2.Statistical analysis of papers in Chinese Journal of Applied Physiology from 2009 to 2013.

OBJECTIVETo analyze the status and papers information of Chinese Journal of Applied Physiology, and provide a guide for authors, readers and editing staffs.
METHODSWe retrieved papers of Chinese Journal of Applied Physiology from 2009 to 2013 from CNKI, and performed a statistical analysis of papers information contained in the text using bibliometrics method. Papers information such as document type, found support, author affiliation type, and discipline classification, etc. was analyzed.
RESULTSSeven hundred and forty two papers including 27 full English articles in total were published in Chinese Journal of Applied Physiology from 2009 to 2013. More papers were supported by national, provincial, and the other foundation, it was about 82.6% of all papers.
CONCLUSIONChinese Journal of Applied Physiology has stable manuscript resources, various discipline papers, and play an important role for development of applied physiology in China.

3.Adeno-associated Virus Serotype 2 Mediated Transduction and Expression of The Human β-Globin Gene in Human Early Fetal Liver Hematopoietic Cells

β-Thalassemia is an inheritance anaemia disease due to the defect in β- globin gene. Gene therapy is considered to be the only method which could cure this disease. Adeno-associated virus type 2 (AAV2) has benn gaining more attention as a vector in human gene therapy for its non pathogenic character and broad host range. Although, the efficacy of recombinant AAV2 (rAAV2) in transducing human hematopoietic stem cells has been investigated by researchers, the results were varied from different laboratory. The view was proposed recently that it may be resulted from helper virus in their packaging system. Respecting this, the packaging system without helper virus was used to produce rAAV2. Human early fetal liver hematopoietic cells not only possess many superior peculiarity compared to hematopoietic cells of bone marrow or cord blood, but also the inherent β-globin gene in the cells is not expressed. Studies on the AAV2 transduction of human fetal liver hematopoietic cells and mediated expression of β- globin gene in vivo were performed and the potential role of AAV2 in β-thalassemia gene therapy was analyzed. The rAAV2 containing a normal human β-globin gene (rAAV2-β-globin) without helper virus contamination were produced. The viral titer, purity and the ability of mediating expression of β-globin gene were detected in vitro. Then, human early fetal liver hematopoietic cells were isolated and were further transducted with the rAAV2-β-globin, followed by transplantation into sublethally irradiated BALB/C nude mice to analyze the β-globin gene expression. The results showed that the high titer and purity of rAAV2-β-globin had the ability of mediating β-globin gene expression in vitro. In 8 recipient BALB/C nude mice, the β-globin gene expression were detected in the 2 mice marrow by RT-PCR. The results suggested that rAAV2 could transduce human fetal liver hematopoietic cells and mediate the β-globin gene expression in BALB/C nude mice, meanwhile the expression level of the gene was still rather low. It is necessary to perform further research on AAV2 biology before applying in β-thalassemia gene therapy.

6.Comparison of Different Extraction Methods of Genomic DNA of Cordia Dicholoma Seeds

Objective To extract high quality genomic DNA of Cordia dicholoma seeds by using different methods;To provide references for researches on genomic DNA of Cordia dicholoma seeds. Methods Genomic DNA of Cordia dicholoma seeds was extracted through improved CTAB method and improved SDS method. Purity and concentration of obtained DNA were detected by spectrophotometry and agarose gel electrophoresis. Results The results of spectrophotometry showed that the purity of genomic DNA obtained through improved CTAB method was better than improved SDS method. Genomic DNA extracted through improved CTAB method was without protein and RNA pollution. The results of agarose gel electrophoresis showed that electrophoresis of genomic DNA obtained through the two methods both had the main belt. However, genomic DNA extracted through improved SDS method degraded more than improved CTAB method. Conclusion Improved CTAB method can obtain relatively high quality genomic DNA of Cordia dicholoma seeds.
8.CT-guided percutaneous interstitial implantation of ~(125)I seeds into the pancreas: an experimental study in pigs

Objective To investigate the feasibility and safety of percutaneous interstitial implantation of ~(125)I seeds into the pancreas of pig under CT-guidance. Methods Twelve healthy pigs were equally divided into 6 groups.~(125)I seed implantation into the pancreatic tail under CT-guidance was performed in pigs of study groups (group A-E), while ghost seeds that contained no radioactive materials were used in the control group (group F). Imaging examination and laboratory tests, including serum amylase, hepatic and renal functions, were conducted before and 1, 7, 15, 21, 30, 60 days after the procedure. Every two pigs (group A-E) were sacrificed each time at 15, 30, 45, 60, and 75 days after treatment, and specimens of pancreas, duodenum, liver, kidney, etc. were collected and sent for pathologic examination. Results The ~(125)I seeds were successfully implanted in all pigs. During the follow-up period, no severe complications occurred. Imaging and pathologic studies demonstrated that in study groups necrosis of pancreatic tissue appeared around the implanted ~(125)I seeds in 15 days, the necrosis area increased significantly in 45 and 60 days, and in 75 days the necrosis size remained quite the same as seen in 60 days. No necrosis was found in the control group (group F) 60 days after treatment. No serious complications, such as effusions, hemorrhage or necrosis of the adjacent duodenum, stomach, liver or kidney, occurred 75 days after the treatment. Conclusion Percutaneous interstitial implantation of ~(125)I seeds into the pig's pancreas under CT-guidance is safe and feasible.
9.Comparison of therapeutic effects between CT-guided ~(125)I seed implantation and chemotherapy with Gemcitabine for the treatment of unresectable pancreatic carcinoma

Objective To compare the therapeutic effects of CT-guided ~(125)I seed implantation with simple Gemcitabine chemotherapy for the treatment of unresectable pancreatic carcinoma. Methods Forty-six untreated patients with unresectable advanced pancreatic carcinoma were randomly divided into two groups: group A (n = 22), treated with ~(125)I seed implantation and group B (n = 24), treated with intravenous chemotherapy using Gemcitabine only. The clinical benefit response (CBR), objective tumor response (OTR), safety and the improvement of living quality were evaluated and analyzed. Results Three months after the treatment, the OTR rate of group A and group B was 45.5% and 8.33% respectively (P < 0.05). The CBR rate of group A and group B was 47.1% and 25% respectively (P < 0.05). No significant difference in PFS between group A and group B (P > 0.05). And also, the incidence of hematological toxicity and complication between two groups were of no significant difference. Conclusion For the treatment of advanced unresectable pancreatic carcinoma, both simple Gemcitabine chemotherapy and ~(125)I seed implantation are able to obtain a moderate objective response, although ~(125)I seed implantation seems to be more effective than Gemcitabine in improving the living quality and survival rate.
10.Effects of acitretin on the level of interleukin-8(IL-8)in the peripheral blood of patients with pustular psoriasis

Objective To investigate the changes of IL-8 in peripheral blood of patients with pustular psoriasis after acitretin treatment.Methods Dual antibody sandwich enzyme-linked immunosorbent assay (ELISA)was performed to measure the IL-8 levels in the peripheral blood of 30 patients with pustular psoriasis and 30 patients with psoriasis vulgaris before and after treatment,and in 30 normal human controls.The relationship between the IL-8 level and disease severity was assessed for patients with pustular psoriasis.Results After acitretin treatment,the condition was improved greatly in patients with pustular psoriasis,together with a significant decrease in the level of IL-8 in the peripheral blood(57.07 ± 12.02 pg/ml vs.96.84 ± 14.68 pg/ml,P < 0.05).Conclusion IL-8 may be involved in the therapeutic mechanism of acitretin in pustular psoriasis.
