Chinese Journal of Clinical Oncology 2014;(1):27-31
The advent of nanoscience and nanotechnology offers unprecedented opportunities in nanomedicine, such as increas-ing therapeutic efficiency and decreasing undesired side effects in cancer treatment. Photodynamic therapy (PDT) is a non-invasive pho-totherapy-based method that is applied in the treatment of cancer and other diseases and has important clinical value. PDT can be com-bined with other therapies to realize the synergistic treatment. The emergence of up-conversion nanomaterials provides a fundamental method to solve the problem of photodynamic therapy of deep tumors. Moreover, the versatile preparation and surface modification methods facilitate the fine-tuning of the emission spectrum of up-conversion nanomaterials and the improvement of the photosensitiz-er's loading capacity. This study reviews the development in design and application of up-conversion nanomaterials for PDT of cancer.

Chinese Journal of Laboratory Medicine 2003;0(08):-
Objective The clinical significance of serum cytokine in kidney transplanted patients have been evaluated by observing the variation of cytokines and biochemical parameters. Methods The serum IL-1?、IL-1?、IL-2、IL-4、IL-6、IL-8、IL-10、VEGF、IFN-?、TNF-?、MCP-1、EGF and 22 items of biochemistry parameters have been followed in 10 patients during 90 days around operation time. Cytokine had been measured by biochips methods in evidence 180 automatic biochips analyzer. The serum biochemical parameters had been measured by Roche diagnostic company reagents kits in HITACHI 7600DDP analyzer. Results Serum IL-6 and IL-8 had reached the highest level [(42.5?42.8) ng/L and (343.5?493.1) ng/L] at 7 days after kidney transplantation, whereas IL-2 and IL-10 were reached the highest level [(16.3 ? 5.7) ng/L and (16.6 ? 24.2) ng/L] respectively at 14 days. Then they decreased gradually, but IL-6 and IL-8 were still higher than normal control. The decreased VEGF, EGF and MCP-1 in kidney failure patient had increased gradually after transplantation. But EGF was still lower than normal control at 90 days after operation. Urea、Cr、Ua and Glu had been restored to normal level after two month of operation and the significant relationship between IL-8 and serum LDH activity have been found( r=0.93,P

OBJECTIVE:To establish a simple and rapid ion-pair-RP-HPLC method for the determination of intracellular concentration of ara-CTP.METHODS:Mononuclearcells were separated from the whole blood, centrifugated and injected for determination on Symmetry C18 column with the mobile phase consisted of 0.1 mol?L-1 KDP buffer solution and 0.01 mol?L-1 tetrabutylammonium hydrogen sulfate(pH=2.7)at a flow rate of 1.0 mL?min-1 with the UV wavelength set at 278 nm.RESULTS:The linear range of ara-CTP was 0.35-9.12 ?g?mL-1(r=0.998 6)with its detection limit at 0.35 ?g?mL-1.The recoveries of ara-CTP at low, middle and high concentrations, and the intra-day and inter-day RSDs were all up to the standards.CONCLUSION:The established method is simple, rapid, stable, reproducible and applicable for the monitoring of intracellular concentration of ara-CTP.

Objective To discuss the clinical application of percutaneous transhepatic cholangiopancreatic drainage (PTCD) combined with CT-guided microwave ablation in treating hepatic hilar cholangiocarcinoma. Methods The clinical data of 29 patients with inoperable Ⅲ and Ⅳ type hilar cholangiocarcinoma complicated by obstructive jaundice, who were admitted to authors’ hospital during the period from December 2012 to August 2014, were retrospectively analyzed. The diagnosis of bile duct adenocarcinoma was confirmed by pathology in all patients. Of the 29 patients, both internal and external biliary tract drainage was employed in 19, external biliary tract drainage in 4, and bilateral (both left and right side) bile duct drainage in 6. CT-guided microwave ablation was carried out when the liver function became improved. A total of 46 procedures of microwave ablation were completed in the 29 patients with a mean of 1.5 times per patient. Hepatic function tests, enhanced CT or MR scan were performed 4-8 weeks after treatment. According to mRECIST criteria the therapeutic results were evaluated; the serum bilirubin levels were recorded; the disease progress and the patient’s survival time were followed up. Results One month after the treatment, complete response (CR) was obtained in 15 patients (15/29, 51.7%), and partial remission (PR) in 17 patients (17/29, 58.6%), with the overall efficacy (CR﹢PR) being 82%. After the treatment, one patient developed hepatic metastasis and another one had pulmonary metastasis. Postoperative serum bilirubin levels showed an obvious decrease in all 29 patients. The 6-month, one-year and two-year survival rates were 68.9% (20/29), 31.0% (9/29) and 6.8% (2/29) respectively; the median survival time was 8.9 months and the overall survival time was 11.7 months. Conclusion For the treatment of hepatic hilar cholangiocarcinoma, PTCD combined with CT-guided microwave ablation is minimally invasive, clinically safe and effective.

Objective The purpose of this study was to investigate the mechanism of action of polypeptide extracts of Eupolyphaga sinensis Walker ( ESW) against oxidative aging. Methods Mice were intraperitoneally injected D-galac-tose for consecutive 20 days to establish an aging mouse model. The model mice were administered with different doses of ESW polypeptide (0, 40, 80, 160 mg/kg/d).The normal activity, movement and anti-stress ability of the mice were ob-served.The activity of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH-PX) in blood and different tissues and the content of glutathione ( GSH) and malondialdehyde ( MDA) of the aging mice were assessed by xanthin oxidase activity measurement and spectrophotometry, respectively. The expression of nuclear factor erythroid 2-re-lated factor 2 (Nrf2) in Caco-2 cells was detected by immunofluorescence. Results Comparing the control and polypep-tide groups, there were significant decreases of body weight gain, organ indexes, anti-stress ability and activity capacity, the activity of SOD, CAT, GSH-PX and the content of GSH, and an increase of the content of MDA in blood and different tissues in the aging mice. With the increasing dose of polypeptide extracts of ESW, the body weight gain, organ indexes of the liver, spleen and kidney were significantly increased, the static and dynamic exercise time was prolonged in the poly-peptide group, and their abilities of hypoxia tolerance and heat tolerance were close to that of normal controls. The SOD, CAT, GSH-PX activity and GSH level in blood and different tissues were significantly increased, but MDA content de-creased.The expression of Nrf2 in Caco-2 cell nuclei was significantly increased in the polypeptide group, close to that of the positive control group. Conclusions The results of our study show that polypeptide extracts of ESW improve the anti-stress and antioxidative capacity in D-galactose-induced mouse models of oxidative aging by initiating Nrf2-ARE antioxidant signaling pathway, therefore, delay the oxidative aging in mice.
