1.Study on Quality Criteria of Fever and Cough Syrup for Children

OBJECTIVE:To establish the quality criteria of fever and cough syrup for children.METHODS:Thin-layer chromatography(TLC) was employed to make qualitative determination of herbba ephedrae,radix scutellariae,radix glycyrrhi-zae in fever and cough syrup for children,and high performance liquid chromatography(HPLC) was used to determine the con-tent of amygdaloside in its chief medicinal material,almond.RESULTS:The spots in the TLC of herbba ephedrae,radix scute-llariae,radix glycyrrhizae were in the same color with those in the chromatograms of control articles.The linear range of amygdaloside was 0.198?g~4.954?g(r=1.0 000).CONCLUSION:The present method is simple,accurate sensitiveness and highly reproductive,which can be used for the quality control of this preparation.
2.EFFECT OF 1?, 25-DIHYDROXY-VITAMIN D_3 ON DIFFERENTIATION AND PROLIFERATION OF A HUMAN MYELOID LEUKEMIA CELL LINE

1 ? 25-dihydroxy-vitamin D3(1? 25 (OH)2 D3)induced differentiation of human promyelocytic leukaemia cells ( HL-60 ) into mature myeloid cells in vitro. The ratio of nuclei to cytoplasma decreased; Their nucleoli reduced; Cells became irrgular and extended the pseudopods. 1 ? 25 (OH ) 2 D3 caused significant increase of nitroblue tetrazolium ( NBT ) reduction and ?-non-specific acid esterase (?-NAE ) and acid phosphatase (ACP ) activities. These data and morphological characteristics suggest that HL-60 cells were conclusively identified as monocytes/macrophages. The histogram of DNA distribution ahylyzed by flow cytometry demonstrated G0 + G1 phase increased and S phase increased and S phase decreased remarkably after treatment with l?, 25 ( OH ) 2 D3 as compared with untreated cells.
3.Molecular imaging of tumor by target microRNAs

MicroRNAs (miRNAs) are a novel class of small noncoding RNAs that regulate gene expression by targeting mRNAs for either cleavage or translational repression.Accumulated studies show that miRNAs aberrantly expressed in tumors are closely related with tumorogenesis and could be potential tumor biomarkers.It will be helpful in the diagnosis,prognosis and(or) predicting treatment response of tumor by imaging miRNAs expression.Several molecular imaging methods,including the optical imaging,radionuclide imaging and multimodality imaging,have been successfully used to visualize miRNAs,miRNAs,a new molecular imaging target,may explore a new field for tumor diagnosis and therapy.
4.The progress of research on optical imaging and radionuclide imaging as intra-operative navigators

Complete surgical resection is very important for the outcome of cancer treatment and prognosis.Radionuclide imaging and optical imaging are effective methods for intra-operative guidance of tumor exploration and resection because of their capability in accurate tumor localization and margin delineation.With the development of more specific molecular probes and more sensitive detectors,advanced imaging methods that combine the advantages of both optical and radionuclide imaging,such as Cerenkov luminescence imaging,will be expected to play a more important role as intra-operative navigating agents for surgical oncology in the near future.
5.Effect of high-flux hemodialysis on advanced oxidation protein products in maintenance hemodialysis patients

Objective To investigate the effect of high-flux hemodialysis (HFHD) on advanced oxidation protein products (AOPPs) in maintenance hemodialysis (MHD) patients.Methods 40 MHD patients (MHD group),50 non-dialytic chronic kidney disease (CKD) patients (CKD group) and 31 healthy subjects undergoing health examination survey received AOPPs detection.After lowflux hemodialysis (LFHD) for 3 months,HFHD was performed in MHD group for 3 months.Plasma AOPPs and other biochemical indicators were tested at the end of 0,3 and 6 months.AOPPs levels were detected in HFHD and LFHD dialysates.Results Plasma AOPPs level was higher in MHD group than in CKD and control groups,and plasma AOPPs level was higher in CKD group than in control group.Compared with before LFHD in MHD group,AOPPs and triglyceride (TG) levels had no significant differences after LFHD for 3 months(t=0.831,-0.191,both P>0.05),while plasma cholesterol (CHO) and β2 MG levels were increased in MHD group after LFHD for 3 months(t=-2.697,10.39,P<0.05 or 0.001).There were significant differences in plasma AOPPs,TG,β2-MG and CHO levels before versus after HFHD for 3 months (t=4.292,2.271,8.297 and 2.295,P <0.001 or 0.05).No AOPPs were detected in HFHD and LFHD dialysates.Conclusions Plasma AOPPs levels are increased in CKD patients.LFHD can not effectively eliminate AOPPs,while long term HFHD can decrease the plasma levels of AOPPs,TG and CHO,and reduce β2-MG accumulation.
7.Quality analysis of 3 species of Radix Paeoniae Alba cultivated in Bozhou by HPLC and LC-MS

Aim: To establish the HPLC and LC-MC methods for the quality analysis of the different species of Radix Paeoniae Alba cultivated in Bozhou. Methods: The LC-MS method was applied to analyze the chemical composition of the cultivated species, the HPLC method was applied to determine the contents of paeoniforin, ben-zoic acid and paeonivayin in crude drugs and processing products. Results: 10 characteristic peaks were identified by LC-MS with reference standards. The significant difference was observed in the chemical compositions of 3 cultivated species. The significant difference was observed in the chemical compositions of 3 cultivated species. The content of paeoniflorin was increased significantly after traditional processing. The results indicated the 4-year-old pubang and 5-year-old xiantiao are the best cultivated species. Conclusion: The methods are simple, rapid and precise to assess and control the quality of different species of Radix Paeoniae Alba cultivated in Bozhou.
8.Effects of STAT3 aberrant activation on malignant transformation of human umbilical cord mesenchymal stem cells when exposed to human breast cancer microenvironment

Objective_To investigate whether hUCMSCs undergo malignant transformation when exposed to MCF-7B breast cancer microenvironment and whether the abnormal activation and over expression of STAT3 play an impor-tant role in this transformation.Methods_The experiment was divided into three groups:blank group ( hUCMSCs were separately cultured),experimental group(hUCMSCs were indirectly co-cultured with MCF-7B breast cancer cells),positive control group(MCF-7B breast cancer cells were separately cultured).Morphology of cells was detec-ted by invertedmicroscope.Cell cycle was detected by flow cytometry.The mRNA expression of STAT3, c-Myc and Bcl-xL was tested by real-time PCR.The protein expression and location of p-STAT3,c-Myc and Bcl-xL were detected by immunofluorescence.The protein expressions of p-STAT3,STAT3,c-Myc and Bcl-xL were also meas-ured by Western blot.Results_The experimental group cells showed typical morphology of the tumor cells.The cells proportion of experiment group in G1 phase was significantly lower than that of the blank group(P<0.05),but which in S and G2 phase were significantly higher than those of the blank group(P<0.05).The mRNA expression levels of STAT3 ,c-Myc and Bcl-xL in experimental group was significantly higher than those in blank group ( P<0.05).p-STAT3,STAT3,c-Myc and Bcl-xL protein were significantly higher than those of the blank group(P<0.05),and they were mainly located in the nuclei.The protein expression of STATS also showed significant changes in experimental group.Conclusions_hUCMSCs trends to malignant transformations when exposed to MCF-7B breast cancer microenvironment.The abnormal activation and over expression of STAT3 are of important factors leading to the malignant transformation of hUCMSCs.
9.Clinical analysis on juvenile dermatomyositis combined with soft-tissue calcification

Objective To analyze the clinical features of juvenile dermatomyositis (JDM) combined with soft-tissue calcification. Methods Forty-seven patients with JDM combined with soft-tissue calcification (soft-tissue calcification group) were retrospectively analyzed, and they were contrasted with 89 patients with non-calcification (non-calcification group). Results The rates of Gotton signe, muscle contracture and joint dysfunction in soft-tissue calcification group were significantly higher than those in non-calcification group:87.23% (41/47) vs. 43.82% (39/89) and 68.09% (32/47) vs. 21.35% (19/89), and there were statistical differences (P<0.05). The dosage of glucocorticoid (conversion of prednisone measuring more than 1.5 mg/kg), rate of using immunodepressant, level of creatine kinase in soft-tissue calcification group were significantly lower than those in non-calcification group:17.02%(8/47) vs. 68.54%(61/89), 25.53%(12/47) vs. 88.76%(79/89), (566.45±240.41) U/L vs. (1 680.12±656.50) U/L, and there were statistical differences (P<0.05). Conclusions The patients with JDM combined with Gotton signe are more prone to soft-tissue calcification. The rate of muscle contracture and joint dysfunction in soft-tissue calcification patients is significantly higher than that in non-calcification patients. For the patients whose creatine kinase are not obviously elevated, they are more prone to soft-tissue calcification. Early active application of glucocorticoid and immunodepressant therapy can reduce or prevent the occurrence or development of late calcium deposition.
10.Analysis of surgical treatment of huge primary liver cancer:report of 86 cases

Objective:To explore the possibility and methods of surgical treatment(ST) for huge primary liver cancer(HPLC).Methods:The clinical data of 86 HPLC were analyzed retrospectively and the main challenges as well as their counter measures during ST are concluded.Results:Among the 86 cases,there were 10 cases which complicated with portal vein(PV),and 5 cases with IVC thrombus respectively;there were 76 cases with HBV cirrhosis,2 HCV cirrhosis,there were 60 and 26 cases of primary and secondary hepatectomy in which included regular right(9 cases),left hemihepatectomy(8 cases)and combined hepatectomy(69 cases).Six cases received PV embolism before surgery.There were 10 and 5 cases of cancer thrombus extraction from PV and IVC respectively.In this group,81 patients were cured,and the perioperative mortality was 5.8%(5/86).The postoperative one—year survival rate for the patients was 77.9%.Conclusions:ST for HPLC is safe and acceptable.Full of preoperative assessment,careful intraoperative exploration,and surgical skill played a key roles.
