1.Cultivation and identification of human colon cancer stem-like cells in the serum-free culture medium in vitro

Objective To generate suspended spheres ,Human colon cancer cell line SW620 cells were acclimated gradually in the free serum culture medium ,1 week later ,a great quantity of spheres were obtained for the further research .Methods SW620 cells and spheres were incubated with the following Abs :anti‐SSEA‐4 Ab and anti‐TRA‐1‐60 Ab and detected with confocal microscopy . SW620 cells and spheres were collected and stained by PI ,Cell cycle distribution was then acquired by flow cytometry .SW620 cells and spheres were transferred into Adipocyte Differentiation Medium and cultured for 7 d and then lipid droplets were counted . SW620 cells and sphere cells were injected subcutaneously into the lateral root of one posterior limb of a nude mouse ,tumor volume was calculated .Results Spheres formed when grown under serum‐free conditions in 7 days ,and the ability of SW620 cells to form spheres was sustained .A few of SW620 adherent cells be cultured in DF+10% FBS clearly showed the expression of pluripotent stem cell markers Ssea‐4(25 .739 ± 7 .62)% ,Tra‐1‐60(27 .742 ± 4 .311)% ,but almost all spheres expressed the 2 markers and there was a significant difference between the two groups in the expression ratio (P<0 .05) .It had been shown that the spheres exhibited a relatively high proportion of cells in G0/G1 phase(84 .19 ± 2 .52)% and low proportion in S phase(7 .18 ± 1 .35)% ;when com‐pared to SW620 cells ,G0/G1 phase(63 .02 ± 6 .73)% ,S phase(20 .89 ± 3 .84)% (P<0 .05) .Adipocytes drops from spheres and ad‐herent cells were calculated under microscope after been cultured in adipocytes liquid for 7 d .The number of adipocytes drops in spheres was (583 .80 ± 77 .69) ,but(169 .20 ± 26 .43)in SW620 adherent(P<0 .05) .2 × 104 sphere cells injected subcutaneously into the lateral root of one posterior limb of a nude mouse fromed transplantation tumor after 4 weeks ,and the tumor size was (2 279 .98 ± 346 .27) mm3 ;2 × 105 adherent cells injected subcutaneously into the lateral root of one posterior limb of a nude mouse fromed transplantation tumor after 4 weeks ,and the tumor size was(889 .75 ± 78 .79) mm3 ,(P<0 .05) .Conclusion In this study , it is successfully observed that SW 620 cells being cultured in serum‐free medium resulted in spheres rapidly and the spheres are se‐rially passaged with relatively high efficiency .The spheres show the characteristics of the cancer stem cell .
3.Analysis of physiological indexes of 128 healthy children receiving cardiopulmonary exercise testing

Objective To study the physiological indexes of children receiving cardiopulmonary exercise testing (CPET) in order to provide some data for establishing the normal CPET reference value in Chinese children.Methods One hundred and twenty-eight healthy children underwent the CPET with the standard scheme.All children were divided into 3 groups according to ages:3-6 years old, >6-10 years old and > 10-14 years old.The physiological indexes of CPET were collected, including maximal oxygen uptake (VO2max/kg) , oxygen uptake at anaerobic threshold (VO2/kg@AT), maximal tolerance power (Pmax), maximal heart rate (HRmax), peak respiratory minute volume to CO2 (peak VE/VCO2), slope of respiratory minute volume to CO2 (VE/VCO2 slope), maximal tidal volume (VTmax) and the maximal minute ventilation (VEmax).The range in normal values for physiological CPET variables was recorded.Results The VO2max/kg was (31.71 ± 7.31) mL/(min · kg), (31.05 ± 5.96) mL/(min · kg), and (34.10 ±8.72) mL/(min · kg) in the 3-6 years old group, >6-10 years old group and > 10-14 years old group of boys.The VO2/kg@AT was (28.32 ± 10.20) mL/ (min · kg), (23.13 ± 9.56) mL/(min · kg), and (22.18 ± 7.72) mL/(min · kg)in the 3-6 years old group, >6-10 years old group and > 10-14 years old group of boys.The VO2 max/kg was (29.27 ± 4.19) mL/(min · kg), (30.67 ± 5.59) mL/ (min · kg) (29.18 ± 6.56) mL/ (min · kg) in the 3-6 years old group, > 6-10 years old group and > 10-14 years old group of girls.The VO2@AT was (26.64 ±9.49) mL/(min · kg) ,(20.53 ±7.66) mL/(min · kg) ,(17.83 ±6.49) mL/(min · kg)in the 3-6 years old group, >6-10 years old group and > 10-14 years old group of girls.VO2max/kg and VO2/kg@AT did not change with age,there was no gender difference in VO2max/kg(P >0.05).VO2/kg@AT of boys was higher than girls in the 3-6 years old group(t =2.351 ,P =0.023).The Pmax,VEmax and VTmax increased with the growth of age(boy: F =49.436,24.290,28.958, all P < 0.001;girl : F =58.287,42.990,28.802, all P < 0.001), but not significant in different gender group(P > 0.05).In boys peak VE/VCO2, VE/VCO2 slope and HRmax were significantly different (F =4.758,P =0.012;F =8.050, P =0.001;F =3.869, P =0.026), however these indexes were not significantly different in girls(all P > 0.05).Conclusions This study provides a set of data for the most important CPET variables in Chinese children.VO2max/kg does not change with age,so it can be used as a valuable parameter in exercise ability evaluation.
4.Protection Mechanism of Neferine in Learning and Memory Function of Rats with Chronic Cerebral Ische-mia

Objective: To investigate the effects of neferine ( Nef ) on the learning and memory function and the expression of Notch1 and SYN in hippocampus in the rats with chronic cerebral ischemia .Methods:Male SD rats (250-300g) were randomly divid-ed into the sham operation group(Sham), the model group(Mod), nimodipine (Nim) positive control group and Nef treatment group with 5 ones in each .The chronic cerebral ischemia ( CCI ) model was established using bilateral common carotid artery ligation ( 2-VO).The rats were orally administered with NS(5 mg· kg-1),Nim(1 mg· kg -1) and Nef (20 mg· kg-1),respectively for 21 days after the first day of operation .The body weight was recorded .The number of hippocampal neurons in the rats was observed by Nissel staining.The learning and memory function was evaluated by Morris water maze test .Notch1 and SYN protein expressions in hippo-campus were detected by IHC staining and Western blot .Results:Compared with the Mod group , Nef group could reverse the reduc-tion of body weight and the number of hippocampal neurons in hippocampus CA 1 region induced by 2-VO, increase the route and the time of platform finding, prolong the escape latency and decrease the number of platform cross on the 21st day, and the effects were bet-ter than those of Nim (P<0.05).Moreover, the down-regulated SYN and Notch1 protein expressions in CCI group were both increase after the Nef treatment on the 21st day.Conclusion:Nef has a protective effect on the function of learning and memory in CCI rats in-duced by 2-VO,which may be related with the strengthened Notch 1 pathway in CA1 region of hippocampus and synaptic plasticity .
5.Progress and obstacles of induced pluripotent stem cell on hematologic disease

Induced pluripotent stem (iPS) cell based on recently developed stem cell reprogramming technique holds great hope for regenerative medicine,in vitro disease modeling and drug evaluation.Recent progress on clinical hematology includes in vitro generation of hematopoietic progenitors and mature blood cells from somatic cells,iPS cells derived from chronic myeloid leukemia cells for the better understanding of the resistance mechanisms of bcr-abl inhibitor imatinib,and moreover,correction the monogenic inherited disease using gene-targeted strategies.However,whether the iPS cells can fully replace human embryonic stem cells still needs further investigation.
6.Clinical observation on treatment of sympathetic cervical spondylopathy by Jiawei Gegen decation

Objective To observe the clinical effect of Jiawei Gegen Decoction on cervical spondylosis of sympathetic.Methods 54 patients with cervical spondylosis from June 2013 to June 2016 were divided into the control group and the study group by odd and even numbers,each 27 cases,the contrel group was treated with Flunarizine Hydrochloride, the study group was treated with Jiawei Gegen decoction,The treatment effect, symptom improvement and adverse reaction rate were evaluated between the two groups.Results Compared with the control group, the treatment effect of the patients in the study group and the improvement of symptoms after treatment were better than the control group,the incidence of adverse reactions was lower in the study group (P <0.05).Conclusion With Jiawei Gegen Decoction for the treatment of sympathetic cervical spondylosis patients, the treatment effect is prominent, at the same time, the symptoms of patients improved better, less adverse reactions, it is worthy of clinical trust.
7.Prominent Lumbar Intervertebral Disc Treated with Shinbone Tube Seal and Dimensional Traction

[Objective] To discuss effective way for Prominent Lumbar Intervertebral Disc.[Method] Apply shinbone tube seal and dimensional traction on 118 cases of the said patients.[Result] After following up for 3~18 months,62 cases were very good,occupying 52%;43 good,36%;10 bad,8%;no effect 3,2%,the effective rate 88%.[Conclusion] It is safe and one of good therapies presently to treat Prominent Lumbar Intervertebral Disc with shinbone tube seal and dimensional traction.
8.Update of percutaneous microwave ablation for renal cell carcinoma

Microwave ablation is widely applied in clinical practice for its safety,minimal invasiveness and definite effectiveness.This technique,however,is still in the initial stage in the treatment of renal tumors.This article introduces the principles and development of the technique,as well as its experimental researches,clinical application and effect evaluation in the treatment of renal cell carcinoma.
9. Chemical constituents from twigs and leaves of Ervatamia hainanensis

Objective: To investigate the chemical constituents in the twigs and leaves of Ervatamia hainanensis. Methods: The chemical constituents were isolated by silica gel, Sephadex LH-20, and ODS columns, as well as RP-HPLC chromatography. Their structures were identified on the basis of physicochemical properties and spectral analyses. Results: Thirteen compounds were isolated and idenified as coronaridine (1), 19-heyneanine (2), 19-epi-heyneanine (3), coronaridine hydroxyindolenine (4), vobasine (5), ursolic acid (6), asiatic acid (7), 8α-hydroxypinoresinol (8), (+)-syringarosinol (9), ervatamisin [(-)-(7R, 7'R, 7″R, 8S, 8'S, 8″S)-4', 4″-dihydroxy-3, 3', 3″, 5-tetramethoxy-7, 9', 7', 9-diepoxy-4, 8″-oxy-8, 8'-sesquineolignan-7″, 9″-diol, 10], toddalolactone (11), threo-2, 3-bis-(4-hydroxy-3-methoxypheyl)-3-methoxy-propanol (12), and β-sitosterol (13). Conclusion: Compounds 6-8 and 10-12 are isolated from the plants of Ervatamia (A. DC.) Stapf for the first time, and compound 9 is firstly reported from E. hainanensis.
