Objective To confirm the expression of Aquaporin2,3,4(AQP2,3,4,water channel protein) in rats'endolymphatic sacs(ES)and kidneys and to investigate and to compare the effects of anti-diuretic hormone(AVP) and DDAVP([deamino-Cys1,D-Arg8]-Vasopressin,V2-receptor agonist)on the expression of AQP2 in rats' ESs and kidneys.Methods 30 healthy Swards white rats were divided into the negative control,AVP group and dDAVP group respectively.The animals were cardiacally perfused.The temporal bones and kidneys were taken out and sectioned by means of the paraffin-embedded technique.The sections of ESs were labeled with fluorescent antibody by immunohistochemical method,and the kidneys' with avidin-biotin-peroxidase complex method(ABC).The expressions of AQP-2 were confirmed in the ESs of the rats while the different effects of the AVP and DDAVP on the ESs and the kidneys were observed.The slides used were analyzed by the image-analyzer and the subsequent data were statistically studied.Results In the cytomembrane and cytoplasm of ESs' epithelia,the constant and clear fluorescent reaction could be observed in the normal control group with the first antibody of AQP2.Significant feeble fluorescent reaction of the first antibody of AQP-2 was revealed in AVP group and DDAVP group.In comparison with the control group,significant stain was found in AVP group and DDAVP group.Noted were also lower gray(P

The concentration of 5 nucleosides, uracil, uridine, guanidine, adenine and adenosine in culture of Paecilomyces hepialid was determined by the developed method of HPLC. The HPLC method was performed on a Waters SunFire C18 (4.6 mm x 250 mm, 5 μm) column with methanol-water gradient elution as the mobile phase. The detection wavelength was 260 nm and the colunmn temperature was controlled at 30 °C. The linear range was 10.00-200.00 mg · L(-1) (r = 0.9994) for uracil, 10.10-202.00 mg · L(-1) (r = 0.9992) for uridine, 10.00-200.00 mg · L(-1) (r = 0.9991) for guanidine, 10.30-206.00 mg · L(-1) (r = 0.9992) for adenine and 10.45-209.00 mg · L(-1) (r = 0.9991) for adenosine, respectively. The RSD of precision was 0.032%, 0.035%, 0.039%, 0.049%, 0.00080%, respectively. The average recoveries of uracil, guanidine, adenine, and adenosine were 97.34%, 99.10%, 101.6%, 98.61% and 100.2% with RSD of 1.3%, 2.1%, 0.96%, 0.95%, and 1.3% respectively. The method showed high sensitivity, good selectivity, linearity and repeatability, which was suitable for the content analysis of 5 nucleosides components in P. hepialid and its extracts.

Objective To test the pathogeny of Vibrio cholerae strains of the epidemic in Jinzhou of Hubei Province.Methods Traditional methods of biochemistry,immobihzation test,string test and typing of blood serum were used to test the 6 strains isolated.The virulence gene which was cholera enterotoxin (ct) was detected by PCR.The whole genome DNA finger print of confirmed strains was analyzed by pulsed field gel electrophoresis (PFGE) after digestion respectively with two enzymes Not Ⅰ and Sfi Ⅰ.The DNA fingerprints were analyzed for clusters.Results The 6 strains of Vibrio cholerae were all O139 by traditional laboratory tests;virulence gene was detected in all 6 strains.The banding pattern was the same in the two maps of PFGE.The results of cluster analysis showed that the similarity coefficient of the six strains was 100％.Conclusion The epidemic of Vibrio cholerae is caused by the same pathogenic bacterium which is O139.

OBJECTIVE:To investigate the characteristics of ARDs induced by ciprofloxacin lactate.METHODS:Using philometric method,65 cases developing drug adverse reactions to ciprofloxacin lactate,reported in domestic journals during the past 2 years,were analysed.RESULTS:Of the ARDs in 65 cases,40 were allergic reactions(61.5%),13 reactions of nervous system(12.3%),3 kidney function damage(4.6%),1 liver function damage,1 cardiovascular reaction(1.5%) and 6 reactions of urinary system.CONCLUSION:The ARDs should be under consideration in choice of ciprofloxalin lactate.

Stressors can be classified as controllable and uncontrollable events.Self-choice behavior plays an important role in the "problem-solution" process of coping when individual faces controllable stressors.As to the uncontrollable stressors there are 2 different coping modes: transformation and compensation coping.This paper reviews the research achievements in the relationship between stress and self-choice behavior.

Objective:To establish an HPLC method for the determination of EDTA-2Na in amphotericin B. Methods: A Waters C18 column(50 mm × 4. 6mm, 5 μm) was used. The mobile phase A was acetic acid solution (1. 5 ml acetic acid was added into 1000ml water, and 41 ml 10% tetrabutylammonium hydroxide solution was added), and the mobile B was acetonitrile with gradient e-lution. The flow rate was 0. 8 ml·min-1 , the column temperature was 30℃, the detection wavelength was 260 nm and the injection volume was 25μl. Results:The results showed that EDTA-2Na in amphotericin B could be detected without any interference. The cal-ibration curve of EDTA-2Na was linear within the range of 0. 92-7. 37μg·ml-1(r=0. 999 9), the LOD was 1. 93 ng·ml-1 and the LOQ was 6. 45 ng·ml-1. The average recovery was 102. 5% (RSD=2. 8%, n=9). Conclusion: The method is simple, selective and accurate. It can be used for the quality control of EDTA-2Na in amphotericin B.

Objective To evaluate the association between C-reactive protein (CRP) blood levels and specific survival time of non-small cell lung cancer (NSCLC) patients treated with chemotherapy.Methods Data from 96 advanced (stage ⅢB/Ⅳ) NSCLC patients were analyzed.All patients were divided into two groups based on the enrolled time as follows the initial treatment group (48 cases) and retreatment group (48 cases).According to 0.6 mg/L and 7.3 mg/L of CRP which were the 1/3 and 2/3 of CRP concentrations,respectively,the 96 patients were divided into low,intermediate and high groups.Kaplan-Meier and Cox proportional-hazard models were used to evaluate the relationship between the CRP level and survival time.Results After adjusting for age,sex,smoking history,histological type and stage of lung cancer,a significant relationship between CRP and survival time was observed (P ＜ 0.05).Such significant differences of survival time were also observed in both of the adenocarcinoma (P ＜ 0.001) and squamous with poorly differentiated (P =0.032) subtypes.On stratification analysis by chemotherapy status,the circulating CRP level in retreatment group was correlated well with survival time (P ＜ 0.001).However,the influence of circulating CRP levels on survival time in initial group did not reach statistical significance (P =0.296).For all patients,the hazard ratio with high CRP levels for NSCLC-specific survival was 1.15 (95 ％ CI 0.82-1.61) compared with that of low CRP levels.The hazard ratio for the initial treatment group and retreatment group were 0.52 (95 ％ CI 0.16-1.74) and 1.77 (95 ％ CI 0.73-4.26),respectively.Patients with high circulating CRP level also responded poorly to chemotherapy.Conclusion A high level of circulating CRP is associated with an inferior response and survival outcome in NSCLC patients treated with chemotherapy.

AIM:To seek a simple and reliable process for extracting effective components in Diedahuoxue film.METHODS: According to uniform design, five factors in extraction were optimized, i.e. ethanol concentration, reflux time, reflux times,ethanol volume and drug particle. The absorption value was taken as evaluating parameter in UV- spectrophotometry. RESULTS: The results showed that the best extracting conditions were 95% ethanol, ten times amount of ethanol, 0.2mm size of particle and reflux time of 1h with 4 repetitions.CONCLUSION:This process is simple and reliable.

Transgenic animals are those in which recombinant DNA technology is used to remove a gene or a specific part of DNA with exogenous DNA, which is hereditable and can be expressed stably. Transgenic rodent technology, first demonstrated by Gordon in 1980,opened the door to the next step in biotechnology and gene regulation, which has been used to reproduce or mimic a part of a human disease in a complex cellular environment by creating in vivo models with germ line transmission of the introduced genetic regulatory elements. Animal models for drug discovery are developing increasingly, which is promoting the research and development new drugs greatly. The use and development of transgenic animals in pharmacological research is reviewed in this article.
