1.MiR-512-3p expression pattern and function in breast cancer

Objective:This study aims to investigate the expression pattern of miR-512-3p in breast cancer and noncancerous paired specimens as well as the effects of miR-512-3p on the proliferation, apoptosis, cell cycle, and cloning of MD-MBA-231 breast cancer cells. The study also aims to identify the miR-512-3p target gene. Methods:Reverse transcription-polymerase chain reaction (RT-PCR) was conducted to quantify the miR-512-3p expression in breast cancer and noncancerous paired specimens. Methylthiazol tetrazolium (MTT) assay, flow cytometry, and clone formation assay were used to characterize the function of miR-512-3p in breast cancer. Target prediction was performed using the TargetScan, PicTar, and miRanda software. The results were validated using RT-PCR and western blot target validation. Results:The relative expression of miR-512-3p in breast cancer specimens was significantly lower than those in normal breast specimens (P<0.05). MTT assay revealed that 48 h after transfection, miR-512-3p significantly repressed the proliferation of MD-MBA-231 cells at a suppression rate of 45.38%and at a concentration of 100 nmol/L. MiR-512-3p increased the percentage of early apoptotic cells in the treatment groups (9.32 ± 0.41)%compared with those in the blank controls (3.1 ± 0.54)%and in the negative controls (2.9 ± 0.39)%(P<0.05). Significant differences were found in the percentages of the G0/G1-and G2/M-phase cells after miR-512-3p transfection compared with those in the controls (P<0.05). In the cloning assay, clone formation was inhibited in the miR-512-3p-transfected groups compared with those in the control groups. RT-PCR and western blot results indicate that miR-512-3p significantly inhibited the c-FLIP mRNA and protein expression. Conclusion:MiR-512-3p expression is relatively decreased in breast cancer specimens compared with those in the normal samples. The negative effect of miR-512-3p on cell proliferation and clone formation and its positive effect on early apoptosis through c-FLIP targeting suggest that miR-512-3p acts as a tumor suppressor gene in breast cancer. Therefore, miR-512-3p may be a new target for the diagnosis and treatment of breast cancer in the future.
2.Research on the Stress Resistance of Saccharomyces cerevisiae Industrial Strains for Molasses

The stress resistance of Saccharomyces cerevisiae industrial strains for molasses to high-concentration ethanol,high temperature,high osmotic pressure,furfural toxicity,phenol toxicity,acetic acid toxicity and G418 toxicity were analyzed by the spot dilution growth assays in this paper. The results showed that the stress resistances among these industrial strains were obviously different. The strains AS2.1189 and AS2.1190 are more resistant to the tested stress factors than any others .The strain 396 is the most resistant to the acetic acid toxicity and G418 toxicity,and the strain 2610 is the most resistant to the high temperature.
3.Analysis on the relevant factors of Hypotension in the patients applying Hemodialysis

Objective To explore the relevant factors of hypotension in the patients applied hemodialysis.Methods 45 patients who had intradialytic hypotension in our dialysis room were selected for the analysis.Results The patients with diabetic nephropathy,the elderly,high temperature,hyponatremia,higher ultrafiltration volume,stepwise ultrafihration and the constant ultrafihration showed higher incidence of hypotension during hemodialysis(x2 =9.87,81.22,171.02,130.12,all P <0.01).Conclusion The primary disease,age of the patient,uhrafiltration volume,temperature,sodium concentration and the ultrafiltration are all the relevant factors for intradialytic hypotension
4.The effect evaluation of continuous care pathway on hypertension control:Evidence from a ru-ral community-based quasi-experiment

Objectives:To test the effect of continuous care on rural hypertension control , we developed a com-prehensive intervention strategy and implemented a community-based quasi-experiment in Southwestern rural China.Methods:The intervention took place in Qianjiang District , Chongqing Municipality from July 2012 to June 2014.4 towns were randomly selected and categorized into 2 groups based on a comprehensive consideration of population and social development level.All the rural hypertension patients in treatment group were intervened with the Multi-institu-tional Continuous Pathway which was consisted of three parts:the Continuous Primary-Care Pathway , the Continuous Clinical Pathway and the Continuous Management Pathway.The patients in the control group served as a blank com-parison using the usual care.Difference-in-differences Model was used to test the effect of blood pressure control in treatment group.Results:853 patients were sampled and investigated using the stratified randomly sampling strategy and 712 of them had been followed up by the end of this program.The potential bias of pilot and patient selection were eliminated through control before and after the intervention.The previous blood pressure trends showed no statis-tically significant difference between groups.The systolic blood pressure in treatment group declined by 10.156 mm-Hg ( P<0.001 ) compared to control group after intervention , and the blood pressure control rate had increased by 27.6% ( P<0.001 ).Other contributing factors besides intervention were family structure , education level and med-ical service availability.Conclusion:The continuous care pathway have a significant marginal positive effect on hyper -tension control besides the national compulsory primary care , and the control rate change of blood pressure is more sen-sitive compared to blood pressure change.The potential contributing factors show that other intervention strategies could be developed to improve the rural hypertension control by adding to the social capital of rural patient , reinforcing the health education and facilitating the village transportation.

6.Study on Bactericidal Activity of Thymopentin Peptide and Its Derived Peptides

OBJECTIVE:To study the bactericidal activity of thymopentin and its derived peptides. METHODS:Agar plate count was adopted to determine the bactericidal activity of thymopentin [arginine(R)-lysine(K)-aspartic acid(D)-valine(V)-tyro-sine(Y),RKDVY],its derived peptide 1 [RKN(agedoite,N)VY] and derived peptide 2(RKKVY)to Gram negative bacterial (Proteusbacillus vulgaris,Escherichia coli) and Gram positive bacterial (Staphylococcus aureus,Enterococcus faecium). There were 15.625-1 000 μg/ml for peptides,102 CFU for bacteria. RESULTS:Three pentapeptides possessed bactericidal activity against Gram negative bacteria. The activities of RKKVY and RKNVY were stronger than RKDVY(P<0.01),there was no significant dif-ference between RKKVY and RKNVY(P>0.05). They also possessed bactericidal activity against Gram positive bacteria,and the activity from strong to weak was RKKVY>RKNVY>RKDVY(P<0.01). CONCLUSIONS:Thymopentin and its derived peptides possess bactericidal activity against Gram negative and positive bacteria,with dose-effect relationship.
7.Removal of Cu,As,and Pb from chrysoidine by supercritical CO_2 extraction

Objective To make an attempt at some conditions on extracting heavy metals in chry-soidine by supercritical CO_2 extration.Methods Taking sodium diethyldithiocarbamate trihydrate(NaDDC?3H_2O) as chelating agent and ethanol as entrainer,the orthogonal test was designed and ICP-MS used to mensurate the contents of Cu,As,and Pb in chrysoidine under different conditions.According to the results,the factors including extracting pressure,temperature,chelating agent dosage,and time were studied.Results When the test sample was 10 g,the optimum condition was that extracting pressure: 25 MPa,the temperature : 60 ℃,chelating agent dosage: 2 g,extracting time: 3 h,and the ethanol dosage: 10 mL.After extracting reaction,the contents of heavy metal in chrysoidine reached United States FDA standard.Conclusion The contents of heavy metal in Chinese herb medicine are notably decreased,and this way provides a new thought and research technique to decrease the contents of heavy metal.
8.Reducing bacterial aerosols by dental procedure with Xipayi mouth rinse or H_2O_2 pre-rinse

Aerosol and splatter formed during dental procedure were collected pre and post rinsing with Xipayi mouth rinse or 1% H2O2,cultured 48 hours with blood agar plates,counted the bacterial colonies.Both groups were effective in reducing bacterial aerosols(P 0.05).Bacterial aerosols by dental procedure can be reduced by using mouthwash pre-operationally,both Xipayi mouth rinse and 1% H2O2 are effective mouthwashes.
9.Early diagnosis and treatment of diabetic macular edema

Diabetic retinopathy ( DR ) is one of the main blinding eye diseases for people over the age of 50, and diabetic macular edema ( DME) is the leading cause of vision loss is DR patients. The early diagnosis and early treatment is important. As OCT and FFA, mfERG, especially the retinal thickness, volume, retinal edema index quantitative indicators such as objective evaluation of macular edema, embodies the new progress of retinal imaging technology in recent years. OCT is a non -contact clinical application in recent years, noninvasive, high resolution of ophthalmic imaging examination, can do it on retinal ultrastructure observation and quantitative analysis, and the technology is relatively mature, become a routine inspection diagnosis of macular edema. Laser photocoagulation, intravitreous injection with Ranibizumab and vitrectomy is nowadays the important means for the treatment of intractable macular edema.
10.Application of joint fixation with breast bracket and body phantom to breast cancer radiotherapy after breast-conserving operation

Objective To apply joint fixation with breast bracket and body phantom to breast cancer radiotherapy after breast-conserving operation.Methods Totally 36 patients after breast-conserving operation underwent joint fixation with breast bracket and body phantom, whose cone beam computed tomography (CBCT) images were obtained with on board imager (OBI) before radiotherapy. The errors between the CBCT images and planning CT images were acquired by matching.Results The setup errors in the directions of left-right (X), head-foot (Y) and belly-back (Z) were (2.6±0.783), (3.0± 1.027) and (1.6 ±0.872)mm respectively, and the percentages that the setup errors were more than 5 mm were 16.5%, 20.7% and 9.1% respectively.Conclusion Joint fixation with breast bracket and body phantom may decrease the setup error in radiotherapy after breast-conserving operation, especially in the belly-back direction, and thus has high clinical values.
