1.Histomorphological studies on hyperplastic prostate of castrated rat caused by androgen.

OBJECTIVESTo investigate prostatic histomorphological changes of rat benign prostatic hyperplasia (BPH) models.
METHODSAfter castrated, SD rats were injected subcutaneously testosterone propionate to induce the BPH. Water substitution method was used to measure prostatic volumes. Prostatic tissue were stained by hemoloxylin and eosin, the morphometric changes of glandular and interstitial tissues were semi-quantified by image analysis system. Multiple linear regression was adopted to analyze the results.
RESULTSIn comparison with normal group, prostatic volumes were significantly enlarged (P < 0.01) with glands expanded and interstitial tissues increased in BPH model. Glandular average diameters, volumes and surface areas in unit volume, as well as glandular circumferences and glandular relative total volumes were all significantly increased (P < 0.01). Glandular counts, density, ratio of glandular surface area to volume. And glandular average curvature were all declined (P < 0.05-0.01), so was interstitial circumference (P < 0.01). But volume density had no changes, and relative total interstitial volumes were obviously increased. Prostatic volume was significantly correlated with glandular relative total volume (r = 0.989, P < 0.001) and interstitial relative total volume(r = 0.789, P < 0.001). Prostatic volume was also correlated significantly with glandular average volume(r = 0.789, P < 0.001).
CONCLUSIONSAction of androgen on rat prostate may mainly lie in glandular epithelial hyperplasia, which manifests enlargement of glandular lumen accompanied by hyperplasia of interstitial tissues.

2.Effects of alcohol intake on penile structure and function in rats.

OBJECTIVETo investigate the effects of alcohol intake on penile structure and function in rats.
METHODSThirty adult male Wistar rats were randomly divided into two groups: control group and alcohol intake group. They were administered with 2 mL of normal saline and 40% alcohol solution respectively through gastric tubes every day. Three months later, the animal model of alcohol intake was evaluated by modified Nayagida's method, and the effects of alcohol on the rats were studied by sexual behavior, the number of apomorphine-induced penile erection, level of testosterone in the sera, and the content of penile smooth muscle.
RESULTSThe scores of animal model of alcohol intake evaluated by Nayagida's method were 0.66 +/- 2.05 in the control group and 9.26 +/- 5.50 in the alcohol intake group (P < 0.05), which indicated that an animal model of alcohol intake was successfully established. Sexual behavior, the number of apomorphine-induced penile erection, testosterone level in the sera, and the content of penile smooth muscle of the alcohol intake group were all statistically different as compared with the control group (P < 0.05).
CONCLUSIONAlcohol intake induces sexual dysfunction in rats, which may be due to the decline of testosterone level in the sera and decline of penile smooth muscle.

3.Oral Testosterone Undecanoate Capsules combined with Qilin Pills for late-onset hypogonadism in males.

Objective:
To investigate the clinical effects of oral Testosterone Undecanoate Capsules (TUC) combined with Qilin Pills (QLP) on late-onset hypogonadism (LOH) in men.
METHODS:
Sixty-three LOH patients meeting the inclusion criteria were randomly divided into a control group (aged [48.4 ± 6.2] yr, n = 32) and an experimental group (aged [47.2 ± 5.6] yr, n = 31) to be treated with oral TUC (80 mg, qd) and TUC + QLP (6g, tid), respectively, both for 3 months. Comparisons were made between the two groups of patients in the IIEF-5 scores, total testosterone (TT) levels, and scores in the Aging Males' Symptoms (AMS) scale before and after treatment.
RESULTS:
After treatment, the patients of the experimental group, as compared with the controls, showed a significantly increased IIEF-5 score (21.7 ± 5.8 vs 15.9 ± 4.7, P <0.05) and TT level ([16.7 ± 2.2] vs [13.1 ± 2.8] nmol/L, P <0.05), but a decreased AMS score (20.7 ± 5.7 vs 31.3±6.5, P <0.05).
CONCLUSIONS
TUC combined with Qilin Pills has a better effect and a lower rate of adverse reactions than TUC used alone in the treatment of late-onset hypogonadism in males.

4.Changes of serum sex hormone levels in male workers exposed to cadmium.

OBJECTIVETo investigate the changes in serum levels of sex hormones in male workers occupationally exposed to cadmium (Cd).
METHODSEighty-five Cd-exposed workers in a cadmium refinery in the south China and 76 local healthy subjects as control were selected in the study. Air samples in the workplaces were collected and detected for Cd concentration. Urinary Cd (UCd) level of the workers was measured by graphite atomic absorption spectrometry (AAS) and adjusted by urine level of creatinine (Cr), as an indicator of Cd-burden in the body of all subjects. Also, their serum levels of testosterone (T), follicle stimulating hormone (FSH) and luteinizing hormone (LH) were determined with radioimmunoassay and enzyme immunoassay, respectively, and dose-effect relationship was evaluated.
RESULTSThe serum testosterone levels in Cd-exposure group with 10.9-21.9 and > 21.9 micro mol/mol Cr were 13.00 and 11.37 nmol/L, significantly higher than that (9.31 nmol/L) in those with 0.0-2.2 micro mol/mol Cr. Significantly more increased level of LH (4.11 and 4.32 U/L) was detected in heavy exposure group in the workshop for electrolysis than in control group (2.52 U/L) and in the group with 0.0-2.2 micro mol/mol Cr of UCd (2.64 U/L). No changes in serum level of FSH were found related to Cd exposure.
CONCLUSIONOccupational Cd exposure could independently contribute to the changes of serum levels of sex hormone in male workers.

5.Cigarette smoke affects sexual function of male rats.

OBJECTIVETo study the effect of cigarette smoke on the sexual function of male rats.
METHODSBased on Ozyurt's smoking model, we equally divided 30 male adult Sprague-Dawley rats into a control and a smoking group, and exposed the latter to cigarette smoke for 60 days. A week before the end of the experiment, we added 5 female rats to each group and observed their mating through 24-hour video surveillance. Sixty days later, all the rats were killed for the determination of the level of testosterone (T) in the plasma and the activity of nitric oxide synthase (NOS) in the corpus cavernosum, and Masson-dyeing image analysis of the penile tissue.
RESULTSCompared with the controls, the rats in the smoking group showed a significant reduction in the times of mating, the level of plasma T (P < 0.05) and the activity of NOS in the penile cavernous tissue (P < 0.05), but a slight increase in the collagen fibers and obvious changes in the blood sinuses.
CONCLUSIONCigarette smoke seriously affected penile erection in the experimental rats. The decrease in plasma T, NOS activity and the area of smooth muscle may be an important mechanism underlying their erectile dysfunction.

6.Testosterone-induced spermatogenic impairment is associated with looser arrangement of spermatogenic cells in rats.

OBJECTIVETo determine whether testosterone-induced intra-testicular testosterone withdrawal and therefore spermatogenic impairment is associated with looser arrangement of spermatogenic cells in rats.
METHODSAdult male SD rats received intramuscular injection of testosterone undecanoate at 19 mg/(kg x 15 d) for 130 days, and then testicular tissue blocks were obtained for the preparation of methacrylate resin-embedded sections and observation of the changes in testicular histology.
RESULTSApart from such changes as impaired spermiogenesis and spermiation, apparently looser arrangement of spermatogenic cells was seen in 11.5% of the seminiferous tubule profiles, with radial cracks (empty spaces) running towards the tubule lumen being formed between lines, bundles or groups of spermatogenic cells (mainly spermatids and spermatocytes).
CONCLUSIONLooser arrangement of spermatogenic cells is one of the key histological changes resulting from intra-testicular testosterone withdrawal in rats.

7.A tentative research of testosterone supplement therapy on male senile dementia.

OBJECTIVESTo understand the hormone levels in male senile dementia patients and the effects of testosterone supplement theraphy (TST) on male senile dementia.
METHODSThe general health condition, mental health condition, hormone levels, and some other aspects were evaluated in 9 patients with male senile dementia without significant contradiction for TST. 9 patients were treated with oral testosterone undecanoate 120 mg per day for six weeks, and relative changes were observed, detected and recorded.
RESULTSIn 9 cases of male senile dementia, the average score was 5.33 in mini-mental state examination (MMSE), 14.56 in Hamilton depression scale (HAMD), 5.0 in geriatric depression scale (GDS), 32.56 in partial androgen deficiency of the aging male (PADAM) measuring form. The volume of testis was small (8.33 ml on average). After 1 week of having finished TST, the levels of serum testosterone (T) dropped significantly (dropped from average 2.88 ng/ml to 2.47 ng/ml, P < 0.05) in 9 patients with TST. General evaluation with clinician's global impression(CGI) for 9 patients showed 2 cases fail and 7 cases with progress (3 of them progress significantly). Average score dropped in HAMD form (10.67) and PADAM form (28.11). Physical strength and psychological status improved significantly, but vasomotor symptoms and sexual function failed response to the treatment.
CONCLUSIONSThe levels of androgen is low, and the symptoms of androgen deficiency are readily seen in male senile dementia. TST can improve life quality of most patients to some extent.

8.Microdissection testicular sperm extraction for patients with non-mosaic Klinefelter's syndrome: An update.

9.Hormonal replacement therapy and aging: Asian practical recommendations on testosterone supplementation.

Profound and diffuse alterations in the production of gonadal and adrenal androgens as well as growth hormone are associated with aging. To convey this concept more appropriately, partial endocrine deficiency in the aging male (PEDAM) was introduced as a term for the phenomenon of hormonal alterations in the aging male. Hormones responsible for some of the manifestations associated with male aging are testosterone, growth hormone, dehydroepiansdrosterone (DHEA), melatonin, thyroid hormones and leptin. Of these, testosterone has been widely investigated and its beneficial and adverse effects on male bodily systems are relatively well established. However, a serious body of confusion and misunderstandings surrounding the diagnosis, treatment and monitoring of men suspected of having androgen deficiency has been raised. Therefore, it is timely to provide practical criteria for diagnosis and treatment to avoid misconception about the use of testosterone in the aging male. To provide an understanding and information of the issues, the following headings are summarized: (1) Important clinical consideration on testosterone supplementation in the aging male; (2) Asian practical recommendations on testosterone supplementation in the aging male.

10.Sodium 4-phenylbutyrate Attenuates High-fat Diet-induced Impaired Spermatogenesis.

OBJECTIVE:
To determine the mitigating effects of sodium 4-phenylbutyrate (4-PBA) on high-fat diet (HFD)-induced spermatogenesis dysfunction.
METHODS:
Male rats (n = 30) were randomly divided into three groups: control, HFD, and 4-PBA (HFD +4-PBA). After 13 weeks, rats were euthanized. Testes and epididymis were harvested for further analysis. Sex hormones were detected, and hematoxylin and eosin staining was performed to examine the histological changes in the testes. Semen samples were collected to evaluate sperm quality. Spermatogenic cell apoptosis was detected by TUNEL assay.
RESULTS:
Compared with the control group, the final body weight and body weight gain were significantly higher in HFD-fed rats, while the testicle/body weight ratios were lower (P < 0.05). In HFD-fed rats, obvious pathological changes in the testicular tissue were observed. Treatment with 4-PBA attenuated HFD-induced histological damage, ameliorated the HFD-induced decrease in serum testosterone (T), and reduced the rate of testicular cell apoptosis (P < 0.05) in obese male rats. Finally, 4-PBA significantly improved semen parameters in HFD rats (P < 0.05).
CONCLUSION
HFD exposure induced detrimental effects on spermatogenesis, semen quality, serum T level, and testicular cell apoptosis in rats. Treatment with 4-PBA ameliorated HFD?induced impaired spermatogenesis via inhibition of apop-tosis in rats. 4-PBA may have therapeutic value in the treatment of obesity?related impairment of spermatogenesis.
