1.Isolation of algicolous fungi and screening of their antitumor activity

Objective To isolate algicolous fungi from algae and obtain antitumor strains.Methods Selective isolation of algicolous fungi from algae was done,and antitumor activity corresponds to their lethiferous effect on brine shrimp and cytotoxicity against K562 cell was determined.Results 221 strains of algicolous fungi were isolated from 28 algae samples collected from intertidal zones of the Yellow Sea.Eight strains showed lethiferous activity and eight showed cytotoxicity.Conclusion The results indicated that algicolous fungi would be one of the important resources to search for new antitumor compounds.
2.Novel cationic liposome loading siRNA inhibits the expression of hepatitis B virus HBx gene.

In order to solve the problem of selection and in vivo delivery problem in siRNA treatment, hepatitis B virus (HBV) HBx gene which could be targeted by siRNA was studied. The siRNA expression plasmid which specific inhibits HBx expression was obtained by in vitro selection via a dual-luciferase plasmid including HBx-Fluc fusion protein expression domain. The selected siRNA expression plasmid was then encapsulated in PEG-modified cationic liposome, which was devoted into pharmacodynamic studies at both cellular and animal level. The results illustrated that the cationic liposome which encapsulated siRNA expression plasmid could effectively inhibit HBx gene expression both in vitro and in vivo.
3.Study on the Quality Analysis Method of Schizonepeta Spike Based on Multiple Feature Components"Quality-quantity"Double Standard

Objective To establish a"quality-quantity"double standard control method of Schizonepeta Spike based on the reference material without relying on multiple reference substances.Methods HPLC technology was used to establish the characteristic atlas of Schizonepeta Spike based on the reference traditional Chinese medicinal materials.The chemical constituents of common peaks were identified by Q-TOF-MS technology,and the authenticity of Schizonepeta Spike was determined by the similarity of characteristic peaks of the reference traditional Chinese medicinal materials and the tested traditional Chinese medicinal materials.The relative content of each characteristic peak in different batches of the tested traditional Chinese medicinal materials was calculated by accurate quantition of pulegone.The"Average value-Standard deviation"was taken as the lower limit of the relative content of characteristic peak chemical components relative to pulegone,and the quality of Schizonepeta Spike was determined according to the lower limit of the relative content of characteristic peak.Results The characteristic atlas and content determination method met the requirements of methodology investigation.A total of 11 common chromatographic peaks were determined,and 6 chemical components were identified,which were luteolin,hesperidin,luteolin-7-O-glucuronide,luteolin,geranyl,pulegone.The similarity between the tested traditional Chinese medicinal materials and reference traditional Chinese medicinal materials was greater than 0.90.The lower limit of relative content of chemical components of Schizonepeta Spike characteristic peak was defined.Conclusion The method can identify Schizonepeta Spike clearly and quickly without many reference substances,and provide reference for quality control of Schizonepeta Spike.
4.Inhibitory effect of berberine on a nude mouse model of cutaneous squamous cell carcinoma and its mechanisms

Objective:To evaluate the effect of berberine on the volume and weight of, cell proliferation and apoptosis in transplanted cutaneous squamous cell carcinoma (cSCC) in nude mice inoculated with A431 cells, and to explore the possible mechanism underlying the inhibitory effect of berberine on cSCC.Methods:A431 cells were cultured, and A431 cell suspension was subcutaneously injected into the back of 20 nude mice to establish a nude mouse model of transplanted cSCC. On day 15 after inoculation, these tumor-bearing mice were randomly and equally divided into 4 groups: low-, medium- and high-dose berberine groups intraperitoneally injected with 10, 20 and 25 mg/kg berberine solution respectively, and control group intraperitoneally injected with sodium chloride physiological solution. The treatment was performed once a day for 35 consecutive days. The tumor volume was measured before, and on days 7, 14, 21, 28 and 35 after the start of treatment. After the end of the experiment, the nude mice were sacrificed, and the tumors were removed and weighed to calculate the tumor growth inhibition rate. Histopathological examination was performed in these transplanted tumors, immunohistochemical study was conducted to determine the expression of Ki67, TUNNEL staining was conducted to determine the number of apoptotic cells in the transplanted tumor tissues, fluorescence-based quantitative PCR and Western blot analysis were employed to determine the mRNA and protein expression of apoptosis-related proteins Bax, Bcl-2, caspase-3 and Ezrin respectively. One-way analysis of variance was used for comparisons among multiple groups, and Dunnett- t test for comparisons of each berberine group with the control group. Results:Along with the increase in the dose of berberine and treatment duration, the tumor growth curve gradually became flat, the tumor growth was inhibited to different degrees in the berberine groups, and the high-dose berberine showed the strongest inhibitory effect on the tumor growth. The tumor growth inhibition rate was 31.05%, 66.68%, 76.49% in the low-, medium-, and high-dose berberine groups respectively, and the tumor weight was significantly lower in the 3 berberine groups than in the control group ( t = 4.07, 6.33, 7.26, respectively, all P < 0.01) . Along with the increase in the dose of berberine, histopathological examination of the transplanted tumors showed that the extent and area of tumor cell necrosis increased, while immunohistochemical study showed that the number of Ki67-positive cells gradually decreased. Moreover, the number of Ki67-positive cells was significantly lower in the low-, medium- and high-dose berberine groups than in the control group (all P < 0.01) , but the number of apoptotic cells was significantly higher in the berberine groups than in the control group ( P < 0.05 or < 0.01) . The mRNA and protein expression of Bax, Bcl-2, caspase-3 and Ezrin significantly differed among the 4 groups (all P < 0.01) . In addition, the mRNA expression of Bcl-2 was significantly lower in the low-, medium- and high-dose berberine groups than in the control group (all P < 0.01) , and the protein expression of Bcl-2 was significantly lower in the medium- and high-dose berberine groups than in the control group (both P < 0.01) ; Bax mRNA expression in the high-dose berberine group and caspase-3 mRNA expression in the medium- and high-dose berberine groups were significantly higher than the corresponding mRNAs in the control group respectively ( P < 0.05 or < 0.01) , and the protein expression of Bax and caspase-3 was significantly higher in the low-, medium- and high-dose berberine groups than in the control group (all P < 0.01) ; Ezrin mRNA expression was significantly higher in the high-dose berberine group than in the control group ( P < 0.01) , but its protein expression was significantly lower in the low-, medium-, and high-dose berberine groups than in the control group (all P < 0.01) . Conclusion:Berberine can inhibit the proliferation of A431 cells and promote their apoptosis in the transplanted cSCC of the nude mice, and then suppress the growth of transplanted cSCC in the nude mice to a certain extent, which may be related to the upregulation of Bax and caspase-3 and downregulation of Bcl-2 and Ezrin.
5.Status and analysis of the capabilities of the provincial level radiological health technical institutions in China from 2014 to 2018

Objective To fully grasp the professional technical capabilities of provincial level radiological health technical institutions, and so as to provide the basis for giving full play to the role of provincial level radiological health technical institutions, exploring and planning the development direction of radiological health technical institutions in China. Methods From September to December, 2019, National Institute for Radiological Protection, China CDC issued questionnaires to 37 provincial level radiological health technical institutions. The basic situation of department setting, number and composition of professional and technical personnel, equipment and radiological health work of the institutions that undertake radiological health related responsibilities were investigated, summarized and analyzed. Results Thirty-seven provincial level institutions participated in this survey, including 24 centers for Disease Control and Prevention, 11 institutions for Occupational Disease Control and Prevention, and 2 other institutions related to radiological health.Most of the institutions were qualified for radiological health related testing or evaluation; Thirty institutions had set up independent radiological health departments. There were 627 people engaged in radiological health professional and technical work, 83.4% of whom were officially on the staffs, and 16.6% are contract staffs.The distribution of gender, age, qualifications, professional title and working years in the radiological health was relatively reasonable, but only 25.8% of the staffs had the radiological education background. From 2014 to 2018, the number of radiological health professional and technical personals inflows and outflows was 164 and 65, respectively. Each institution undertakes 14.6 radiological health responsibilities in average. The annual workload of provincial level institutions was large, but there were relatively few laboratories and instruments for radionuclide measurement and internal radiation measurement. Conclusion The provincial level radiological health technical institutions still need to strengthen the capability building and make up the short board, play the leading and guiding role in provincial regions, so as to promote the further development of the national radiological health work.

7.Comparative analysis on radiological health technical services in China in 2021 and 2022

ObjectiveTo analyze the quality and status of technical service items of radiation health technical service institutions (RHTSI) in China. Methods A total of 608 and 622 RHTSIs with radiation health technical service qualifications from 31 provinces, autonomous regions, and municipalities in the years of 2021 and 2022 were selected as the research subjects. The data of quality monitoring of radiological health technology services, comparison of radiological health testing capabilities, and investigation of the current status of technical support institutions were collected to analyze the status of technical service items which was conducted by RHTSIs from 2021 to 2022. Results A total of 622 RHTSIs in China obtained technical service institution qualifications in 2022, an increase of 14 from 608 in 2021. While a total of 404 of 622 RHTSIs conducted radiological health technology services, a decrease of 11.4% from 456 in 2021. A total of 241 804 technical service items were conducted in 2022, an increase of 39.7% from 173 064 in 2021. The median number of technical service items by non-health system RHTSIs was higher than that of the health system RHTSIs in 2021 and 2022, (203 vs 40 items, 215 vs 55 items, all P<0.01). The number of technical service institutions and technical service items conducted in different regions from high to low were the eastern, central, and western regions in 2021. The number of technical service institutions in different regions was highest in the eastern region, followed by the central and western regions, while the number of technical service items from high to low were the eastern, western, and central regions in 2022. The number of provincial, municipal, and county-level health system RHTSIs decreased by 6.5%, 26.3%, and 27.3%, respectively, in 2022 compared with 2021. The number of technical service items conducted by provincial health system RHTSIs increased by 48.6%, while those conducted by municipal and county-level health system RHTSIs decreased by 13.8% and 21.3%, respectively. Conclusion Although the number of RHTSI conducting technical services decreases in 2022 compared with 2021, the number of technical service items conducted increases. Non-medical RHTSI undertake the majority of technical service items. Within the medical institutes, the number of technical service items conducted by provincial RHTSI is higher than that of municipal and county-level RHTSI.

In this paper, the notice of "Opinions of the State Administration for Market Regulation on further promoting the reform of qualification accreditation of inspection and testing institutions" (No. 206 [2019] which issued on October 25, 2019 has aroused our reflection to concern the development trend of qualification accreditation of radiological health technical service institutions. In order to adapt to the changes under the new situation and avoid the risks of technical review that may be faced in the qualification accreditation of radiation health technical service institutions in the future and to discussed the ways that radiological health technical service institutions can be used to confirm the testing ability in the qualification certification process.
9.Research progress on the intervention of traditional Chinese medicine in renal interstitial fibrosis based on PI3K/Akt signaling pathway

Renal interstitial fibrosis(RIF) is the main pathological manifestation of chronic kidney disease. Due to the complexity of the mechanism, there is no specific treatment for RIF in clinical practice. The abnormal activation of the phosphatidylinositol 3-kinase (PI3K)/protein kinase B(Akt) signaling pathway and the activation of downstream target genes are key drivers of RIF induction and progression. Traditional Chinese medicine has the characteristics of precise efficacy and minimal toxic side effects, and the occurrence and development of RIF can be regulated by multiple targets and mutual coordination. This review focuses on the PI3K/Akt signaling pathway and summarizes the potential targets and regulatory mechanisms of traditional Chinese medicine in the treatment of RIF. It is found that various effective ingredients (such as sinomenine, mangiferin, coumarin derivates from Hydrangea paniculata, etc.) and formulas (such as Fushengong decoction, Qi-Bang-Yi-Shen formula, etc.) of traditional Chinese medicine can inhibit fibroblast proliferation, improve inflammation and oxidative stress, maintain mitochondrial stability, and slow down ferroptosis through this pathway, thereby delaying the occurrence and progression of RIF.
10.Investigation and analysis of the current situation of radiation health technical services in China

Objective To investigate the current situation of radiological health technical service institutions and their technical services in China, and to provide a basis for better utilizing radiological health technical service resources and strengthening institutions’ capability building. Methods From October to December 2021, we conducted quality monitoring, ability comparison, capability building surveys, and other investigations on radiation health technical services to collect information on the qualifications and technical services of radiation health service institutions. The data were pooled and analyzed using SPSS software. Results By the end of 2021, there were 608 radiological health technical service institutions across China, with 47.0% of them located in the eastern region, and the percentage of institutions in the health system (42.6%) was less than that in the non-health system (57.4%). Institutions of grade A offered 62.3% of technical services, while institutions of grade B provided 37.7% of technical services; technical services provided by the non-health system and the health system accounted for 84.2% and 15.8%, respectively; institutions in the health system at the province, prefecture, and county levels offered 37.1%, 50.6%, and 12.3% of technical services, respectively. Conclusion In recent years, the proportion of institutions in the non-health system has increased significantly, but the proportion of institutions in the health system with radiological health qualifications is small, and the development is uneven in the numbers of institutions, technical services, and professional and technical personnel in the eastern, central, and western regions and at the province, prefecture, and county levels in China. It is necessary to further encourage and support health system institutions in strengthening capability building and applying for radiological health qualifications, and to increase construction and investment in the western region and at the district/county levels, in order to improve the overall capability and level of radiological health technical services in the country.
