1.Investigation of the effect of VEGF antisense oligodeoxyribonucleotides in ischemic retinopathy

Objective To investigate the effect of suppression of ischemia induced retinal neovascularization by VEGF antisense oligodeoxyribonucleotides. [WT5”HZ]Methods [WT5”BZ]Mouse models of hyperoxia induced ischemic retinopathy were established. Retrobulbar injections were performed with VEGF antisense oligodeoxyribonucleotides or NS in 4 groups:normal control and various doses respectively. The nuclei of new vessel buds extending from the retina into the vitreous in different groups were counted and compared under the light microscope. Results There were plenty of new vessel buds in the eyes of mice in hyperoxic condition., while the number of the nuclei of new vessel buds is less in the murine eyes with retrobulbar injection of VEGF antisense oligodeoxyribonucleotides,especially the nuclei were redused with 59.3% in eyes with large dose. Conclusion The proliferation of retinal new vessel may be suppressed by using the retrobulbar injection of VEGF antisense oligodeoxyribonucleotides.
2.THE MORPHOLOGICAL AND CELLULAR DYNAMIC CHANGES IN ARSENIC TRIOXIDE-TREATED RAT LIVER CANCER

Objective To study the efficiency of the treatment of experimental hepatocellular carcinoma in rats with arsenic trioxide and to elucidate the possible mechanism.Methods Wistar rats were fed with diethylnitrosamine (DEN) to induce HCC, then treated with As2O3. The histological changes in liver tissue were observed under microscope, and the cellular dynamic parameters were studied by flow cytometry. Results Treatment with As2O3 caused HCC cells death via both apoptotic and non-apoptotic mechanisms when the dose was high (5 mg/kg), the necrosis was seldom and apoptosis was common when the dose was appropriate (1 mg/kg). Proliferation index (PI) decreased sharply in high-dose (5 mg/kg) group (P<0.01), but not in other two (1 mg/kg, 0.2 mg/jg) groups (P>0.05). However, S phase fraction (SPF) decreased dramatically in all three groups (P<0.01). Although apoptosis of HCC cells was common in all three groups, it reached the top only when the dose (1 mg/kg) was appropriate (P<0.001), and it was obviously accompanied with accumulation of cells in G2/M (G2/M restriction). Conclusion These date demonstrate that arsenic trioxide induces apoptosis of rat HCC cells, and it is closely associated with G2/M restriction when apoptosis reaches the top. The data also suggest that arsenic trioxide can inhibit cell proliferation, which is dose-dependent and time-dependent. The fact that continuous intermittent i.p. injection of arsenic trioxide can also be effective may afford a novel way to use the drug more safely.
3.A manufacture method of schistosomiasis epidemic maps based on Google Earth

Objective To make various schistosomiasis epidemic maps based on Google Earth. Methods The various ele-ments for schistosomiasis epidemic maps were marked in the Google Earth platform by adding the place mark,path,polygon, overlay and so on. Results Various schistosomiasis epidemic maps were produced and saved,such as the schistosomiasis epi-demic area map of the city,the map of Oncomelania hupensis snail distribution in the town,and the schematic map of snail envi-ronments. Conclusion The schistosomiasis epidemic maps based on Google Earth are clear and visual. The production process is very simple and easy to learn. It is suitable for the use in the grass-root schistosomiasis control stations.

7.Establishment of Oncomelania hupensis snail database based on smartphone and Google Earth

Objective To establish an Oncomelania hupensis snail database based on smartphone and Google Earth. Meth?ods The HEAD GPS software was loaded in the smartphone first. The GPS data of the snails were collected by the smartphone. The original data were exported to the computer with the format of KML/KMZ. Then the data were converted into Excel file for?mat by using some software. Finally the results based on laboratory were filled and the digital snail data were established. The data were converted into KML and then were showed by Google Earth visually. Results The snail data of a 5 hm2?beach along the Yangtze River were collected and the distribution of the snails based on Google Earth was obtained. The database of the snails was built. The query function was implemented about the number of the total snails the living snails and the schistosome infected snails of each survey frame. Conclusion The digital management of the snail data is realized by using the smartphone and Google Earth.
8.Expression of COX-2 and VEGF in non - Hodgkin lymphoma and its clinical significance

Objective To investigate the expression and significance of COX-2 and VECF in NHL Methods The expression of COX-2 and VECF in 28 cases of NHL was detected by immunohistochemistry, and 22 patients with benign lymphadenopathy were chosen as controls. Results Positive rates of COX-2 and VEGF were 82.1 %(23/28) and 78.6 %(22/28) in NHL respectively, significantly higher than that in the control (P <0.01). The expression rate of COX-2 was positively correlated with that of VEGF in tissues of NHL (χ~2 =10.50, P<0.01). Conclusion The expression of COX-2 and VEGF are high in NHL COX-2 and VEGF might have a common induction pathway, and selective COX-2 inhibitor might be a new drug for treatment of NHL in the future.
9.Efficacy analysis of 48 cases with newly diagnosed acute promyelocytic leukemia

Objective To evaluate clinical treatment method and efficacy of newly diagnosed acute promyelocytic leukemia (APL),and analyze the relevant factors about the long-term survival.Methods The clinical data of 48 patients with newly diagnosed APL were analyzed retrospectively.All of them used alltrans retinoic acid (ATRA) combined with anthracycline as induction remission therapy.After induction remission,ATRA combined with chemotherapy was used as consolidation therapy,and ATRA,arsenic trioxide and conventional chemotherapy alternated as maintenance therapy.Short-term efficacy was analyzed.Patients were followed up,and the rates of overall survival (OS) and disease-free survival (DFS) were analyzed.Long-term efficacy was analyzed by COX proportional hazards regression models univariate analysis.Results The complete remission (CR) rate was 87.5%(42/48) in all 48 patients with APL.The time from treatment beginning to CR was (30.7 ± 4.6) d.Age was the only factor affecting the rate of CR.The rates of 1-year,3-year and 5-year OS were (87.5 ± 4.8)%,(85.4 ± 5.1)% and (78.3 ± 6.7)% in 48 patients with APL.The rates of 1-year,3-year and 5-year DFS were (97.6 ±2.4)%,(93.9 ±4.2)% and (89.5 ± 5.9)% in 42 patients with CR.COX proportional hazards regression model univariate analysis result showed that the patient' s age,gender,lactate dehydrogenase,diffuse intravascular clotting,risk stratification and bone marrow abnormalities promyelocyte ratio had no correlation with the rate of DFS (P >0.05).Conclusions ATRA combined with anthracycline as induction remission therapy,after induction remission ATRA combined with chemotherapy as consolidation therapy,and ATRA,arsenic trioxide and conventional chemotherapy alternated as maintenance therapy can get a higher rate of CR and long-term survival in patients with newly diagnosed APL.It is worthy of clinical application.
