1.Influence on Nasal Sinuses Mucosa Epithelium GR and IκBα Expression for Chronic Rhinosinusitis Model Treated with Bi-Yuan-Shu Oral Liquid
World Science and Technology-Modernization of Traditional Chinese Medicine 2014;(1):98-102
This study was aimed to investigate the influence of Bi-Y uan-Shu (BYS) Oral Liquid on glucocorticoid re-ceptor (GR) and nuclear factor IκBα expression of nasal sinuses mucosa epithelium among chronic rhinosinusitis (CRS) models in order to explore its therapeutic mechanism for CRS from the anti-inflammatory reaction aspect. One hundred New Zealand rabbits were selected and randomly divided into the normal group, sham operation group, model group, BYS group, and clarithromycin group, with 20 rabbits in each group. After the CRS model was established, no intervention was given to the normal group, sham operation group, or model group. The intragastric administrations of BYS (1.5 mL·kg-1·d-1) and clarithromycin (25 mg·kg-1·d-1) were given for 14 days, respectively. The nasal sinuses mucosa was taken after the treatment. And HE stain was used to observe its pathological changes. Western Blotting was used in the detection of nasal sinuses mucosal epithelium cytoplasm GR and IκBα expression. The results showed that there were obvious nasal sinuses mucosa inflammatory cell infiltration, chronic inflammation changes, and obvious hyperplasia of glandular organs and goblet cells. Compared with the normal group, the GR expression was obviously reduced (P< 0.01). And the IκBα expression was obviously increased (P< 0.01). After the intragastric administration of BYS, the nasal sinuses mucosal epithelium was repaired with no obvious inflammatory cell infiltration, glandular organs, or goblet cells. Compared with the model group, the GR expression was obviously increased (P< 0.01). The IκBα expression was obviously decreased (P<0.01). It was concluded that BYS can promote GR expression to inhibit inflammation. Meanwhile, it can restrain IκBα expression to prevent the over inhabitation of IκBα on NF-κB. It can dynamically regulate the balance of the nasal sinuses mucosa epithelium inflammation.

Objective To study the mechanism of paraquart-induced renal injury in rats, the expression of heine oxygenase-2(HO-1) and inducible nitric oxide synthase (iNOS).Methods 84 adult healthy Spragne-Dawley (SD) rats (female and male in half) were randomly divided into two groups, the control group (group A,n=42) and the paraquart group (group B,n=42).The rats in group B were treated with paraquart (25 mg/kg) intraperitoneally, the rats in group A were treated with the same dose of normal saline. Its histopathological change, the expression of HO-1,iNOS and evaluate the mRNA expression of HO-1 were observed. Results ①In group A,the tissue structure was clear without abnormal. In group B,there were obvious lesions in the renal tubule of cortical part, including cellular swelling, the canula narrow, the mesenchymal congestion and edema. These pathologic changes gradually reached the peak on the 1st day,and did not relieved till the end ofthis study.②In group A,there was no or weak expression of iNOS.At the 3rd hour, the expressions of iNOS in group B were observed in the kytoplasm of renal tubular epithelial cell and glomemlar endothelial cell of cortical part.It reached the peak on the 1st day and continued at a high level after that.③ In group A,there was no or weak expression of HO-1 and HO-1 mRNA.At the 3rd hour, the expressions of HO-1 in group B were observed in the membrane and kytoplasm of renal tubular epithehal cell of cortical part, immunohistochemistry score(IHS) was higher, the expression of HO-1 mRNA increased, there was a statistical significance between group B and group A(P<0.05).It reached the peak on the 1st day,and then decreased, there were 110 statistical difference between group B and group A on the 5th day(P>0.05).Conclusions HO-1 and iNOS ale involved in the procedures of renal injury, but its mode of action and regulating path still need further study.

Objective To observe the influence of ascites in severe acute pancreatitis (SAP) on the function and viability of peritoneal macrophages in order to investigate the role of peritoneal macrophages in pathophysiological alteration and secondary pancreatic infection in SAP. Methods After the ascites of SAP models treated peritoneal macrophages for 1,3,6,12 and 24 hours in vitro, neutral red phagocytosis, cell viability and TNF secretion of peritoneal macrophages were determined respectively. Results The phagocytosis, cell viability and TNF secretion of macrophage all decreased with the treating time prolonged in the tests. Conclusions The ascites of SAP decreased the phagocytosis, viability and TNF secretion of peritoneal macrophages, and was one of the factors to promote secondary pancreatic and peripancreatic infection as well as bacterial translocation of gut.

Objective To construct a bait vector containing human glucocorticoid receptor(GR) ligand binding domain (LBD) in yeast two hybrid system in order to screen its cDNA library Methods PCR was used to amplify GR LBD fragment from the fetal liver cDNA library with the primers designed in accordance with the sequence in GenBank GR LBD The product was inserted into pGEM(r) T Vector Systems After verified with restriction endonuclease digestion of SmaⅠ/SalⅡ, the vector was inserted into the "bait plasmid" pGBKT7 (named as pGBKT7 GR LBD) After confirmation with restricted endonuclease digestion and sequence analysis, the plasmid was transformed into the yeast cell AH109, and the transformants were selected on SD/ Trp plates The interaction between GR and SRC 1 was tested by ? gal activity with yeast two hybrid system Results The amplified product of 893 bp was inserted into pGEM(r) T Vector and proven to be successful with double restriction enzyme digestion Sequence analysis revealed that the sequence was correctly inserted into pGBKT7 with a right reading frame AH109 [pGBKT7 GR LBD] grew on SD/ Trp plates, but not on the other selective media GR could interact with SRC 1 Conclusion The bait plasmid pGBKT7 GR LBD constructed expresses GR LBD correctly, and can't activate the transcription of reporter gene alone in yeast two hybrid system
