1.miR-155 is conductive to chondrogenic differentiation of bone marrow mesenchymal stem cells

BACKGROUND:It is discovered recently that miRNA is a new regulator that is able to have an impact on gene expression and miRNA contributes to proliferation, differentiation and self-renewal of pluripotent stem cels.
OBJECTIVE:To investigate the mechanism by which miR-155 regulates chondrogenic differentiation of bone marrow mesenchymal stem cels.
METHODS: Sixty healthy Sprague-Dawley aged 12 weeks were randomized into study group and control group, with 30 in each group. Under anesthesia, rats were sacrificed to harvest bone marrow of the lower limbs. Then bone marrow mesenchymal stem cels were isolated, cultured, and transfected with miR-155 mimics in the study group and a negative control sequence in the control group. After chondrogenic induction, RT-PCR was used to detect the expressions of Sox9, Colagen II, Aggrecan and Colagen X gene, and western blot assay to detect the expression of Sox9 and Runx2 proteins.
RESULTS AND CONCLUSION:Compared with the control group, the mRNA expressions of Sox9, Colagen II and Aggrecan were higher, but the mRNA expression of Colagen X was lower in the study group (P < 0.05); the protein expression of Sox9 was higher, but the protein expression of Runx2 was lower in the study group (P < 0.05). These findings indicate that miR-155 promotes the chondrogenic differentiation of bone marrow mesenchymal stem cels and moreover, it can suppress the hypertrophy trend of bone marrow mesenchymal stem cels differentiating into chondrocytes.

4.Temporal regulation of transcription factor Mef2c by histone acetylases during cardiogenesis.

OBJECTIVETo observe the temporal modification of transcription factor Mef2c by histone acetylases (HATs) P300, PCAF, and SRC1 during cardiogenesis and to provide a basis for investigating the pathogenesis of congenital heart disease.
METHODSThe normal heart tissues from embryonic mice (embryonic days 14.5 and 16.5) and neonatal mice (postnatal days 0.5 and 7) were collected. The binding of P300, PCAF, and SRC1 to Mef2c gene and level of histone H3 acetylation in the promoter region of Mef2c were evaluated by chromatin immunoprecipitation assays. Meanwhile, real-time PCR was used to measure the mRNA expression of Mef2c.
RESULTSP300, PCAF, SRC1 were involved in histone acetylation in the promoter region of Mef2c during cardiogenesis in mice, and binding of P300, PCAF, and SRC1 to the promoter of Mef2c varied significantly in different stages of cardiogenesis (P<0.01). The level of histone H3 acetylation and mRNA expression of Mef2c in the promoter region of Mef2c also varied significantly in different stages of cardiac development (P<0.01). The levels of acetylated H3, Mef2c mRNA, and HATs (P300, PCAF, SRC1) changed over time. They were highest on embryonic day 14.5 (P<0.01), decreased gradually with cardiac development, and were maintained at low levels after birth.
CONCLUSIONSThe mRNA expression of Mef2c varies during cardiogenesis in mice, which indicates that Mef2c plays an important role in the process of cardiac development. Meanwhile, histone acetylation in the promoter region of Mef2c is regulated temporally by HATs P300, PCAF, and SRC1.

5.Repair of postburn hand using scarred skin and an abdominal flap in patients with severe burns.

OBJECTIVETo introduce a method for repairing and restoring hand function of the patients suffering from large surface area burn with scarce normal skin for reconstruction.
METHODSSeven patients (ten hands) were treated in our department from April 1994 to February 2001. The TBSA involved with second- or third-degree burns was 85%-96%. All the hands had severe scar contracture on the dorsum and lost most of their function. A scarred skin flap, based on the ulnar border of the hand was elevated integrally on the dorsum. A random-pattern abdominal flap at a less-scarred area was designed and elevated to cover the defect of the hand while the scarred skin flap of the hand was transferred to the donor site of the abdominal flap.
RESULTSAll patients were followed for 0.5 to 4 years postoperatively. The range of motion of the metacarpophalangeal joint and the space capacity of the first web were greatly improved. All patients regained self-care ability.
CONCLUSIONSThis method is simple and has satisfactory results. Under the circumstances where normal skin was not available for reconstruction, the function of the burned hand could be greatly improved by this method.

6.Bioinformatics analysis for mmu-miR-3475-3P in cardiac development

Objective To investigate the mmu-miR-3475-3P possible participating in biology process and signal pathway.Methods The expression of mmu-miR-3475-3P in the mature mouse heart and in the embryonic mouse heart was measured by qRT-PCR.The target genes were predicted through comprehensively using the common microRNA on-line databases such as TargetScan,miRDB and miRanda,and then the obtained targeted genes were performed the gene function annotation and signal pathway analysis.Results There was significant difference between the expression of mmu-miR-3475-3P in the embryonic mouse heart and the expression in the mature heart.Bioinformatics analysis by using TargetScan,miRDB and MiRanda on miR3475-3P revealed that microRNA was likely to regulate 441 target genes.Conclusion mmu-miR-3475-3P is highly expressed in the embryonic mouse heart.The target genes predicted by mmu-miR-3475-3P are enriched in multiple signal pathways and cellular biological processes.
7.Improving effects of mesylate cinepazide on neurological function of focal cerebral ischemia in rats

Objective To investigate the effect of mesylate cinepazideon on focal cerebral ischemia infarction in rats .Methods Sprague -Daw-ley(SD) rats were randomly divided into six groups:sham group, model group, control group ( cinepazide maleate 29 mg? kg-1 ) , three doses test groups (mesylate cinepazideon,45,22.5,11.25 mg? kg -1).Twenty-four hours after administration , Zea -Longa method was used to assessment the neurological disfunction scores .Triphenyltetrazolium chlo-ride ( TTC) staining was used to measure cerebral infarction size .Histo-pathology were determined with hematoxylin - eosin staining. Results Compared with model group , the infarction area were signifi-cantly reduced in high , middle test groups ( 45 , 22.5 mg? kg -1 ) and control group.And neurologic function were improved in high , middle doses test groups.Conclusion Infarction area was reduced and neuro-logical function was improved on nerve cells after administration .
8.Protective effect of Rhubarb flavonoids on rats with diabetic nephropathy

Objective To explore the renal protective mechanism of Rhubarb flavonoids in diabetic rats. Methods Sixty SD rats were assigned into six groups: blank group, model group, control group ( Huangkui capsule, 675 mg? kg-1 ) , high, middle, low doses of test groups(Rhubarb flavonoids, 42,21,10 mg? kg -1).The microalbumin-uria, serum glucose, blood urea nitrogen, creatinine, total cholesterol and triglyceride levels were determined at the time of 90 days after administration.And renal tissues was observed by light microscopy. Results Compared with the model group, the rats in high, middle test groups ( 42, 21 mg? kg-1 ) significantly reduced the blood glucose, blood urea nitrogen, serum creatinine, urinary albumin and improve glomerular pathological structure of rat, whereas the low dose group were not significantly changed.Conclusions Rhubarb flavonoids has signifi-cant effects on rats with diabetic nephropathy for renal protective.
9.A method of measuring the displacement of the distal radioulnar joint on the three-dimensional CT imaging.

OBJECTIVESTo establish a clinical method for measuring the displacement of the distal radioulnar joint (DRUJ) precisely irrespective of ulnar variance, and to derive normal population translation references with palmar and dorsal stress.
METHODSThirty-seven normal distal forearms were scanned with computed tomography using an apparatus designed by Pirela-Cruz. Each extremity was scanned in two positions: maximal ulnar palmar and dorsal stress. The digital imaging and communications in medicine (DICOM) CT images were then imported into Mimics 10.0 for three-dimensional reconstruction. On the DRUJs 3D images, choose the most prominent point of the palmar and dorsal margins of the sigmoid notch and the excavate ulna fovea as the reference points A, B and C. A perpendicular line was then drawn from the point C to a line connecting points A and B with the intersection D. Calculate the ratio of AD/AB and DB/AB. Two observers measured all the DRUJs independently and one repeated the measurements one month later to determine the interobserver and intraobserver reliability.
RESULTSThe mean ratio values of palmar (AD/AB) and dorsal (DB/AB) translation were 0.39 ± 0.07 and 0.37 ± 0.07, and the normal references (x(-) ± 2 s) were from 0.25 to 0.50 and from 0.23 to 0.50, respectively. No significant differences were observed in terms of positions, genders and dominant hands. The intraclass correlation coefficient (ICC) values for interobserver and intraobserver reliability (DB/AB, AD/AB) were 0.84, 0.80, 0.93 and 0.92, respectively.
CONCLUSIONSThis new method could accurately measure the displacement of DRUJs with acceptable reliability, even with ulna positive or negative variance. Instability of DRUJ may be indicated when AD/AB is less than 0.25 or BD/AB is less than 0.23.

10.Cystic tumour of the atrioventricular node: a case report and review of the literature.

Cystic tumour of the atrioventricular node is a rare primary cardiac tumour that can cause complete heart block and sudden death. Here, we describe a male case aged 42 years who suddenly died without a medical and family history of cardiac illnesses. After detailed macroscopic and microscopic examinations, a cystic mass was found in the atrioventricular nodal region. The small lesion was less than 1 cm in diameter, and consisted of small and large cystic spaces and tubular structures lined by flat, cuboidal or squamous epithelium. Immunohistochemical staining revealed the tumour epithelium positive for epithelial membrane antigen, carcinoembryonic antigen, antigen epitopes AE1/AE3, cytokeratins CK5/6 and CK7, but negative for calretinin, HBME-1, Wilms' tumor 1, factor VIII, chromogranin, synaptophysin or smooth muscle actin, suggesting an endodermal rather than mesothelial origin.
