1.Antitumor effects of losartan in EAC mice

AIM: To study the antitumor e ff ects of losartan in EAC mice. METHODS: The inhibitory rates of t umor growth and the ratio of extending viability were observed in EAC mice in th ree groups given losartan, 5-FU and NS, respectively. RESULTS: Comparing with the control groups, the inhibition to tumor growth was 46.3 % in losartan ( 12.5 mg?kg -1 ) group. But losartan did not lengthen life time in EAC mice. The inhibition to tumor growth was was 31.5 % in 5-F U (5 mg?kg -1 ) group. CONCLUSION: Losartan can inhibit the tumor growth of EAC, but not lengthen life time of the animals.
2.Bone loss (osteoporosis)induced by retinoic acid in mice and its dose-dependent effects

AIM To study the dose dependent effects on osteoporosis induced by retinoic acid(RA) and to analysis the metabolic characteristics of bone. METHODS Three dosages of retinoic acid were given to the mice for 2 weeks while the body weight(BW) were measured once a week. At the end, femoral dry weight, Bone hydroxyproline, calcium content, Phosphor, Magnesium, Zinc, Manganese and Sulphonium of femora were measured. RESULTS Compared with normal control mice, three dosages of retinoic acid can induce the decrease of femoral dry weight and the short transverse diameter of femoral, Bone hydroxyproline content decreased remarkably, and so did the bone mineral. The Bone HOP and Bone Ca content was coincident with the femoral dry weight, but the lessening of Bone P, Bone Mg, Bone Zn and Bone Mn became inverse ratio with the femoral dry weight, particularly that of the Bone Zn. CONCLUSION Three dosages of retinoic acid in mice can induce osteoporosis with the lessening of bone mass, which characteristics were the losing of Bone HOP and the Bone Ca content in geometric proportion. The levels of Bone P, Bone Mg, and Bone Zn had lost remarkably in the early time, the Bone Zn may play importance function in osteoporosis model induced by retinoic acid.
3.Suppression of the expression of hPOT1 gene in human gastric cancer cell line SGC-7901 with vector-based RNA interference technique

Objective To construct the expression vector of siRNA targeting the human protection of telomeres (hPOT1) gene for the observation of the inhibitory effect of the vector on hPOT1 gene expression in gastric cancer cell line SGC-7901, and to offer a ground work for further study on the roles of hPOT1 in growth and proliferation of gastric cancer cells. Methods 64 base-pair oligonucleotide for small interfering RNA expression targeting hPOT1 gene was designed and chemically synthesized. After annealing, double oligonucleotides were inserted into the downstream of H1 RNA promoter of pSUPER to recombine psiRNA-hPOT1 plasmids, and transiently transfected into gastric cancer cell line SGC-7901 with liposome-mediated transfection after restriction enzyme and sequencing analysis. Semi-quantitative RT-PCR was used to detect the expression levels of hPOT1 gene. Result Recombinant psiRNA-hPOT1 vector was confirmed by sequencing analysis. The results demonstrated that 64nt had been inserted into the expected site. Furthermore, the inserted sequence was exactly correct. The recombinant psiRNA-hPOT1 vector could significantly suppress the hPOT1 expression in gastric cancer cell line SGC-7901. Conclusion The constructed siRNA expression vector of hPOT1 gene psiRNA-hPOT1 recombinant plasmid can block the expression of hPOT1 gene in gastric cancer cell line SGC-7901.

5.Study of adiponectin expression in placenta and its correlation with preeclampsia

Objective To investigate the expression of adiponectin in placenta and its correlation with preeclampsia.Methods Placental tissues were collected from normal term pregnancies(normal pregnancy group,n=20),mild preeclampsia(mild preeclampsia group,n=12)and severe preeclampsia (severe preeclampsia group,n=22).The expression of adiponectin protein and the intensity of its mRNA in placenta were detected using immunohistochemistry and RT-PCR,respectively.Integral optical density (IOD)which represents the expression level of adiponectin protein,and the ratio of adiponectin cDNA PCR products to β-actin cDNA PCR products which represents the intensity of transcription of adiponectin mRNA in placenta were analyzed.Results (1)The expression of adiponectin protein was observed in cytoplasm of placental cytotrophoblasts and syncytiotrophoblasts among three groups.There was no significant difference in adiponectin protein expression between maternal side and fetal side of placenta in three groups(all P>0.05);(2)The expression of adiponectin protein in placenta in severe preeclampsia group(30 984 ±14 604)was significantly lower than that of mild preeclampsia group(58 360±8910,P<0.01)and of normal pregnancy group(53 246±17 554,P<0.01).There was also no significant difference in the expression of adiponeclln protein in placenta between term delivery and preterm delivery in severe preeclampsia group(38 890±20 386 vs 29 319±8997,P>0.05),however,the expression of adiponectin protein in placenta in term delivery of severe preeclampsia group was significantly lower than that ofterm delivery of normlal pregnancy group(38 890±20 386 vs 53 246±17 554,P<0.05);(3)The expression of adiponectin mRNA was detected in placental tissues among three groups also.The intensity of transcription of adiponectin in placenta in severe preeclampsia group(1.0±0.2)was markedly lower than that of mild preeclampsia group(2.9±0.8,P<0.05)and normal pregnancy group(3.3±1.1,P=0.000).Conclusion The expression of adiponectin decreases in placenta tissues of severe preeclampsia,indicating that the abnormal expression of adiponectin may be involved in the pathogenesis of preeclampsia.
6.The protective effect of synthetic cordyceps Sinensia Sacc powder on the liver grafts of rats

Objective To investigate the protective effect of the extractions of synthetic Cordyceps Sinensia Sacc powder on cold preserved injured tissues of rat livers. Methods The extractions of synthetic Cordyceps Sinensia Sacc powder were added into LR at certain proportion. Having been preserved for 6 or 12 hours, the liver grafts of rats were reperfused for 30 minutes. And then, ATP, ADP, AMP in livers were measured and added up to total adenine nucleotide (TAN). Besides, AST, ALT, MDA, and ET in the effluent of the reperfusion were assayed. Pathological sections were studied and apoptosis index was detected by TUNEL method. Results The values of ATP, TAN in the experimental group preserved for 12 hours were significantly higher than the control group (P0.05), but the mean of the former was higher than the latter. The experience of "Zhenjing Anshen" acupuncture method originally created by professor WANG Fuchun for treatment of insomnia was introduced in this paper. From aspects of insomnia pathogenesis, theoretical foundation, characteristics of acupoint selection, needing methods, needing time, etc., the experience of Professor WANG Fuchun for treatment of insomnia was explained. The "Zhenjing Anshen" acupuncture method proposed, for the first time, "new three layers" method of acupoint selection, including Sishencong (EX-HN 1), Shenmen (HT 7), Sanyinjiao (SP 6). This method presents the principles of acupoint selection along meridian, acupoint selection based on essence-qi-spirit, harmony of yin and yang. The acupuncture manipulation is emphasized, and treating time (the period of the day from 3 pm to 5 pm) is focused on; acupoint selection is simple but essential, and acupoint combination is scientific, which receives notable therapeutic effect in clinic.

9.Directional differentiation of bone marrow mesenchymal stem cells into oligodendrocytes induced by the combination of various neurotrophic factors

BACKGROUND: Myelination following axonal regeneration is a key factor affecting the recovery of spinal cord injury. Oligodendrocyte survival directly affects the myelination following axonal regeneration. OBJECTIVE: To investigate the feasibility of differentiation of rat bone marrow mesenchymal stem cells (BMSCs) into oligodendrocytes induced by neurotrophic factors. DESIGN, TIME AND SETTING: The cell molecular biology in vitro study was performed at the Laboratory of Department of Orthopaedics, Tongji Hospital from September 2006 to June 2007. MATERIALS: A total of 5 Sprague Dawley rats aged 2-4 weeks, of both gender were selected. Bilateral femur and tibia bone marrow was obtained to harvest BMSCs. METHODS: At passage 4, BMSCs were incubated in serum-free medium, supplemented with N2, 20 ng/mL basic fibroblast growth factor, 20 ng/mL epidermal growth factor for 48 hours, and incubated in medium containing 500 ng/mL insulin-like growth factor I and N2 for 3 days. MAIN OUTCOME MEASURES: Morphological changes were observed using an phase contrast microscope. Semiquantitative RT-PCR was utilized to detect specific marker mRNA expression of oligodendrocytes. Using neuron marker anti-microtubule-associated protein, astrocyte marker anti-glial fibrillary acidic protein, oligodendrocyte marker anti-galactocerebroside, anti-myelin basic protein antibody, immunocytochemical staining was performed to detect the positive rate of the differentiation of BMSCs into oligodendrocytes. RESULTS: Morphological changes in BMSCs during the differentiation into oligodendrocytes: After the induction, a majority of BMSCs presented the morphological characteristics of oligodendrocytes. Cytoplasm retraction towards nucleus, cell process extension towards outwards, and strong refraction were found. With the prolongation of time, several cell processes connected and formed a typical net-shape structure. Specific marker mRNA expression of oligodendrocytes: Following induction, specific strap of myelin basic protein mRNA and galactocerebroside mRNA could be detected. Positive rate of oligodendrocytes: During induction, the positive rates of galactocarebroside, myelin basic protein and microtubule-associated protein were 65%, 45% and 10%, respectively. CONCLUSION: The combination of epidermal growth factor, basic fibroblast growth factor and insulin-like growth factor can effectively promote the directional differentiation of BMSCs into oligodendrocytes.
10.Enhancement of ginsen-stem-leaves to the antitumor effect of 5-FU in mice inoculated S180

AIM: To observe the enhancement of ginsen stem leaves (GSL) to the antitumor effect of 5 FU in mice inoculated S180. METHODS: The different combinations of GSL and 5 FU were used to examine the inhibition rates of tumor growth in KM mice inoculated S180. RESULTS: The combination of GSL (15 mg?kg -1 ) and 5 FU (1.5 mg?kg -1 ) obtained 26.90% of inhibition rates of tumor growth (P
