1.Role of PPARα/PGC-1αin doxorubicin induced mouse dilated cardio-myopathy

[ ABSTRACT] AIM:To investigate the changes of peroxisome proliferator-activated receptors ( PPAR)α/peroxi-some proliferator activated receptor coactivator 1 alpha ( PGC-1α) in doxorubicin ( DOX) induced dilated cardiomyopathy ( DCM) and its effect on the energy metabolism and myocardial function in mice .METHODS:Forty mice were randomly divided into 4 groups:control group, DOX group, PPARαinhibitor group and PPARαagonist group.The DCM model was established by injection of DOX.The protein levels of PPARα/PGC-1αwere detected.The PPARαinhibitor and PPARαagonist were used 2 weeks beforeinjection of DOX.The contents of adenine acid and phosphocreatine ( Pcr) in the mito-chondria were measured by high-performance liquid chromatography ( HPLC) .The ANT activity was analyzed by the atrac-tyloside-inhibitor stop technique.The changes of the echocardiography and hemodynamics were also observed.RESULTS:DOX induced DCM model was successfully established.The protein levels of PPARαand PGC-1αin control group were significantly higher than those in DOX group (P<0.05).Both of the high-energy phosphate contents and the transport ac-tivity of ANT were decreased in DOX group (P<0.05), and the hemodynamic parameters were disordered (P<0.01). Compared with DOX group, PPARαinhibitor pre-treatment significantly reduced the PPARα/PGC-1αexpression.Mean-while, high-energy phosphate contents in the mitochondria and the ANT transport activity of the mitochondria decreased, as well as the left ventricular function ( P<0.05) .On the other hand, PPARαagonist significantly increased the expression of PPARαand PGC-1α, and improved the transport activity of ANT.In addition, the hemodynamic parameters were amel-iorated, but the high-energy phosphate contents of the mitochondria did not significantly change.CONCLUSION:PPARα/PGC-1αplays an important role in the regulation of ANT transport activity in dilated cardiomyopathy induced by DOX, and the activation of PPARα/PGC-1αhas protective effects on the DCM induced by DOX.
2.Investigation and analysis of an extensive skin injury to the back caused by accidental irradiation in interventional procedure

Objective:To carry out investigation and analysis of an extensive skin radiation injury to the back accidentally caused by interventional procedure and to explore the problems faced in the event with emphasis on avoiding the reoccurance of similar events in the future.Methods:The data were collected by consulting the patient′s detailed medical history, collecting and analyzing clinical diagnosis and treatment data, tracking and observing their clinical manifestations and signs. The patient′s peripheral blood samples were also collected, together with the biological dose estimated and the equipment data collected on the site of the interventional treatment hospital.Results:The whole body dose to the patient was estimated to be 0.95 Gy. The typical values of kerma rate of radiation incident on the body surface due to fluoroscopic procedures were 373.5 mGy/min in subtraction modality and 47.8 mGy/min in fluoroscopy modality, respectively. The annual effective dose to the interventional radiologist was 20.51 mSv due to his operation in long-time radiation exposure conditions, higher than 3.09 mSv for other interventional radiologists with similar workload in the same department. The whole body and local clinical manifestations of the patients were in line with radiation injury. No clear diagnosis has been obtained in several hospitals, nor can obvious treatment outcomes be obsevered.Conclusion:Combined with the biological dose estimation result and clinical manifestations, the case was diagnosed as degree Ⅳ skin radiation injury. Radiation injury is closely related to whether the operation is conducted according to the standard and the output dose of X-ray machine. Non-specialized hospitals should strengthen clinical diagnosis and treatment of radiation injury.
3.Analysis of the eye lens dose and annual effective dose to some interventional radiation workers in Xinxiang city

Objective:To analyze the eye lens dose and annual effective dose to interventional radiation workers in some hospitals of Xinxiang city from 2020 to 2022, and to ascertain the dose to interventional radiation workers.Methods:By using TLDs, the eye lens dose Hp(3) and annual effective dose Hp(10) were monitored for three consecutive years in six hospitals in Xinxiang city. The lens doses and annual effective doses to intervention radiation workers in different years in different-level hospitals and departments were analyzed. Results:From 2020 to 2022, a total of 117 people were monitored. The left eye lens dose range was 0.12-164.24 mSv, and the right eye lens dose range was 0.07-51.64 mSv. The average annual dose was 8.56 mSv for left eye lens and 4.49 mSv for right eye lens The average annual dose distribution in the MDL-5 mSv range for the left and right eye lens was 60.68% and 73.50%, respectively. 9.41% (11 people) of the left eye lens doses exceeded 20 mSv. The annual effective doses range was 0.11-31.27 mSv, with average annual dose of 2.56 mSv. The proportion of average annual effective doses mainly distributed in the range of MDL to 1.25 mSv was 52.14%, with 2.56% annual effective dose exceeding 20 mSv. There was no significant difference in left and right eye lens dose and annual effective dose between the tertiary hospitals and the secondary hospitals in three years ( P>0.05). Compared with different departments, the cumulative per capita dose in three years was statistically significant (left eye H=11.42, right eye H=13.72, annual effective dose H=25.94, P<0.05). The lens dose and annual effective dose in neurology department were lower than those in cardiology department and comprehensive intervention department ( Zcardiology department=-3.33, -3.78, -4.83, P<0.05; Zcomprehensive intervention department=-2.71, -2.63, -4.39, P<0.05). Conclusions:Most of the annual equivalent dose and annual effective dose to eye lens of the interventional radiation workers in Xinxiang city meet the national limits, but some of them have higher doses and exceed the national limits. It is suggested that the routine and continuous monitoring of eye lens doses to interventional radiologists should be strengthened while routine monitoring of annual effective dose, and attention should be paid to the eye lens and annual effective dose to interventional radiologists in secondary hospitals to improve the awareness of protection.
4.Sensitive and Rapid Detection of Giardia lamblia Infection in Pet Dogs using Loop-Mediated Isothermal Amplification.

Giardia lamblia is recognized as one of the most prevalent parasites in dogs. The present study aimed to establish a loop-mediated isothermal amplification (LAMP) assay for rapid and specific detection of G. lamblia from dogs. The fecal samples were collected and prepared for microscopic analysis, and then the genomic DNA was extracted directly from purified cysts. The concentration of DNA samples of G. lamblia were diluted by 10-fold serially ranging from 10(-1) to 10(-5) ng/microl for LAMP and PCR assays. The LAMP assay allows the amplification to be finished within 60 min under isothermal conditions of 63degrees C by employing 6 oligonucleotide primers designed based on G. lamblia elongation factor 1 alpha (EF1alpha) gene sequence. Our tests showed that the specific amplification products were obtained only with G. lamblia, while no amplification products were detected with DNA of other related protozoans. Sensitivity evaluation indicated that the LAMP assay was sensitive 10 times more than PCR. It is concluded that LAMP is a rapid, highly sensitive and specific DNA amplification technique for detection of G. lamblia, which has implications for effective control and prevention of giardiasis.

5.Tanshinone attenuates myocardial injury via activating JAK2/STAT1 pathway in a murine model of viral myocarditis

Objective To explore the effects of tanshinone and JAK2/STAT1 signaling pathway related mechanism in CVB3-induced myocarditis in murine.Methods A total of 110 inbred male Balb/c mice which were 4 to 6 weeks-old were randomly divided into five groups:normal control (N,n =10),myocarditis control (C,n =25),tanshinone group (T,15 mg · kg-1 · d-1,i.p.,n =25),janus kinase 2 inhibitor AG490 group (A,10 mg · kg-1 · d-1,i.p.,n =25),T + A group (H,n =25).Myocarditis was induced by 0.5 ml 10-9.51 TCID50/ml CVB3 i.p.injection for 10 days in group C,T and H.Myocardial histopathologic changes were observed and phospho-STAT1 expressions were detected by immunohistochemistry and Western blot analysis.The levels of serum cardiac troponin Ⅰ were detected with chemiluminescence immunoassay.Results (1) Compared with group C,the histopathologic scores were significantly higher in group A and H (3.35 ±0.57 and 3.34 ±0.54 vs.2.12 ±0.39,P <0.01),but lower in group T (1.40 ± 0.34 vs.2.12 ± 0.39,P < 0.01).(2) The expression of p-STAT1 protein was similar in group A and H compared to group N (P > 0.05),but was significantly lower than that in group C (0.017 ± 0.010 and 0.020 ± 0.010 vs.0.246 ± 0.010,P < 0.01).The expression of p-STAT1 protein was significantly higher in group T than in group C (P < 0.01).(3) The levels of serum cardiac troponin Ⅰ in group C,A,T and H were significantly higher than in group N ((0.42 ±0.06),(1.17 ±0.25),(0.23 ± 0.05) and (1.04 ±0.19) μg/L vs.(0.02 ±0.01) μg/L,all P <0.01).The levels of serum cardiac troponin Ⅰ were significantly higher in group A and H compared with group C ((1.17 ±0.25) and (1.04 ± 0.19)μg/L vs.(0.42 ±0.06)μg/L,P < 0.01),but were significantly lower in group T than in group C ((0.23 ±0.05) μg/L vs.(0.42 ±0.06) μg/L,P <0.01).(4) There was a negative correlation between the expression level of p-STAT1 and the histopathologic scores(y =-4.503 x + 3.371,R2 =0.738,P < 0.01),but a positive correlation between the levels of serum cardiac troponin Ⅰ and the histopathologic scores(y =1.935x + 1.165,R2 =0.766,P < 0.01).Conclusion Tanshinone could attenuate myocardial injury via upregulating the JAK2/STAT1 signaling pathway in this murine viral myocarditis model.
6.Research advances in the prevention and treatment of hepatic ischemia-reperfusion injury with traditional Chinese medicine components

Hepatic ischemia-reperfusion injury (HIRI) is a very common complication of liver transplantation, liver resection, and shock. At present, many studies have been conducted on HIRI, but there is still a lack of drugs for radical treatment in clinical practice. Many factors, such as related cells, molecular mechanisms and signaling pathways, reactive oxygen species and oxidative stress response, nitric oxide, and mitochondria, mediate the development and progression of HIRI, which leads to the decline of patients' quality of life and even endangers their life safety. Based on the pathogenesis of HIRI and related articles, this article summarizes the research advances in the prevention and treatment of HIRI with traditional Chinese medicine components, so as to provide theoretical support for basic research and clinical research on HIRI.
7.Construction and internal validation of a nomogram for predicting the risk of positive prostate biopsy in MRI-negative patients

【Objective】 To establish a nomogram model for predicting the risk of positive prostate biopsy in MRI-negative patients, and to perform the internal validation. 【Methods】 We retrospectively analyzed the clinical data of 197 MRI-negative patients who underwent prostate biopsy at our hospital, analyzed the independent predictors of positive prostate biopsy with univariate and multivariate logistic regression analysis, constructed the nomogram model and conducted internal validation. 【Results】 Multivariate logistic regression analysis showed age (P=0.003), digital rectal examination (DRE)(P=0.005), total prostate-specific antigen (tPSA) (P=0.001) and prostate volume (PV)(P<0.001) were independent risk factors of MRI-negative but prostate biopsy-positive results. The nomogram model based on all variables was established. The area under the receiver operating characteristic (ROC) curve (AUC) was 0.862, which was greater than that of tPSA (AUC=0.739), PV(AUC=0.711) and DRE(AUC=0.666) (all P<0.05). The average absolute error of the model was 1.1% after 500 internal resampling, indicating that the prediction of positive prostate biopsy was consistent with the actual situation. 【Conclusion】 The age, DRE, tPSA and PV were independent predictors of positive prostate biopsy in MRI-negative patients. The nomogram model has a good prediction performance.
8.Relationship between objective sleep quality and cognitive function in remission depression patients with insomnia

ObjectiveTo discuss the relationship between objective sleep quality and cognitive function in remission depression patients with insomnia. MethodsA total of 47 patients with depression in remission in Beijing Huilongguan Hospital were enrolled. Insomnia Severity Index (ISI) and the sleep efficiency calculated by the first four items of Pittsburgh Sleep Quality Index (PSQI) were defined as insomnia. Among the selected patients, 22 subjects were classified into insomnia group, and 25 subjects in non-insomnia group. The objective sleep status of all subjects was evaluated by polysomnography (PSG), and the cognitive function was evaluated with the Repeatable Battery for Assessment of Neuropsychological Status (RBANS). A regression model was established to explore the relationship between objective sleep quality and cognitive function in remission depression patients with insomnia. ResultsThere was statistically significant difference in immediate memory factor scores of RBANS between depression patients in remission with and without insomnia (t=-2.102, P<0.05). The sleep efficiency (Z=-2.113, P<0.05), the proportion of N3 sleep stage (Z=-3.175, P<0.01) and the duration of N3 sleep stage (Z=-3.196, P<0.01) in insomnia group were all lower than those in non-insomnia group, and the nocturnal awakening (Z=-2.058, P<0.01), the proportion of N1 sleep stage (Z=-2.399, P<0.05) and the duration of N1 sleep stage (Z=-2.463, P<0.05) were higher than those of the insomnia group. Within insomnia group, the increased proportion of REM sleep was correlated with the improvement of attention function (β=0.953, 95% CI: 0.123~2.038, P<0.05), and the increase of nocturnal awakening was correlated with the decline of delayed memory (β=-0.173, 95% CI: -0.325~-0.024, P<0.05). ConclusionObjective sleep is associated with cognitive impairment in remission depression patients with insomnia, the proportion of REM sleep has a positive impact on attention function, and frequent nocturnal awakening at night has a negative impact on delayed memory.
9.Comparison of Total Protein Content Difference of Poria cocos from Different Producing Areas

OBJECTIVE:Compare the difference of total protein content of Poria cocos from different producing areas. METHODs:Using bovine serum albumin (BSA) as control ,0.4 mol/L sodium hydroxide solution as exctraction solution , Coomassie brilliant blue G- 250 as chromogenic reagent ,visible spectrophotometry at 595 nm was used to determine the contents of total protein of P. cocos ;cluster analysis was used to classify 34 batches(S1-S34)of P. cocos from different producing areas. RESULTS:The linear range of BSA was 1.45-17.40 μ g/mL (r=0.999 6). RSDs of precision ,stability (20 min) and reproducibility tests were all lower than 3%;recoveries were 100.14%-104.26%(RSD=1.43%,n=9). The contents of total protein in 34 batches of P. cocos from different producing areas were 0.388 4%-1.129 7%. The results of cluster analysis showed that among 34 batches of P. cocos ,the total protein content of P. cocos produced in Yingshan county of Hubei province (S2,S3) was higher than 1%,clustered into one category ;the total protein contents of P. cocos produced in Hubei ,Yunnan,Anhui and Hunan(S1,S5-S10,S12,S13,S16,S17,S19-S21,S23-S25,S28,S30,S31)were 0.653 5%-0.946 1%,clustered into one categony,and the remaining batch content were 0.388 4%-0.601 2%,clustered into one category. CONCLUSIONS :Established method is suitable for the content determination of total protein content of P. cocos . The protein content of P. cocos from Yingshan county of Hubei province is the highest ,followed by Yunnan and Anhui in 34 batches of P. cocos from different producing areas.
10.Study on “Qi-invigorating”Effect and Its Mechanism of Total Saponins of Astragalus membranaceus on Rats with Qi-deficiency

OBJECTIVE:To study “Qi-invigorating”effect and its possible mechanism of total saponins of Astragalus membranaceus on rats with Qi-deficiency ,and to provide reference for elucidating the material basis of “Qi-invigorating”effect of A. membranaceus . METHODS :Forty male Wistar rats were randomly divided into normal group ,model group ,positive control group [Buzhong yiqi pills ,4.5 g/(kg·d)],A. membranaceus total saponins high-dose and low-dose groups [ 252,28 g/(kg·d),by the amount of total saponins] according to body weight ,with 8 rats in each group. Except for normal group ,the model of Qi-deficiency was made in other groups by the method of “diet disorder+fatigue ”. At the same time ,administration groups were given relevant medicine intragastrically ,and normal group and model group were given constant volume of water,once a day ,for consecutive 21 days. After last administration ,the general situation of rats was observed ;the body weight ,spleen index and thymus index of rats were detected ;weight-bearing swimming time was recorded ;the levels of spleen T lymphocyte subsets CD 3 and CD 4,the levels of ATP and ADP in liver tissue ,serum levels of ALB ,RBC and HBG in blood as well as the serum levels of SOD,MDA,lactate,LDH,CK,IL-2,IL-12 and TNF-α were all detected. RESULTS:Compared with normal group ,body weight,thymus index ,spleen index ,weight-bearing swimming time ,the level of spleen T lymphocyte subsets CD 3,ATP,ADP, ALB,IL-2 and IL- 12 were decreased or shortened significantly in model group (P<0.05 or P<0.01). The levels of MDA , lactate,CK and TNF-α were increased significantly (P< 0.05). Compared with model group ,body weight ,spleen index,weight-bearing swimming time ,the level of spleen T lymphocyte subsets CD 3 and the levels of ATP ,ADP,ALB, RBC and IL- 2 were increased significantly or prolonged(P<0.05);while the levels of MDA ,lactate,CK and TNF-α were decreased significantly in A. membranaceus total saponins high-dose group(P<0.05 or P<0.01).Weight-bearing swimming time ,the levels of ATP ,ADP and IL- 2 in A. membranaceus total saponins low-dose group were increased significantly or prolonged (P<0.05 or P<0.01),while the levels of MDA ,lactate,CK and TNF-α were decreased significantly (P<0.05 or P<0.01). Compared with positive control group ,spleen index ,spleen T lymphocyte subsets CD 3,weight-bearing swimming time and ATP level of A. membranaceus total saponins high-dose group were increased significantly or prolonged (P<0.05 or P<0.01),while MDA levels of A. membranaceus total saponins high-dose and low-dose groups were decreased significantly (P<0.05 or P<0.01). CONCLUSIONS :A. membranaceus total saponins can reduce the body ’s accumulation of blood lactic acid ,the activity of CK ,the level of lipid peroxide and regulate immunity to tonify Qi ,delay fatigue and improve exercise ability.
