1.Imaging features of anterior cruciate ligament reconstruction graft insufficiency

ObjectiveTo investigate the imaging features of anterior cruciate ligament(ACL) graft insufficiency.Methods X-Ray and MR imaging examinations in 24 consecutive patients who had ACL reconstructive graft insufficiency were retrospectively evaluated for tunnel position,osteoarthrosis and its related complications.Follow-up arthroscopy showed 16 graft tears and 8 graft laxities.Fisher exact test was used to compare tunnel malpositions,the proportion of graft tear on MRI and osteoarthrosis between graft tear group and graft laxity group.ResultsTwo malpositions of tibial tunnel and 3 malpositions of femoral tunnel were seen in graft tear group.Three-malpositions of tibial tunnel and 4 malpositions of femoral tunnel were seen in graft laxity group.The proportion of tibial or femoral malposition showed no significant difference between the two groups(P =0.289,P =0.167).In graft tear group,15 complete graft tears were diagnosed correctly,1 partial tear was misdiagnosed as normal on MRI.In graft laxity group,4 grafts were diagnosed as normal and 4 were considered as graft tear on MRI.A significant difference was seen between the two groups (P = 0.028) in the proportion of graft tear diagnosed on MRI.Fourteen osteoarthrosis were seen in graft tear group and 5 in graft laxity group.No significant difference was seen between the two groups ( P =0.289) in the proportion of esteoarthrosis.Conclusion The proportions of tunnel malposition and osteoarthrosis showed no significant difference between the graft tear group and graft laxity group.Most graft tears can be diagnosed accurately on MRI,but some cases of graft laxity may be misdiagnosed for graft tear.

3.The expression of S-100 protein, CD1a, CD83 and Ki-67 in oral Langerhans cell histiocytosis.

OBJECTIVETo study clinicopathological features, diagnosis, differential diagnosis of oral Langerhans cell histiocytosis (LCH), retrospective clinicopathologic study was carried on and a variety of immune phenotype were detected.
METHODSThe clinicopathological features of 29 cases of oral LCH were analyzed. The immunohistochemical staining of S-100 protein, CD1a, CD83 and Ki-67 were used in above cases by immunohistochemical streptavidin-biotin peroxidase (SP) and Elivison two-step method. Statistical analysis was adopted for the results.
RESULTSOf the 29 cases of LCH, the expression of S-100 protein and CD1a were positive in 24 cases and negative in 5 cases, so 5 cases were excluded from the diagnosis of LCH. Among 24 cases of LCH, 15 patients were male and 9 were female. The median age was 7.50 years. 14 lesions were in the mandible, 5 were in the maxilla and 5 involved the mandible and maxilla. 9 cases were in stage I, 13 in stage II and 2 in stage III, according to Bartnick classification. Immunohistochemistry showed all 24 cases staining for S-100 protein and CD1a were positive. Comparing with maxillofacial lesions involved soft tissue, Ki-67 positive rate was lower and CD83 positive rate was higher in maxillofacial single bone lesion.
CONCLUSIONThe immunohistochemical staining of S-100 protein and CD1a are important for the diagnosis of LCH. Maxillofacial bone single LCH might have lower proliferative activity and a higher state of maturity. Maxillofacial LCH involved soft tissue might have a higher proliferative activity and a lower state of maturity.

4.Application of a method of improving the quality of sampling in review to determine the light areas of endemic fluorosis in quality control

ObjectiveTo study an application of a method of improving the quality of sampling in review to determine the light areas of endemic fluorosis(referred to as endemic fluorosis) in quality control. Methods Of 15 light endemic fluorosis township(town), six were randomly sampled, and the prevalence of dental fluorosis in 22 village primary school children aged 8 to 12 were reviewed to determine the improved quality of sampling in Xuyong county Sichuan province. ResultsSix townships(towns) were selected by simple random sampling from 15 endemic fluorosis townships(towns) for review inspection in Xuyong country. A total of 22 villages were verified, accounting for 22.7% of the total 97 villages verified. Of the 416 children for review inspection of dental fluorosis, 383 children were positive. The consistent rate of children' s dental fluorosis was 92.07%, and the verification to be slight villages was up to 21 endemic villages, accounting for 95.45%. ConclusionsThe application of a method of improving the quality of sampling can improve the efficiency of quality control, and improve the accuracy. It is a novel quality control method.
5.Study on Th9 cells and Bregs cells in immunological pathogenesis of ulcerative colitis and their correlation with disease activity of patients

Objective:To explore the role of Th9 cells and Bregs cells in immunological pathogenesis of ulcerative colitis(UC)and their correlation with disease activity.Methods:A total of 105 patients with UC who were admitted to Hai'an People's Hos-pital from June 2020 to March 2022 were prospectively included as observation subjects(UC group).Among them,48 cases were in disease remission(remission UC group)and 57 cases were in disease activity(active UC group).According to the severity of the pa-tients'conditions in active UC group,subgroups were divided:mild active group(n=20),moderate active group(n=24)and severe active group(n=13).In addition,50 healthy volunteers during the same period were selected as control group.The proportion of Th9 and Bregs cells in the peripheral blood of patients between the two groups were analyzed by flow cytometry,and the serum levels of IL-9,IL-10,IL-35 and C-reactive protein(CRP)were detected by ELISA.The correlation between Th9 and Bregs cell levels and disease ac-tivity and inflammation degree were also analyzed by Pearson correlation analysis.Results:The proportion of Bregs cells and Th9 cells in patients with UC were significantly higher than those of control group.The CAI score,the proportion of Bregs cells and Th9 in active UC group were higher than those in remission UC group(P<0.05).The ratios of Th9 cells and Bregs cells in patients with severe active UC were significantly higher than those in patients with moderate and mild active UC groups,and the ratios in moderate active group were higher than those in mild active group(P<0.05).The levels of CRP and IL-9 in patients with UC in the active and remission stages were significantly higher than those in control group and higher in active stage than in the remission stage(P<0.05).IL-10 and IL-35 levels in the active and remission stages were significantly lower than those in control group,and the levels in the active stage were lower than those in the remission stage(P<0.05).The levels of CRP and IL-9 in severe active group were higher than those in mild and moderate active groups,and the levels in moderate active group were higher than those in mild active group(P<0.05).The levels of IL-10 and IL-35 in severe active group were significantly lower than those in moderate and mild active groups,and the levels in moderate active group were lower than those in mild active group(P<0.05).Pearson analysis showed that Th9 and Bregs cells were positively correlated with disease activity,CRP and IL-9 levels,and negatively correlated with IL-10 and IL-35 levels(P<0.05).Conclusion:The proportion of Th9 and Bregs cells in peripheral blood of patients with active UC are significantly increased,which are closely related to the occurrence and development of the disease and expected to become good indicators for auxiliary clinical early diagnosis and evaluation of the disease.
6.Mucinous adenocarcinoma of salivary gland: a case report.

OBJECTIVE:
To investigate the killing effects of VP(3) on nasopharyngeal carcinoma cell line CNE-2.
METHODS:
Plasmid expression vector pcDNA3.1(-) CMV.VP(3)-His was constructed and identified by Kpn I/EcoR I endonuclease analysis, and then sequenced to verify successful insertion in the sense direction of VP(3) gene. pcDNA3.1(-) CMV.VP(3)-His and pcDNA3.1(-)-His expression plasmid was transiently transfected into nasopharyngeal carcinoma cell line CNE-2 . VP(3) protein expression was detected by Western blotting. MTT assay was used to determine the killing effects of VP(3) gene on nasopharyngeal carcinoma cell line CNE-2.
RESULTS:
Endonuclease analysis and sequencing confirmed the recombinant plasmid contained the complete VP(3) CDS sequence. Western blotting detected a 14.03 kD protein expression from the transfected cells, which was the expecting band of VP(3) gene. The growth of CNE-2 cells that expressed VP(3) gene was inhibited,while the growth of CNE-2 cells that did not express VP(3) gene was not inhibited.
CONCLUSION
VP(3) gene can kill nasopharyngeal carcinoma cell CNE-2.

8.Study on the role of miR-567 in proliferation,migration and cell cycle of NSCLC through regulation of CDK8 and its clinical relevance

Objective To investigate the role of microRNA(miR)-567 in the proliferation,migration and cell cycle of non-small cell lung cancer(NSCLC)through regulation of cyclin dependent kinase 8(CDK8)and its clinical relevance.Methods Tumor tissues and adjacent tissues of 40 NSCLC patients were collected,and the expressions of miR-567 and CDK8 were detected by real-time quantitative fluorescent PCR(qRT-PCR).miR-NC mimic,miR-567 mimic,oe-NC,and oe-CDK8 were transfected into A549 and H1975 cells.The ex-pressions of miR-567 and CDK8 were detected using qRT-PCR.Cell proliferation was detected by CCK-8 method,and cell migration was detected by Transwell assay.Cell cycle changes were detected by flow cytome-try.The targeting of miR-567 and CDK8 was detected by luciferase reporter gene assay.Results In the tumor tissues of NSCLC patients,the expression of miR-567 was decreased,while the expression of CDK8 was in-creased,and the two were negatively correlated(P<0.05).In A549 and H1975 cells,miR-567 mimic group was compared with miR-NC mimic group,the expression of miR-567 was increased,the expression of CDK8 was decreased,the proliferation and migration levels of cells were decreased,the proportion of G1 phase was increased,and the proportion of S phase was decreased.The fluorescence intensity of miR-567 mimic group was lower than that of miR-NC mimic group in normal CDK8.miR-567 mimic+oe-CDK8 group was compared with miR-567 mimic+oe-NC group,the expression of CDK8 was increased,the proliferation and migration levels of cells were increased,the proportion of cells in G1 phase was decreased,and the proportion of cells in S phase was increased.Conclusion miR-567 can inhibit NSCLC proliferation and migration by targeting CDK8 expression and controlling tumor cell arrest in the S phase.
9.Influences of tenuigenin on learning and memory ability and neurotransmitters in senescence-accelerated mice

Objective To discuss the improving effect of tenuigenin on learning and memory ability and its influence on cerebral monoamine neurotransmitters in senescence-accelerated mice prone 8(SAMP8).Methods The senescence-accelerated mice(SAMP8)were chosen as subjects,and then randomly divided into the model group,tenuigenin group and Donepezil group.SAMR1 were chosen into the the control group.After intragastrical treatment for 3 months,the learning and memory ability and the levels of 5-hydroxytryptamine(5-HT),5-hydroxyindoleacetic acid(5-HIAA),dopamine(DA)and norepinephrine(NE)were detected respectively by applying Morris water maze and high performance liquid chromatography(HPLC).Results In the tenuigenin group the hidden platform test showed that the escape latency was,respectively,(43.56±10.87)s,(41.37±9.71)s,(37.62±10.05)s and (41.60±13.61)s from the 2nd to 5th day,and the reversed test showed that the escape latency was (37.10±14.04)s on the 9th day,which were all shortened compared with those in the model group,and the difference was statistically significant(P<0.05).In the tenuigenin group the level of 5-HT,5-HIAA,DA or NE was,respectively,(5.95±0.93)mmol/L,(4.43±1.08)mmoL/L,(2.61±0.87)mmol/L and(9.44±3.76)mmol/L,which all increased compared with those in the model group,and the difference was statistically significant(P<0.05).Conclusion Tenuigenin can improve the learning and memory ability of SAMP8,and the effect may related to that it can regulate the level of cerebral monoamine neurotransmitters.
10.Study on the in-hospital diagnosis and treatment time in patients with ischemic stroke in Hebei Province

Objective:To investigate the in-hospital diagnosis and treatment time for patients with acute ischemic stroke in Hebei Province.Methods:The data of in-hospital diagnosis and treatment of acute ischemic stroke in Hebei Province were collected and analyzed, and then compared with the NINDS recommended time. Methods The data of in-hospital diagnosis and treatment of acute ischemic stroke in Hebei Province were collected and analyzed, and then compared with the NINDS recommended time.Results:The median time in hospital diagnosis and treatment was significantly longer than the NINDS recommended time (104 min vs. 60 min, P<0.001). The median time from completing the cranial CT scan to getting the CT report differed significantly to the NINDS recommended time (30 min vs. 20 min, P<0.001). The median time from getting the CT report to obtaining treatment was 43 min, which was significantly longer than the NINDS recommended 15 min ( P<0.001). The median time of in-hospital diagnosis and treatment for emergency service system (EMS) patients was 101 min, which was shorter than that for non-EMS patients (104 min, P=0.01). The median time of in-hospital diagnosis and treatment in Tertiary Hospital was 105 min, which was significantly longer than that in Secondary Hospital 99 min, ( P<0.05). Conclusions:The in-hospital emergency treatment delay in Hebei Province was relatively serious for patients with acute ischemic stroke. The time between obtaining the head CT report to beginning thrombolytic therapy is the most important factor in hospital delay. EMS can shorten in-hospital delay for acute ischemic stroke. Compared with the tertiary hospital, the secondary hospital has shorter in-hospital delay time.
