OBJECTIVETo explore the expression of p-p38 MAPK protein and the number of astrocytes expressing p-p38 MAPK in CA1 hippocampus in rats during the induction of brain ischemic tolerance induced by intermittent hypobaric hypoxia (IH) preconditioning.
METHODSThirty healthy adult male Wistar rats were randomly divided into 6 groups (n = 5 in each group): sham 0 min group, IH + sham 0 min group, sham 7 d group, IH + sham 7 d group, Ischemia (Is) 7 d group, and IH + Is 7 d group. Neuropathological evaluation was performed by thionine staining in CA1 hippocampus in rats. The expression of p-p38 MAPK in CA1 hippocampus was observed by immunohistochemical staining. And the number of astrocytes expressing p-p38 MAPK was observed by immunofluorescent double labeling.
RESULTSThe results showed that IH preconditioning induced brain ischemic tolerance successfully. At the same time, IH preconditioning obviously up-regulated the expression of p-p38 MAPK protein in CA1 hippocampus, and also increased the number of astrocytes expressing p-p38 MAPK.
CONCLUSIONIt might be concluded that IH preconditioning induced brain ischemic tolerance by up-regulating the expression of p-p38 MAPK protein in pyramidal neurones and astrocytes.

Objective To investigate the role of nuclear factor-kappa B (NF-κB)signaling pathway in propofol-induced suppression of up-regulation of inducible nitric oxide synthase (iNOS) gene expression in LPSstimulated RAW264.7 cells. Methods RAW264.7 cells were purchased from cell bank of Chinese Academy of Sciences and cultured in DMEM culture medium containing 10% fetal bovine serum. The cells were seeded in 6 cm diameter dishes (3 ml/dish) or in 6-well plates (2 ml/well) with a density of 5 × 105/ml and randomly divided into 3 groups ( n =18): normal control group (group C),group LPS (group L)and group LPS + propofol (group LP).The cells were incubated with LPS 1 μg/ml in groups L and LP.Propofol 50μmol/L was added to the culture medium at 2 h before LPS in group LP.Cells were harvested at 30 min after being stimulated with LPS.Phosphorylation of IκB kinase(p-IKK) and NF-κB activity were detected by Western blot. The expression of iNOS mRNA was determined after 6 h exposure of the cells to LPS.Results LPS significantly up-regulated the expression of p-IKK and iNOS mRNA and increased NF-κB activity in group L as compared with group C.Propofol pretreatment significantly attenuated the effects of LPS on p-IKK,iNOS mRNA expression and NF-κB activity. Conclusion NF-κB signaling pathway is involved in the propofol-induced suppression of up-regulation of iNOS mRNA expression in LPS-stimulated RAW264.7 cells.
5. Important role of Rho/ROCK2 pathway in the development of Alzheimer’s disease
Journal of International Pharmaceutical Research 2016;43(1):33-38
Rho/ROCK pathway is a ubiquitous singling pathway in organisms, and is involved in many biological processes. In the brain of Alzheimer’s patients, the activities of Rho and Rho associated coiled coil forming protein kinase(ROCK)are up-regulated, which is accompanied by the elevation of Aβ42 level, and the abnormal change of the morphology and function of neuronal processes, suggesting that the occurrence and development of Alzheimer’s disease(AD)is associafed with the overexpression and excessive activation of Rho or ROCK. Rho/ROCK2 pathway is considered a target pathway for the prevention and treatment of AD, and Rho or ROCK2 also becomes an important target for AD drug development. Numerous studies have revealed that suppressing the expression or decreasing the activity of Rho or ROCK2 can reduce Aβ42-induced neurotoxicity, protect neurons, and slow down the occurrence and de velopment of AD. Therefore, specific inhibition of ROCK2 has an important significance for the repair of central nervous system damage and the treatment of AD. This article reviews several effects of Rho/ROCK2 pathway on the development of AD.

Objective To investigate the imaging diagnosis of acetabular dysplastic coxarthrosis. Method The imaging of the acetabular dysplasia was analyzed in 51 patients (87 hips). All but four patients were women. The age ranged from 22 to 78 years, and the mean age was 42.6 years. Pelvis radiographs were studied in all cases, and CT scan was performed in 18 cases and MRI in 10 cases. Results The fundamental signs on the plain film included shallow acetabulum, increased obliquity and insufficient coverage of the femoral head by the acetabulum. The CE angles were determined in all the cases, ranging from -10? to 30?, mean 12.9?. The charp angles ranged from 35? to 67?, mean 45.3?. The secondary osteoarthritis were revealed in 75 hips (84.2%). 54 hips (62.1%) had cyst like lesions in weight bear area of the acetabulum or femoral head. 19 hips had subluxation of femoral head. CT and MRI displayed small cystlike lesions and forward shift of the femoral head, which could not be shown on plain film, respectivly in 13 and 10 hips. Conclusion The cyst like lesion of subarticular region is a common sign in acetabular dysplastic coxarthrosis. CT or MRI can show the early osteolytic lesion and forward subluxations.

Objective To evaluate the feasibility and clinical efficacy of laparoscopic nephron-sparing surgery without renal wound suturing. Methods From December 2012 to February 2014,9 patients (4 male,5 female) with renal tumor underwent laparoscopic nephron-sparing surgery without suturing in our hospital. The mean age in those patients was 56 years (range 42-72 years). The mass size ranged from 1.1to 3.8 cm (mean 2.4 cm).The location of tumor included 2 in left kidney,7 in right kidney.Meanwhile,6 tumors were found in the middle part of kidney,2 tumors were demonstrated in the lower part of kidney, and 1 tumor located in the upper part of kidney. The imaging examination confirmed that all tumors protruded the renal surface and infiltrated into the renal parenchyma about 0.1-1.2 cm,which could be defined as the peripheral renal tumor without the invasion of collective system. The retroperitoneal approach was used in all patients with 3 trocars.During the operation, the renal artery was exposed and fats surrounding or on the surface of the tumor were resected. The renal artery was occluded.Tumor excision was then performed using scissors. An incision was made at the point 0.5 cm away from the margin of the tumor, extending deeply to completely cut the tumor.Meanwhile, bleeding was controlled with bipolar diathermy. Then, the clamping of the renal artery was removed and the retroperitoneal pressure was reduced in order to check the site of bleeding. If the bleeding occurred severely, the kidney wound was sutured with 2-0 absorbable sutures.Otherwise, the coagulative glue was sprayed on the wound surface. Results In all the patients, the procedures were done without conversion to open surgeries. The renal wound suturing appeared in one case because the tumor infiltrated into the deep of the kidney and the bleeding could not be completely controlled by bipolar diathermy.Eight of 9 cases were successfully performed without suturing. The mean time of renal artery occlusion was 8 min (range 3-18 min).In our patients, the mean operation time was 120 min (90-180 min),and the mean blood loss was 60 ml (10-200 ml).None of the patients received blood transfusion during the surgery. The mean postoperative hospital stay was 7 days (5-9 days). No complications, such as urine leakage, bleeding occurred after the operation. All the patients were followed up with no recurrence of tumor. Conclusions Laparoscopic nephron-sparing surgery without renal wound suturing is feasible and safe for patients with the small and the depth of infiltration less than 0.5 cm tumor in the kidney.

The rate of corneal graft rejection is still high for high-risk keratoplasty although immune suppression drug is routinely used. The role of traditional Chinese medicine in corneal transplantation is concerned gradually.Heijingpaichitang on the prevention and treatment of rats with high-risk corneal allograft rejection needs further study. Objective This study was to investigate the inhibitory effect of heijingpaichitang on high-risk corneal transplantation immune rejection in rats. Methods Sixteen female SD rats were used as the donors and 32female Wistar rats were served as recipients. The high-risk corneal trasplantation models were established by corneal suture in 32 Wistar rats, and then homogeneity variant SD-Wistar corneal transplantation was performed. The recipients were randomized into model control group, cyclosporinc A (CsA)group, heijingpaichitang group and CsA +heijingpaichitang group.CsA,heijingpaichitang and CsA + heijingpaichitang was orally administered 4 days after operation once per day for 15 days, and normal saline solution was used at the same way in the model control group.Ocular anterior segment reaction was examined under the slit lamp and corneal opacification, edema and neovasculation were scored based on Larkin' s criteria.Rejection index of the corneal graft was recorded and the graft survival time was calculated. The pathological examination of the corneal graft was carried out in all rats, and the inflammatory cells in the corneas and CD4+ cells in the periphery blood were assayed using flow cytometry. The use of the animals complied with ARVO Statement. Results Corneal graft rejection occurred in 10 days after operation in the model control group,12-13 days in the CsA group and heijingpaichitang group and 22 days in the CsA +heijingpaichitang group.Compared with model control group, the scores of the corneal opacification, corneal edema and neovascularization were significantly lower in the CsA group, heijingpaichitang group and CsA+heijingpaichitang group (P<0.05),and all the scores were declined in the CsA+ heijingpaichitang group compared with CsA group and heijingpaichitang group(P<0.01),but no significant differences were seen in the scores between the CsA group and heijingpaichitang group(P>0.05).The mean survival time of grafts was (10.38 ±1.69)days in the model control group,(22.50 ± 3.07) days in the CsA + heijingpaichitang group, with the significant difference (t =-9.790,P =0.000).The pathological examination of graft showed that the lymphocytes and new blood vessels were less in the CsA+heijingpaichitang group compared with CsA group and heijingpaichitang group 15 days after operation.Flow cytometry verified that the number of lymphocytes in graft,CD4+cells and CD4+/CD8+ in periphery blood were significantly lower in the heijingpaichitang group,CsA group and CsA+heijingpaichitang group compared with model control group (P<0.05).Conclusions Heijingpaichitang can inhibit immune rejection to certain extent in high-risk corneal transplantation rat and has a similar effect to 0.1% CsA.Heijingpaichitang and 0.1% CsA have a synergistic effect.
