1.Screening for epigenetically masked genes in Kashin-Beck disease by microarray

Objective We used the DNA methylation microarrays to investigate the differential methylation genes and loci sites in Kashin-Beck disease (KBD),to study the relationship between DNA methylation and KBD pathogenesis.Methods Totally 12 KBD adults and 12 healthy adults were selected and peripheral blood samples were collected and DNA was extracted.Illumina 450K bead-chip was applied to detect methylation status in KBD and healthy controls.Aberrant hyper-methylated sites were filtrated according to the P value after correction and methylation differences,together with GenomeStudio soft.Screened genes were validated using bisulfite sequencing polymerase chain reaction (BSP) technology.Results A total of 484 948 loci sites were analyzed and compared,93 differential methylated loci were found by comparing KBD and normal people,including 34 hypermethylated sites and 59 hypomethylated sites.There were 50 genes corresponding to the loci,43 genes not reported in literature.According to gene ontology analysis,the genes were involved in the immune response,antigen processing,phosphate and phosphoric acid metabolism and phosphorylation and the process of metal ions in combination.However,in the verification test using BSP method,there was no significant difference in methylation rate in human leukocyte antigen (HLA)-DRB1 between the case and the control group (48% vs 70%,x2 =3.688,P > 0.05).Conclusions The high and low differentially methylated sites in peripheral blood DNA of KBD patients are significantly different from those of the health control.HLA-DRB1 locus is not significantly different between the BSP verification test and methylation chip.
2.Association study on personality and plasma cortisol level in the patients with generalized anxiety disorder

Objective To study the correlation of personality characteristics and plasma cortisol levels in the patients with generalized anxiety disorder.Methods According to Diagnostic and Statistical Manual of Mental Disorders (DSM-Ⅳ), 38 patients with generalized anxiety disorder and 39 normal controls were assessed with Hamilton anxiety scale (HAMA) , generalized anxiety disorder scale (GAD-7) , penn state worry questionnaire (PSWQ) , symptom checklist-90(SCL-90) and eysenck personality questionnaire (EPQ).Electrochemiluminescence was used to test plasma cortisol concentration.Results The EPQ-N score in the patients was significantly higher than that in normal controls(t=14.307, P<0.01).Furthermore,N score in the patients showed a significantly positive correlation with HAMA total score, SCL-90 total score, somatization score, interpersonal sensitivity score, depression score and anxiety score (r=0.492-0.977, P<0.01).Plasma cortisol levels in patients with generalized anxiety disorder((668.50±60.22) nmol/L) were significantly higher than those ((345.35±58.91) nmol/L) in normal controls (t=23.802, P<0.01).Plasma cortisol concentrations displayed a significantly positive correlation with N score, GAD-7, PSWQ, HAMA total score, SCL-90 total score, somatization score, interpersonal sensitivity score, depression score, and anxiety score (r =0.501-0.995, P< 0.01).Conclusion Plasma cortisol levels in the patients with generalized anxiety disorder are higher than those in normal controls.Plasma cortisol levels are related to the mental health.Patients with generalized anxiety disorder tend to be neuroticism.
3.Investigation on Clinical Pharmacist’s Work Status in West China Hospital of Sichuan University

OBJECTIVE:To investigate the work status of clinical pharmacists in West China Hospital of Sichuan University and to provide rational suggestions for the development and improvement of clinical pharmacy. METHODS: The status of hospital clinical pharmacy and training of clinical pharmacy were studied by a questionnaire survey on 26 clinical pharmacists and an interview on 9 trainees majored in clinical pharmacy. RESULTS: The major tasks for hospital clinical pharmacists were to participate in clinical rounds, conduct pharmacy rounds alone, participate in patients’ treatment and outpatient pharmaceutical care, listen to case discussion, participate case consultation, coordinate with doctors to make medication plan, collect adverse drug reactions and give lecture among physicians etc. The pharmacists were welcome by most of the clinical departments. CONCLUSIONS: Clinical pharmacy in West Hospital of Sichuan University achieved a good beginning, but to achieve a further progress, the focus of clinical pharmacists’ work should be shifted to provide patient-centered pharmaceutical technical service, meanwhile, the support offered and the importance attached by the government as well as the efforts of clinical pharmacists are indispensible.
4.Effects of Different Processing Methods on the Contents of Oleanolic Acid and Ursolic Acid in Chaenomeles Medicinal Materials

Objective To investigate effects of different processing methods on the contents of oleanolic acid and ursolic acid in chaenomeles medicinal materials. Methods The contents of oleanolic acid and ursolic acid in samples processed by different processing methods in producing area were determined by HPLC with Agilent HC-C18 (2) chromatographic column (4.6 mm×250 mm, 5 μm), mobile phase of methanol-water-acetic acid-triethylamine (89∶11∶0.1∶0.05), detection wavelength of 210 nm, column temperature at 25 ℃, the flow rate of 1.0 mL/min. Results The total contents of oleanolic acid and ursolic acid in the samples prepared through 4 different processing methods were 0.86%, 0.93%, 1.29%, and 1.47%, respectively, showing that different processing methods in producing area had a significant effect on contents of oleanolic acid and ursolic acid in chaenomeles medicinal materials. Conclusion This study provides a basis for processing methods of chaenomeles medicinal materials in producing area.
5.Principles and skills of designing opening speech in epidemiological courses in the vision of internet

Internct has provided abundant resources for teaching,but it also brings two challenges,information overload and knowledge fragmentation.Under the environment of Internct,only by efficient course designs,can teachers deal with these two new challenges,and attract students' attentions and motivate their learning interests.Among teaching designs,the design of teaching ‘opening speech’ is quite important.During the course construction of Epidemiology in China Medical University,design strategies on opening speech were explored;the principles of opening speech design were determined.And the opening speeches were designed and cumulated for the teaching modules of Epidemiology.Furthermore,the skills of opening speech design were summarized.

This paper analyzed the status of the medical students'volunteer services in China and abroad , dis-cussed the feasibility and necessity of medical students'volunteer service .The influence factors , elvaluation and ef-fectiveness of the medical students'volunteer services were sumed up , and its development tendency was also dis-cussed .The following suggestions were put forward , namely improving the volunteer service participation , making the voluntary service a routine work , aiming to giving references for medical schools and volunteer organizations car-rying out the medical students'volunteer services .
7.Effects of bisoprolol combined with rosuvastatin on endothelial function and inflammation in patients with coronary slow flow

Objective To investigate the effects of bisoprolol combined with rosuvastatin on endothelial function and inflammation in patients with coronary slow flow (CSF). Methods Ninety CSF patients treated from August 2014 to October 2015 were randomly divided into control group, statin group and combined group, thirty cases in each group. The control group was given conventional therapy (aspirin 100 mg/d and isosorbide mononitrate 60 mg/d), statin group was given rosuvastatin 10 mg/d on the basic of control group, while the combined group was given bisoprolol 5 mg/d on the basic therapy of statin group. The serum concentrations of nitric oxide (NO), endothelin-1(ET-1), high-sensitivity c-reactive protein (hs-CRP) and interleukin-6 (IL-6) were detected before treatment and 8 weeks after treatment. The improvement of patients with angina pectoris was evaluated. Results After eight-week treatment, the NO levels were significantly increased in combined group and statin group, while the ET-1, hs-CRP and IL-6 levels were significantly decreased than those before the treatment (P<0.05). At the same time, comparing with the statin group and control group, the NO level was increased in combined group (P<0.05), while the ET-1, hs-CRP, and IL-6 levels decreased significantly (P<0.05). There were significant differences in the effective rates between the combined group (90.0%) and the statin group (83.3%), which were higher than those in control group (56.7%). Conclusion Bisoprolol combined with rosuvastatin can improve the endothelial function and anti-inflammatory in the treatment of CSF.
8.Clinical application value of IGRA for screening Tibetan patients with pulmonary tuberculosis

Objective To compare the sensitivity and specificity of interferon-gamma release assay(IGRA) in the Tibetan and Han patients with pulmonary tuberculosis(TB),and to investigate its clinical application value for screening Tibetan patients with pulmonary TB.Methods Sixty-four Tibetan cases of pulmonary TB,41 Tibetan cases of non-pulmonary TB,40 Han cases of pulmonary TB and 38 Han cases of non-pulmonary-TB in our hospital from April 2013 to April 2015 were selected and performed IGRA.T cells infected by mycobacterium tuberculosis were detected by ELISA.The difference between Tibetan abd Han nationalities was compared.The clinical value of IGRAfor screening Tibetan patients with pulmonary TB was evaluated by calculating the statistical indicators.Results The sensitivity,specificity,positive predictive value and negative predictive value of IGRA for diagnosing Tibetan pulmonary TB were 90.62%(58/64),80.49%(33/41),87.88%(58/66) and 84.62%(33/39) respectively,which for diagnosing Han patients with pulmonary TB were 92.50%(37/40),84.21%(32/38),86.05%(37/43) and 91.43%(32/35) respectively.The sensitivity and specificity of IGRA for diagnosing Tibetan and Han pulmonary TB had no statistical difference(χ2=0.005,P=0.944;χ2=0.018,P=0.893).Conclusion IGRA has high value for early diagnosis of Tibetan pulmonaty TB and is conducive to screening of early pulmonary TB.
9.Using random allocation book for random allocation concealment in a clinical trial

Objective Random allocation concealment is important in ensuring high-guality randomized controlled clinical trial.In this paper , we aimed to design a more convenient and effective two couplet random allocation method through improving the random allocation concealment technology which uses the envelope method . Methods According to the randomized sequence , a two-couplet random allocation should be prepared for every subject and bound into a book in order , which named random assignment book with cover and instructions .The first couplet is to collect the enroll information and the second couplet is to show the allocation informa -tion.The serial numbers of the two couplets which around sealant are the same , and leave blank at the same areas of the two couplets for signing the enter information .The content of the first couplet can be completely copied to the second couplet .In order to avoid expo-sing the allocation information in advance , the back of the second couplet should be black-printed. When the subjects are sure qualified, the researchers selected the corresponding two couplet in a sequential order .Exposing the allocation infromation in the second couplet, and the subjects will be allocated to the group designated on the second couplet .This random allocation book should be entrusted to professional printing mechanism ,printing according to the random assignment sequence table and in the strict quality control .For the multicenter clinical trials , it can be printed in parts . Results The method has been implemented in nearly ten randomized controlled trials and obtained the consistent high praise . Conclusion The random allocation book method is easy to produce , simple operation and convenient in monitoring , which can effectively achieve the random allocation concealment in a clinical trial as well as having a sig -nificantly practical value in ensuring the quality of randomization .
10.Single nucleotide polymorphism of estrogen metabolizing genes CYP17、CYP19 and breast cancer susceptibility

Objective To investigate the relationship between single nucleotide polymorphism in estrogen metabolizing genes CYP17、CYP19 and breast cancer susceptibility.Methods A case-control study was performed.PCR-base restriction fragment length polymorphism(PCR-RFLP)and short tandem repeat polymorphism(STRP)assay were used to detect the single nucleotide polymorphism of CYP17、CYP19 in 213 breast cancer cases and 430 matched controls.Resuits CYP17 A2/A2 genotype was found in 6.7%of breast cancer cases,which was significantly higher(P<0.05)than that in controls(2.4%);the frequency of A2 allele of CYP17 was 16.2%in breast cancer cases,which was significantly higher(P<0.05)than that in controls(10.6%).There Was alSO significant difference in the frequency of(TTTA)10allele of CYP19 which was 12.4%in breast cancer cases and 8.2%in controls(P=0.02).Conclusions The allele of CYP17 A2 and CYP19(TTTA)10 and CYP17 A2/A2 genotype were positively associated with the susceptibility of breast cancer.
