working stress of neurology nurses and take targeted measures to cope with these factors,relieve the stress of neurology nurses and improve the nursing quality.
5.Investigation of sources of work stress of neurology nurses in new work pattern and its influence factors

Objective To investigation work stress and its influence factors of neurology nurses in new work pattern.Methods 90 neurology nurses in a grade Ⅲ class A hospital was picked with the convenience sampling method and surveyed with a questionnaire for basic information,nurses stressor,social support and coping styles.Results The overall score work stress of neurology nurses was (2.41±0.43).Major sources of work stress were nursing profession and work (3.03 ± 0.58),work load and time allocation the social support (2.90 ± 0.64 ),working environment and resources (2.53 ± 0.86 ),patients caring ( 2.36 ± 0.54 ) and management and interpersonal relationships ( 1.67 ±0.57).Social support and coping styles scored as (43.94 ±6.22) and (54.43 ± 9.92) respectively.Both social support and coping styles showed positively correlation with the work stress of the neurology nurses( t=7.187,7.071,respectively ;P < 0.01).Better support from the society reduced the stress of the nurses.Conclusions Quality of nursing care services demonstration project is a good starting point for reforming the nursing work.Managers should seize this opportunity to fully understand the work stress status of neurology nurses and effectively provide measures to ease the pressure of nurses,improve the quality of care,and ensure the sustainability "quality care service".
6.Imaging diagnosis of osteopoiknosis: a report of 6 cases in four generations of one family and 3 sporadic cases.

OBJECTIVETo analyze the imaging features of osteopoikilosis and its diagnosis knowledge.
METHODSThe imaging data of 9 patients with osteopoikilosis were analyzed retrospectively, including 6 familial cases and 3 sporadic cases. In 6 familial cases,there were 4 males and 2 females with an average age of 28 years old ranging from 10 to 63 years. Clinical manifestations of 1 familial case were left knee pain and limitation of activity for 3 years, and other 5 cases without clinical manifestation. In 3 sporadic cases, there were 2 males and 1 female with an average age of 33.7 years old ranging from 25 to 44 years. Three sporadic cases had obvious injury history with following up from 6 to 12 months. All imaging results of 9 cases were observed.
RESULTSThe imaging data of 6 familial osteopoikilosis showed the multiple round or oval nodes within bone with clear margins, uniform density, different size. The occurrence of the hyperostotic spots preferentially localized in the epiphyses and metaphyses of the long bones, and carpus and tarus. X-ray features of 3 sporadic osteopoikilosis were similar to that of 6 familial cases and for 6 to 12 months follow-up X-ray features were unchanged.
CONCLUSIONThe imaging features of osteopoikilosis are relatively specific such as the multiple mottling dense focal within bone with clear border and bilateral symmetry, and the focus located on cancellous bone and the diaphyses usually is unaffected. The imaging is a valuable examination for the accurate diagnosis of osteopoikilosis.

7.Biocompatibility of Genipin cross-linked type I collagen with human adipose-derived stem cells in vitro

BACKGROUND:Low toxicity of Genipin has certain species and cellspecificity. Biocompatibility of Genipin cross-linked type I colagen with human adipose-derived stem cels is essential for construction of
tissue-engineered adipose.
OBJECTIVE:To investigate the bbiocompatibility of Genipin cross-linked type I colagen with human adipose-derived stem cels.
METHODS:Human adipose-derived stem cels were isolated and cultured to the third generation, and the cels were seeded on Genipin cross-linked type I colagen scaffold. MTT assay was used to evaluate the adhesion and proliferation of cels on the scaffold, and the toxic effects of Genipin cross-linked type I colagen on human
adipose-derived stem cels. Optical microscopy and scanning electron microscopy were utilized to observe the adhesion and growth process of human adipose-derived stem cels on the scaffold as wel as the morphological changes of cels.
RESULTS AND CONCLUSION:Human adipose-derived stem cels could adhere to the scaffold immediately
after seeded and increase gradualy on the scaffold, with the average adhesion rate of 86.5%. Optical microscopy and scanning electron microscopy showed that human adipose-derived stem cels adhered wel on the scaffold. The cels increased gradualy over time, and could migrate into the scaffold, and distribute evenly with the passage of time when observed with optical microscopy. The result showed Genipin possesses very low cytotoxicity to the cels, and the outstanding biocompatibility is found between the cels and scaffoldin vitro after cross-linked with Genipin.
8.Inhibitory effect of chlorogenic acid on senescence of human skin fibroblasts

[Abstract ] Objective To evaluate the inhibitory effect of chlorogenic acid on senescence of human skin fibroblasts (HSFs). Methods Fibroblasts isolated from human foreskin were treated with 1 mmol/L glyoxal in vitro to develop a model for cellular senescence. In order to select effective concentrations of chlorogenic acid, some HSFs were treated with 1 mmol/L glyoxal alone or in combination with chlorogenic acid at different concentrations (5, 10, 20, 40, 80 μmol/L)for 3 days, with those receiving no treatment serving as the blank control group. Then, methyl thiazolyl tetrazolium (MTT)assay was performed to evaluate the proliferative activity of HSFs. Some HSFs were divided into 5 groups to be cultured alone(blank control group), or treated with 1 mmol/L glyoxal(glyoxal group)or the combination of 1 mmol/L glyoxal and chlorogenic acid at effective concentrations of 10, 20 and 40 μmol/L (glyoxal + chlorogenic acid groups). Senescence associated β-galactosidase (SA-β-gal)staining and real-time fluorescence-based quantitative PCR were conducted to determine the percentage of senescent cells and expression level of p16INK4a mRNA respectively. Statistical analysis was carried out by one-way analysis of variance followed by the least significant difference(LSD)-t test. Results Compared with the blank control group, the glyoxal group showed significantly decreased cellular proliferative activity of HSFs (55.65% ± 2.00% vs. 100% ± 6.90%, P < 0.01), while chlorogenic acid increased the proliferative activity of HSFs in a dose-dependent manner, and the increase reached a peak at 40 μmol/L. Concretely speaking, the glyoxal + 10-, 20-, 40-, 80-μmol/L chlorogenic acid groups all significantly differed from the glyoxal group in cellular proliferative activity (60.75% ± 1.32%, 67.65% ± 1.90%, 75.71% ± 3.25% and 75.69% ± 2.38% vs. 55.65%± 2.00%, all P < 0.05), but no significant difference was observed between the glyoxal group and glyoxal + 5-μmol/L chlorogenic acid group or between the glyoxal + 40-μmol/L chlorogenic acid group and glyoxal + 80-μmol/L chlorogenic acid group (both P > 0.05). Therefore, 10 - 40 μmol/L was selected as the effective concentrations of chlorogenic acid. The glyoxal group showed significant increases in the percentage of senescent (SA-β-gal-positive)cells (35.65% ± 2.24% vs. 13.00% ± 2.22%, P < 0.01)and expression level of p16INK4a mRNA (2-ΔΔCt: 1.00 ± 0.06 vs. 0.26 ± 0.05, P <0.01)compared with the blank control group, while the glyoxal + 10-, 20-, 40-μmol/L chlorogenic acid groups showed significantly decreased percentage of senescent cells (31.50% ± 2.13% , 22.31% ± 3.11% and 19.32% ± 3.01%respectively)and expression level of p16INK4a mRNA (2-ΔΔCt: 0.88 ± 0.08, 0.73 ± 0.06 and 0.68 ± 0.04 respectively) compared with the glyoxal group (all P < 0.05). Additionally, the percentage of senescent cells decreased with the increase in chlorogenic acid concentrations in the glyoxal + chlorogenic acid groups. Conclusion Chlorogenic acid can protect HSFs from glyoxal-induced senescence.
9.Construction of tissue engineered adipose using human adipose stem cells with chitosan-modified silk fibroin

BACKGROUND:Based on the original advantages of silk fibroin, positive charged water-soluble chitosan modified silk fibroin is modified on surface and could improve celladhesion on the scaffolds. OBJECTIVE:To verify the biocompatibility of chitosan-modified silk fibroin with human adipose-derived stem cells (hADSCs), and feasibility of constructing tissue engineered adipose in vitro. METHODS:The hADSCs at passage 3 were seeded on chitosan-modified silk fibroin at the concentration of 1×107/L, as the experiment group;at the same cellconcentration, hADSCs were seeded in 96-wel plates as the control group. MTT tests were performed to evaluate the adhesion, growth and proliferation of hADSCs on chitosan-modified silk fibroin. Then hADSCs were implanted on the chitosan-modified silk fibroin scaffolds at the concentration of 1×109/L. The hADSCs seeded onto chitosan-modified silk fibroin complexes were respectively cultured with adipogenic differentiation medium and ordinary high-glucose DMEM. The complexes were stained with oil red O, and detected with RT-PCR after cultured 14 days. RESULTS AND CONCLUSION:The hADSCs adhered to and proliferated on the scaffolds. After cultured with adipogenic differentiation medium for 14 days, oil red O staining demonstrated that there were amount of mature adipocytes on the scaffold. The peroxisome proliferator activated receptorγ2 was positively expressed. The chitosan-modified silk fibroin possessed excellent biocompatibility in vitro. The co-cultured hADSCs could be induced to mature adipocytes successful y.
