Objective To test the in vitro effect of the extract of Herba taraxaci on Demodex folliculorum. Methods Active Demodex folliculorum were obtained from patients with moderate to severe demodex infestation. Herba taraxaci and Radix stemonae were extracted respectively with 80% ethanol under 85℃, and a preparation with a concentration of 200mg/ml was made. The extractions were used in vitro to examine the anti-mite activity by observing time of killing mites. Physiological saline and Radix stemonae extraction served as blank control and positive control respectively. PH value of Herba Taraxaci extract was noted. Skin irritation test of normal and wounded skin and acute toxicity test were carried out with rabbit shin, and Herba taraxaci and 75% ethanol were served as experiment and control medicine. Results Motion and morphology of the mites considerably changed with the effect of Herba taraxaci extract. Aim: To establish an analytical method for the simultaneous determination of norfloxacin,ofloxacin,pefloxacin,ciprofloxacin,lomefloxacin,danofloxacin,enrofloxacin,sarafloxacin and difloxacin in eggs using solid-phase extraction-LC-MS/MS.Methods: Egg samples were deproteinized with acetonitrile,followed by defatting with hexane.Then the samples were processed by solid-phase extraction and analyzed by LC-MS/MS using an electrospray source.The separation was carried out on a Shimadzu Shim-pack VP-ODS C_(18) column,with a mobile phase consisting of acetonitrile-0.1% formic acid(13: 87).Results: The validated method was proved to be of high specificity,accuracy and sensitivity.Conclusion: The established method is suitable for the routine residual monitoring of fluoroquinolones.

Objective In order to find the polymorphism site applicable to efficient genetic diagnosis on Haemophilia A in Han Chinese Population in Wenzhou.Methods With the method of polymerase chain reaction(PCR) and polyacrylamide gel electrophoresis(PAGE),288 of X chromosomes from 96 men and 96 women were detected on the polymorphism of BCL I in the intron 18 of FV Ⅲ gene.Results The gene frequency of the polymorphic site BCL I was 34.38% in Han Chinese population in Wenzhou.43.75% women were heterozygous and the polymorphism information content(PIC) was 0.4512.Conclusion For Han Chinese population in Wenzhou,the BCL I genetic site has enough information,being one of the genetic markers with high polymorphism,applicable to the screening for carrier and prenatal diagnosis of Haemophilia A in Wenzhou.
7.Relationship between EGFR and KRAS mutations and clinicopathologic features of non-small cell lung cancers

Purpose To explore the relationship between the mutations of epidermal growth factor receptor ( EGFR) and KRAS genes and clinicopathological characteristics in patients with non-small cell lung cancers (NSCLC). Methods Clinical samples from 431 NSCLC patients were obtained for EGFR and KRAS gene analysis. PCR based direct DNA sequencing was used to investigate mutations in exon 18-21 of EGFR gene and codon 12 and 13 of exon 2 of KRAS gene. Results The overall EGFR mutation rate of primary NSCLC was 53. 6% (231/431) in this study cohort and eight cases showed double EGFR mutations. Mutation rates in female and male were 65. 2% (122/187) and 46. 9% (98/209), respectively. The mutation rate was higher in patients with non-smokers and adeno-carcinoma and adenosquamous carcinoma subtypes than in their counterparts (P<0. 05), with the percentage of 57. 2% (124/216), 60. 3% (199/330), 42. 9% (6/14), respectively. In squamous cell carcinomas and other subtypes, EGFR mutation rates were 11. 6% (5/43) and 11. 1% (1/9), respectively. The EGFR mutation types included exon 18 point mutations (4. 0%, 9/227), exon 19 deletion mutations (4. 5%, 101/227), exon 20 insert or point mutations (9. 7%, 22/227) and exon 21 point mutations (41. 4%, 94/227). Activating mutations of KRAS gene were detected in 7. 8%(31/396) of NSCLC. Twenty-eight patients showed codon 12 mutations ( G>T, G>A, G>C) , and three patients had codon 13 mutations ( G>A, G>T) . Most of these mutations were G to T transversion (64. 5%, 20/31). Conclusion Polymerase chain reaction-direct sequencing is a reliable and effective method for the detection of the EGFR and KRAS gene mutation in NSCLC patients. The mutation rate of EGFR is higher in Chinese patients, especial-ly in non-smoking female patients with adenocarcinoma.

Objective To evaluate the clinical application of sweep pattern visual evoked potential in assessing visual acuity (SPVEP-A)of amblyopic children.Design Prospective clinical study.Participants 80 patients(148 case)of amblyopic children and 26 children(52 case)with normal vision.Methods The correlation were analysed of SPVEP-A and E acuity in 80 amblyopic children and of SPVEP-A and PVEP-A in 26 children of normal visual.SPVEP-A was recorded with UTAS-E 3000(LKC)using gratings of 25 differ- ent spatial frequency from 0.2 to 18.8cpd as stimulus.The response were averaged and DFT on the monitor display.SPVEP-A was de- termined by extrapolate to 0 response amplitude.Both transient PVEP-A and SPVEP-A were examined in 26 normal children.The ob- jective visual acuity in PVEP were determined with the highest spatial frequency which evoked a recognizable response.Main Out- come Measures SPVEP-A,PVEP-A and E acuity.Results SPVEP-A were from 0.35 to 0.9 among 80 patients with corrected E acu- ity from 0.1 to 0.8.The correlation coefficient of SVEP-A-I with E acuity was 0.602 .The correlation coefficient of SPVEP-A.between reproducibility detection was 0.448.The correlation between SPVEP-A and E acuity in amblyopic slightly was higher than middle and severe amblyopia(r=0.773 vs r=0.590).In 26 normal children a correlation coefficient of 0.679 was obtained between E acuity anti PVEP-A but 0.424 between E acuity and SPVEP-A.Higher evaluation or lower evaluation of E acuity was found in SPVEP-A in this study.Acuity was upostimated in lower scope of E acuity and underestimated in higher scope of E acuity.The reproducibility and vari- ation were not satisified.Conclusion The sweep VEP is a objective method of amblyopic screening.There is significant positive corre- lation between SPVEP-A and E,but its variability is more great,so we should improve its stability and positive rate uheriorly.
9.Correlation analysis between mental health and work engagement of nurses

Objective:To investigate the status of mental health and work engagement of clinical nurses, to explore the correlation between them, and to provide scientific basis for improving the mental health and increase work engagement levels of nurses.Methods:From August to September 2018, 1 140 nurses in Taihe Hospital of Shiyan City of Hubei Province · Affiliated Hospital of Hubei University of Medicine were selected as survey objects by convenient sampling method, and the mental health and work engagement of nurses were investigated by Symptom Checklist-90 (SCL-90) and Utrecht Work Engagement Scale (UWES-9), correlation analysis was used to explore the relationship between mental health and work engagement, and multiple linear regression was used to analyze the influencing factors of work engagement in clinical nurses.Results:The score of SCL-90 of clinical nurses was 123.01±38.14 and the score of UWES-9 was 3.53±1.40. Correlation analysis showed that the nurse with total scores of mental health and work engagement, vitality, dedication, and concentration were negative correlation( r values were -0.250--0.218, P<0.01). Multiple linear regression analysis showed that age, depression, and paranoia were the influencing factors of clinical nurses work engagement ( t values were 4.682, -3.211, -2.715, P<0.01). Conclusions:The mental health level of nurses is good, and their work engagement level is average. Mental health is closely related to work engagement. Nursing managers should take effective measures to improve the mental health and increase work engagement levels of nurses.
10.Optimization of the Water Extraction Technology of Punica granatum Peel and Flowers in Muzha Uighur Medicine Suppository

OBJECTIVE:To optimize the water extraction technology of Punica granatum peel and flowers in Muzha supposito-ry. METHODS:Using total tannin,ellagic acid contents as evaluation indexes,extration time,times,the amount of solvents as in-vestigation factors,L9(34)orthogonal test was adopted to optimize the water extraction technology of P. granatum flowers and peel. And verification test was conducted. RESULTS:The optimal extraction technology of total tannin and ellagic acid in P. granatum peel and flowers was as follow as 10-fold water reflux extraction for 3 times with 2 herbs,0.5 h every time. The average content of total tannin and ellagic acid extracted from 3 batches of herbs was 151.69 mg/g (RSD=1.15%,n=3),24.59 mg/g (RSD=1.41%,n=3)in the verification test,respectively. CONCLUSIONS:The optimized extraction technology is stable,feasible,and can be used for extracting the total tannin and ellagic acid in P. granatum peel and flowers.
