1.Study on the mechanism of apoptosis of mouse kidney cells in dietary zinc deficiency during the growth period

Objective To observe the cell apoptosis, oxidative stress reaction and expressions of Bcl-2 and Bax in kid?ney of dietary zinc deficiency mice during growth period, and discuss the mechanism of renal cell apoptosis induced by zinc deficiency. Methods Thirty weaning male mice were randomly divided into zinc-deficient group and zinc-adequate group, and 15 mice for each group. Zinc-deficient group was fed with zinc deficiency diet (0.85 mg/kg), while zinc-adequate group was fed with enough zinc diet (30 mg/kg). The TUNEL method was applied to observe the cell apoptosis, and the apoptotic in?dex was measured. The content of SOD and MDA were detected to observe the oxidative stress reaction in kidney. The ex?pression levels of Bcl-2 and Bax protein were detected by Western blot assay. Results Compared with zinc-adequate group, the cell apoptosis and oxidative stress reaction were increased in zinc-deficient group. The expression of Bcl-2 de?creased, and the expression of Bax increased. The ratio of Bcl-2 and Bax declined in kidney of zinc deficiency mice. Conclu?sion Diet zinc deficiency in growth period may result in the decreased antioxidase, the increased oxidative stress reaction, and the changed Bcl-2 and Bax expression, which promote the occurrence of cell apoptosis in kidney.
2.Advances in Study on Functional Magnetic Resonance Imaging in Irritable Bowel Syndrome Comorbid with Anxiety and Depression

Anxiety and depression are common conditions among patients with irritable bowel syndrome(IBS)which may induce or aggravate intestinal symptoms. Psychological factors are considered to be involved in the pathogenesis of IBS via both central and peripheral pathways,and functional magnetic resonance imaging(fMRI)revealed that several brain areas,such as anterior cingulate cortex and prefrontal cortex,are associated with IBS,anxiety and depression. This article reviewed the use of fMRI for studying the correlation of IBS brain activity with anxiety and depression and its role in the central pathogenesis of IBS.
3.Quality inspection of clinical research in traditional Chinese medicine.

Abstract: Beginning with 4-level quality control measures of clinical research in traditional Chinese medicine (TCM), we elaborated the implementation process and demands of quality control measures of each level, including quality control, monitoring, auditing, and inspection. On the basis of joint inspection experience of 41 projects of the "Prevention and Treatment of Difficult and Complicated Diseases of TCM" plan of the "11th Five-year National Key Technology R&D Program", we analyzed the ensuring effect of 4-level quality control system and joint inspection model, and then pointed out the existing problems in the executing process of quality control system at different levels and joint inspection model. Finally we investigated what should be revised in the quality control system and joint inspection model, thus providing the theoretical support for quality inspection improvement of TCM clinical research.
4.Analysis of risk factors for postoperative nausea and vomiting in spinal anesthesia patients

Objective To investigate the risk factors for postoperative nausea and vomiting (PONV) in spinal anesthesia patients.Methods A total of 841 patients received spinal anesthesia were visited after operation.Data were analyzed using univariate analysis and multivariate Logistic regression to identify risk factors related to PONV.Results PONV occurred in 94 patients (11.2%,94/841).Univariate analysis showed that PONV was unrelated with gender,age,ASA classification,anesthesia mode (P > 0.05),related with operation department (P =0.026),body mass index (P =0.020),education level (P =0.000),history of previous surgery anesthesia (P =0.005),history of PONV (P =0.000),history of kinesia (P =0.002),smoke (P =0.019),intraoperative using of tramadol (P =0.018).Multivariate analysis showed that operation department (OR =4.039,95% CI 1.331-12.259,P=0.048),education level (OR =3.504,95% CI 1486-8.260,P=0.015),history of PONV (OR =5.113,95% CI 1.790-14.606,P =0.002),intraoperative using of tramadol (OR =5.316,95% CI 1.091-25.908,P =0.039) were identified as independent risk factors for PONV.Conclusions The independent factors associated with PONV following spinal anesthesia include operation department,education level,history of PONV,intraoperative using of tramadol.Identifying patients who are at high risk for PONV will enable the formation of more timely management project.
5.Clinical studies on the combiantion of single lower limb traction and massage therapy for the lumbosacral tunnel syndrome

Objective To observe the therapeutic effect of single lower limb traction plus massage for the lumbosacral tunnel syndrome.Methods The selected 80 candidate patients were divided into the observation group with 38 patients and treatment group with 39 patients. The observation group received single lower limb traction plus massage and the treatment group received pelvic traction plus massage. Both groups therapy last 5 days, The Japanese orthopaedic association scores (JOA) was evaluated for subjective symptoms, signs, functional activity and bladder function score. The therapeutic effect was evaluated.Results The total effective rate (94.7% vs. 82.1%;χ2=12.160,P=0.007) in the observation group was significantly higher than that in the control group. O, The scores of JOA at one week after treatment (20.2 ± 2.0 vs. 18.3 ± 2.8;t=3.419, P<0.01) and two weeks after treatment (24.4 ± 1. 4vs. 22.6 ± 3.0;t=3.359,P<0.01) in the observation group were significantly higher than those in the control group.Conclusions The single lower limb traction plus massage can improve the clinical symptoms of patients with lumbosacral tunnel syndrome, and be superior to the pelvic traction plus massage.
6.Predictive Value of Obesity Index to the Metabolic Syndrome among Tianjin Adults

Objective To explore the predictive value of obesity-related index for metabolic syndrome(MS)among Tianjin adults. Methods A total of 522 adults who participated in a physical examination were included in this cross-sectional study. Participants 'gender and age were record-ed,height,weight,waist circumference(WC)and hip circumference were measured,and TC,TG,HDL-C and FPG were tested. The receiver op-erating characteristic(ROC)curves were generated to identify the optimal measurement of obesity for the prediction of metabolic risk in this popu-lation. Results The area under the ROC curve(AUC)value for waist-to-height ratio(WHtR)predicted the maximum value of MS,followed by WC and BMI,and WHR was the lowest. The optimal cut off value of WHtR was 0.509. There was significant difference in the ability of BMI be-tween different age groups and WC to predict MS. In the groups of<40 years old and≥60 years old,BMI had higher predictive value for MS than WC. Conclusion WHtR has the best predictive value for evaluating the MS risk compared to BMI ,WC or WHR alone among Tianjin adults. The predictive value of BMI and WC for evaluating the MS risk can be improved by the method of age stratification.
7.Expression and clinical significance of HIF-1αin breast cancer

Objective To detect the expression of hypoxia-inducible factor-1α (HIF-1α) in breast can-cer and to investigate the feasibility of HIF-1α as a new therapy target for breast cancer. Methods 56 cancer tissue specimens of women with primary breast cancer admitted from Jan. 2013 to Dec. 2013 in the Fourth Hos-pital of Hebei Medical University Breast Center were selected. Immunohistochemistry was used to detect the ex-pression of HIF-1α protein in cancer tissues. The relation between HIF-1α protein expression and the clinico-pathological parameters was analyzed. Results The positive expression rate of HIF-1α protein in breast cancer was 73.2% (41/56) and its high expression rate was 41.1% (23/56), which were significantly higher than those in benign breast tumor group (P=0.000). HIF-1α protein expression was positively correlated with breast tumor size, stage, histological grade, lymph node metastasis and the expression of HER2, and it was negtively correlated with ER. Conclusions High expression of HIF-1α in breast cancer was significantly associated with poor prog-nosis. HIF-1α protein is also a risk factor for breast cancer as well as HER2, and they may have a synergistic role in the progression of breast cancer.
8.Effect of Benidipine-atorvastatin Combination Therapy on Inflammatory Markers and Flow-mediated Dilatation in Elderly Patients with Isolated Systolic Hypertension

Objective The effect of combined treatment of statins and calcium channel blockers(CCB) in elderly patients with isolated systolic hypertension(ISH) are not well understood.The purpose of the present study was to assess the additive effect of benidipine-atorvastatin combination therapy in old patients with ISH.Methods Ninety patients with ISH were randomized to receive: placebo(n=15),benidipine treatment(4-8 mg/d,n=25),atorvastatin(20 mg/d,n=25) and benidipine plus atorvastatin treatment(4-8 mg/d+20 mg/d,n=25) for 5 months.Serum concentrations of IL-6,IL-8,hsCRP,flow-mediated dilatation of the brachial artery,urine microalbumin,left ventricular mass index(LVMI),carotid intima-media thickness(CIMT),BP and plasma lipids profiles were examined before and after treatment.Results After 5-months of treatment,the combination treatment significantly reduced IL-6(IL-6 reduction magnitude,combination group: 6.5+6.3 ng/L vs benidipine group: 3.1?3.2 ng/L vs atorvastatin group: 3.3?2.3 ng/L,P
9.Influence of Oxygen-Induced on Expression of Monocyte Chemoattractant Protein-1 in Retinopathy of Newborn Rats

Objective To explore the relationship of monocyte chemoattractant protein-1(MCP-1)and oxygen-induced retinopathy(OIR).Methods Thirty-six newborn SD rats were divided into 2 expanded litters,18 of which were exposed to 50 mL/L oxygen and then 10 mL/L oxygen in alternating 24-hour periods(experiment group),an additional 18 rats as control rats were raised simultaneously in room air(control group).On postnatal 14 days(P14),oxygen-exposed rats were removed to room air.The eyeballs of 6 rats from each group were enucleated and fixed by formaldehyde at postnatal 14 days(P14),postnatal 17 days(P17),and postnatal 21 days(P21)respectively,and then cross sectioned.The nuclei of proliferative retinal vessels were counted through the crosssections to measure the average retinal capillary density index(RCDI)when stained with hematoxylin and eosin(HE)under light;the expression of MCP-1 was measured by immunohistochemistry.Results There existed obviously differencc between 2 groups in the field of both RCDI and the expression of MCP-1 at the same time point [t(P14)=6.69 P=0.001,t(P17)=3.43 P=0.006,t(P21)=2.37 P=0.039;t(P14)=40.45,t(P17)=43.44,t(P21)=17.45 Pa=0],when RCDI and the expression of MCP-1 were compared among the different time points with in the same group,there existed obviously difference among three time points in the experiment group(F=17.74 P=0.0001;F=421.5 P=0),but no differencc in control group(F=0.016 P=0.984;F=0.006 P=0.994).There existed positive correlation between the expression of MCP-1 and the value of RCDI in experiment group(r=0.822 P=0).Conclusions Neovascularization resulting from OIR occurs before room air recove-ry.MCP-1 is upregulated and subsequently downregulated in OIR.Neovascularization in the OIR model appear to be associated with increased retinal MCP-1.
10.Adherence test of Staphylococcus epidermidis on the surface of different material intraocular lenses

Background Postoperative endophthalmitis following intraocular lens(IOL)implantation is still one of the most feared complications of cataract surgery.Bacterial adhesion to IOLs during their insertion is a prominent etiological factor.The adhesion characteristics of bacteria to IOL are very vital for the prevention of endophthalmitis after cataract surgery.Objective The present study was to observe the in vitro adherence ability of Staphylococcus epidermidis to different intraocular lenses(IOLs)and compare the results in bacterial counting between scanning electron microscopy(SEM)photographs and quantitative cultures. Methods Five types of IOLs,including hydrophobic acrylic IOL,polymethylmethaerylate(PMMA)IOL,heparin-surface-modified(HSM) PMMA IOL,silicone(SI)IOL and hydrophilic acrylic IOL,were put into S.epidermidis(ATCC 12228)suspension for 1 hour.The bacterial adhesion numbers on the IOL surfaces were counted by quantitative cultures and scanning electron microscopy(SEM) photographs. Results Quantitative culture counting of viable adherent bacteria released by sonication showed that hydrophobic acrylic IOL and PMMA IOL were more likely for bacteria to attach.The number of bacteria on the five types of IOL surfaces showed significant differences(F=100.084,P=0.000).No significant differences were found in the number of bacteria between hydrophilic acrylic IOL and HSM-PMMA IOL (t=2.285,P=0.052)with the quantitative culture method.Direct counting of adherent bacteria in SEM photographs revealed that there were significant differences in bacterial adhesion numbers among difierent IOL material groups,with the numbers from high to low as follows:Hydrophobic IOL>PMMA IOL>SI IOL>Hydrophilic IOL>HSM-PMMA IOL(F=118.065,P=0.000).The counting method by SEM method was superior to that by quantitative cultures (t=5.019,P=0.000). Conclusion The bacterial adhesion ability varies upon the difference of IOL materials.Less bacterial adhesion is found on hydrophilic acrylic IOL and HSM-PMMA IOL,implying that the use of IOLs made from these two materials during surgery could diminish the incidence of postoperative endophthalmitis and intraocular inflammation associated with IOLs implantation.
