2.Expression and significance of c-myc and CD24 in colorectal cancer

Objective To determine the expression of c-myc and CD24 in colorectal carcinoma,colo-rectal polyp and normal mucosa,and to explore the role and correlation of them in the carcinogenesis of colorec-tal carcinoma. Methods The expression of c-myc and CD24 in colorectal carcinoma(n = 60),colorectal ade-nomatous polyp(n = 45),colorectal hyperplastic polyp(n = 15)and the adjacent non-cancerous tissue(n =30)was observed by immunohistochemical assay. Results The positive rate of c-myc in colorectal carcinoma were 73. 3% ,significantly higher than that in colorectal adenomatous polyp 44. 4%( χ2 = 9. 016 8,P <0. 01),colorectal hyperplastic polyp 13. 3%(χ2 = 18. 215 9,P < 0. 01)and adjacent non-cancerous tissue 6. 7%(χ2 = 25. 133 0,P < 0. 01);the positive rate of CD24 in colorectal carcinoma was 76. 7% ,significantly higher than that in colorectal hyperplastic polyp 6. 7%(χ2 = 25. 133 0,P < 0. 01)and adjacent non-cancerous tissue 3. 3%(χ2 = 43. 107 4,P < 0. 01). c-myc expression in colon cancer was significantly correlated with cancer site(χ2 = 8. 352 3,P < 0. 01),lymph node metastasis(χ2 = 4. 275 1,P < 0. 05),differentiation (χ2 = 4. 115 3,P < 0. 05)and TNM stage(χ2 = 5. 739 9,P < 0. 05). CD24 expression in colon cancer was significantly correlated with cancer size(χ2 = 9. 333 6,P < 0. 01),lymph node metastasis(χ2 = 7. 693 0,P <0. 01),differentiation(χ2 = 5. 870 0,P < 0. 05)and TNM stage(χ2 = 4. 498 7,P < 0. 05). There was a pos-itive correlation relationship between CD24 and c-myc in colorectal carcinoma tissue(χ2 = 10. 824 9,r = 0. 39, P < 0. 05). Conclusion The expression of c-myc and CD24 are high in colorectal cancer,having a significant correlation with some of the clinicaopathological features. c-myc is likely to act as a downstream target gene of CD24 signaling pathway,whose expression is probably regulated by CD24 in colorectal carcinoma tissue.

Objective To evaluate the imaging characteristic and the value of application of breath hold two dimensional fast imaging employing steady state acquisition(FIFSTA) sequence, for aortic dissection(AD) magnetic resonance many time phases imaging. Methods To analyze retrospectively the manifestation of 5 cases with aortie dissection diagnosed, Bakey type Ⅰ,Ⅱ, Ⅲ, in 1, 1 and 3, all were examined using ECG triggered, two dimensional FIESTA sequence during end-expiratory breath hold. Scan with the long axes of aortic position(oblique position parallel with aortic arch) and transitional position. The results were analyzed by the soft of workstation AW4.2. Results The display rate of the true lumen and the false lumen of AD were 100% ,the display rate of membrane in aorta was 100% ,4 cases showed the hole of intimal tear,2 cases with thrombus,2 cases with aortic regurgitation,2 cases with ejecting in the hole of intimal tear. Conclusion Need not any contrast media, quickly acquired imaging, high contrast to noise ratio, least artifact,and especially show membrane in aorta and for the hole of intimal tear of aortic dissection(AD) magnetic resonance many time phases imaging with two dimensional FIESTA sequence.
4.Nitrogenous chemical constituents of Weiceng

Objective To investigate the nitrogenous chemical constituents of Weiceng .Methods Weiceng extract was subjected to various column chromatography and spectroscopic methods were used for the elucidation of compounds .Results Seventeen compounds were isolated and identified as cyclo-(Leu-Tyr)(1), cyclo-(Phe-Tyr) (2), cyclo-(Pro-Gly) (3), L-Pyroglutamic acid methyl ester (4), uracil (5), thymine (6), N-acetylphenylalanine (7), tyrosine (8), phenylala-nine (9), phenylalanine methyl ester (10), N-methyl leucine (11), isoleucine (12), valine (13), leucine (14), glutamic acid (15), glycine (16) and aspartic acid (17).Conclusion All the seventeen compounds are isolated from Weiceng for the first time .Before this study , cyclopeptides 1-3 have never been isolated from soy bean or its products .
5.Expression and significance of CD24 in colorectal cancer

Objective To determine the expression of CD24 in colorectal carcinoma, and to explore the relationship between CD24 and the clinicopathological features of colorectal carcinoma.Methods The expression of CD24 in 62 cases of colorectal carcinoma, 47 cases of adenomas, 15 cases of colorectal polyps and 30 cases of the adjacent non-cancerous tissues were observed by immunohistochemical assay.The relationship between CD24 and the clinicopathological features was analyzed.Results The positive rates of CD24 in colorectal carcinoma 72.6% and adenomas 63.8% were significantly higher than those in colorectal hyperplastic polyps 13.3% (x2 =17.83, P =0.00;x2 =11.61, P =0.00) and adjacent non-cancerous tissues 6.7% (x2 =35.15, P =0.00;x2 =24.64, P =0.00).The expression of CD24 in colorectal carcinoma had a significant correlation with the tumor diameter (x2 =5.48, P =0.02), tumor differentiation (x2 =8.86, P =0.00), Duke staging (x2 =11.47, P =0.00) and lymph node metastasis (x2 =8.92, P =0.00).Conclusion The expression of CD24 is high in colorectal carcinoma, having a significant correlation with the size of tumor, degree of differentiation, Duke stage and lymph node metastasis.
6.Effects of general anesthesia and epidural anesthesia on coagulation function during total knee replacement in the elderly

BACKGROUND:In perioperative period of total knee replacement in elderly patients, it is crucial to maintain the normal function of blood coagulation. However, many factors may influence coagulation function of patients in perioperative period. Of them, anesthesia is an important factor. Different anesthesia methods wil produce different effects on blood coagulation. Appropriate anesthesia methods should be selected in the clinic to maintain the stability of coagulation function. OBJECTIVE:To explore the effect of application of general anesthesia and epidural anesthesia in elderly knee replacement and the effects on the function of blood coagulation. METHODS:A retrospective analysis was performed on clinical data of 135 elderly patients after total knee replacement in Dongying Hospital of Shandong Provincial Hospital Group from September 2012 to September 2013. Al patients were divided into control group (67 cases;general anesthesia) and observation group (68 cases;epidural anesthesia) according to the mode of anesthesia. Coagulation indexes and D-dimer levels were observed before anesthesia, 6 hours after anesthesia, and 1 day after replacement in both groups. The incidence of deep venous thrombosis was measured and compared between the two groups in 12-month fol ow-up.
RESULTS AND CONCLUSION:Through the statistics and comparison, no significant difference was detected in blood coagulation indexes at different time points in the two groups (al P>0.05). However, significant differences in D-dimer levels were detectable between the two groups at 6 hours after anesthesia and in the morning at 1 day after replacement. D-dimer levels were significantly lower in the observation group than in the control group (al P<0.05). The incidences of deep venous thrombosis were 3%and 21%in the observation and control groups, respectively, showing significant differences (P<0.05). These results suggest that epidural anesthesia during elderly totak knee replacement obtained good effects, and could maintain stable coagulation function.
7.The clinical relevance between the serum level of pregnancy-associated plasma protein-A and degree of acute coronary syndrome

Objecitve To investigate the relationship between the level of pregnancy-assoiated plasma protein-A(PAPP-A) and high-sensitivity C-reactive protein(hs-CRP) in patiente with acute coronary syndrome(ACS) and the role of PAPP-A in predicting the stability of atherosclerotic plaques.Methods A total of 43 patients were selected as ACS group,25 patients as stable angina(SA) and 13 persons as control.ACS group ansisted of 20 patients with unstable angina(UA) and 23 patients with acute myocardial infarction(AMI).The peripheral venous blood samples of the both control and angina groups were drawn on the morning of the following day after hospitalization, and of the AMI group were drawn 24 hours after episode.The concentration of PAPP-A was detected by enzyme-linked immune sorbent assay(ELISA),and the concentration of C-reactive protein(CRP) was detected by scatter turbidimetry method.The concentration of PAPP-A and CRP in each group was compared.Results The plasma hs-CRP and PAPP-A levels in patients with AS increased significantly compared with those in UA and healthy volunteers; PAPP-A was significantly related with hs-CRP(r=0.308);using the method of multiple regression and correlation PAPP-A had linear relationship with hsCRP in all of the cases.Conclusion PAPP-A can play a role in evaluating the clinical stability of patients with coronary disease,and may serve as one of the biomarkers of vulnerable plaques.
8.Relationship between mast cells and renal interstitial fibrosis in interstitial nephritis

Objective To explore the potential relationship between tryplase-positive mast cells (MCs) infiltration and renal interstitial fibrosis in acute interstitial nephritis (AIN) and chronic interstitial nephritis (CIN). Methods Renal biopsy specimens from patients with AIN (n=11) and CIN (n=16) were studied and 11 patients in minimal change diseases (MM)were as controls. Histochemistry and immunohistochemistry staining assay were applied to delect the expression of tryptase, proteinase-activated receptor-2 (PAR-2), TGF-?1 and collagen type I (Col I )in the renal tissues. Immunofluo-rescence double-staining assay was used to assess the relationship among MCs, PAR-2-positive cells, and TGF-?1-positive cells in the renal interstitium respectively. Results MCs in AIN and CIN were significantly increased compared with those in controls and were mainly scattered in the fibrotic areas of renal interstitium. Objective To study the effect of adenoviral-delivered short hairpin RNA (shRNA) target against permeability glycoprotein (Pgp) as a new drug in anti-epileptic drug resistance epilepsy treatment and to evaluate its efficiency. Methods MDR Sprague-Dawley (SD) rat estrocyte model was induced by Coriaria Lactone (CL), mainly over-expressing mdrlb. To reverse the drug resistance, astrecytes were treated with constructed replication deficient adencvirus AdS-EGFP-shRNAI-U6 delivering short hairpin (shRNA) target agianst mdrlb gene. Total RNA and protein were extracted from the infected cells, mdr1 b level was detected by Quantitative Real-time PCR whereas Pgp by Western blot, Rhodamine123 (Rho123) efflux ratio by Flow Cytometry. Results AdS-EGFP-shRNA1-U6 was succesfully constucted with high virus titer of 6×1010 pfu/ml. The interference efficency of AdS-EGFP-shRNA1-U6 agianst mdrlb in rat astrecyte model was about 94%. The Rho123 efllux ratio was about 15. 8%, significiently lower than control group which was 56. 2% (F = 127.5, P < 0. 05). Conclusions Pgp over-expression has been successfully suppressed and MDR has been reversed, which may provide a premising approach for refractory epilepsy remedy.
