Objective:To quantify the tryptase and T cell immunoglobulin mucin domain-3(TIM-3)double positive mast cells in hu-man chronic periodontitis tissue using double immunofluorescence staining.Methods:25 healthy controls,28 chronic mild periodontitis and 30 chronic advanced periodontitis patients were included.The gingival specimens were stained with HE for histology,and with double immunofluorescence staining for the identification of tryptase and TIM-3 double positive mast cells in gingival tissue.Results:In chronic periodontitis tissue the degree of gingival inflammation was significantly increased,the densities(cells/mm2 )of tryptase and TIM-3 double positive mast cells were significantly increased(P<0.05),in addition,that in chronic advanced periodontitis group was significantly higher than in the chronic mild periodontitis group(P<0.05).Conclusion:Tryptase and TIM-3 double positive mast cells has the similar tendency as the severity of periodontitis inflammation in human periodontitis tissue.Tryptase and TIM-3 double positive mast cells may play an important role in human chronic periodontitis.

Objective To understand the situation of eosinophils absolute value and percentage in the population of occupational health examination in Changsha City ,and to analyze the distribution situation of physical examination abnormal results of eosino‐phils absolute value(EOS) and percentage(EOS% ) .Methods The blood cell analysis results and physical examination abnormal manifestations in 22 135 cases of occupational health examination were statistically analyzed .Results Among cases of EOS absolute value and EOS% increase ,the majority had the changes of EKG abnormality ,fatty liver ,etc .the minority were reactive and clonal increase ,meanwhile the cases of unknown cause eosinophils increase also accounted for a large proportion .Conclusion It is sugges‐ted that a large number of experiments and clinical studies should be carried out for further investigate the significance of eosino‐phils increase in the occupational health examination .

Objective To explore the surgical indication,diagnosis and treatment of suspected pancreatic cystic lesions.Methods To-tally 341 patients were admitted into our hospital because of pancreatic cystic lesions from October 2010 to October 2015.Except the 278 confirmed cases,the clinical data of the rest 63 patients with vague diagnosis were retrospectively analyzed.Results The surgical indication of pancreatic cystic lesions were:the lesion diameter was more than 4 cm;the imaging diagnosis was malignant;the lesion was combined with obvious clinical symptoms which can not explain with other diseases;there were asymptomatic pancreatic cystic lesion and main pancre-atic duct dilation without surgical risk factors;the follow-up showed that the desease is in development.Endoscopic ultrasonography guided fine needle aspiration is a useful supplement for more accurate diagnosis.However,it is an uncertain diagnostic value currently.Conclusion Diameter of lesion,uncertain imaging diagnosis,severe abdominal symptoms,obstructive jaundice and abnormal serum tumor markers are the surgical indication for patients.

Objective To verify the indicator reference value ranges of blood cells analytic report by quoting the WS/T 405-2012 industry standard in Changsha area and to establish the reference value ranges of research parameters suitable for local area.Methods The blood cells analytic results in 3138 cases of reference group were analyzed according to the program of the Formulation of Reference Intervals in Clinical Laboratory Detection Items and the results were compared with the WS/T 405-2012 industry standard.Results WBC,Neu#,Mon%,MCHC,PLT,MPV,PCT,P-LCC,P-LCR and InR‰ in the report parameters and WBC-D,PDW-SD in the research parameters had no statistical differences between males and females(P>0.050);WBC,Neu#,Mon%,MCHC and PLT had no difference between genders,the indicators were narrowed compared with the reference value ranges in standard;the reference value ranges of Neu%,RBC,HGB,MCV and MCH in the male blood cells analytic indicators were narrowed,the lower limits of Bas%,Lym#,Mon# and HCT reference value ranges were elevated,the upper limits of Bas%,Lym#,Mon# and HCT reference value ranges were elevated,while the upper limits of Eos# and Eos% reference value ranges were decreased,the Lym% lower limit was decreased;Neu%,Lym#,Mon# and RBC in the female blood cells analytic indicators were narrowed,the lower limits of Bas# and Bas% were increased,the lower limits of Eos%,Lym%,HGB,MCV and MCH were decreased,the upper limits of Eos#,Eos%,Lym%,HGB,MCV and MCH were decreased and the HCT upper limit was elevated.Conclusion The Changsha area should have the reference value ranges of selective quoted standards and establishes the reference value ranges suitable for local area or laboratory.

Objective To explore the dynamic changes and significance of heart rate variability(HRV)during the initial phase after acute exposure to Tibet plateau by combining time,frequency domain and non-linear analysis.Methods Eighty-six healthy young men of Han nationality underwent short-term(5 min)heart rate variability(HRV)monitoring at 560 meter above sea level(plain),then were transported to Tibet plateau(3675-meter above sea level)by airplane.There,all subjects were divided into 3 groups,and A gorup(n=12)underwent HRV monitoring on the 2nd day after arrival,B group(n=48)on the 3rd day,C group(n=26)on the 4th day.Results Compared with the data obtained at plain,the standard deviation of normal to normal intervals(SDNN),high frequency(HF)and total power(TP)decreased significantly(P50 ms(PNN50)and normalized high frequency(HFnu)also decreased obviously(P

AIM:To investigate the effect of short-chain acyl-CoA dehydrogenase ( SCAD) on collagen expres-sion and proliferation of rat cardiac fibroblasts and to explore the relationship between SCAD and cardiac fibrosis . METHODS:The model of proliferation and collagen expression of rat cardiac fibroblasts induced by angiotensin II was es -tablished.After treatment with siRNA-1186, the expression of SCAD at mRNA and protein levels , fatty acids beta oxida-tion rate, ATP, the enzyme activity of SCAD and free fatty acids in the rat cardiac fibroblasts were determined . RESULTS:The mRNA and protein expression of SCAD was decreased in the rat cardiac fibroblasts induced by angiotensin II compared with the control cells , and the expression of collagen I and collagen III was significantly upregulated .Com-pared with negative control group , SCAD expression and activity , fatty acid beta-oxidation rate and ATP significantly de-creased in siRNA-1186 group, but the content of free fatty acids were obviously increased in the rat cardiac fibroblasts , and the expression of collagen I and collagen III was significantly up-regulated.CONCLUSION:The expression and synthesis disorder of collagen may be triggered by down-regulation of SCAD .SCAD may be a promising therapeutic target for myocar-dial fibrosis .

The result would provide some gist to choice the suitable pattern of bilingual teaching for other subject of our college.

Objective To evaluate the clinical efficacy and safety of progesterone combined with nifedipine in the treatment of upper ureteral calculi.Methods A total of 94 patients with upper ureteral calculi werc divided into control group and treatment group,47 cases in cach group.Patients in control group were given anisodamine hydrochloride injection 5-10 mg by intramuscular injection,qd.Patients in treatment group received progesterone injection 20 mg by intramuscular injection,bid and nifedipine tablets 10 mg by under tongue,tid.All patients were treated for 14 d.The clinical efficacy,C-reactive protein(CRP),interleukin-6 (IL-6) and adverse drug reactions were compared between the two groups.Results The clinical efficacy in treatment group was 95.74％ (45/47 cases),had significant difference with that in control group,which was 82.98％ (39/47 cases,P ＜0.05).The levels of CRP and IL-6 in treatment group were (2.17 ± 0.25) mg · L-1,(4.72 ± 0.51) ng · L-1,had significant difference with those in control group,which were(3.45 ±0.42) mg· L-1,(8.34±0.89) ng · L-1 (P ＜0.05).The main adverse drug reactions in treatment group were dry mouth,blurred vision and rashes,the incidence rate was 6.38％ (3/47 cases).The main adverse drug reactions were dry mouth,rapid heart beat,difficulty urinating and rashes in control group,the incidence rate was 10.64％ (5/47 cases,P ＞ O.05).Conclusion Progesterone combined with nifedipine in the treatment of upper ureteral calculi was effective with high safety.

Objective To summarize effect of application of Damage Controlled Surgery (DCS) in abdominal serious and key points in nursing.Methods We retrospectively reviewed clinical data of 76 patients treated by DCS from 2008.1 to 2010.12.Key points of nursing:( 1 ) first stage:nursing in simplifying surgery;(2) second stage:nursing in resuscitation; (3) third stage:nursing in determining surgery.Results DCS was performed in all the 76 patients.Then 30 patients recovered and 42 patients underwent operations again.The cure rate was 94.7％ (72/76).The mortality rate was 5.1％ (4/76),which had nothing to do with DCS.Conclusions The effect of DCS surpasses the conventional operation in serious abdominal diseases.A reasonable application and nursing care of DCS can reduce patient' s postoperative complications and death rate.These nursing measures are assurance of putting DCS in action successfully for patients.

Objective Sperm screening is an essential step in IVF procedures. The swim-up method, an assay on sperm motility, is used clinically to select the ideal sperm for subsequent manipulation. However, additional parameters, including acrosome reaction capability, chemotaxis, and thermotaxis are also important indicators of mammalian sperm health. To monitor both sperm motility and chemotaxis simultaneously during sperm screening, we designed and constructed a microdevice comprising a straight channel connected with a bi-branch channel that mimics the mammalian female reproductive tract. Methods The width and length of the straight channel were optimized to select the motile sperm. Cumulus cells were selectively cultured in the bi-branch channel to generate a chemoattractant-forming chemical gradient. Sperm chemotaxis was represented by the ratio of the sperm swimming towards different branches. Results The percentage of motile sperm was improved from ( 58. 5 ± 3. 8 ) % to ( 82. 6±2.9)% by a straight channel 7 mm in length and 1 mm in width. About 10% of sperm were found chemotactically responsive in our experiment, which is consistent with previous studies. Conclusion The combined evaluation of both sperm motility and chemotaxis was achieved for the first time, and the motile and chemotactically responsive sperm can be easily enriched on a lab-on-a-chip device to improve IVF outcome.
