1.Research progress in lncRNAs and the diagnosis, prognosis, and drug resistance of gastric cancer

Long non-coding RNAs (lncRNAs) are noncoding RNAs with lengths exceeding 200 bp. These lncRNAs cannot code complete proteins but control gene expression. Recent studies have shown that lncRNAs are closely related to the diagnosis, prognosis, and drug resistance of gastric cancer. This article summarizes the recent progress of studies in China and abroad on lncRNAs relative to the diagnosis, prognosis, and drug resistance of gastric cancer.

ObjectiveTo study the relationship between the susceptibility of mutiple sclerosis (MS) and single nucleotide polymorphisms (SNP) rs6897932 of interleukin-7 receptor alpha (IL-7RA) in Asian.MethodsThe SNP rs6897932 in the IL-7RA gene was genotyped by real time polymerase chain reaction (PCR) using TaqMan SNP Genotyping Assays. 78 cases with MS and neuromyelitis optica (NMO), 187 patients with non-NMO MS and 158 healthy controls were enrolled.ResultsThe frequencies of both the C allele and the CC genotype of SNP rs6897932 in the IL-7RA gene in non-NMO MS patients were higher than those of healthy controls(89.3% vs. 79.8%, OR=2.12, 95%CI:1.38-3.25 P<0.01; 78.6% vs. 63.3%, OR=2.13,95%CI:1.32-3.43, P<0.01).However, there was no significant difference in the frequency of either the C allele or the CC genotype between control group and NMO patients.ConclusionsIL-7RA gene is one of susceptibility gene for MS in Asian. C allele was presumed as a risk factor of MS while T allele might be a protective factor.

6.Relationship between CYP2D6*10 gene polymorphism and metoprolol therapeutic effect for hypertension

Objective: To study the relationship between CYP2D6*10 gene polymorphism and metoprolol therapeutic effect for hypertension. Methods: A total of 60 patients with essential hypertension (EH) received metoprolol 47.5mg/d for 3d. After 3d the plasma metoprolol concentration after oral 2h was measured. Polymorphism of CYP2D6*10 gene was detected by PCR-RFLP. According to results of gene detection, the patients were divided into CC genotype group (wild type homozygote, fast metabolism type, n=14), CT genotype group (heterozygous mutation, intermediate metabolism type, n=25) and TT genotype group (homozygous mutation, slow metabolism, n=19). Metoprolol dosage was adjusted according to CYP2D6*10 genotype. After one week, plasma concentration of metoprolol after oral 2h was measured again, and mean heart rate and blood pressure were measured before and after gene-directed therapy. Results: Before gene-directed therapy, compared with CC and CT group there was significant increase in plasma concentration of metoprolol [(26.57±19.40) ng/ml vs. (23.88±12.86) ng/ml vs. (64.74±32.94) ng/ml, P<0.01] in TT group; compared with TT group, there were significant rise in mean systolic blood pressure [mSBP, (132.84±13.40) mmHg vs. (144.14±14.28) mmHg], mean diastolic blood pressure [mDBP, (76.95±9.07) mmHg vs. (81.36±7.33) mmHg] and mean heart rate [mHR, (69.13±11.83) times/min vs. (76.66±7.33) times/min] in CC group, P<0.05 all. After gene-directed therapy, there were no significant difference in plasma concentration of metoprolol, mSBP, mDBP and mHR among all groups, P>0.05 all; Compared with before gene-directed therapy, there was significant increase in plasma concentration of metoprolol, and significant decrease in mSBP, mDBP and mHR in CC group (P<0.05). There were no significant difference in blood pressure and heart rate between before and after treatment in CT group and TT group (P>0.05). Conclusion: CYP2D6*10 gene polymorphism affects metoprolol metabolism and its therapeutic effect on hypertension, gene-directed therapy can significantly improve drug therapeutic effect and reach ideal therapeutic goal in short time.
7.Therapeutic effect of bisoprolol with gene targeting therapy for hypertension

Objective:To explore therapeutic effect of bisoprolol with CYP2D6 and CYP3A4 gene targeting therapy for hypertension .Methods :A total of 100 cases with essential hypertension (EH) were selected ,among them 20 ca-ses were randomly chosen to receive routine medication (bisoprolol 5 mg/d ,routine treatment group ) ,the other 80 patients were equally divided into group A (CYP2D6*10) and group B (CYP3A4*1G) .According to detection results of CYP2D6*10 and CYP3A4*1G gene polymorphism ,they were dived into group ACC (n=13) ,group ACT (n=14) ,group ATT (n=10) ,group BCC (n=21) ,group BCT (n=17) and group BTT (n=0) .Bisoprolol dosage was adjusted according to genotypes :group ACC and group BCC received 10 mg/d ,group ACT and group BCT received 5 mg/d ,group ATT and group BTT received 2.5 mg/d .Blood drug concentration ,blood pressure and heart rate were compared among above groups after two weeks .Relationship between plasma concentration of bisoprolol and gene polymorphism of drug metabolism enzyme was evaluated .Results:Comparison between related groups with same bi-soprolol dosage :there were no significant difference in blood drug concentration ,heart rate and blood pressure be-tween ACT and routine treatment group , P>0.05 all;compared with routine treatment group ,there was significant rise in blood drug concentration [ (33.5 ± 19.1) ng/ml vs .(50.13 ± 23.21) ng/ml] ,significant reduction in heart rate [ (71.4 ± 5.16) times/min vs .(66.5 ± 6.04) times/min] and blood pressure [ (127.22 ± 10.44/82.4 ± 7.27) mmHg vs .(119.48 ± 11.97/71.2 ± 10.30) mmHg] in BCT group , P<0.05 all .Conclusion:Because gene possesses polymorphism ,therapeutic effect of bisoprolol treating hypertension is differing ;the gene targeting therapy may significantly improve therapeutic effect for hypertension .
8.Pre-L-methionine feeding strategy for S-adenosyl-L-methionine fermentative production.

The yield of S-adenosyl-L-methionine (SAM) on high-cell-density fermentation by saccharomyces cerevisiae is mostly affected by the feeding strategy of pre-L-methionine. The mutant strain SAM0801 that could accumulate more SAM was used in this study. Six high-cell-density fermentation experiments in 5 L fermentor were investigated to get the optimal feeding time and amount of L-methionine. The results showed that when 40 g L-methionine was added in the fermentor after 30 h fermentation, a dry cell weight of 100 g/L was achieved. Under this condition, after 58 h fermentation, both the dry cell weight and the yield of SAM reached the maximum, 168 g/L and 14.48 g/L respectively.

9.Thoracoscopelungcancer resection with non tracheal intubation anesthesia

Objective To evaluate the feasibility and safety of thoracoscopic lung cancer surgery under non-tracheal intubation anesthesia. Methods Twenty patients with peripheral lung cancer were enrolled in experimental group and control group. Then monitored and recorded Systolic pressure (SBP), diastolic pressure (DBP), mean arterial pressure (MAP), heart rate (HR), electrocardiogram (ECG), heart rate (HR), Oxygen saturation (SpO2), Final moisture CO2 partial pressure (PETCO2), central venous pressure, invasive arterial blood pressure and blood glucose and the related complications like sore throat, hoarse voice, nausea and so onin such time points: before induction (T0), induction of intubation (T1), operation (T2), and sudden removal (T3) of the two groups. Results The laryngeal mask group was given a smaller stimulus to the cardiovascular system during anesthesia.The time of feeding, the exhaust, the time of getting out of bed, the average hospitalization day, the reduction of hospitalization expenses, pharynx, respiratory and cardiovascular complications were shorter and less than intubation group. Conclusion The laryngeal mask ventilation intravenous anesthesia with thoracic vagal nerve block in the thoracoscopic lobectomy is simple, safe, no intubation-related complications and single lung ventilation lung injury, in line with surgery -anesthesia overall minimally invasive development concept, worthy of clinical promotion.
10.Changes of IL-1,IL-6,TGF-?,Fas and FasL expressed by Leydig cell in testis infection

Objective:To study the effect of immune regulation of the rat Leydig cells in testis infection.Methods:UU was injected into the bladders of rats,which mimics the ascending infection pathway,while using culture medium injection as control.After 1,2,3 w respectively,the rats were sacrificed to observe the pathological alterations in testis by histological examination.Leydig cell were also separated from testis,the comparation of levels of IL-1,IL-6,TGF-?,Fas and FasL mRNA expression among control and UU infected groups was made by RT-PCR.Results:Compared with control,in UU infected group,the levels of IL-1,IL-6,TGF-? increased,the level of Fas decreased,the level of FasL increased.Conclusion:Leydig cell can play the role of immune regulation,with the expressing changes of IL-1,IL-6,TGF-?,Fas and FasL in the infection of testis in rat.
