1.Interventional chemoembolization through hepatic artery and superior mesenteric artery for primary hepatocellular carcinoma:a control study of 21 cases

Objective To investigate the clinical efficacy, the toxicity and side reactions of interventional chemoembolization with FOLFOX4 regimen through both hepatic artery and superior mesenteric artery, i.e. dual access technique, in treating primary hepatocellular carcinoma. Methods Between November 2010 and March 2013 at authors’ hospital, a total of 21 patients with advanced primary hepatocellular carcinoma (the study group) were treated with FOLFOX4 regimen by using dual access interventional technique. FOLFOX4 regimen included hepatic arterial infusion of 5-fluorouracil 400 mg/m2, hepatic arterial chemoembolization with iodipin and oxaliplatin 85 mg/m 2, intravenous administration of calcium folinate 200 mg/m2 IV on the first and second day, trans-superior mesenteric artery continuous infusion (lasting for 22 hours) of 5 -Fuorouracil 600 mg/m2 on the first and second day. During the same period other 21 patients with primary hepatocellular carcinoma were selected (used as the control group) to receive conventional hepatic arterial chemoembolization. In both groups, the treatment was repeated after 4-6 weeks. The therapeutic effect and the toxicity and side reactions were evaluated after the second treatment. Results The effective rate for the study group and the control group was 61.9% and 28.6% respectively, and the median survival time for the study group and the control group was 14.7 months and 9.4 months respectively. The differences in the effective rate and the median survival time between the two groups were statistically significant (P = 0.030 and P = 0.034). The occurrence of toxicity and side reactions, such as digestive tract reactions and the damage of liver function, in the study group were strikingly lower than those in the control group. Conclusion Through dual approach of hepatic artery and superior mesenteric artery catheterization, interventional chemoembolization with FOLFOX4 regimen is outstandingly effective for primary hepatocellular carcinoma, meanwhile, the side effects are very slight.
2.The analysis of misdiagnosis and mistreatment in Budd-Chiari syndrome with hepatic vein obstruction

Objective To investigate the clinical symptoms and imaging features of Budd-Chiari syndrome with hepatic vein obstruction (HVBCS) and the reasons of mistreatment. Methods Thirteen patients with HVBCS were misdiagnosed and mistreated as inferior vena cava (IVC) obstruction,including 8 patients treated with stent implantation in IVC once and 5 patients with balloon dilatation. After analysis of the clinical symptoms,signs and imaging features; hepatic vein obstruction was further confirmed by digital subtraction angiography (DSA)in all patients. Results All patients had variable degrees of portal hypertension and no apparent symptoms of IVC obstruction. CT or / and MRI showed obvious caudate lobe enlargement and DSA showed IVC narrowing with external compression. All patients were undertaken hepatic vein angiography including 4 with PTV and 9 with hepatic vein stent implantation. All patients' clinical symptoms and signs completely disappeared or markedly improved after the procedure. Conclusion The stenosis of IVC in HVBCS,caused by compression compensatory hypertrophy of hepatic caudate lobe can be cured by hepatic vein angioplasty which is the most correct and effective method.
3.Study on the in vitro Cell Uptake and Toxicity of Resibufogenin-loaded PLGA-TPGS Nanoparticles

OBJECTIVE:To study the in vitro uptake of Resibufogenin(RBG)lactic acid glycolic acid copolymer-water solu-ble vitamin E (PLGA-TPGS) in human liver cancer HepG2 cells,mouse ascites-type lymphatic metastasis of tumor HCa-F cells, and the toxicity on HepG2 cells. METHODS:RCPTN loading RBG and coumarin-6(C6)were prepared. Fluorescent inverted mi-croscope was used to observe the in vitro uptake by RCPTN HepG2,HCa-F cells. It was divided into negative control group,blank PLGA-TPGS nanoparticles(EPTN)group,5-fluorouracil solution(FS)group,RBG solution(RS)group,RBG/PLGA nanoparti-cles(RPN)group and RPTN group. WST-1 was conducted to investigate the optical density at 450 nm wavelength of HepG2 cells after 24,48,72 h incubated by FS,RS,RPN and RPTN with different final concentrations (1.25,2.5,5,10,20 μg/mL);the cell viability (CV) and half inhibitory concentration (IC50) were calculated. RESULTS:RCPTN distributed around the nucleus of HepG2,HCa-F cells. CV was decreased by RBG concentration increased in RPN group and RPTN group,and decreased by time prolonged;compared with FS group,CV in RPTN group was decreased(P<0.05 or P<0.01). IC50 of HepG2 cells incubated by FS,RS,RPN and RPTN was decreased by time prolonged,ordered by RS>FS>RPN>RPTN;IC50 incubated by RPN and RPTN for 48,72 h was obviously less than that of FS and RS(P<0.05 or P<0.01). CONCLUSIONS:RPTN can deliver RBG in-to HepG2,HCa-F cells,showing inhibition effect on HepG2 cells which is stronger than RPN,RS and FS.
4.Cassae-type diterpenes from seeds of Caesalpinia minax.

Fifteen cassaen-type diterpenes were isolated from the 95% ethanolic extract of the seeds of C. minax through various chromatographic techniques. Their structures were identified on the basis of spectroscopic data as pulcherralpin (1), caesalpinin ML (2), chamaetexane C (3), chamaetexane D (4), 6β, 18-diacetoxycassan-13, 15-diene (5), neocaesalpin K (6), neocaesalpin MP (7), neocaesalpin M (8), neocaesalpin Q (9), neocaesalpin P (10), neocaesalpin R (11), caesaldekarin D (12), caesaldekarin A (13), caesaldekarin b (14), 3β,6α-diacetoxyvouacapane (15). Among them, compounds 14, 9-11 were isolated from this plant for the first time.

5.The expression and clinical significance of multi-genes in breast cancer

Expression of MDR1 had a positive ~correlation with mutant p53 accumulation and HER2 expression(P0.05 ).In univariate analyses,TNM staging, axillary lymph node metastasis, mutant p53 accumulation, and HER2 over-expression were negatively correlated with DFS and OS, and MDR1 over-expression significantly reduced OS but not DFS. In multivariate analysis, axillary lymph node metastasis, over-expression of MDR1 and HER2 were independent risk factors for prognosis. Conclusions ~Induction of multidrug resistance and poor response to chemotherapy and endocrinotherapy may be the chief reasons for poor prognosis of breast cancer with mutant p53 accumulation, and HER2 and MDR1 over-expression. ~Determination of the above genes′expression in breast cancer tissue can be of use in deciding the degree of ~malignancy , metastasis phenotype and prognosis of brest cancer. Increasing anthracycline dose may increase the ~overall response rate to chemotherapy and improve prognosis in patients with mutant p53 accumulation, HER2 and MDR1 over-expression, especially HER2 over-expression.
6.HBsAg/HBsAb double positive hepatitis B virus infection model in vitro and in vivo.

The pathogenesis of HBsAg (+)/HBsAb (+) double positive hepatitis B virus infection was investigated by simulating HBsAg/HBsAb coexistence in vitro and establishing HBsAg/HBsAb double positive model in vivo. Eukaryotic expression plasmids PCI-SY, PCI-adw, PCI-adr, PCI-ayw, which expressed S gene product of different serotypes, were constructed and transfected into HepG2 cells. Recombinant proteins were purified from the transfected cells. At the same time, HBsAg mouse antiserum was obtained by immunizing mice with PCI-SY plasmid. HBsAg/HBsAb coexistence was simulated using these antigens and antiserum. Furthermore, the expression plasmids expressing different serotypes of S gene product including PCI-adw, PCI-adr, and PCI-ayw were injected into mice via tail vein. HBsAg and HBsAb in mice sera were tested at the first and 7th day respectively after antigen plasmids injection. Both in vitro simulation and in vivo animal models demonstrated that HBsAg antigen and HBsAb of the same serotypes could not coexist, but HBsAg antigen and HBsAb of different serotype could coexist. HBsAg/HBsAb double positive hepatitis B virus infection could be due to infection of viruses of different serotypes.
7.Effect of TSLC1 gene on proliferation, invasion and apoptosis of human hepatocellular carcinoma cell line HepG2.

The recombinant plasmid pCI-TSLC1 carrying TSLC1 gene was stably transfected into human hepatocellular carcinoma cell line HepG2. Cell proliferation was analyzed by MTT assay. The ability of migration was determined by transwell and FACSort flow cytometry was used to detect the cell cycle distribution and apoptosis. Western blotting revealed that H4 expressed higher amounts of TSLC1 protein than H15 and H0 did. The growth of TSLC1-transfected cells was significantly suppressed in vitro, and the ability of migration was reduced as well. The re-expression of TSLC1 could induce cell apoptosis. It was concluded that TSLC1 strongly inhibited the growth and ability of migration of HepG2 cell line in vitro and also induced apoptosis, suggesting that TSLC1 could reduce the tumorigenicity of human hepatocellular carcinoma cell line HepG2 in vitro, which provided a basis for further exploring the roles of TSLC1 in hepatocellular cellular carcinoma.

8.Influence of Bupleurum Liver-Coursing Powderon behavior,neuronal apoptosis and autophage in hippocampus of depressive rats

Objective To observe the effect of Bupleurum Liver-Coursing Powder on the neuronal apoptosis and autophage in hippocampus of depression model rats, and to explore the mechanism of its anti-depression effect. Methods 60 male SD rats were randomly divided into control group,model group and drug intervention group (n=20).The depression model was produced by giving the rats chronic unpredicted mild stress.The rats in model group and control group received the same volume of normal saline,and the rats in drug intervention group received Bupleurum Liver-Coursing Powder solution on the basis of model.The depressional behavior was examined using sucrose preference test,tail-suspension test and Morris water maze.Flow cytometry was employed for the detection and quantification of the apoptotic cells in the hippcampus. The expressions of LC-3 and Beclin 1 were detected using Western blotting method. Results The pencertages of sucrose preference of the rats in model group and drug intervention group were significantly lower, while the tail-suspension immobility time and the excape latency time were higher than those in control group (P<0.05);the apoptotic rates of cells,the ratios of LC3-Ⅱ/LC3-Ⅰand the expression levels of Beclin-1 were higher than those in control group (P<0.05).Compared with model group,the percentage of sucrose preference of the rats in drug intervention group was increased (P<0.05),the tail-suspension immobility time and the excape latency time were decreased (P<0.05),and the apoptotic rate of cells,the ratio of LC3-Ⅱ/LC3-Ⅰand the expression level were also decreased (P<0.05).Conclusion Bupleurum Liver-Coursing Powder has significant anti-depression effect, which may be related to inhibiting the apoptosis and degrading the autophage of neuros.
9.Investigation of Capillaria hepatica infection in rodents from Wuhan section of Yangtze River marshland

Objective To understand the prevalence of Capillaria hepatica in rodents from Wuhan section of the Yangtze River marshland. Methods Rodents were trapped in Jiang an section of Wuhan marshland of the Yangtze River. The livers of the rodents were examined for pathological changes by unaided eyes and the liver tissues were examined for the eggs of C. hepati-ca by a microscope. Results According to the natural conditions the investigation was carried out in 6 survey areas. Each sur-vey area was placed with 60 mousetraps and all 360 mousetraps were recovered. A total of 31 rodents rodent density 8.61%were captured and examined including 24 Apodemus agrarius 3 Rattus norvegicus 4 Sorex caecutiens and C. hepatica eggs were found in 1 R. norvegicus 1/3 and not found in A. agrarius and S. caecutiens. Conclusion This study has documented a prevalence of C. hepatica in rodents from Wuhan section of the Yangtze River marshland where is a natural epidemic focus of ca-pillariasis hepatica.
10.Comparison in purification methods of the recombinant human cardiac troponin I

Objective To compare the two kinds of purification method for purifying recombinant human cardiac troponin I(cT-nI)to obtain the stable cTnI and promote the study of cTnI diagnosis standardization.Methods The cTnI inclusion body was ob-tained by the ultrasonic broken engineering,after washing by 2% Tritonx-100,2M urea,dissolved in 8M urea,then purified by the column refolding on CM-FF and the dilution refolding respectively.The cTnI yields were compared between the two kinds of meth-od and the stability at 4 ℃,20 ℃,-80 ℃ and on the freeze-dried condition was compared.Then the purification method to effi-ciently obtain the stable cTnI was established.Results The protein about 2 mg and 1.4 mg could be obtained by CM-FF on the col-umn refolding and the dilution refolding from 0.1 g of wet inclusion body,respectively.The former method had the short cycle and high efficiency.The cTnI purified by the column refolding on CM-FF was more stable at 4 ℃,20 ℃,-80 ℃ and on the freeze-dried condition.Conclusion The column refolding on CM-FF is more stable and highly efficient in purification of cTnI than the dilution refolding.
