1.Effect and mechanism of tissue transglutaminase-mediated chemoresistance in cancer cells
Cancer Research and Clinic 2013;25(9):643-645
TG2 is a calcium-dependent enzyme that catalyzes the formation of covalent bonds between free amine groups in one protein and protein-bound glutamines of another, creating highly cross-linked protein complexes.TG2 is ubiquitous and most diverse member of the transglutaminase family of enzymes.TG2 are implicated in cell differentiation,receptor-mediated endocytosis, cell adhesion, and induction of apoptosis.Recently,a growing body of literature suggests that it could be closely related to the development of drug resistance. Its role in cancer biology is briefly reviewed in this paper.
2.The clinical efficacy of recombinant human thrombopoietin for the treatment of post-chemotherapy throm- bocytopenia of adult patients with acute myeloid leukemia
The Journal of Practical Medicine 2017;33(16):2711-2713
Objective To study the effect of recombinant human thrombopoietin(rh TPO)for the treatment of post-chemotherapy thrombocytopenia in adult patients with acute myeloid leukemia(AML). Methods A total of 60 newly diagnesed AML patients ,law-and Intermediate-Rist with post-chemotherapy thrombocytopenia were randomly divided into experimental group (30 cases,rh TPO treatment)and control group(30 cases,no rhTPO treatment). Comparisons were made on the blood platelet recovery degree, the days of the blood platelet counting less than 20 × 109/L,the recovery days of the blood platelet counting more than 50 × 109/L,and incidence of adverse reac-tions of experimental group. Results After treatment, blood platelet recovery level between the experimental group and the control group has no difference(P>0.05). The days of the blood platelet counting less than 20 × 109/L and the recovery days of the blood platelet counting more than 50 × 109/L of experimental group were less than the control group separately(P < 0.05). The experimental group had low incidence of adverse reactions(6.7%). Conclusion rhTPO can not significantly increase the level of platelet recovery of AML patients after chemotherapy ,but can obvi-ously shorten the time of platelet recovery without obvious adverse reactions. The specific time needs further studied.
3.Effects of Ilexonin A on acute hypoxic pulmonary vasoconstriction in rats
Chinese Journal of Pathophysiology 1986;0(01):-
Acute hypoxic pulmonary vasoconstriction (HPV) plays an important role in physiological regulation. The purpose of this study was to explore the effects of Ilexonin A on hemodynamics and HPV, and their relation to prostaglandins (PG_(?)) and leukotrienes (LT_(?)).The results show that the high pulmonary vascular resistance induced by Ilexonin A in normoxic rats may be partially caused by the increased LTs and have no relation to PGs. Ilexonin A may potentiate HPV at least by two ways: replacing the production of PGs (especially PGI_2) in hypoxic Wistar rats, and increasing LTs production.
4.Pharmacokinetics of antibody-drug conjugates and their research progress in the treatment of gastrointestinal malignancies
Acta Pharmaceutica Sinica 2023;58(9):2632-2639
The morbidity and mortality of gastrointestinal malignancies are the highest in the world. For patients with poor response to conventional chemotherapy, new treatment methods are urgently needed. In recent years, under the background of precision medicine, antibody-drug conjugates (ADCs) with high tumor specificity and potent toxicity have become a hot research spot in the field of biomedicine. However, due to the complex structure and mechanism of ADCs, its pharmacokinetic research is facing great challenges which are the biggest resistance to the development of ADCs at present. In this case, it is of great significance to understand the pharmacokinetic properties of ADCs and make use of it to improve the efficacy of ADCs in the treatment of gastrointestinal malignancies. Based on the basic composition and mechanism of ADCs, this review summarizes the pharmacokinetic properties of ADCs, discusses its recent advances in the treatment of gastrointestinal malignancies, in order to provide more references for follow-up research on ADCs.

Acta Pharmaceutica Sinica 2015;50(6):755-759
With development of bio-technique, more and more proteins were applied as clinical approaches. However, the protein homogeneity, especially the N-glycosylation limited the further research and application of these protein drugs. The analysis method for N-glycans is believed to be critical in protein drugs development. To enhance the N-glycans isolation efficiency and accelerate the pretreatment, a new strategy was built on ultrafiltration-devices. New methods increased the isolation efficiency of N-glycans containing N-acetylglucosa mine with 10%-20%. The degrading of N-glycans containing sialic acids was also minimized with this method. 20%-100% more N-glycans with sialic acids were isolated. The pretreatment was finished within 30 min. Coupled with HPLC-HRMS, an effective and reliable strategy designed for protein drugs N-glycans analysis were developed.
