1.Research on the effectiveness of WPBL teaching method in medical humanities English course under the background of "New Medical Science"

Objective:To explore whether the introduction of web problem based learning (WPBL) teaching method in medical humanities English class under the background of new medical science is helpful to the course learning and the improvement of students' humanistic quality.Methods:Taking 602 students from Batch 2017 clinical medicine in Xi'an Medical University as the experimental group and 610 students from Batch 2018 as the control group, the research lasting a semester was carried out around eight modules. The control group adopted the traditional teaching mode, while the experimental group used the WPBL mode for teaching relying on the Blue Pigeon Cloud Network Language Learning Platform. After teaching, through comparing the final exam results and issuing self-made questionnaires, the teaching effects of the two methods on students' self-study ability, thinking ability, teamwork ability and knowledge application ability were compared. The data were statistically processed by SPSS 20.0.Results:The final average score of the experimental group (62.13±2.11) was higher than that of the control group (60.42±2.03), with significant differences ( P < 0.05). According to the statistical analysis of the questionnaire, there were differences between the two groups in evaluating the teaching effect, and the proportion of the experimental group was over 60%, significantly higher than that of the control group. 93.3% of the students in the experimental group were satisfied with the WPBL teaching method, while only 43.0% of the students in the control group were satisfied with the traditional teaching method, showing a significant difference between them. The evaluation of students in the experimental group were higher than the control group in terms of self-study ability, data access ability, knowledge exploration ability, language expression ability and problem finding and solving ability. 91.1% of students in the experimental group believed that the introduction of WPBL teaching mode in the course would help to improve their medical humanistic quality, which was significantly higher than the proportion of students in the control group. Conclusion:The empirical study shows that the introduction of WPBL teaching method in medical humanities English course can not only enable students to master learning skills, but also strengthen students' humanistic quality, thus promoting the development of medical education.
3.Regulation of Pituitary Tumor Cell Prolieration by Epidermal Growth Factor and Genistein

Recombinant human epidermal growth factor (rhEGF, 20 ng/ml) and Genistein (740 μmol/ml) were added to the cells from 12 cases of human pituitary tumors and co-cultured in vitro. The 3H-thymidine uptake of tumor cells was measured by scintillation counter and tyrosine kinases activity was detected by using immunocytochemical method. The results showed that 3H-thymidine uptake and tyrosine kinases activity were increased in 10 out of 12 cases of human pituitary tumor by EGF and both declined by Genistein. It was suggested that EGF had a strong mitogenic effect on majority of pituitary tumors. Genistein could abolish the effects of EGF promoting the proliferation of the tumor cells, inhibit the growth of the tumor and might be useful in pituitary tumor treatment.
4.Feasibility of acute hypervolemic hemodllution with hyuroxyethyl starch in the elderly patients during major surgery

There was no significant difference in the volume of solution infused, blood loss, CO, SV, CVP and SVR between the two groups. Conclusion Age is not a primary factor negating the use of AHH. Under combined general-epidural anesthesia AHH with 6% hydroxyethyl starch (15 ml?kg-1 ) can be well tolerated by the elderly patients without cardiac and pulmonary disease.
5.Expression of Epidermal Growth Factor, Transforming Growth Factor-αand Epidermal Growth Factor Receptor in Human Pituitary Tumors

Objective:To explore the mechanism of growth factor autocrine stimulation in human pituitary tumors. Method: Immunohistochemical analyses of EGF、TGF-αand EGFR were studied on paraffin-embeded sections of 30 pituitary tumors. Results: Expression of EGFR and its ligand EGF、TGF-αhas a variable stained cells intensity,density and type. EGFR and the ligands expressed in majority of pituitary tumors.Conclusion:The EGF autocrine stimulation exists in pituitary tumors,and tyrosine kinases inhibitors may be useful for pituitary tumors treatment.
6.Shear stress-regulated vascular endothelial permeability is mediated by Rho GTPases

Dysfunctions of vascular endothelial permeability are related to a number of human diseases such as atherosclerosis, high blood pressure, stroke, inflammation, cancer, diabetes-induced retinopathy, macular edema and so on. Shear stress is an important mechanical force that affects vascular endothelial cells. It plays a particular role in permeability regulation. Rho GTPases is a family of small G proteins which act as cell signal molecules. They are assumed to mediate the regulation of the permeability of vascular endothelial cells. In this paper review is given on how shear stress regulates the permeability of vascular endothehal cells as well as the in-fluence of Rho GTPases on the role in molecular mechanism. It is suggested that shear stress-regulated vascular endothelial permeability is mediated by Rho GTPases.
7.Protective effect of isoflurane against lung ischemia-reperfusion injury in rats

Objective To investigate the effect of isoflurane administered before ischemia on polymorphonuclear neutrophil (PMN) infiltration and expression of adhesion molecules in the lung injured by ischemia-reperfusion.Methods One-hundred and twenty male SD rats weighing 250-350 g were randomly divided into 4 groups ( n = 30 each) :Ⅰ sham operation group (S) ;Ⅱ I/R group in which hilum of left lung was clamped for 45 min and then undamped for reperfusion; Ⅲ Iso + I/R in which 1 MAC isoflurane was inhaled for 30 min before ischemia and Ⅳ Iso + S in which 1 MAC isoflurane was inhaled for 30 min without I/R. The animals were anesthetized with intraperitoneal pentobarbital 50 mg?kg-1 then tracheostomized and mechanically ventilated with 100% O2(VT= 10-15 ml?kg-1) . PaCO2 was maintained at 35-45 mm Hg. Right jugular vein and left carotid artery were cannulated for BP monitoring, blood sampling and fluid and drug administration. Anesthesia was maintained with ketamine 10 mg?kg-1?h-1 and vecuronium 0.1 mg?kg-1?h-1. 1 MAC isoflurane (1.38% in rats) was inhaled for 30 min before hilum of left lung was clamped with an atraumatic clamp. Left lung ischemia was maintained for 45 min then the left lung was released for reperfusion. MAP was monitored and blood gases were analyzed during experiment. The animals were killed at the end of 45 minute ischemia and at 30, 60 and 120 min reperfusion and left lung was removed for: (1) determination of W/D lung weight ratio, myeloperoxidase (MPO) activity and expression of ICAM-1 mRNA; (2) light and electron microscopic examination; (3) broncho-alveolar lavage (BAL). BAL fluid (BALF) was collected and the number of cells, percentage of PMN and total protein concentration in BALF and the expression of CD18 on PMN surface were determined. Results The W/D lung weight ratio, MPO activity and expression of ICAM-1 mRNA in the lung tissue, the percentage of PMN and TP concentration in BALF and the expression of CD18 on PMN surface were all significantly increased during reperfusion in I/R group but isoflurane pretreatment significantly attenuated the I/R induced increases. Histological examination showed that the I/R induced lung injury was also ameliorated by isoflurane pretreatment. Conclusion Inhalation of isoflurane before ischemia could protect the lungs against I/R injury by inhibiting the PMN infiltration and expression of ICAM-1 mRNA and CD-18.
8.The cost-effect analysis of tele-wound treatment platform on chronic lower limb ulcers in remote district

Objective To investigate the cost-effect of tele-wound treatment platform with cellular phone terminator and traditional outpatient treatment on chronic lower limb ulcers in remote district.Methods Thirty seven cases with chronic lower limb ulcers from the key hospitals in the district of west Hunan Province were collected by cluster sampling,and randomly divided into the intervention group (25 cases)and the control group (12 cases).They chose remote wound treatment platform intervention with cellular phone terminator and traditional dressing change in clinic.All patients received a 1.5-year follow-up,or cure and amputations were set as the endpoint of the study.The cost-effect,including the incidence of complications in the course of treatment,number of switches,treatment time,the cost of treatment,as well as medical human resources consumption was analyzed and compared between the two groups.Results The incidence of complications in the course of treatment in the intervention was (76.0%) significantly lower than the control group(91.7%).There was a significant increase in the dressing times and the duration of treatment; and there was no significant difference in the cost of treatment and medical human resources consumption between two groups.Conclusions Compared to the traditional outpatients wound treatment,the multimedia message-based tele-wound treatment platform unlikely increased medical human resources consumption and the cost of treatment,and may reduce the incidence of complications and the treatment time.
9.Protective effect of Zinc and Salvia on the immune system caused by noise in rats

Objective To observe the protective effect of the zinc gluconate combined with Salvia injection on noise induced serum immunoglobulin(IgG,IgM,IgA) and immune molecules (IL-2,IL-4) in rats.Methods 50 female SD rats were randomly divided into control group,noise group,zinc gluconate noise group(plus zinc group),Salvia injection noise group (plus Salvia group) and Zinc gluconate oral liquid and Salvia injection noise group(combined group),10 rats in each group.Except the control group,rats in the other groups were continuously exposed to high frequency steady noise for two weeks.The concentrations of serum immunoglobulin (IgG,IgM,IgA) and immune molecules (IL-2,IL-4) after the intervention of noise were compared.Results Serum levels of IgG,IgM,IL-2,IL-4of the control group were significantly higher than the noise group (all P < 0.05),plus zinc group (all P < 0.05),plus Salvia group(all P <0.05),the differences were statistically significant.Serum level of IgA of each group was not statistically different compared with control group (all P > 0.05).Serum levels of IgG,IgM,IL-2,IL-4 in the noise group were significantly lower than those in the control group (all P < 0.05),combined group (all P < 0.05),the differences were statistically significant.Serum levels of IgG,IgM,IL-2 in plus zinc group were significantly higher than the noise group(all P < 0.05).Serum IgA concentration of each group showed no significant difference compared with noise group(all P > 0.05).Conclusion Zinc gluconate oral solution and Salvia injection have protective effect on immune system affected by noise in rats,the protective effect of combination of zinc gluconate oral solution and Salvia injection is stronger than the individual.
10.Expression and Preliminary Research on the Soluble Domain of EV-D68 3A Protein.

To understand the structure of the soluble region of Enterovirus 68 3A protein, we construct a prokaryotic expression vector expressing the soluble region of EV-D68 3A protein, and identify the forms of expression product after purification. The EV-D68 3A(1-61) gene was amplified by PCR and then cloned into the expression vector pET-28a-His-SUMO. The recombinant plasmid was transformed into Escherichia coli BL21 induced by IPTG to express the fusion protein His-SUMO-3A(1-61). The recombinant protein was purified by Ni-NTA Agarose and cleaved by ULP Protease to remove His-SUMO tag. After that, the target protein 3A(1-61) was purified by a series of purification methods such as Ni-NTA, anion exchange chromatography and gel filtration chromato- graphy. Chemical cross-linking reaction assay was taken to determine the multiple polymerization state of the 3A soluble region. A prokaryotic expression vector pET28a-His-SUMO-3A(1-61) expressing the solution region of EV-D68 3A was successfully constructed and plenty of highly pure target proteins were obtained by multiple purification steps . The total protein amount was about 5 mg obtained from 1L Escherichia coli BL21 with purity > 95%. At the same time, those results determined the homomultimer form of soluble 3A construct. These data demonstrated that the expression and purification system of the soluble region of 3A were successfully set up and provide some basic konwledge for the research about 3A crystal structure and the development of antiviral drugs targeted at 3A to block viral replication.
