Objective To apply a one-step loop-mediated isothermal amplification(LAMP) assay for rapid detection of Escherichiaco-li .Methods According to the Genbank of Escherichiacoli lacZ(accession number:M74750) ,a set of four specific primers were designed for the Escherichiacoli LAMP assay and optimized the reaction .The results were judged with naked eyes and electrophoretic analysis .Results Genomic DNAs from 13 bacterial strains including Escherichia coli strains were amplified using LAMP ,and no amplicon was observed in the other bacterial strains .The detection limits of LAMP assay was 15 cfu/mL .Conclusion LAMP assay is extremely rapid ,cost-effective , specific and highly sensitive and has potential usefulness for clinical application .

Heart failure is becoming an increasingly common and significant problem in the field of pediatric cardiology,β-blockers as one of the most important cardiovascular drags in the pharmacopoeia,can not only obviously relieve symptoms of severe heart failure,but also reduce cardiovascular events and allcause mortality significantly,the efficacy is effective and high security.This article reviewed β-blockers in the treatment of children heart failure mechanism,the clinical application evaluation,the basic principles and relevant issues in use process,the appropriate choice and usage according to the clinical indications.

Objective To investigate the effect of budesonide inhalation combined with azithromycin in treatment of children with mycoplasma pneumonia and the corresponding clinical nursing pathway.Methods From February 2010 to 2011,100 cases of children patients with mycoplasma pneumonia (all with budesonide inhalation combined with azithromycin) were randomly divided into the experimental group and the control group with 50 patients in each group.The experimental group adopted clinical nursing pathway,while the control group underwent routine care.After one or three-month follow-up,forced vital capacity (FVC),forced vital capacity 1s (FEV1),peak expiratory flow values (PEF) and other pulmonary functional indexes and clinical efficacy were compared between two groups.Results The clinical effective rate of the experimental group was 94％,higher than the control group.FVC,FEV1,PEF changes in the experimental group were better than the control group.Conclusions Budesonide inhalation combined with azithromycin in treatment of mycoplasma pneumonia in children has good effect,coupled with the clinical nursing pathway can effectively improve lung function,reduce the incidence of adverse reactions during treatment,significantly improve the prognosis,should be used widely in clinic.

Malnutrition is significantly correlated with the high mortality and other poor outcomes of stroke;in turn,stroke can increase the incidence of malnutrition.Old age and swallowing dysfunction are independent risk factors of post-stroke malnutrition.The sequential nutritional support with enteral nutrition and parenteral nutrition can reduce or avoid the complications of both malnutrition and stroke,and therefore should be considered for stroke patients,especially those who are critically ill.

Risk factors related to malnutrition were prospectively studied in 82 cases with acute ischemic stroke in our hospital during December 1,2007 and December 31,2008. The incidences of malnutrition on the first and ( 14 ± 1 )th day were 18% (15/82) and 30% (25/82). Study showed that age ( P = 0. 007 ), dysphagia ( P = 0. 043 ), malnutrition on admission ( P = 0. 003 ) were risk factors related to malnutrition on the ( 14 ± 1)th day after admission; there was no interaction between dysphagia and the other two factors, dysphagia was an independent risk factor related to malnutrition after acute ischemic stroke.

HES solutions have notable effects on ameliorating hemodynamic and hemorrheological parameters, as well as unique capacity to prevent and reduce capillary leak. Compared with other colloid solutions, HES is more effective, safe and economical. It is an ideal volume expander.

After the practical experiences with embedded subject service for research in nursing science in Library , The First Affiliated Hospital of Chongqing Medical University was described , such as constructing and recommending resources for nursing staff , helping them to search and read related resources , providing training service of how to write and submit papers , the models of embedded subject service for research in nursing science were summarized , such as laying equal stress on construction and recommendation of resources , systematic service in the embedding processes , and tackling problems by groups in coordination .

Objective To establish a rapid ,sensitive direct method for detecting Escherichia coli (E .coli) in the positive blood culture bottles by using the loop‐mediated isothermal amplification (LAMP) .Methods Based on the lacZ (accession number :M74750) gene sequence of E .coli in the gene sequence GenBank by the National Institutes of Health (NIH) ,4 specific primers (2 inner primers and outer primers) were designed ,the lacZ gene was amplified and the amplification reaction was optimized .The sen‐sitivity and specificity of this method were verified and its comparison with the traditional culture method was conducted .Results Sixty‐eight strains of E .coli were detected in 300 positive blood culture bottles by adopting the LAMP method .The designed prim‐ers had good sensitivity and good specificity for the amplification of E .coli .The sensitivity and specificity of LAMP all reached 100% compared with the traditional method ,but the traditional method needed 3 d to get the results ,while LAMP only needed 1 h . Conclusion LAMP is extremely rapid for detecting E .coli with low cost ,strong specificity and high sensitivity and is suitable for clinical application .

Objective To examire the effects of hydroxyethyl starch (HES 200/0.5 6%) on lung capillary permeability in endotoxemic rats.Methods Forty-two male Wistar rats weighing 250-300 g were randomly divided into 7 groups with 6 animals in each group: group 1 served as control; group 2 received lipopolysaecharide (LPS) 6mg?kg-1 intraperitoneally (i.p.); in group 3-6 1 min after LPS 6 mg?kg-1 i.p. HES 3.75 ml?kg-1 (group 3), 7.5 ml? kg-1 (group4), 15 ml? kg-1 (group 5) or 30 ml?kg-1 (group 6) was infused i. v. at a rate of 0.2 ml ? min-1 ; group 7 received only HES 30 ml ? kg . The animals were anesthetized with urethane. Right external jugular vein was cannulated for continuous intravenous infusion. Animals were sacrificed at 2 h and 4 h after LPS challenge respectively. Lung capillary permeability, lung wet/dry weight ratio and lung PMN accumulation were measured at 4 h after LPS challenge and nuclear factor Kappa B (NF-?B) activation in the lungs at2 h after LPS challenge.Results HES 3.75 ml?kg-1 and 7.5 ml ? kg-1 significantly reduced LPS-induced increase in lung capillary permeability, lung wet/dry weight ratio and lung PMN accumulation ( P

Background Retinal transplantation is a new approach to the treatment of retinal degeneration diseases,and how to avoid or reduce the immune rejection after transplantation is a problem.In experimental studies on retinal transplantation,C57BL/6 mice are often used as donors and rd mice serve as recipients.Studies showed that Fas ligand (FasL) protein induces the apoptosis of Fas+ inflammatory cells by FasL/Fas signal pathway,speculating that FasL protein is associated with immune rejection after transplantation.Objective The aim of this study was to investigate FasL protein expression change in retinas of C57BL/6 mice and rd mice with aging and reveal the immune characteristics of the mouse retinas.Methods The frozen sections of eyeball from C57BL/6 mice and rd mice at the age of postnatal (PN)-0 week,PN-1 week,PN-2 week,PN-3 week and PN-4 week were prepared.The expression of FasL protein in the mouse retinas was examined by immnofluorescense technique.Images were acquired by fluorescence microscope and analyzed semi-quantitively by software from laser scanning confocal microscope as the fluorescence intensity (FI).The results were compared among different strains of mice.Results The retina developed imperfectly in PN-1 week C57BL/6 mice and FasL protein was positively expressed in the whole retina.In PN-2,3 and 4 week C57BL/6 mice,retinas finished the development with 10 layers,and retinal pigment epithelium (RPE),inner segment (IS),outer limiting membrane (OLM),outer plexiform layer (OPL),inner nuclear layer (INL),inner plexiform layer (IPL) and ganglion cell layer (GCL).Retinal structure and expression of FasL protein in the whole retina of rd mice in the PN-1 week were similar with C57BL/6 mice,however,ONL cells of rd mice were evidently decreased with aging.The RPE,OPL,INL,IPL and GCL expressed the FasL protein in PN-2,PN-3 and PN-4 week rd mice.The mean FI of FasL protein in the RPE layer was 184.199±16.747,186.797±7.904 and 184.319± 18.795 in rd mice of PN-2 week,PN-3 week and PN-4 week,which were significantly higher than 160.402±22.851,160.995 ±22.799 and 105.787 ± 17.676 in C57BL/6 mice (t =-3.360,P =0.002;t'=-4.277,P =0.000;t =-12.175,P=0.000).There were not significant differences in the mean FI of FasL protein in IPL between C57BL/6 mice and rd mice at ages of PN-2 week,PN-3 week and PN-4 week (all at P＞0.05).Conclusions The cells of retinal ONL are gradually decreased with the development of rd mice,and FasL protein expression intensity in RPE is evidently enhanced in rd mice compared with C57BL/6 mice.
