1.RATIO OF NEURON AND GLIA DIFFERENTIATED FROM THE EMBRYONIC HUMAN NEURAL STEM CELLS AT DIFFERENT TIME IN VITRO

Objective To compare the ratios of neuron and glia differentiated from the embryonic human neural stem cells(hNSCs) at different times in vitro. Methods The cells from hippocampus of aborted human embryo tissue(8-12 weeks after conception) were mechanically dissociated and cultured with DMEM/F 12 medium containing h-EGF,h-bFGF,h-LIF.The NSCs were induced to differentiate by 1%FBS and identified by Musashil,?-Ⅲ Tubulin,GFAP and Galc immuno-cytochemical staining at different times. Results The cell spheres were Musashil positive at the 12th hour after differentiation.On the 2nd day after differentiation,a few cells exhibited ?-Ⅲ Tubulin and GFAP immunoreactivities,but no Galc positive cells.The number of ?-Ⅲ Tubulin immuno-positive cells reached the highest on the 7(th) day and the percent was 48.2%.The number of GFAP immuno-positive cells increased from the 2nd to the 23rd day and the positive ratio reached 65.3%.A few Galc immuno-positive cells were detected with time passing.Conclusion 1% FBS may promote human NSCs to differentiate into three cell types of central nervous system,and the ratios of differentiated neuron and glia are significantly different at various times.
2.PTCH2 gene alterations in keratocystic odontogenic tumors associated with nevoid basal cell carcinoma syndrome

T)of PTCH2 were identified and 9 polymorphisms (3 of which were novel) were determined in the present series. Conclusion: Although not as frequent as PTCH1 mutations, PTCH2 germline mutations were detectable in a subset of NBCCS patients with KCOTs. The pathogenetic role of these PTCH2 mutations is yet to be clarified.

5.Preparation of acteoside and forsythoside B reference substances from Lamiophlomis rotata

Objective To establish a method for the isolation and preparation of forsythoside B and acteoside reference substances from Lamiophlomis rotata.Methods Forsythoside B and acteoside in L.rotata were isolated and purified by macroporous resin,Sephadex column,and preparative HPLC.Results Analysis with HPLC showed the content of the prepared acteoside and forsythoside B reached to 98.93 and 99.91%,respectively.Conclusion This method is effective for the high purity of prepared acteoside and forsythoside B.It can be used as reference substances for the qualitative and quantitative analyses of Chinese herbal medicine.
6.QUANTITATIVE STUDY ON THE CAPILLARIES OF THE HUMAN THALAMUS AND ITS ADJACENT STRUCTURES

The brains from four newborns were used in this study. After continuous artery-vein injection with coloured materials, the brains were embedded in nitrocellulose. and coronal sections of 500?m, 100?m and 30?m in thickness were prepared in alternative and successive sequence. The 500?m sections were cleared in wintergreen oil and mounted, and the 100?m and 30?m sections were stained with thionin. The diameter and density of capillaries in thalamus and its adjacent structures were measured by means of Leitz MPV-Tasplus multifunction image analyser. All the data were dealt with variance and correlation analysis statistically. The capillary density of the thalamus and its adjacent structures varied obviously. The value in putamen, cellular layers of lateral geniculate body, anterior thalamic nucleus and subthalamic nucleus was the highest; while the value in dorsomedial nucleus, ventral lateral nucleus, pulvinar nucleus, ventral posterior lateral nucleus, lateral posterior nucleus, centromedian nucleus, medial geniculate body, ventral anterior nucleus and internal medullary lamina was the intermediate; however, the value in internal capsule and fibrous layers of lateral geniculate body was the lowest. The capillary diameter of ventral anterior nucleus and internal capsule was the widest; that of putamen and cellular layers of lateral geniculate body was the smallest; the rest was the intermediate. In the same measured structures, negtive correlation was shown between the capillary diameter and density.
7.THE DISTRIBUTION OF INTRATHALAMIC VESSELS IN HUMAN BRAIN

The course, distribution of the intrathalamic vessels on 8 sides of newborn brains were studied by means of the continuous artery-vein infusion, Spalteholz's clearing and Nissl's staining. The brains were all cut into the coronal sections of 500 ?m, 100?m and 30?m thickness in alternate and successive order. The main arteries supplying thalamic nuclei were: 1. The thalamoperforating artery; 2. The geniculothalamic artery; 3. The medial posterior choroidal artery; 4. The medial inferior pulvinar artery; 5. The lateral ventricular choroidal artery. The thalamic veins opened respectively into the internal cerebral vein, the veins of the lateral ventricle, the veins of the interpeduncular fossa and the basal vein. There were three patterns of course relation between thalamic arteries and veins. They were solitariness, accompanying and one vein surrounded by several arteries.

10.Comparison of cerebral oxygenation inpatients undergoing gynecological laparoscopic surgery under sevoflurane-versus propofol-based anesthesia

Objective To compare the cerebral oxygenation in the patients undergoing gynecological laparoscopic surgery under sevoflurane-versus propofol-based anesthesia.Methods Fifty adult patients,aged 18-56 yr,weighing 55-72 kg,of American Society of Anesthesiologists physical status Ⅰ or Ⅱ,with hemoglobin≥ 110 g/L,undergoing elective gynecological laparoscopic surgery under general anesthesia,were equally and randomly divided into sevoflurane group (group S) and propofol group (group P) using a random number table.Anesthesia was induced with iv midazolam 0.04 mg/kg,sufentanil 1.5-2.0 μg/kg,rocuronium 0.9 mg/kg,and propofol 1.5-2.0 mg/kg.Anesthesia was maintained with inhalation of sevoflurane (2.0%-2.6%) in group S,or with target-controlled infusion of propofol (plasma concentration 2.5-4.0 μg/ml) in group P.The concentrations of sevoflurane and propofol were adjusted to maintain bispectral index value at 40-50.In both groups,remifentanil was given by targetcontrolled infusion (target plasma concentration 2 ng/ml),and vecuronium 0.06-0.08 mg/kg was injected intermittently to maintain muscle relaxation.The regional cerebral oxygen saturation (rSO2) was recorded after induction and at 10 min before pneumoperitoneum (T1),at 10 min of pneumoperitoneum in the Trendelenburg position (T2),and at 10 min after the end of pneumoperitoneum (T3).The occurrence of cerebral oxygen desaturation was recorded during surgery.Results The rSO2 was significantly higher at T2,3 in group S than in group P (P<0.05).Compared with the value at T1,no significant change was found in rSO2 at T2,3 in group S (P>0.05),and the rSO2 was significantly decreased at T2,3 in group P (P<0.05).The incidence of cerebral oxygen desaturation was 12% in group P,and cerebral oxygen desaturation was not detected in group S.Conclusion Sevoflurane-based anesthesia produces no effect on rSO2,however,propofol-based anesthesia can decrease rSO2 in the patients undergoing gynecological laparoscopic surgery,and sevoflurane-based anesthesia is more suitable for gynecological laparoscopic surgery.
