3.Activation of Rho Kinase in Lung Tissue of Hypoxic Pulmonary Hypertension in Rats in Different Stages and Its Significance

Objective To study the expression of Rho kinase and its functional activation in lung tissue from hypoxic pulmonary hypertension(HPH) rat model,and the effects of fasudil on HPH.Methods Seventy-two male Spraque-Dawley rats were randomly divided into the control group,hypoxic model group,and fasudil-intervention group[group with hypoxia and fasudil for 15 mg/(kg?d)],respectively.Mean pulmonary arterial pressure(mPAP) and right ventricle hypertrophy index(RVHI) were measured.Expression of Rho kinase mRNA and protein were examined by reverse transcriptase-polymerase chain reaction(RT-PCR) and Western blot,respectively.The phosphorylation of binding subunit of myosin phosphatase(MBS)-a substrate of Rho kinase was detected by Western blot and defined,as the mark of functional activation of the kinase.Results The expression of Rho kinase mRNA in hypoxic model group was markedly upregulated even before the onset of the third day after the experiment(HPH),and it was much lower in rats of fasudil group than that of hypoxic model group.The phosphorylation of MBS was significantly higher in hypoxic model group than that in control group,and it was positively correlated with the mPAP and RVHI(all P
4.Distribution and antimicrobial resistance of pathogens causing lower respiratory tract infection in patients in a hospital, 2013-2015

Objective To explore the distribution and antimicrobial resistance of pathogens causing lower respiratory tract infection in patients, and provide basis for rational choice of antimicrobial agents in clinic.Methods All bronchoalveolar lavage fluid specimens in a hospital from January 1, 2013 to December 31, 2014 were performed culture, antimicrobial susceptibility testing of isolated strains was performed with Kirby-Bauer method.Results A total of 999 strains were isolated from 7 702 bronchoalveolar lavage fluid specimens, 398 (5.17%), 326 (4.23%), and 275(3.57%) strains were bacteria, fungus, and Mycobacterium respectively.The main bacteria were Pseudomonas aeruginosa(P.aeruginosa, n=97), Acinetobacter baumannii(A.baumannii, n=87), Klebsiella pneumoniae(K.pneumoniae, n=62), Staphylococcus aureus(S.aureus, n=44), and Haemophilus influenzae (n=28);the main fungi were Candida albicans (n=161), Aspergillus fumigatus (n=41), and Aspergillus flavus(n=38);the main Mycobacterium were Mycobacterium tuberculosis(n=271).Antimicrobial susceptibility testing results showed that resistance rates of P.aeruginosa to the commonly used antimicrobial agents (except ticarcillin/clavulanic acid and levofloxacin) were all<30.00%.Resistance rates of A.baumannii to ampicillin/sulbactam, imipenem, amikacin, gentamicin, and tobramycin were all >80.00%, but to the other commonly used antimicrobial agents were 36.84%-60.53%.Among 62 strains of K.pneumoniae, 20 were extended-spectrum β-lactamases(ESBLs)-producing strains.Antimicrobial resistance rates of ESBLs-producing strains were obviously higher than non-ESBLs-producing strains.Among 44 S.aureus strains, 26 were methicillin-resistant S.aureus (MRSA), resistance rate of S.aureus to penicillin was 97.73%, to vancomycin, teicoplanin, and linezolid were all 0, to the other antimicrobial agents were 9.09%-61.36%.Conclusion Bacteria is the major pathogen causing lower respiratory tract infection in hospitalized patients, but fungus and Mycobacterium can not be ignored.Resistance rates of P.aeruginosa to commonly used antimicrobial agents is relatively low, but resistance of A.baumannii is more serious.
6.The expression and clinico-pathological correlation analysis of erbB-2 and erbB-3 in salivary adenoid cystic carcinoma

Objective Through detecting the expression of erbB-2 and erbB-3 of epithelial growth factor receptor family in clinical specimens of salivary adenoid cystic carcinoma, to explore the role of erbB-2 and erbB-3 in the carcinogenesis and development of salivary adenoid cystic carcinoma. Methods Thirty-four specimens of adenoid cystic carcinoma were tested with immunohistochemical assay. Then the relationship between the expressions of erbB-2, erbB-3 and the gender, age of patients, TNM staging, tumor size and nerve invasion were analyzed.Results The positive expression ratios of erbB-2 and erbB-3 were 64. 7% and 32.4% respectively. And 29.4% of tumors were positively stained both by erbB-2 and erbB-3. The expression of erbB-3 was significantly correlated with TNM staging of tumors (P = 0. 026). The correlationship between the expressions of erbB-2 and erbB-3 was also found. Conclusions erbB-3 and erbB-2 may play important roles in the carcinogenesis and development of salivary adenoid cystic carcinoma. The positive expression of erbB-3 was associated with the biological behavior of tumors and prognosis of patients, and might become one of markers to evaluate the prognosis of patients.
7.Changes in Neuropeptide Y Y1 Receptor mRNA Level in the Infarcted Myoc ardial Tissues of Rats

Aim To investigate the change in neuropeptide Y(NPY) Y1 receptor level in infarcted myocardium tissues of (MI) rats. Method MI was induced by ligating the left descending anterior coronary artery (LAD) in the heart of rats. The techniques of reverse transcription polymerase chain reaction (RT-PCR) were used to identify the exist of NPY Y1 receptor mRNA in myocardial tissues. The technique of semi-quantitative PCR wa s used to observe the change of NPY Y1 receptor mRNA level. Results NPY Y1 receptor mRNA distributed in the heart tissues of rat. Compared with sham operated rats ,the NPY Y1 receptor mRNA level both in infarction area and non-infarction area was increased significantly after MI fo r 1d and 3d. The NPY Y1 receptor mRNA level in the heart tissues of sham-op erated rats was also significantly increased compared with that in control rats . Conclusion These results suggest that MI may result in increase NPY Y1 receptor level in the heart tissues of rats. Stre ss stimulation such as surgery may also increase the NPY Y1 receptor level in the heart.
8.Clinical outcome of FLT3-ITD (+) acute myeloid leukemia patients treated with allogeneic hematopoietic stem cell transplantation

Objective To study the clinical outcome of patients with fns-like tyrosine kinase-3internal tandem duplication (FLT3-ITD) positive acute myeloid leukemia (AML) treated with allogeneic hematopoietic stem cell transplantation (allo-HSCT) and to explore the potential prognostic factors to patients' survival including transplant types or disease status.Methods A total of 314 AML patients in our center from October 2006 to October 2012 were retrospectively analyzed,among whom,54 patients were defined with FLT3-ITD positive.Survival rates and treatment-related mortality were further analyzed.Results For all 54 FLT3-ITD positive patients,the 3-year overall survival (3-OS) rate was 56% and 3-year leukemia-free survival (3-LFS) rate was 47%.The outcome of haplo-identical HSCT was similar as that of sibling donors(3-OS rate:60% vs 54% ; 3-LFS rate:54% vs 45%,respectively).There were 47 patients who received transplantation in first complete remission(CR1).The other 7 patients were of disease relapse or in CR2 before transplantation.Not surprisingly,patients in CR1 had better prognosis than those in nonCR1.Conclusions Allo-HSCT is an effective treatment for AML patients with FLT3-ITD positive mutation.The survival outcome of haplo-identical HSCT was comparable with that of sibling donors.Relapse of AML was the dominant factor related to the mortality of FLT3-ITD positive AML patients after allo-HSCT.

In this study,a series of new sildenafil analogues 11-27 possessing a guanidine group were synthesized to investigate their PDE5 inhibitory activity and selectivity using[~3H]cGMP SPA kit in vitro and efficacy in the rat model of erection.Most of the compounds showed potent activity against PDE5,and more importantly,several compounds exhibited higher PDE5 selectivity over PDE6 than that of sildenafil.Structure-activity relationship of these sildenafil analogues was also discussed.Within this series of compounds,compound 15(IC_(50) =1.7 nmol/L)not only exhibited more potent PDE5 inhibitory activity than that of sildenafil (IC_(50) = 6.5 nmol/L),but also showed functional efficacy in the rat model of erection.
