Objective To investigate the effects of Tianshu capsule(TSC) on the plasma levels of ?-endorphin(?-EP) and 5-Hydroxytryptamine(5-HT) and c-fos expression of brain tissue in the rat with migraine.Methods The rat migraine model was established by subcutaneously administration of nitroglycerin.The intragastric administration dose of TSC was divided into three groups as follows:high dose(7.5 g/kg),middle dose(3.75 g/kg) and low dose(1.88 g/kg).TSC was administrated respectively in therapeutic and prophylactic model.The control group and model group were administrated corresponding dose of normal saline.The plasma levels of ?-EP and 5-HT in rat were measured by radioimmunoassay and HPLC respectively;the distribution of ?-EP,5-HT and c-fos in rats periaqueductal grey were evaluated by immunohistochemistry.Results Compared to model group,the plasma levels of ?-EP were increased remarkably in high dose TSC therapeutic and prophylactic groups,middle dose prophylactic group(all P

Objective To investigate the effects of Tianshu capsule on vasoactive substances in blood plasma or brain and hemodynamics in migraine animal models. Methods After subcutaneously administration of nitroglycerin (NTG) and tube feeding Tianshu capsule, the radioimmunoassay or spectrophotometry was used to measure the plasma contents of calcitonin gene related peptide (CGRP), nitric oxide (NO) and nitric oxide synthase (NOS) in rats.The distribution of NOS1, CGRP in rat nucleus tratus spinalis nervi trigemini was detected by immunohistostaining, and the changes of internal carotid artery flow velocity in rabbit were measured by transcranial Doppler.Results The levels of NO, NOS and CGRP were elevated in the plasma of migraine rats. Aim To investigate the protective effects of on focal cerebral ischemic-reperfusion(I/R)injury in rats.Methods The model of rat middle cerebral artery occlusion(MCAO)was induced to observe the change of praxiology of rats,infarction percentage,water content,histology of the rat brains.The levels of superoxide dismutase(SOD),malondialdehyde(MDA),reduced glutathione hormone(GSH),Na+,K+-ATPase,Ca2+-ATPase,nitric oxide(NO)were also measured.Results Salidroside at different dosages(24,12,6 mg?kg-1)could obviously decrease cerebral function score,cerebral infarct size and water content,and repair pathological injury of focal cerebral I/R in rats.MDA,LD,NO contents in brain tissue were significantly decreased and activities of GSH,SOD were significantly improved.Na+,K+-ATPase,Ca2+-ATPase at dosages of 24,12 mg?kg-1 were also significantly improved.Conclusion Salidroside had protective effects on injuries of cerebral I/R.
5.Constituents in Desmodium blandum and their antitumor activity

Objective To investigate the chemical constituents in Desmodium blandum and their antitumor bioactivity.Methods Various chromatographic techniques were employed for the isolation and purification of the constituents by butanol at 50 ℃ including silica gel, Sephadex LH-20.The compounds structures were elucidated by spectral analyses(IR,UV,NMR,and MS).Then their cytotoxic activity was studied.Results They were identified as N,N-dimethyltryptamine (Ⅰ),5-methoxy-N,N-dimethyltryptamine(Ⅱ),citrusinol(Ⅲ),yukovanol(Ⅳ),(Z)-1-(4-hydroxy-2,3-dimethoxyphenyl)-3-(4-hydroxyphenyl) propene(Ⅴ),(Z)-1-(3-hydroxy-2,4-dimethoxy phenyl)-3-(4-hydroxy-3-methoxy-phenyl) propene(Ⅵ),methyl protocatechuate(Ⅶ),katuranin(Ⅷ).Conclusion Eight compounds are isolated from the stems of D.blandum for the first time.In the MTT antitumor experiments,compounds Ⅰ and Ⅲ—Ⅵ have the cytotoxic activity to KB cell.

This paper has analysed the particularity of practice and teaching of oral and maxillofacial surgery.Binding with the change of medical pattern in recent years,the change of relation of doctors and patients as well as the present of "the People's Republic of China Practical Doctor Law"and "Rules of Sitling Malpractice",it has discussed that we should simultaneously think highly of raising to practice ability, reinforcing education of medical ethics, emphasizing legal consciousness and deepening ethics idea during practice teaching of oral and maxillofacial surgery,which can ensure well development of interns.
7.RNA interference inhibiting permeability glycoprotein over-expression in pharmaco-resistance rat astrocytes model

Objective To study the effect of adenoviral-delivered short hairpin RNA (shRNA) target against permeability glycoprotein (Pgp) as a new drug in anti-epileptic drug resistance epilepsy treatment and to evaluate its efficiency. Methods MDR Sprague-Dawley (SD) rat estrocyte model was induced by Coriaria Lactone (CL), mainly over-expressing mdrlb. To reverse the drug resistance, astrecytes were treated with constructed replication deficient adencvirus AdS-EGFP-shRNAI-U6 delivering short hairpin (shRNA) target agianst mdrlb gene. Total RNA and protein were extracted from the infected cells, mdr1 b level was detected by Quantitative Real-time PCR whereas Pgp by Western blot, Rhodamine123 (Rho123) efflux ratio by Flow Cytometry. Results AdS-EGFP-shRNA1-U6 was succesfully constucted with high virus titer of 6×1010 pfu/ml. The interference efficency of AdS-EGFP-shRNA1-U6 agianst mdrlb in rat astrecyte model was about 94%. The Rho123 efllux ratio was about 15. 8%, significiently lower than control group which was 56. 2% (F = 127.5, P < 0. 05). Conclusions Pgp over-expression has been successfully suppressed and MDR has been reversed, which may provide a premising approach for refractory epilepsy remedy.
8.Effect of Nimodipine Liposomes for Injection on Focal Cerebral Ischemia/Reperfusion Injury in Rats

Objective:To investigate the effect of nimodipine liposomes for injection(NOLI)on focal cerebral ischemia/reperfusion(I/R)injury in rats.Methods:Seventy SD rats were divided into NDLI 1.00 mg/kg,NDLI 0.50 mg/kg,NDLI 0.25 mg/kg,nimodipine 1.00mg/kg,solvent 10 mL/kg,sham-operation and ischemic model groups.The model of middle cerelral artery occlusion in rat was replicated.The behavioral scores in rats were assessed in all groups.The infarct volume,brain water content,biochemical indices of brain homogenate and histology were detected.Results:1he NDLI 1.00mg/kg,0.50 mg/kg and 0.25 mg/kg groups could significantly improve the behavior scores in focal cerebral ischemic rats,reduce the volume of cerebral infarction,decrease the brain water content,improve the activities of Na+,K+-ATPase,Ca2+-ATPase,glutathione(GSH)and superoxide dismutazse(SOD)in brain tissues,reduce conteras of malondialdehyde(MDA),lactic acid(LA)and nitric oxide(NO),and improve histo logical injury.Conclusions:NDLI has the protective effect on focal cerebral ischemia/reperfusion injury in rats.
9.Experimental study on the spatial distribution of X-ray in the X-ray room and the rational use of the results

Objective To study the radiation dose distribution in the X-ray room,and provide the strategy of radiation protection for the medical staff and the patient’s nursing who had to enter the room while the X-ray was exposing.Methods The thermolumi-nescent dosemeters(TLDs)was placed around the center of the X-ray tube with the same level of the bed.Then,exposure parame-ters,including the X-ray tube voltage value and the field of view,were changed for different groups while exposing.All of the TLDs were taken back to the lab for analysis.Results The differences between the two groups which had the same distance in different di-rections were statistically significant (P <0.01).With the same radiographic condition and direction,the radiation dose on the site of 10 cm from X-ray tube center was the maximum,while the site of 120 cm was the minimum.With the same radiographic condition and distance,the radiation dose on the anode side of the X-ray tube in the room was relative lower,while the site behind the X-ray tube was relative higher.With the same voltage value,distance and direction,the same sites that had the smaller FOV(34 cm×34 cm) received lower radiation dose than those with larger FOV(52.6 cm× 52.6 cm).Meanwhile,the sites with the voltage of 70 kV re-ceived the lower radiation dose than that with the voltage value of 120 kV.Conclusion In the X-ray room,the medical staff and the patient’s nursing can choose the area on the right side(anode side),keep far away from the X-ray tube center,avoid the rear of the X-ray tube and the cathodic direction of the X-ray tube to reduce the radiation dose.
10.Screening of proteins interacting with RSK4 in breast cancer by affinity purification and liquid chromatography/mass spectrometry

Background and purpose:As a tumor suppressor gene, ribosomal S6 kinase 4 (RSK4) plays important roles in inhibiting cell proliferation, migration and inducing cell apoptosis. However, the proteins interacting with RSK4 are still unknown. This study aimed to screen proteins interacting with RSK4 in breast cancer cell line MDA-MB-231 by lfag-tag affnity puriifcation and LC-MS/MS (liquid chromatography/mass spectrometry).Methods:The pcDNA3.1/EGFP-RSK4-Flag eukaryotic expression vector was constructed by inserting full lengthRSK4 gene into vector pcDNA3.1/EGFP-Flag. And then the recombinant plasmids were transferred into MDA-MB-231 cells. Real-timelfuorescent quantitative polymerase chain reaction (RTFQ-PCR) and Western blot were used to detect the expression of RSK4 in MDA-MB-231 cells. Affnity puriifcation and LC-MS/MS were applied to screen proteins interacting with RSK4, and the related action mechanism of RSK4 with its interacted proteins was detected based on bioinformatics gene ontology (GO) and ingenuity pathway analysis (IPA).Results:Twenty-four proteins, such as serine/threonine-pro-tein kinase 38 (STK38)/serine/threonine-protein kinase 38-like (STK38L), MOB kinase activator 2 (MOB2) and protein arginineN-methyltransferase 5 (PRMT5), were successfully identiifed by Flag-tag affnity puriifcation followed by LC-MS/MS analysis, which probably interacted with RSK4. Bioinformatics analysis of the identiifed proteins suggested the proteins interacting with RSK4 were involved in diverse biological pathways, such as apoptosis and cell migration. Conclusion:According to bioinformatics results of proteins interacting with RSK4 identiifed by affnity puriifcation and LC-MS/MS, biological networks of RSK4 are involved in apoptosis and migration in breast cancer cells.
