OBJECTIVETo study the effect of Buyang Huanwu decoction (BYHWD) inducing angiogenesis on the neuroblast migration from the subventricular zone and its mechanisms after focal cerebral ischemia.
METHODThe middle cerebral artery occlusion (MCAO) was performed to mice for 30 minutes to establish the model. The rats were divided into sham group, model group, BYHWD group and endostatin group. BYHWD (20 g x kg(-1), ig) and endostatin (10 μg, sc) were administered 24 h after ischemia once a day for consecutively 14 days. At 14 d after ischemia, the density of micro-vessel and the number of neuroblasts in the ischemia border zone were determined by immunofluorescence staining. The mRNA and protein expression of cell-derived factor-1 (SDF-1) and brain-derived neurotrophic (BDNF) were examined by real-time PCR and Western blot.
RESULTCompared with the model group, BYHWD significantly increased the density of micro-vessel and the number of DCX positive cells in the ischemia border zone (P < 0.01), and significantly increased the SDF-1 and BDNF mRNA and protein expression (P < 0.01). Compared with BYHWD group, endostatin significantly reduced the density of micro-vessel and the number of DCX positive cells in the ischemia border zone (P < 0.01), as well as the SDF-1, BDNF mRNA and protein expression (P < 0.01).
CONCLUSIONBYHWD could promote the neuroblast migration from the subventricular zone via inducing angiogenesis after cerebral ischemia, the mechanism may be correlated with up-regulating the expression of SDF-1 and BDNF.

AIMTo study the metabolic kinetics of MN9202 in Beagle dog liver microsome.
METHODSBeagle dog liver microsomes were prepared by using ultracentrifuge method. After incubating 0.4 micromol x L(-1) MN9202 with 1 g x L(-1) microsomes for 30 min at 37 degrees C, the reaction was terminated by adding 0.5 mL alkalization. The RP-HPLC was used to determine the drug in the incubation mixture. The Michaelis-Menten parameters Km, and Vmax in Beagle dog liver microsomes were initially estimated by analyzing Lineweave-Brurk plot. Various selective CYP inhibitors were used to investigate their inhibitory effect on the metabolism of MN9202.
RESULTSThe Km, Vmax and CLint of MN9202 were (22.6 +/- 8.0) micromol x L(-1), (0.54 +/- 0.17) micromol x g(-1) x min(-1) and (0.0242 +/- 0.0009) L x g(-1) x min(-1), respectively. The metabolism of MN9202 was significantly inhibited by ketoconazole (Ket) and troleandomycin (Tro) in Beagle dog liver microsomes. Tranylcypromine (Tra) could inhibit the metabolism of drug as well. While other inhibitors showed little inhibitory effect on the metabolism of MN9202.
CONCLUSIONIt was shown that CYP3A and CYP2C19 were involved in MN9202 metabolism. The inhibitors of human CYP3A and CYP2C19 may have potential interaction with MN9202, and this can reduce the metabolism rate and increase the toxicity of MN9202.

OBJECTIVETo investigate the characteristics of seroconversion of HBV NAT screening-positive crowd from blood donors in Dongguan city and provide reference for the safety of blood transfusion and disease prevention.
METHODSWith retrospective survey, Nucleic acid testing (NAT) was used to analyze 28800 HBsAg-negative samples by ELISA from blood donors in Dongguan city from August, 2006 to August, 2007 with Roche Cobas AmpliScreen systems; and follow-up research including NAT for HBV-DNA, ELISA for HBsAg and multiple factors analysis for HBV infection was carried out on HBV NAT screening-positive crowd.
RESULTS10 positive pooling were screened from 28800 samples; after further detection, 2 of these positive pooling were HBV-DNA negative and 8 HBV-DNA positive samples were found. The 10-week follow-up research on these 8 blood donors showed that 6 were HBV-DNA positive and HBsAg-negative at 2 weeks, 4 weeks, 6 weeks, 8 weeks, 10 weeks respectively, 1 was not HBsAg positive until 2 weeks and was positive on follow-up, and considered in "window period", 1 was HBV-DNA negative, HBsAg-negative on follow-up. Of these 8,7 were not only migrant laborers with poor condition of work, life and health but also in high risk of secondary infection for HBV, in addition they had little idea of therapy or prevention measures of HBV infection and the other 1 was HBV-DNA negative, HBsAg-negative on follow-up, who was in better condition than the above 7 donors.
CONCLUSIONNAT is more sensitive than ELISA in screening HBV, but the probability of being false positive of NAT can not be ignored at the same time. On the hand, only screening HBsAg for HBV is relative limitation in high infection region of China. Some factors would have effect on the serum conversion of blood donors including the quality of work and life, therapy or prevention measures.

OBJECTIVE: To analyze the prevalence and influencing factors of work-related musculoskeletal disorders(WMSDs) in civil aviation flight attendants. METHODS: A total of 810 flight attendants from three civil aviation airlines in China were selected as research subjects using the convenient sampling method. The revised Musculoskeletal Disorders Investigating Questionnaire was used to investigate the prevalence of WMSDs in various parts of the body in the past year. RESULTS: The total prevalence of WMSDs in flight attendants in this survey was 64.4%(522/810). The prevalence of WMSDs in various parts of the body from high to low was: neck(48.0%), shoulder(38.6%), lower back/waist(26.0%), upper back(19.8%), feet(15.1%), knee(14.0%), hip and leg(11.0%), hand and wrist(9.0%) and elbow(5.1%). The results of multivariate logistic regression analysis showed that working with an uncomfortable posture and the shortage of staff in the work sector were risk factors for neck WMSDs(all P<0.05). The protective factors were sufficient rest time and voluntary decision when to take a break during work(all P<0.05). Carrying heavy objects >20 kg, working in uncomfortable posture and shortage of staff were risk factors for shoulder WMSDs(all P<0.05). Working in uncomfortable posture and repeated an operation every minute were risk factors for lower back/waist WMSDs(all P<0.05), and sufficient rest time was its protective factor(P<0.05). CONCLUSION: The prevalence of WMSDs in civil aviation flight attendants is high, and the neck, shoulder and ower back/waist are the most commonly affected part of the body. The main influencing factors are poor ergonomics and work organization.

OBJECTIVETo evaluate the feasibility, efficacy and safety of the percutaneous coronary intervention (PCI)guided by computed tomography (CT) coronary angiography derived roadmap and magnetic navigation system (MNS).
METHODSDuring June 2011 and May 2012, thirty consecutive patients receiving elective PCI were enrolled, coronary artery disease was primarily diagnosed by dual-source CT coronary angiography (DSCT-CA) at outpatient clinic and successively proved by coronary artery angiography in the hospital. Target vessels from pre-procedure DSCT-CA were transferred to the magnetic navigation system, and consequently edited, reconstructed, and projected onto the live fluoroscopic screen as roadmap. Parameters including characters of the target lesions, time, contrast volume, radiation dosage for guidewire crossing, and complications of the procedure were recorded.
RESULTSThirty patients with 36 lesions were recruited and intervened by PCI. Among the target lesions, sixteen were classified as type A, 11 as type B1, 8 as type B2, 1 as type C. The average length of the target lesions was (22.0 ± 9.8) mm, and the average stenosis of the target lesions was (81.3 ± 10.3)%. Under the guidance of CT roadmap and MNS, 36 target lesions were crossed by the magnetic guidewires, with a lesion crossing ratio of 100%. The time of placement of the magnetic guidewires was 92.5 (56.6 - 131.3) seconds. The contrast volume and the radiation dosage for guidewire placement were 0.0 (0.0 - 3.0) ml and 235.0 (123.5 - 395.1) µGym(2)/36.5 (21.3 - 67.8) mGy, respectively. Guidewires were successfully placed in 21 (58.3%) lesions without contrast agent. All enrolled vessels were successfully treated, and there were no MNS associated complications.
CONCLUSIONSIt is feasible, effective and safe to initiate PCI under the guidance of CT derived roadmap and MNS. This method might be helpful for the guidewire placement in the treatment of total occlusions.
