1.Genetic characteristics of the wild measles viruses isolated in the North, Middle and High-Plateax regions

17 wild-type measles strains were isolated from both sporadic cases and 4 outbreaks in the Northern, Centre, and Highland areas of Vietnam in 2000-2002. The sequence analysis of 450 nucleotides of the most variable COOH- terminus of N gene showed that established in the year 2000 with the representative of MVi/Beijing.CHI/9411, genotype H2 (Clade H) continued circulating in the North and Highland of Vietnam. This study also revealed that wild-type measles strains circulating in the Center fell into genotype HI (Clade H) with the representative of MVi/Hunan.CHI/93. It was the first time that genotype HI was discovered In Vietnam. The difference within each genotype is bigger, nevertheless, no more than 3.0% at nucleotide level. The wild-type strain isolated in Lai Chau belonging to genotype D5 (Clade D) has imported into Vietnam from Thailand was under consideration.

2.Some mutations in CYP21 gene causing congenital adrenal hyperplasia due to 21-hydroxylase deficiency, the relationship between genotype phenotype and the carriers in family.

Background: Impaired synthesis of adrenal hormones because of steroid 21 - hydroxylase deficiency is one of the common inborn errors of metabolism. The disease is caused by mutations in CYP21 gene and inherited as an autosomal recessive trait.Objectives: This study aims to detect some mutations in CYP21 gene as well as study the relationship between genotype - phenotype and the carriers in patients' family. Subjects and method:43 patients with classic 21 - hydroxylase deficiency and 10 patients' parents were analysed CYP21 gene by using PCR techniques with specific primer pairs. The data was collected and analysed by EpiInfo 6.04 and other common medical statistic method. Results:Among 43 children patients of congenital adrenal hyperplasia due to 21-hydroxylase deficiency, the rate between male and female was equavalent. The most frequent mutation causing steroid 21 - hydroxylase deficiency was deletion 8bp of exon 3. Then, was splicing site mutation of intron 2. The mutation of exon 1 (Pr030Leu) was also detected. About genotype - phenotype relationship, nearly all deletions 8bp (80%) and 12 splicing mutations (87.5%) were associated with salt - wasting phenotype. Conclusion: Parents were the carriers corresponding to the autosomal recessive rule.

3.Knowledge and practice of health workers of HIV/TB at Diagnostic Counseling and Testing (DCT) sites

Background: The HIV/TB pandemic is a serious danger for humans. In Vietnam, many NGOs recommended that the supply of DCT services for TB patients is essential in HIV/TB prevention. Assessing knowledge and practice of HIV/AIDS preventive tasks for health workers in a context that tuberculosis combined with AIDS is rising, is imperative. Objectives: (1) To describe the knowledge and practice of health workers about the DCT model on HIV/TB patients. (2) To evaluate the knowledge and practice of HIV prevention of occupational exposure at DCT sites in Ha Noi. Subjects and method: A cross - sectional study carried out on all 30 health workers who provided HIV/ TB at 10 DCT sites of TB units in Ha Noi. Results and conclusions: The percentage of health workers, that had full knowledge about DCT methodology was low (<50%), and about all 3 groups of knowledge was very low (21.4%). 82.5% - 100% of health staffs had implemented completely the process of HIV/TB at DCT sites but not very skillfully. All of them paid attention to general prevention and standard preventive methodology, but their practices were not synchronous and correct. Less than 50% of the health workers had full knowledge about the assessment of exposure risk and treatment after exposure.

5.Characterization of Two New Records of Mucoralean Species Isolated from Gut of Soldier Fly Larva in Korea.

While surveying the diversity of fungi of the order Mucorales, two isolates, EML-PUKI12-1 and EML-PUKI06-1, were obtained from the gut of soldier fly larvae inhabiting the bulrush at a pond located in the Chonnam National University Arboretum, Gwangju, Korea. The isolates were confirmed as Mucor irregularis and Mucor fragilis species, respectively, based on the morphological characteristics and phylogenetic analysis of rDNA internal transcribed spacer region. Such mucoralean species belonging to undiscovered taxa has not previously been described in Korea.

A seed-borne fungus, Curvularia sp. EML-KWD01, was isolated from an indigenous wheat seed by standard blotter method. This fungus was characterized based on the morphological characteristics and molecular phylogenetic analysis. Phylogenetic status of the fungus was determined using sequences of three loci: rDNA internal transcribed spacer, large ribosomal subunit, and glyceraldehyde 3-phosphate dehydrogenase gene. Multi loci sequencing analysis revealed that this fungus was Curvularia spicifera within Curvularia group 2 of family Pleosporaceae.

During fungal diversity surveys of the order Eurotiales in Korea, two fungal strains, EML-DG33-1 and EML-NCP50, were isolated from samples of rat dung and fig tree leaf collected at a garden located in Gwangju in 2014. To complete the National Species List of Korea, it is a prerequisite to verify whether many questionable species, which were previously recorded but not confirmed, indeed present in Korea. Herein, the isolates were confirmed as undescribed species, Paecilomyces variotii and Talaromyces amestolkiae based on the combination of morphological and phylogenetic analyses of multigenes including the rDNA internal transcribed spacer, β-tubulin, and RNA polymerase II subunit 2.

8.Effects of Compounds from Physalis angulata on Fatty Acid Synthesis and Glucose Metabolism in HepG2 Cells via the AMP-activated Protein Kinase Pathway

The ability of the total extract from Physalis angulata; three fractions after partitioning with n-hexane, ethyl acetate (TBE), and water; and four withanolides (compounds 1 – 4) to phosphorylate 5'-adenosine monophosphate-activated protein kinase (AMPK) and acetyl-CoA carboxylase (ACC) in HepG2 cells was evaluated. The TBE fraction (50 μg/mL) activated p-ACC and p-AMPK expression most strongly. Compounds 1 – 4 (10 μM) upregulated p-ACC expression at different levels. Compound 4 induced the most significant changes in p-AMPK expression, followed by 1 and 2. Sterol regulatory element-binding proteins (SREBPs) play a functional role in the transcriptional regulation of the lipogenic pathway, including fatty acid synthase (FAS) and ACC. The effects of compounds 2 and 4 (10 μM) on FAS and SREBP-1c expression under high glucose conditions (30 mM) in HepG2 cells were evaluated further. Both dose-dependently inhibited FAS and SREBP-1c expression as well as lipid accumulation (1 – 10 μM) were compared to high-concentration glucose control, which upregulated FAS and SREBP-1c. These results suggest that compounds 2 and 4 upregulate AMPK, suppress FAS and SREBP-1c, and have potential effects on glucose and lipid metabolism.
9.Characterization of Two New Records of Zygomycete Species Belonging to Undiscovered Taxa in Korea.

During a biodiversity survey of undiscovered taxa in Korea, two zygomycetous fungal strains were isolated. The first strain, EML-FSDY6-1 was isolated from a soil sample collected at Dokdo Island in the East Sea of Korea in 2013, and the second strain, EML-DG-NH3-1 was isolated from a rat dung sample collected at Chonnam National University garden, Gwangju, Korea in 2014. Based on the morphological characteristics and phylogenetic analysis of the internal transcribed spacer, 18S and 28S rDNA, actin and translation elongation factor-1α genes. EML-FSDY6-1 and EML-DG-NH3-1 isolates were confirmed as zygomycete species, Absidia pseudocylindrospora and Absidia glauca, respectively. Neither species has previously been described in Korea.

Using dilution plating method, 47 fungal isolates were obtained from a soil sample collected from Dokdo in the East Sea of Korea in 2013. In this study, two fungal isolates, EML-MFS30-1 and EML-DDSF4, were confirmed as undescribed species, Metarhizium guizhouense and Mortierella oligospora in Korea based on current classification system using multi loci including rDNA internal transcribed spacer, large subunit, small subunit, and beta-tubulin (BTUB) genes. Herein, detailed morphological descriptions on characters of the undescribed fungal species as well as their molecular phylogenetic status are provided with comparisons to related species.
