1.EFFECT OF 1?, 25-DIHYDROXY-VITAMIN D_3 ON DIFFERENTIATION AND PROLIFERATION OF A HUMAN MYELOID LEUKEMIA CELL LINE

1 ? 25-dihydroxy-vitamin D3(1? 25 (OH)2 D3)induced differentiation of human promyelocytic leukaemia cells ( HL-60 ) into mature myeloid cells in vitro. The ratio of nuclei to cytoplasma decreased; Their nucleoli reduced; Cells became irrgular and extended the pseudopods. 1 ? 25 (OH ) 2 D3 caused significant increase of nitroblue tetrazolium ( NBT ) reduction and ?-non-specific acid esterase (?-NAE ) and acid phosphatase (ACP ) activities. These data and morphological characteristics suggest that HL-60 cells were conclusively identified as monocytes/macrophages. The histogram of DNA distribution ahylyzed by flow cytometry demonstrated G0 + G1 phase increased and S phase increased and S phase decreased remarkably after treatment with l?, 25 ( OH ) 2 D3 as compared with untreated cells.
2.Study on Quality Criteria of Fever and Cough Syrup for Children

OBJECTIVE:To establish the quality criteria of fever and cough syrup for children.METHODS:Thin-layer chromatography(TLC) was employed to make qualitative determination of herbba ephedrae,radix scutellariae,radix glycyrrhi-zae in fever and cough syrup for children,and high performance liquid chromatography(HPLC) was used to determine the con-tent of amygdaloside in its chief medicinal material,almond.RESULTS:The spots in the TLC of herbba ephedrae,radix scute-llariae,radix glycyrrhizae were in the same color with those in the chromatograms of control articles.The linear range of amygdaloside was 0.198?g~4.954?g(r=1.0 000).CONCLUSION:The present method is simple,accurate sensitiveness and highly reproductive,which can be used for the quality control of this preparation.

Complete surgical resection is very important for the outcome of cancer treatment and prognosis.Radionuclide imaging and optical imaging are effective methods for intra-operative guidance of tumor exploration and resection because of their capability in accurate tumor localization and margin delineation.With the development of more specific molecular probes and more sensitive detectors,advanced imaging methods that combine the advantages of both optical and radionuclide imaging,such as Cerenkov luminescence imaging,will be expected to play a more important role as intra-operative navigating agents for surgical oncology in the near future.
4.Molecular imaging of tumor by target microRNAs

MicroRNAs (miRNAs) are a novel class of small noncoding RNAs that regulate gene expression by targeting mRNAs for either cleavage or translational repression.Accumulated studies show that miRNAs aberrantly expressed in tumors are closely related with tumorogenesis and could be potential tumor biomarkers.It will be helpful in the diagnosis,prognosis and(or) predicting treatment response of tumor by imaging miRNAs expression.Several molecular imaging methods,including the optical imaging,radionuclide imaging and multimodality imaging,have been successfully used to visualize miRNAs,miRNAs,a new molecular imaging target,may explore a new field for tumor diagnosis and therapy.
5.Effect of high-flux hemodialysis on advanced oxidation protein products in maintenance hemodialysis patients

Objective To investigate the effect of high-flux hemodialysis (HFHD) on advanced oxidation protein products (AOPPs) in maintenance hemodialysis (MHD) patients.Methods 40 MHD patients (MHD group),50 non-dialytic chronic kidney disease (CKD) patients (CKD group) and 31 healthy subjects undergoing health examination survey received AOPPs detection.After lowflux hemodialysis (LFHD) for 3 months,HFHD was performed in MHD group for 3 months.Plasma AOPPs and other biochemical indicators were tested at the end of 0,3 and 6 months.AOPPs levels were detected in HFHD and LFHD dialysates.Results Plasma AOPPs level was higher in MHD group than in CKD and control groups,and plasma AOPPs level was higher in CKD group than in control group.Compared with before LFHD in MHD group,AOPPs and triglyceride (TG) levels had no significant differences after LFHD for 3 months(t=0.831,-0.191,both P>0.05),while plasma cholesterol (CHO) and β2 MG levels were increased in MHD group after LFHD for 3 months(t=-2.697,10.39,P<0.05 or 0.001).There were significant differences in plasma AOPPs,TG,β2-MG and CHO levels before versus after HFHD for 3 months (t=4.292,2.271,8.297 and 2.295,P <0.001 or 0.05).No AOPPs were detected in HFHD and LFHD dialysates.Conclusions Plasma AOPPs levels are increased in CKD patients.LFHD can not effectively eliminate AOPPs,while long term HFHD can decrease the plasma levels of AOPPs,TG and CHO,and reduce β2-MG accumulation.

Dexamethasone is a synthetic glucocorticoid that is widely used in clinical due to its multiple pharmacological effects. Recently, dexamethasone is increasingly utilized in anti-cancer therapy. It is frequently used to prevent side effects of chemotherapy such as nausea, vomiting and pain, as well as to increase the anti-tumor activity of the cancer chemotherapeutic agents as a chemosensitizer and to inhibit tumor growth as an anti-cancer agent in some certain cancers. Dexamethasone produces the effects in anti-inflammation, anti-angiogenesis, control of estrogen activity and so on, by binding to glucocorticoid receptor to regulate gene expression of some important bio-signal molecules. Those signal pathways could interfere with the transcription of various factors which can regulate proliferation, invasion and metastasis of tumors.

8.Protective effects of total salvianolic acids on cerebral hypoxia in mice

Aim The protective effect of total salvianolic acids (Sal) on cerebral hypoxia inmice was studied. Methods Acute cerebral hypoxia was induced by sodium nitrite scand decapitation.The effect of Sal on acute cerebral hypoxia in mice and neuronalhypoxia injury induced by sodium dithionite in primary cultures were ob-served. Results Sal in the doses of 10, 20 mg?kg-1 iv protected mice against theacute cerebral hypoxia and inhibited the production of lipid peroxidation in brain tis-sue of mice caused by cerebral hypoxia. Sal in the doses of 1~10 ?g? L-1 reduced therate of cell death and the content of MDA and lowered LDH content in extra-cellularbathing media in oxygen deprived cortical cultures.Conclusions Sal protects miceagainst cerebral hypoxia by suppressing the generation of lipid peroxide.
9.Study on correlations between contents of gastrodine and ecological factor in gastrodia rhizome vegetatively

Objective: To study the effect of ecological factors on contents of gastrodine, preliminary discuss the relations between the soil ecological factor and the quality of gastrodine. Methods: To collected wild and cultivated gastrodia rhizome and growth soil sample from the different habitat in different harvesttime. To examine microorganism quantity, the contents of soil organic matter, the contents of soil metallic, the soil pH value and the contents of gastrodine in the soil, and analyze the relations between the soil ecological factor and the quality. Results: In collection samples, it’s showed negative correlation (P0.05). Conclusion: The bacteria/fungus constitution condition, the contents of soil organic matter and the soil pH value were important ecological factor that affect the gastrodia quality.
10.The influence of dexmedetomidine intrathecal injection on formation of chronic neuropathic pain in rats

Objective To study the effect of dexmedetomidine(DEX)intrathecal administration on formation of chronic neuropathic pain induced by chronic constriction injury(CCI)in rats. Methods One hundred and twenty healthy adult male Sprague-Dawley(SD)rats were divided randomly into four groups(each n=30):sham group, model group,30μg/kg and 60μg/kg DEX intrathecal injection group(D30 group,D60 group). The CCI model was installed by left sciatic nerve ligaturing,sham surgery was done by exposing the sciatic nerve without ligation,and 30 μg/kg,60 μg/kg DEX(each,10 μl)and 10 μl normal saline were given intrathecally in D30,D60 and model groups respectively,all kinds of injection being once a day for 7 days. Hind paw mechanical contraction reflex threshold(MWT),heat-shrinkable reflex latency(TWL)and the score of motor dysfunction of hind extremity in rats were recorded on 1 day before ligation and 1,3,7 days after operation,and before ligation and 1,7,14 and 21 days after operation,the contents of tumor necrosis factor-α(TNF-α),interleukin(IL-1βand IL-6)in intumescentia lumbalis were measured by enzyme linked immunosorbent assay (ELISA). Results Compared with before operation,there were no significant differences in values of MWT,TWL,the motor function evaluation,TNF-α, IL-1β,and IL-6 at each time points(all P>0.05);however,with the prolongation of time,MWT and TWL were decreased,and the motor function evaluation,TNF-α,IL-1β,and IL-6 were increased in all the other groups. Compared with those of the sham group,MWT and TWL were declined,and the motor function evaluation,TNF-α, IL-1β,and IL-6 were elevated in model group. Compared with those of model group,30μg/kg and 60μg/kg DEX could significantly raise MWT(g)and TWL(s),obviously improve motor function and remarkably decrease the contents of TNF-α(pg/mg),IL-1β(pg/mg)and IL-6(pg/mg)in the spinal cord of CCI rats from 1 day after operation. And the changes in 60 μg/kg DEX group were more significant than those in 30 μg/kg DEX intrathecal injection group〔postoperative 1 day MWT:39.3±1.3 vs. 20.3±2.2,TWL:10.9±0.4 vs. 8.2±1.1,motor function score:2.00±0.00 vs. 2.00±0.75,TNF-α:33±7 vs. 125±18,IL-1β:108±12 vs. 203±34,IL-6:156±39 vs. 405±75,all P<0.05〕. Conclusions The DEX intrathecal administration has certain degree of dose-dependent therapeutic effect on hyperalgesia in CCI rat models. The mechanism is related to the amelioration of inflammatory reaction at the lumbar segment of spinal cord.
