1.Tissue and Induction Expression Profiles of Rice Phospholipid Hydroperoxide Glutathione Peroxidase at Protein Level

Proteins arc the major molecules performing life activities, and their spatial and temporal expression profiles in organisms are very important for understanding their accurate functions. Phospholipid hydroperoxide glutathione pcroxidase (PHGPx) is a unique antioxidant enzyme that directly reduces lipid hydroperoxides in biomembranes and plays a vital role in defense against mernhrane peroxidation damage. The protein expression profiles of rice PHGPx (OsPHGPx) were investigated in different rice tissues and under various stress treatments by using Western blot analysis. The results showed that in mature rice plants, OsPHGPx was mainly distributed in leaves, especially flag leaves, and in rice seedlings, OsPHGPx was detected in shoots and leaves. Moreover, OsPHGPx expression in rice seedlings could be markedly induced by H2O2 and NaCI, but weakly influenced by several plant hormones. Time- and dose-dependent effects were observed in both H2O2 and NaCI treatments, and the strongest induction was observed when rice seedlings were treated with 0.5 mmol/L H2O2 for 12 h or 500 mmol/L NaCI for 24 h. Additionally, dimethylthiourea, a H2O2 trap, inhibited H2O2-enhanced expression of OsPHGPx, but did not impair the enhanced effect of NaCI, implying that NaCl-induced OsPHGPx expression was not mediated by H2O2. These results will contribute greatly to further study the exact physiological function of OsPHGPx in rice.
2.Cost-effectiveness analysis of o-phthalaldehyde and glutaraldehyde as endoscope disinfectors

Objective To compare the durability and economic value of o-phthalaldehyde(OPA)and glutaraldehyde as endoscope disinfectors in continuous use of cleaning machine.Methods Compare the maximal number of endoseopes which could be disinfected by the two disinfectors at its minimal effective concentration,as well as the corresponding costs.The cost-effectiveness of two disinfectors were evaluated with Revenue-Incremental Cost Analysis.Results The number of endoscopes disinfected by OPA were 1.56 times as compared with glutaraldehyde,and an additional 2.95 yuan could be gained with OPA at the cost of 1 yuan.Conclusion When cleaning machine is full loaded,OPA is more durable than glutaraldehyde,and Can gain more benefit at the same cost.
4.High-resolution ultrasonography for distinguishing benign from malignant thyroid nodules

Objective The extensive use of high-resolution thyroid ultrasonography has markedly improved the detection rate of thyroid nodules ( TNs) .However, the ultrasonographic features of benign and malignant TNs usually overlap and have yet to be de-fined.This study aimed to investigate the ultrasonographic features and risk factors of malignant TNs by comparing their ultrasonograph-ic manifestations with those of benign TNs. Methods A total of 119 TNs (63 benign and 56 malignant) in 64 patients were includ-ed in this retrospective study.The gender, age, location of the TNs, and other variables were subjected to univariate analysis, the risk factors of malignant TNs were screened by multivariate logistic regression analysis, and the predictive value of the logistic regression model was assessed using the ROC curve. Results Multivariate logistic regression analysis indicated that the risk factors of malig-nant TNs in ultrasonography were irregular shape (OR:6.166, 95%CI:1.681-22.617), micro calcification (OR:3.593, 95%CI:1.800-7.169), and A/T ratio≥1 (OR:17.577, 95%CI:1.739-166.468).ROC curve analysis showed the area under the curve (AUC) to be 0.922 (95%CI:0.868-0.976). Conclusion Such ultrasonographic manifestations as irregular shape, micro-calcification, and A/T ratio≥1 are valuable in identifying the nature of TNs.
5.High glucose augments stress-induced apoptosis in endothelial cells

Hyperglycemia has been identified as one of the important factors involved in the microvascular complications of diabetes, and has been related to increased cardiovascular mortality. Endothelial damage and dysfunction result from diabetes; therefore, the aim of this study was to determine the response of endothelial cells to stressful stimuli, modelled in normal and high glucose concentrations in vitro. Eahy 926 endothelial cells were cultured in 5 mmol/L or 30 mmol/L glucose conditions for a 24 hour period and oxidative stress was induced by exposure to hydrogen peroxide (H2O2) or tumour necrosis factor- α (TNF- α ), following which the protective effect of the glucocorticoid dexamethasone was assessed. Apoptosis, necrosis and cell viability were determined using an ELISA for DNA fragmentation, an enzymatic lactate dehydrogenase assay and an MTT assay, respectively. High glucose significantly increased the susceptibility of Eahy 926 cells to apoptosis in the presence of 500 μmol/L H2O2, above that induced in normal glucose (P<0.02). A reduction of H2O2- and TNF- α -induced apoptosis occurred in both high and low glucose after treatment with dexametha-sone (P<0.05). Conclusion high glucose is effective in significantly augmenting stress caused by H2O2, but not in causing stress alone. These findings suggest a mechanism by which short term hyperglycemia may facilitate and augment endothelial damage.
6.Pathogen distribution and drug resistance in pneumonias among immunocompromised hosts

Objective To provide evidence for diagnosis and treatment by investigating the pathogens and drug resistance of immunocompromised host (ICH)pneumonias.Methods Statistical method was used to analyze retrospectively the data of pathogens and drug resistance of ICH pneumonia treated in our hospital in 2014. Results We confirmed 187 cases of ICH pneumonia by pathogen study of 324 patients.Gram-negative bacillus (68.42%)was the main pathogen of ICH pneumonia.Acinetobacter baumanii (AB)accounted for the first place. Staphylococcus aureus infection was still the first of Gram-positive coccus.Conclusion Gram-negative bacillus was the main pathogen of ICH pneumonia in our hospital and the isolated bacteria show strong antibiotic resistance. Therefore,it is necessary to strengthen dynamic monitoring of pathogens in the secretions from ICH pneumonia patients’ lower respiratory tract and drug resistance.It is suggested that clinicians make anti-infection treatment cover drug-resistant bacteria.
7.Isoflavone genistein protects high glucose-induced human aortic endothelial cell apoptosis through estrogen receptor-mediated pathway

Objective The aim of this study was to determine if isoflavone genistien has protective effects against high glucose-induced cell apoptosis in human aortic endlthelial cells,and investigate the possible mechanism for this protection.Methods Human aortic endothelial cells subjected to normal (5mmol/L) or high glucose (25mmol/L) were treated with genistein at 0,50,100nmol/L.Parallel experiments were performed with 100nM 17b-estradiol,and also in the presence and absence of the pure anti-estrogen ICI-182,780 (100nmol/L).The effects on cell apoptotic DNA fragmentation were determined using cell death ELISA,and the effects on cellular proliferation were determined using tritiated thymidine incorporation assay.Estrogen receptor expression was detected by Taqman quantitative PCR.Results Genistein at 100nmol/L significantly reduced high glucose-induced DNA fragmentation,and reversed cell DNA synthesis inhibition (P<0.001) after 24 hours' incubation.The effect of genistein was completely blocked by ICI-182,780administration.Estrogen receptor beta,but not alpha was found to be expressed in these cells.Conclusion Isoflavone genistein shows protection against high glucose-induced cell damage through estrogen receptor beta,reducing apoptotic DNA damage and protecting from the inhibition of cell proliferation.
8.The study on optimization of inner environment of a negative pressure isolator for transportation of infectious disease patient

Objective Study and determine the suitable negative pressure condition for a negative pressure isolator. Method The filter efficiency, pressure, capabilities of inner environment and physiological parameters of the user were tested in different negative pressure conditions. Results The different negative pressure conditions influence above mentioned capabilities of the isolator considerably except for the filter efficiency Conclusion Under the conditions of -30Pa~-50Pa,those are optimized the capabilities of inner environment and physiological parameters of the user.
9.Genotype analysis for measles vaccine strain and wild-type viruses of H1 genotype by RT-PCR-RFLP

Objective To establish RT-PCR-RFLP method for studying the genotype of wild mea-sles virus strains isolated from Tianjin area from 2002 to 2008. Methods Isolations of measles virus were carried out by tissue culture method from urine and throat swab specimens collected from suspected cases. RNA were extracted from the virus specimens. The 594 bp fragment of C terminal of the N (nucleoprotein) gene was amplified by one-step RT-PCR, then the PCR products were digested with Bcn I , separated on agarose gel electrophoresis and then analyzed by the method of RFLP (restriction fragment length polymor-phism). In addition, above results were compared with DNA sequencing. Phylogenetic tree was plotted based on the results for the genetic relationship and distance analysis. Results Sixty-nine measles virus strains were isolated from 189 specimens from 2002 to 2008, of which the C terminals of N gene were all de-tected positive. Among the 69 strains of measles virus isolates, 98.55% (68/69) belonged to Hla sub-geno-type which was the predominant sub-genotype, and only one strain (1.45%) belonged to H1b sub-genotype by RFLP analysis which was in accordance with the results by DNA sequencing method. Phylogenetic tree analysis indicated the H1a sub-genotype measles virus strains should be further divided into 2 clades, and the variation fluctuated between 0.2% and 3.8%. There were transmission chains caused by different virus strains co-cireulation. Conclusion A genotype, H1a and H1b sub-genotype can be identified by RT-PCR-RFLP assay specically based on the restriction enzyme Bcn I .The RT-PCR-RFLP assay can be a rapid, simple, accurate and efficient method for large-scale surveillance of measles virus strains in China.
10.Clinicopathology, prognosis and EG-1 mRNA in breast cancer

Objective To investigate the correlation between the expression of EG-1 in breast cancer and the clinicopathological factors and prognosis of breast cancer.Methods EG-1 mRNA expression in 72 malignant and 18 benign breast tissues were evaluated by RT-PCR method,and its correlation with clinical characteristics and prognosis were retrospectively analyzed.Results EG-1 expression level was higher in malignant tissue than in the corresponding benign breast tissue(71%vs.24%,P<0.05).EG-1 expression was significantly correlated with lymph node metastasis and protein VEGF (P<0.05)and was not significantly with age,menopause,tnmor size,grade,hormone status,protein Her2 and pS3.The metastastic rate and recurrence rate in EG-1 positive patients was higher than that in EG-1 negative patients.Multivariate Cox model showed that EG-1 positive was an independent factor affecting overall survival and disease free survival.Conclusions EG-1 expression was up-regulated in breast cancer and significantly correlated with lymph node metastasis and protein VEGF.
