OBJECTIVE:To provide reference for rational application of typeⅡpsychotropic drugs. METHODS:The applica-tion of type Ⅱ psychotropic drugs in our hospital during 2012-2014 was analyzed statistically in respects of consumption sum,con-sumption amount,DDDs,DDC,etc. The trend of the application of type Ⅱ psychotropic drugs and the reasonability of medication structure were evaluated in our hospital. RESULTS:9 type Ⅱ psychotropic drugs were used in our hospital during 2012-2014,in-cluding 5 oral preparations and 4 injections. The consumption sum of drugs during 2012-2014 were 22 551.33 yuan,31 489.95 yu-an and 57 742.87 yuan,increasing by 39.64% in 2013 and 83.37% in 2014. The number of type Ⅱ psychotropic drugs increased year by year,and the amount of estazolam was the largest and took up the first place in 3 years. DDDs of estazolam also occupied the first place in the list of DDDs. DDC of Phenobarbital tablet was the smallest. CONCLUSIONS:Type Ⅱ psychotropic drugs mainly are oral drugs and reasonable in structure in our hospital during 2012-2014. The consumption sum and amount of type Ⅱpsychotropic drugs increase reasonably year by year.

OBJECTIVE: To investigate the effects of dexmedetomidine(Dex) preconditioning on neuroapoptosis in hippocampus induced by isoflurane and the expression of CRMP2 proteins in neonatal rats. METHODS: Sixty neonatal rats at postnatal day 7 were assigned randomly into control group, Dex preconditioning control group, isoflurane group and isoflurane with Dex preconditioning groups which at dose of 25, 50 and 75 μg·kg-1. Rats in groups of isoflurane or control were exposed 0.75% isoflurane or air for 6 h. Rats in groups of Dex preconditioning were injected intraperitoneally with different doses of Dex 20 min before exposure and rats in other groups were injected intraperitoneally with 150 μL saline. At the end of exposure, the hippocampus of some rats were separated and proteins expression of cleaved caspase-3, phospho-CRMP2 (p-CRMP2), CRMP2 were detected by Western blot (n=6); other rats were perfused and their brains were embedded by paraffin, the neuronal apoptosis in hippocampus CA1 was detected by TUNEL staining 2 h after exposure (n=4). RESULTS: The number of TUNEL positive cells in hippocampus CA1 of rats in isoflurane group was increased by 434.99% (P<0.001) when compared with control group, Dex at dose of 75 μg·kg-1 significantly inhibited the increase of isoflurane-induced TUNEL positive cells by 71.87% (P<0.001). The expression of cleaved caspase-3 protein in isoflurane group increased by 90.20% (P<0.001) compared with control group, while Dex at dose of 25, 50 and 75 μg·kg-1 reversed isoflurane-induced increase of cleaved caspase-3 by 30.54% and 53.71%(P<0.05)and 96.71% (P<0.001)separately. Isoflurane increase the ratio of p-CRMP2/CRMP2 in the hippocampus by 119.41% (P<0.001), and didn't influence the expression of CRMP2; while Dex at dose of 50 and 75 μg·kg-1 inhibited isoflurane-induced increase of p-CRMP2/CRMP2 ratio by 63.75% (P<0.01) and 77.70% (P<0.01) and increased the expression of CRMP2 by 44.30% (P<0.05) and 62.13% (P<0.01). CONCLUSION: Dex Attenuates isoflurane-induced neurodegeneration through inhibitory effect of hippocampal neuroapoptosis in neonatal rats, decrease the ratio of p-CRMP2/CRMP2 and increase the expression of CRMP2 may be one of the mechanisms for neuroprotection and this Conclusion may provide the theoretic foundation for reasonable selection of anaesthetics.

[Objective]To investigate the effect of a single subcutaneous dose of an antisense oligonucleotide(ASO) on particle-induced osteolysis.[Method]The murine calvaria osteolysis model was utilized in ICR mice.Bone resorption was measured with the toluidine blue staining.Osteoclasts were detected by tartrate resistant acid phosphatase(TRAP) staining assay and were quantified by a TRAP quantification kit.[Result]Bone resorption was 0.347 ? 0.09 mm2 in animals with particle implantation,and decreased to 0.123 ? 0.05 mm2 and 0.052 ? 0.02 mm2 after ASO treatment in low and high doses,respectively.The bone resorption was reestablished in animals given an additional TNF-?.The number of osteoclasts in animal calvaria treated with ASO was reduced obviously compared with those untreated animals and the quantification results indicated that about 90% osteoclastgenesis was suprressed by the ASO.Additionally,the osteoclastgenesis was reestablished by the addition of TNF-?.[Conclusion]An antisense oligonucleotide targeting an inflammatory factor,TNF-?,has been to suppress the osteolysis induced by particle for the first time.This new finding holds a great promise.It is a therapeutic strategy for the component loosening.

Objective To investigate the causes of fever in systemic lupus erythematosas (SLE)patients and identify the characteristics of the fever caused by SLE fever infection after steroid therapy.Methods From January 2002 to May 2007, 487 SLE hospitalized patients with fever were admitted and their data were retrospectively studied. The clinical manifestations and medications were followed up. A case control design was carried out to identify the differences between fever caused by lupus and infection. Results Among all the 487 patients, 265 had infection, 206 had fever that was caused by SLE per se, fever caused by both lupus and concurrent infection was confirmed in 8 patients, malignancy and miscellaneous were the causes of fever in 4 respectively. The most common sites of infection were respiratory tract (61.3%), skin and mucosa (10.9%) and central nervous system (9.1%). The main microorganisms isolated were bacteria (77.4%), fungi (13.5%) and virus (7.8%). Prednisene ≤ 100 mg/d could suppress fever caused by lupus in 81.0% patients, usually within 1 to 5 days. In the case control study, lower C3, C4 and CH50, anemia, lymphocytosis, higher immunoglobulin levels, elevated anti-dsDNA antibodies, elevated anti-nuclear antibodies and higher SLEDA1 score were associated with SLE fever. Long SLE duration, old age, elevated CRP levels,leukocytosis, neutrophilia, high accumulated doses of prednisone and ever use of immunosuppressive medications were factors associated with infection. Conclusion The fever of in-patient SLE patients is mainly caused by infections and disease flare. Moderate dose of steroid therapy is effective to control the fever caused by lupus flare. The fever caused by SLE has different clinical features and laboratory results when compared to that caused by infection although both responses to steroid therapy.

Objective To study the mechanism of FTY720 on type Ι diabetic rats. Methods The typeⅠdiabetes rat model was established by feeding male SD rats with high energy urea and injecting into the abdominal cavity with low dose cephalosporins (STZ, 30 mg/kg). In the following, the treated rats were divided into two groups: model group and FTY720 group. Another group of untreated rats was assigned as normal control group. FTY720 group was given 1 mg/kg FTY720, the normal control group and model group was given the equal amount of water. Results The cardiac function of FTY720 group was recovered markeyly. FTY720 activated the expression of vascular endothelial cells S1P1 , diabetes and reduced the expessions of S1P3 and PKCβ Ⅱand it restored the migration ability of diabetes cardiovascular endothelial cell , as well as the abnormally elevated cardiovascular endothelial cells induced by high sugar permeability. Conclusion The S1P1 and S1P3 cut is an important reaction pathway to the complications of diabetes and cardiovascular dysfunction. FTY720 may reduce the damage to core blood vessels caused by diabetes , and pathological angiogenesis which functionally depends on the PKCβⅡ signaling pathways. Therefore, FTY720 may provide a potential therapy for cardiovascular function impaired by diabetes.

OBJECTIVE: To study the induction and growth of Magnolia officinalis Rehd.et Wils.var.biloba Rehd.et Wils. callus and to compare the content of active components in callus from different sources.METHODS: The factors influencing the induction and growth of Magnolia officinalis Rehd.et Wils.var.biloba Rehd.et Wils. callus were studied by single factor analysis and orthogonal design. The contents of active components in callus from different sources were determined by HPLC. RESULTS: In the various explants, the apical buds and stem fragments exhibited the best ability of dedifferentiation to form callus, followed by pistils. The optimum culture medium of the callus induction was B5+NAA0.1 mg?L-1+6-BA1.0 mg?L-1, while the culture medium B5+NAA0.5 mg?L-1+6-BA4.0 mg?L-1 markedly improved the growth of the callus; after secondary culture of the callus induced by the WPM basic medium, the total content of Magnolol plus Honokiol ranged from 0.167 9% to 0.234 4%, but the induction rate of the callus was low at 33.08%～48.65%. Under the culture condition of MS+NAA0.5 mg?L-1+6-BA1.0 mg?L-1,the rate of callus induction was 62.12% and the total content of Magnolol plus Honokiol after secondary culture was 0.168 6%. CONCLUSION: The results serve as reference in the tissue culture of Magnolia officinalis Rehd.et Wils.var.biloba Rehd.et Wils and the screening of the high production cell line.

Objective:To analyze the effect and safety of blood glucose fluctuation in brittle diabetes treated with intensive insulin therapy combined with sodium-glucose transporter 2 inhibitor.Methods:Ninety-eight patients with brittle diabetes in Shenzhen Longhua District People's Hospital were selected from May 2017 to May 2019. They were divided into two groups by random number table, 49 cases in each group. The control group was treated with intensive insulin therapy combined with saglitine, while the observation group was treated with intensive insulin therapy combined with sodium-glucose transporter 2 inhibitor. Following up for 12 weeks, the plasma glucose and blood glycemic variations, insulin dosage, plasma 8-iso-prostaglandin F2α, hypersensitive C-reactive protein and safety related indicators were compared between the two groups before and after treatment.Results:Compared with the control group, the standard deviation of blood glucose [(1.8±0.5)mmol/L vs (6.5±1.4)mmol/L, t=7.235], large amplitude glycemic excursions [(6.5±1.1)mmol/L vs (17.3±4.7)mmol/L, t=13.446], postprandial glucose excursion [(1.2±0.4)mmol/L vs (9.2±2.0)mmol/L, t=8.921], inter-quartile range [(3.7±1.1)mmol/L vs (12.4±4.2)mmol/L, t=7.003], means of daily difference [(1.5±0.4)mmol/L vs (4.6±0.8)mmol/L, t=4.537] in the observation group were significantly decreased ( P<0.05), and the levels of plasma 8-iso-prostaglandin F2α [(7.8±1.2)ng/L vs (13.6±2.3)ng/L, t=4.882], hypersensitive C-reactive protein [(5.2±1.3)mg/L vs (8.7±1.3)mg/L, t=4.406], insulin dosage [(30.9±10.2)U/d vs (42.3±13.4)U/d, t=5.726] and body mass index [(18.3±1.2)kg/m 2 vs (21.0±2.3)kg/m 2, t=4.135] also decreased significantly ( P<0.05), and the incidence of hypoglycemic events [16.3%(8/49) vs 36.7%(18/49), χ 2=9.697] and severe hypoglycemia [0 vs 14.3%(7/49), χ 2=7.268] decreased significantly ( P<0.05). Conclusions:Combination of dapagliflozin and intensive insulin therapy can significantly improve glucose metabolism in brittle diabetes, and reduce insulin dosage and hypoglycemic events.

To investigate the alkaloid constituents from the trunk barks of Winchia calophylla. The constituents were isolated and purified by column chromatography over silica gel and Sephadex LH-20 columns. The structure of the isolated compound was elucidated by MS and NMR spectra, and then confirmed by a single-crystal X-ray crystallographic analysis. The isolated compound was also evaluated for its in vitro cytotoxicity against the proliferation of seven kinds of human cancer cell lines (lung cancer: A549, breast cancer: MCF-7, prostate cancer: PC-3, glioma: U87MG, multiple myeloma: U266, MM1. S, and MM1. R). A new monoterpenoid indole alkaloid, identified as calophyline B (1), was isolated from the ethanol extract of the trunk barks of W. calophylla. Compound 1 is a new compound, which shows no cytotoxicity against the proliferation of the above human tumor cells.

Objective To constitute a database of chronic wounds for characterizing the patients with chronic wounds.Methods Based on the guideline of chronic wounds defined by international wound healing society,86 outpatients with chronic wounds were selected between January and October in 2013 to set up a wound database using the wound information management system.The parameters including etiology,age distribution,mean time of fracture healing and wound healing time posterior to different treatments were analyzed.Results (1) Statistical difference existed for cause distribution (P ＜0.05).Top three causes were trauma (29％),varicosity (26％) and pressure sore (19％).(2) Age distribution varied significantly among the patients with various causes of chronic wounds (P ＜ 0.05).(3) Statistical difference was observed in healing time of various types of chronic wounds (P ＜ 0.05).Diabetic wound and varicose ulcer took the longest healing time,with average 116.36 days and 88.48 days respectively.(4) Patients treated with novel dressings had shorter healing time than those managed with traditional dressings (P ＜ 0.05).Conclusions Trauma,varicosity and pressure ulcer are relatively common among the outpatients.Etiology of chronic wounds is strongly related to age and novel dressings contributed to a shortened cause of the disorder.
