1.Cloning of human REG ? cDNA and construction of the stable transfective cell line

Objective:To construct the stable transfective cell line with the eukaryotic expression vector for human REG?cDNA to laid the foundation for further study of the function of REG?.Methods:REG?cDNA was obtained by RT-PCR of total RNA extracted from the human breast cancer cell MCF-7,and it was digested by the restriction endonuclease EcoRⅠand EcoRⅤbefore connection withPcDNA3.1.The recombination with forward insert were slelected by restriction endonuclease digestion and confirmed correct by sequencing and then transfected into HBL-100 cell by using lipofectamine2000.The stable transfected cell line was selected in medium containing antibiotic(G418).Results:The result of immunocytochemistry and RT-PCR of total RNAextracted from the stable transfected cell line showed that there existed the overexpression of REG?cDNA in it.Conclusion:The construction of the eukaryotic expression vector for REG?was successful,and the stable transfected cell line for overexpression REG? was obtained by G418 selection.This work may pave the way for further study of the functions of REG?.
2.IL-16 in Senile Chronic Obstructive Pulmanary Disease (COPD) and Its Clinical Significance

OBJECTIVE To investigate the role of IL-16 in the process of airway inflammation of chronic obstructive pulmanory disease(COPD) patients.METHODS The sputum of 60 COPD elderly was collected in the period of exacerbation and alleviation.Serum level of IL-16 was tested using ELISA.RESULTS IL-16 level was higher in severe [(4240.7?296.36,1737.59?367.81)pg/ml] and moderate [(3125.87?286.35,1564.34?478.99)pg/ml] COPD patients in exacerbation and alleviation period than in mild patients(173.75?31.26,129.27?23.89)pg/ml and IL-16 level decreased after effective therapy.CONCLUSIONS IL-16 participates in the procedure of airway inflammation.Surveillence of dynamic variation of IL-16 can reflect the severity of airway inflammation,but it can not offer more diagnostic value.

PTEN gene is considered to be the first discovered tumor suppressor gene with dual specificity phosphatase activity.The research has shown that the increasing popularity of PTEN rests on its important roles in cancer cell growth,differentiation,signaling transduction and cell apoptosis.Besides,the mutation of PTEN is related to cancer initiation and progression.The recent advances on PTEN gene in malignant tumors will be reviewed in this paper.
4.DSG biological information infrared liver disease therapy instrument improves liver microcirculation in mice

Objective To investigate the effect of DSG biologic information infrared liver disease therapy instrument(BILT liver disease therapy instrument)on liver microcirculations in mice.Methods Liver fibrosis was induced in 40 SPF mice which were randomized into 4 groups:without irradiation group,coht light irradiation group,infrared light irradiation group and BILT group(10 in each group).Twenty healthy mice were taken as the control.Liver blood flow of each group was observed by laser-Doppler flow instrument,and blood flow rate of liver microcirculation in each group was observed by intravital microscopy.Pathological examination and serum biochemistry were also performed.Results In fibrosis mice,the liver blood flow in average of BIUF group Was higher than that of without irradiation group(P=0.004),while comparing with without irradiation group there were no significant differences in liver blood flows of the cold light irradiation group and infrared light irradiation group(P=0.713 and 0.465).Red blood cell velocity of microvessels in average of the healthy group(without irradiation)wag higher than that in the without irradiation group offibrosis mice(P=0.001),and it in the BILT group(fibrosis mice)Was also higher than those in other groups of fibrosis mice(P=0.004,0.020 and 0.030).For fibrosis mice,the average AST in BILT group was lower than that in the without irradiation group(P=0.027),and the difference in hydroxyproline(Hyp)levels between the groups was not of statistical significance(P=0.433).Conclusion BILT liver disease therapy instrument can improve the liver microcir-culation,reduce AST level and improve liver pathological state in mouse with fibrosis.

Long-term potentiation(LTP)is a typical model of synaptic plasticity which has great relationship with learning and memory mechanisms:The induction of LTP depends on the activation of the calcium channels and LTPs generated by calcium ions influx through different channels play different roles in memory.This paper reviews the progress of the research on LTPs induced by activations of N-methyl-D-aspanate receptor(NMDAR)channels and voltage-dependent calcium channels(VDCC).Finally,it gives an example of new forms of low frequency stimulation for induction of LTP.
6.Standardized analgesic in total knee arthroplasty perioperative application

Objective To study the standardization of analgesia in total knee replacement (TKA),and to assess its efficacy and safety of perioperative period application.Methods 64 cases of total knee arthroplasty cases were selected,with 30 cases of male,and 34 cases of female(Aged 61 -78),included 28 cases on the left side,and 36 cases on the right side.According to the random of prefabricated CARDS (1 :1 parallel design of experiment) patients were divided into observation group and control group,with 32 cases in each group(17 cases were women, 15 cases were male,).The observation group were treated with standardized analgesia,the control group were treated with traditional postoperative controlled intravenous analgesia (PCIA)pump.48 h after surgery,the different time points after waking up general anesthesia visual analogue scale (VAS),giving pain sleepers auxiliary analgesic drug application were recorded.VAS score and analgesic drug usage and complications (nausea,vomiting,respiratory depression,motor nerve paralysis,etc.)incidence index etc.of the two groups of patients were compared.Results 64 patients were included in the study,the differences of the two groups of patients in age [observation group:(68.22 ± 0.18),the control group:(69.69 ±0.17),t =5.767,P =0.8237)],gender(15 cases were men,17 cases were women),body mass index [BMI,observation group:(23.38 ±0.19),the control group:(23.94 ±0.14),t =2.339, P =0.082)],the American society of anesthesiologists (ASA)classification (both for 10 cases of ASA grade I,ASA grade II 22 cases),had no statistical significance (P >0.05).And different time points after surgery anesthesia to VAS score had significant difference:[(control group:6 h resting 2.888 ±0.049 /ankle pumps 2.941 ±0.028,12h resting 4.731 ±0.028 /ankle pump 5.747 ±0.033,24h resting 6.859 ±0.033 /ankle pump 7.175 ±0.03,48h resting 4.14 ±0.03 /ankle pumps 5.209 ±0.025.observation group:6h resting 1.316 ±0.024/ankle pumps 1.884 ± 0.023,12h resting 2.05 ±0.02 /ankle pump 3.060 ±0.02,24h resting 2.35 ±0.02 /ankle pump 3.934 ±0.018, 48h resting 2.119 ±0.015 /ankle pumps 2.317 ±0.017.t value resting/ankle pump:6h,17.80 /28.75.12h,76.67 /69. 14.24h,116.9 /91.95.48h,61.11 /95.51.P <0.01)].Analgesic drug usage (control:55.97 ±0.25,the observation group:19.53 ±0.19,t =113.1,P <0.01),the incidence of complications (control:0.45 ±0.00,observation group:0.06 ±0.00;t =169,P <0.01 ),and other aspects of the differences were statistically significant (P <0.05 ). Conclusion Standardized analgesia on the perioperative analgesic effects of total knee arthroplasty is better than PCIA.It can significantly reduce the extra analgesic drug usage,decrease the incidence of complications,and has the characteristics of safe and efficient.

9.Treatment of serious attrition with defect of dentition in old people

Objective:To summarize the expierence in the prostheti c treatment of serious attrition with defect of dentition in old people. Methods: Removable denture with occlusal pad was one timely prepa red for 32 people aged 58~80 years old with serious attrition and defect of de n tition. 6 of the patients had temporomandibular disturbance (TMD). The time of f ollow up visit of the patients ranged from 2.5 months to 5 years.Res ults: The masticatory function of the patients was improved obviousl y .Unilateral mastication was achieved. The symptom of articulation was all rel ieved.The occlusal relationship was not changed or occlusal vertical dimension ( OVD) was appropriately increased in the patients without TMD. The symptom of TMD did not appear after using denture in patiends with TMD.Conclu sions:During occlusal reconstruction in old people with serious attri tion and defect of dentition, the recovery of OVD and occlusal relationship is v ery important.
10.Preparation and Curative Effects of Different Compound Scald Preparations for Deep Ⅱ? Scald

OBJECTIVE:To prepare different compound scald preparations and to investigate their repairing effects for deep Ⅱ? scald in rabbits.METHODS:The deep Ⅱ? scald model was established in depilated rabbits by infrared radiation.Then the rabbits in the trial groups were treated with the same drug at different dosage and different forms of medication:compound scalded gels,compound scalded ointments and the compound scalded films,with Moist Exposed Burn Ointment as positive control and 0.9% NaCl aqueous solutions blank.After a certain time of treatment,the contraction of the scald skin was observed and the average value of the diameter of the scald skin was calculated.RESULTS:In three trial groups and Moist Exposed Burn Ointment positive control group,no scald infection occurred,and the healing surfaces of the scalds were all smooth and even.However,the healing time was different between 3 trial groups and Moist Exposed Burn Ointment positive control group,with compound scalded gels showing the best efficacy in that the scald surfaces were free from infection,the healing surfaces were smooth and even,and the contracted diameter of the scalded skin was significantly lower than in other groups,showing significant differences as compared with other groups(P
