1.Basic fibroblast growth factor-transfected bone marrow mesenchymal stem cell transplantation for acute kidney injury

BACKGROUND:It has been confirmed that basic fibroblast growth factor (bFGF) can promote the growth, proliferation, differentiation and functional expression of most cels derived from neuroderm and mesoderm. OBJECTIVE:To investigate the effect of bFGF-transfected bone marrow mesenchymal stem cel transplantation in rats with acute kidney injury. METHODS: bFGF genes were transfected into bone marrow mesenchymal stem cels via an adenovirus vector, and then expression of bFGF in transfected cels was identified using RT-PCR technology. Rat models of acute kidney injury were prepared by clipping bilateral renal pedicles, and then randomized into three groups (n=20): rats were given injection of bone marrow mesenchymal stem cel suspensionsvia tail vein as negative transfected group, those given injection of bFGF-transfected bone marrow mesenchymal stem cel suspensionsvia tail vein as bFGF-transfected group, and the others given injection of DMEMvia tail vein as model group. Four weeks later, levels of serum creatinine and urea nitrogen were detected, expressions of connective tissue growth factor and growth factor in renal tissues were detected by Western blot assay, and morphology of renal tissues was observed using hematoxylin-eosin staining. RESULTS AND CONCLUSION:bFGF genes were successfuly transfected into bone marrow mesenchymal stem cels. Compared with the model group, the levels of serum creatinine and urea nitrogen were significantly reduced in bFGF-transfected and negative transfected groups, especialy in the bFGF-transfected group (P < 0.05), while expressions of connective tissue growth factor and transforming growth factor in renal tissues in bFGF-transfected and negative transfected groups were significantly weakened in these two groups (P < 0.05), but there were no significant differences between the bFGF-transfected group and negative transfected group (P > 0.05). Besides, renal tissues necrosis and inflammatory reactions were mitigated in the negative transfected group; renal tubules with normal outlines and no overt necrotic cels could be found in the bFGF-transfected group. These findings show that bFGF-transfected bone marrow mesenchymal stem cel transplantation plays a better role in acute kidney injury repair in rats.
2.The effect of “T”incision on distant stereovision of cataract patients with high cornea astigmatism

Objective To evaluate the effect of “T”incision on distant stereovision of cataract patients with high cornea astigma-tism .Methods 40 age-related cataract patients (40 eyes) with high cornea astigmatism for surgery in our hospital were divided into two groups :A(20 eyes) an B(20 eyes) .The A groups received superior(11 :00) clear corneal incision(CCI) .The B groups besides the superior(11:00) CCI ,for surgery on the steepest meridian ,an additional couple of “T”incisions were made to release the preex-isting corneal astigmatism .The operation eyes were all implanted Rayner foldable posterior chamber intraocular lens .The UCVA , corneal astigmatism ,distant stereovision were observed at 3 months after operation .Results The UCVA in each group after sur-gery was all better than that of preoperation ,difference being statistically significant (P<0 .01) .the difference between Group A and Group B was significant(P<0 .05);In Group A ,there were no significant difference on the corneal J0 and P before and after op-eration(P>0 .05) ,The corneal J0 and P in group B after surgery were lower than that of preoperation .difference being statistically significant(P<0 .01) .difference between Group A and Group B(P<0 .01) at 3 months postoperation was statistically significant (P<0 .01);The stereopsis in each group after surgery was all better than that of preoperation ,difference being statistically significant (P<0 .01) .the difference between Group A and Group B postoperation was significant (P<0 .05) .Conclusion the“T” incisions on the steepest corneal meridian can improve the patient′s visual function and distant stereovision .

BACKGROUND: Recently, with the increasing popularity of dental implant and orthognathic surgery, more and more people pay attention to the structure of mandibular canal and adjacent tissues. However, there are few reports addressing specimen measurements at home and abroad. OBJECTIVE: To measure the anatomical data of mandibular canal and adjacent tissue of mandible specimen to provide reliable reference data and anatomical evidence for alveolus surgery, tooth implantation, and orthognathic surgery. DESIGN, TIME AND SETTING: A controlled observation was performed at the Laboratory of Anatomy, Jilin Medical College between March 2007 and September 2007. MATERIALS: Twenty complete mandible specimens. METHODS: Sixteen adult, ex vivo mandibles with complete dentition and 4 edentulous mandibles were measured in every dental position of sagittal profile and posterior mental foramen in terms of molar, alveolar crest, and mandibular canal. Data were statistically analyzed. MAIN OUTCOME MEASURES: Mental foramen location, horizontal distance from anterior border of mental foramen to anterior border of mental canal, mandibular foramen location, distances from mandibular canal to molar root tip, lingual bone plate of mandible, inferior border of mandible, mandibular buccal bone plate, and alveolar ridge crest. RESULTS: Mandibular canal, located in the inferior region of the mandible body, ran towards the lingual side and was close to the inferior border of mandible and most close to the mandibular molar root tip. It turned towards posterolateral and buccal side in front of mental foramen and then passed through mental foramen. The distance of mandibular canal between edentulous mandible and alveolar ridge crest was obviously shortened. CONCLUSION: Multi-section observation and measurement of ex vivo mandible specimens provide precise, reliable evidence for preoperative design of orthognathic surgery, facilitate surgeons to better formulate surgical proposals, and prevent some complications, including intraoperative massive hemorrhage, interior alveolar nerve injury, mandibular angle fracture, and mental bone lateral wall perforation.
6.Comparison of problem-based learning and lecture-based learning for clinical teaching in de-partment of cardiovascular medicine

Objective To compare the effect between problem-based learning and lecture-based learning for clinical teaching in the department of cardiovascular medicine. Methods Totally 110 five-year-program cardiovascular interns from June 2011 and June 2012 were selected. They were randomly divided into the PBL group(n=55) and LBL group(n=55). PBL and LBL teach-ing methods were applied in the two groups respectively. Teaching effects were evaluated by exam and questionnaire investigation. SPSS 15.0 was used to do data processing; t test was used to compare the average score of two groups;chi-square test was used to process the results of the questionnaire. P<0.05 signifies sta-tistically significant differences. Results There were statistical differences in examinational average score between PBL group and LBL group ((87.89 ±5.39) vs. (82.63 ±5.26), P<0.05). PBL group had significantly higher satisfaction rate in motivating study interests , deepening understanding of theoretical knowledge, cultivating self-learning ability, training verbal expression and developing clin-ical thinking, etc(P<0.05). Conclusions PBL teaching method demonstrates advantages in teaching of cardiovascular medicine and enhances the teaching effect. But the PBL teaching method should be improved in basic knowledge teaching, cultivation of teachers' ability and case selection.
7.Heme oxygenase decreases the generation of ROS in AngⅡ induced proliferation and hypertrophy of cultured vascular smooth muscle cells

AIM:To investigate the role of heme oxygenase (HO) in AngⅡ induced proliferation and hypertrophy of cultured vascular smooth muscle cells. METHODS:(1) Western blotting analysis was carried out to detect protein level of HO-1 in the tissues. (2) [3H]-TdR,[3H]-leucine incorporation was measured in cultured vascular smooth muscle cells. 8.Effect of heme oxygenase on vascular remodeling in renal hypertension

AIM: To investigate the effect of heme oxygenase on vascular remodeling in renal hypertension. METHODS: Male Wistar rats were randomly divided into sham-operated, 2K1C (two-kidney one-clip) and hemin-induced groups. Four weeks after the treatments, the thickness of aortic media and HO enzymatic activity of the aorta were determined. Immunohistochemical staining was carried out to detect protein of HO-1 in the aorta. RESULTS: The blood pressure in 2K1C renal hypertension rats started to increase two weeks after the surgery and stabled at a high level at the 4th week. Hemin, an inducer of HO-1, markedly inhibited the increase in blood pressure. Aortic medium thickness of the 2K1C rats at 4th week was 27 5% thicker than that in the sham-operated rats. The thickness of aortic medium of the hemin-induced rats was 16 1% less than that in 2K1C group. At the 4th week after operation, protein level and enzymatic activity of HO-1 in aorta were higher than that in 2K1C group compared to those in the sham-operated group. CONCLUSION: Renal hypertension caused vascular remodeling and the activation of HO-1. HO-1 induction decreased the blood pressure of renal hypertension and reduced vascular remodeling.
9.Therapeutic effect of intracoronary versus intravenous bolus tirofiban in elderly patients with acute coronary syndrome

Objective To analyze the therapeutic effect of intracoronary versus intravenous bolus tirofiban on myocardial perfusion and major cardiovascular events (MACE) in elderly patients with non-ST-segment elevation acute coronary syndrome (NSTE-ACS) and explore the optimal route of tirofiban application. Methods From July 2009 to July 2010, 120 NSTE-ACS patients undergoing percutaneous coronary intervention (PCI ) were consecutively enrolled in this study. They were randomly divided into two groups: intracoronary (60 cases) versus intravenous (60 cases) bolus tirofiban. Thrombolysis in myocardial infarction (TIMI) flow, TIMI myocardial perfusion grade (TMPG) and MACE 30 days after PCI were observed. Results The incidence of TIMI flow and TMPG 3 grade in intracoronary group were higher than in intravenous group [53(88.3%) vs. 38(63.3%); 53(88.3%) vs. 40(66.7%), respectively, both P<0.05]. However, MACE incidence and bleeding complications during hospital 30 days after PCI had no significant difference between the two groups [1 (1.7%) vs. 0; 3(5.0%) vs. 5(8.3%)], which were not statistically significant (P>0.05). Conclusions Intracoronary bolus tirofiban before PCI more effectively increases coronary blood flow and myocardium blush than intravenous route in elderly NSTE-ACS patients.

Objective To study the effect of expression of RNA interference targeting protein kinase B(PKB) gene transfection mediated by nanoparticles(NP) on intimal hyperplasia(IH) in vein grafts of rats.Methods Nanoparticle PKB short hairpin RNA(shRNA) gene complex was prepared with PLGA and PVA.Autogenous vein graft model was established in 72 rats by transplanting internal jugular vein to carotid artery.The models were randomly divided into 3 groups:(1) PKB shRNA group: PKB-shRNA gene mediated by NP were transfected into the veins before anastomosis.(2) Empty vector group: the veins were transfected by empty vector mediated by NP.(3) Control group(no transfection).The grafted veins were harvested 3,7,14 and 28 days after the operation respectively.The mRNA and protein expression of PKB were determined by Northern blot and Western blot.IH was observed by HE and Verhoeff stain.The presence of apoptotic VSMC was detected by TUNEL stain.Results Compared to empty vector group and control group,PKB shRNA group had less expression of mRNA and protein of PKB gene(P
