AIM:To observe the influences of artesunate(AS)on murine bone marrow hematopoietic stem and progenitor cells.METHODS:Murine bone marrow hematopoietic stem and progenitor cells were cultured in vitro with different concentration of AS.The numbers of erythrocyte colony-forming unit(CFU-E)or burst-forming unit(BFU-E)in liquid or semisolid culture were counted through light microscope.Apoptosis of cultured cells and the changes of mitochondrial membrane potential were detected by flow cytometry(FCM)analysis,meanwhile,DNA ladder gel electrophoresis were conducted.RESULTS:AS could significantly inhibit the formation of CFU-E and BFU-E(P
2.Double Contrast Diagnosis of Small and Micro Gastric Carcinoma at the Beginning of Early Gastric Carcinoma

Objective:To improve the diagnostic accuracy of double contrast radiography of small and micro gastric carcinoma.Methods:The X-ray findings of small gastric carcinoma(9 cases)and micro-gastric carcinoma(3 cases) proved by surgical pathology were analyzed and compared with pathologic finding.1 misdiagnosed cases and 1 missed cases were analyzed.The double contrast films of acute gastric ulcer were reviewed and differentiated with SGC and MGC.Results:8 cases were examined by the double contrast procedure before gastroscopy,4 were diagnosed small gastric carcinoma,1 was diagnosed micro-gastric carcinoma,1 was misdiagnosed ulcer and 1 was missed,1 was suspected of being carcinoma.4 were examined by the double contrast procedure after gastroscopy,2 were small gastric carcinoma and 1 was micro-gastric carcinoma,which accorded with gastroscopy finding.One of micro-gastric carcinoma missed by gastroscopy was detected by DC.Conclusion:The imaging method of double contrast examination is the most effective one in detecting and diagnosing early gastric cancer.The detecting rate should be obviously increased by combining with gastroscopy closely. 3.Factors Related with Activity of Daily Living in Patients with Schizophrenia

Objective To explore the activity of daily living in patients with schizophrenia and its related factors. Methods 200 patientswith schizophrenia were investigated with the questionnaire about general condition, the Positive and Negative Syndrome Scale (PANSS)and Activity of Daily Living Scale (ADL) when hospitalized. The relationship between the scores of ADL and sexuality, age, education level,marital status, profession before the disease attacking, household, disease type, course of the disease, frequency of attacking and hospitalizationor scores of PANSS was analyzed. Results The score of ADL was significantly above the normal level (t=32.206, P=0.000) in theschizophrenics. The score of ADL was not significantly different among the patients in different gender, education level, marital status, profession,household and the frequency of hospitalization (P>0.05), but was significantly different among the ages (F=3.851, P=0.023) and diseasetype (F=19.793, P=0.000). The score of ADL positively correlated with the course of the disease (r=0.161, P=0.023), frequency of attacking(r=0.396, P=0.000) and the difference value between the numbers of attacking and hospitalization (r=0.425, P=0.000), so as the totaland each factors scores of PANSS (P=0.000), but did not correlated with frequency of hospitalization (r= 0.097, P=0.174). By multiple regressionanalysis, only 3 factors score of PANSS, the frequency of attacking and the difference value between the numbers of attacking andhospitalization entered into the regression equation (F=53.630, P=0.000). Conclusion The patients with schizophrenia generally fall in activityof daily living. The more serious the disease, the more frequent the attacking and the more the difference value between the numbers ofattacking and hospitalization, the poorer the activity of daily living was.
4.Influence of Admission Mode on Quality of Life of Patients with Chronic Schizophrenia

Objective To compare the influence of different admission modes on quality of life of patients with chronic schizophrenia.Methods 200 patients with chronic schizophrenia were randomly assigned to discontinuous group and continuous group with 100 cases in each group. All patients were treated with atypical antipsychotic drugs. A discontinuous hospitalization combined with home treatment was given to the discontinuous group and a continuous hospitalization to the continuous group for 1 year. The Positive and Negative Syndrome Scale (PANSS) and the Schizophrenia Quality of Life Scale (SQLS) were respectively used to evaluate the patient's condition and quality of life before and after the intervention. Results The intervention methods on scores of PANSS and SQLS in both groups had a change trend with time. The scores of PANSS and SQLS were significantly lower in the end of 6 months and 1 year after intervention than before in both groups (F=2119.513, 390.981, P=0.000). There was a interaction effect between intervention method and time in both groups on the scores of PANSS and SQLS (F=17.660, 11.709, P=0.000). There was significant difference between 2 groups on the score of PANSS (F=19.673,P=0.000). The decrease range of those scores was greater in the discontinuous group than in the continuous group (P=0.000). Conclusion A discontinuous hospitalization combined with home treatment is more beneficial than a continuous hospitalization for the patients with chronic schizophrenia in controlling the syndrome and improving or keeping the quality of life.
5.Cost-effectiveness Analysis of Rehabilitation of Intermittent Hospitalization on Chronic Schizophrenia

Objective To analyze the cost-effectiveness of the rehabilitation of intermittent hospitalization combined with home treatmenton chronic schizophrenics. Methods 100 patients with chronic schizophrenia were randomly assigned to study group and controlgroup, 50 cases in each group. All patients in both groups were treated with atypical antipsychotic drugs. An intermittent hospitalization combinedwith home treatment was given to the study group and a continuous hospitalization to the control group for a year. Positive and NegativeSyndrome Scale (PANSS), Activities of Daily Living Scale (ADL) and Schizophrenia Quality of Life Scale (SQLS) were used to evaluatethe patient's condition and activity of daily living and quality of life before and after the intervention. In the meantime, a cost-effectivenessanalysis was made. Results The interventions had significant influence on the scores of PANSS, ADL and SQLS in both groups. Thescores of PANSS, ADL and SQLS were significantly lower 1 year after the intervention than before in both groups (FB= 540.577, 318.148,326.751, all P=0.000). There was a significant interaction effect between intervention method and time in both groups on the scores ofPANSS, ADL and SQLS (FAB=13.710, 10.918, 6.578, P=0.000, 0.001, 0.012). The decrease value of those scores was significantly greater inthe study group than in the control group (t=3.703, 3.304, 2.565, P=0.000, 0.001, 0.012). The ratio of cost-effectiveness was larger in thestudy group than in the control group (t=7.035, P=0.000). Conclusion Intermittent hospitalization combined with home treatment is morebeneficial for the patients with chronic schizophrenia in controlling the syndrome, keeping or improving the activity of daily living and thequality of life. Therefore its rehabilitation benefit is better than that of continuous hospitalization.
6.Isolation and characterization of cancer stem cell-like sphere-forming cell subpopulation from the human nasopharyngeal cell line CNE-2

Objective: The present study aimed to isolate and characterize a cancer stem cell-like sphere-forming cell subpopula-tion. Methods: By using a spheroid culture stem cell-conditioned medium, we isolated a subgroup of cancer stem-like cells from naso-pharyngeal cancer cell lines. Chemotherapy resistance was analyzed by using methyl thiazolyl tetrazolium, and clone-forming capabili-ty was determined by using softer agar clone formation assay. Finally, we verified the expression of the stemness-specific gene andβ-catenin by using immunocytochemistry staining and RT-PCR. Results: The lower-differentiated nasopharyngeal cancer lines con-tained more sphere-forming cells, whereas sphere-forming cells were not observed in the high differentiated nasopharyngeal cancer line CNE-1. Compared with CNE-2, CNE-2S (sphere-forming cells derived from CNE-2) exhibited higher chemotherapy resistance and clone-forming ability. Interestingly, the stemness genes BMI-1, ABCG2, and ALDH1 exhibited higher expression in CNE-2S than in CNE-2. β-catenin, a vital transcription factor of the WNT pathway related to stem cells, exhibited higher expression in CNE-2s cellular nucleus and plasma than in CNE-2. Conclusion: We isolated a subgroup of stem-like nasopharyngeal cancer sphere-forming cells. This discovery paves the way for the development of therapeutic strategies aimed at eradicating tumorigenic subpopulations in nasopharyn-geal cancer.
7.Expression and significance ofβ-catenin in gastric adenocarcinoma and the metastasis of lymphnode

Objective To explore the expressions ofβ-catenin in gastric adenocarcinoma primary tumors and metastatic lymph nodes,and to determine if it is associated with infiltration degree and whether it can provide the basis for gastric cancer metastasis.Methods The expressions ofβ-catenin were detected in 156 patients with gastric adenocarcinoma specimens,40 cases with the corresponding lymph nodes and 12 cases of normal gastric tissues by the method of immunohistochemistry.The expressions ofβ-catenin in gastric adenocarcinoma and corre-sponding lymph node metastases were analysed.The relationship between the expression of β-catenin and the clinical pathological features of gastric adenocarcinoma was ascertained,and difference between them was observed.Results The positive expression rate ofβ-catenin in normal gastric tissue membrane was 100.0%(12/12).The expression rate ofβ-catenin was 29.5%(46/156)in adenocarcinoma cell membrane,and it was 10.0%(4/40)in metastatic lymph nodes cell membrane.The positive expression ofβ-catenin was not associated with patient′s age(χ2 =2.160,P=0.142),gender (χ2 =1.229,P=0.268),but it was associated with tumor infiltration degree (χ2 =4.032,P=0.045 ),degree of tumor differentiation (χ2 =6.093,P=0.048),tumor stage (χ2 =4.591,P=0.032),and metastasis lymph nodes (χ2 =4.485,P=0.034).The incidence of abnor-mal or loss expression ofβ-catenin in gastric adenocarcinoma metastatic lymph nodes was higer than that in gastric adenocarcinoma (χ2 =6.362,P=0.012).Conclusion The expression of β-catenin will be a great help to judge the biological behaviors such as the malignant degree of tumor and the capacity of tumor invasion and metas-tasis.
8.Synthesis and preliminary evaluation of 18F-AIF-NOTA-G-TMTP1 for PET imaging on nude mice bearing high-metastatic potential hepatoma cells

Objective To synthesize 18F-AlF-NOTA-G-TMTP1 and evaluate its potential for PET imaging on nude mice bearing high-metastatic potential hepatoma cells.Methods NOTA-G-TMTP1 was synthesized by the standard Fmoc-solid phase synthetic protocols and radiolabeled with 18F using NOTA-AlF chelation method.The nude mice models bearing low-metastatic potential HCC97L and high-metastatic potential HCCLM3 xenografts were established separately.The tumor-targeting characteristics of 18F-AlF-NOTA-G-TMTP1 were assessed by microPET/CT and biodistribution assay.Results NOTA-G-TMTP1 was labeled with 18F in one step with (25±6)% labeling yield (n=5).The radiochemical purity of 18F-AlF-NOTA-G-TMTP1 was more than 95% with a specific activity more than 11.1 GBq/μmol.The octanol/water partition coefficient (logP) for 18F-AlF-NOTA-G-TMTP1 was-3.166±0.022.The tumor to muscle ratios were 1.8± 0.4 and 4.7±0.2 at 35 min post injection for HCC97L and HCCLM3,respectively.The uptake of 18F-AlF-NOTA-G-TMTP1 in HCCLM3 tumor was inhibited (61.4%) by unlabeled G-TMTP1.Conclusion 18F-AlF-NOTA-G-TMTP1 has been successfully synthesized.It shows specific uptake by tumor induced by the high-metastatic potential hepatoma cells.
9.Effects of Guiqi Yiyuan Ointment on Immune Function in 12C6+ Beam Bystander-effect Rats

Objective To observe the effects and mechanism of Guiqi Yiyuan Ointment on immune function in 12C6+ beam bystander-effect rats.Methods Totally 60 Wistar rats were randomly divided into blank group,model group,positive control group,and TCM low-,medium-,and high-dosage groups,with 10 rats in each group.TCM low-,medium-,and high-dosage groups were given 1.2,2.4,and 4.8 g/kg Guiqi Yiyuan Ointment for gavage,and the blank group,model group and positive control group were given equal dose of sodium chloride solution for gavage for 14 days.Positive control group received intraperitoneal injection of amifostine 30 minutes before modeling.On the 15th day,except the blank group,the other groups were given 4 Gy 12C6+ beam of single irradiation on the right lung.After irradiation,each group was given gavage for 3 days.The general condition of rats was observed,peripheral blood leukocyte(WBC),red blood cell(RBC),hemoglobin(Hb),platelet(PLT)levels were detected,ELISA was used to detect the contents of serum interleukin(IL)-4 and interferon-γ(IFN-γ),lung and spleen organ indices were calculated,HE staining was used to observe the pathological morphology of left lung and spleen tissues,and flow cytometry was used to detect the proportion of CD4+ and CD8+ T lymphocytes in the spleen,qPCR was used to detect the expressions of T-bet and GATA-3 mRNA in right lung and spleen tissues.Results Compared with the blank group,the model group rats showed a decrease in peripheral blood WBC,an increase in RBC,Hb,PLT,and an decrease in serum IL-4 and IFN-γ contents;the organ indices of left lung and spleen was significantly reduced,the alveoli were deformed,the alveolar walls became thicker,the alveolar cavities shrank,the lung tissue was significantly bleeding,the spleen tissue structure was severely damaged,the splenic sinus cavity became narrower,and the splenic cord showed fibrous hyperplasia;the proportion of CD4+ T lymphocytes and CD4+/CD8+ ratio in spleen tissue of the model group decreased,while the proportion of CD8+ T lymphocytes increased,the expression of T-bet mRNA in the left lung and spleen tissues decreased,while the expression of GATA-3 mRNA increased,with statistical significance(P<0.05).Compared with the model group,the positive control group and TCM medium-and high-dosage groups showed an increase in peripheral blood WBC,an decrease in RBC,Hb,PLT,an increase in serum IL-4 and IFN-γ contents;the index of left lung and spleen organs increased in TCM high-dosage group;the pathological damage of left lung and spleen tissues in each treatment group alleviated to varying degrees,while the proportion of CD4+ T lymphocytes and CD4+/CD8+ ratio in spleen tissue of the positive control group and TCM high-dosage group increased,the proportion of CD8+ T lymphocytes decreased,the expression of T-bet mRNA in left lung and spleen tissues increased,and the expression of GATA-3 mRNA decreased,with statistical significance(P<0.05).Conclusion Guiqi Yiyuan Ointment can alleviate the lung and spleen injury of rats induced by 12C6+ beam radiation.Its mechanism is related to regulating the proportion of CD4+,CD8+ T lymphocytes and the expression of T-bet and GATA-3,and recovering cellular immune function.
10.Stereological study of the placenta in patients receiving different vasopressors for hypotension during cesarean section.

OBJECTIVETo study the effects of dopamine and phenylephrine for treatment of hypotension during cesarean section under combined spinal epidural anesthesia (CSEA) on the stereology of the placenta.
METHODSForty puerperants undergoing cesarean section under CSEA were randomly divided into dopamine group and phenylephrine group. Ropivacaine (16 mg) was administered immediately after spinal anethesia. Blood pressure was maintained near the baseline by adjusting the drug infusion rate. Fetal blood gas, Apgar score, and placental villus microvascular stereological changes were observed during the operation.
RESULTSThe microvascular density was significantly lower in dopamine group than in phenylephrine group (P<0.05). Phenylephrine group showed significantly lower umbilical artery blood pH than dopamine group (P<0.05). The Apgar score and blood pressure were comparable between the two groups (P>0.05). Compared to the baseline, both of the two groups showed significantly lowered heart rate during the operation (P<0.01).
CONCLUSIONDopamine is associated with the risk of fetal acidosis. Phenylephrine is helpful for preventing hypotension by increasing placental blood flow and improving oxygen supply to ensure maternal and fetal safety during cesarean section.
