1.Effects of IGF1 on proliferation and migration of endothelial cells on high glucose

Objective To investigate the effects and the possible mechanism of insulin like growth factor-1 ( IGF1 ) on proliferation and migration of human umbilical vein endothelial cells ( HUVEC) in high glucose. Methods Cultured HUVEC were divided into 5 groups( control group, high glucose group, hypertonic group, high glucose+IGF 1 group, high glucose+IGF1+ AZD5363 group) . The proliferation was detected by methyl thiazolyl tetrazolium ( MTT) , the migration was detected by transwellchamber. The mRNA expressions of serine/threonine kinase( Akt) and FoxO1 were detected by real-time PCR. The protein expressions of FoxO1/p-FoxO1 and Akt/p-Akt were detec-ted by immunohistochemisty. Results Compared with the control group, the rate of cell survival in high glucose group decreased significantly(P<0. 05), the transitional was decreased significantly(P<0. 05), the mRNA ex-pression of FoxO1 was increased significantly( P<0. 05 ) , the protein expressions of FoxO1/p-FoxO1 and Akt/p-Akt were increased. Compared with high glucose group, the cell survival rate and migration rate in high glucose+IGF1 group were increased significantly, the mRNA expression of FoxO1 was decreased significantly(P<0. 05), the protein expressions of FoxO1/p-FoxO1 and Akt/p-Akt were decreased. Compared with high glucose group, the cell survival rate and migration rate in high glucose+IGF1+AZD5363 group had no significant difference, the mR-NA expression of FoxO1 was decreased significantly ( P<0. 05 ) , the protein expressions of FoxO1/p-FoxO1 had no significant difference. The mRNA expression of Akt in five groups had no significant difference. Conclusion IGF1 can improve the proliferation and migration of endothelial cell in high glucose. It shows that IGF1 acts through Akt to promote FoxO1 phosphorylation.
2.Literature Analysis on Syndrome Types and TCM Prescriptions of Behcet's Disease

Objective To analyze syndrome types and TCM prescriptions of Behcet's disease;To summarize the regularity of TCM syndrome differentiation and treatment principles of Behcet's disease.Methods Related literature in CNKI and VIP database from Jan. 1990 to Oct. 2013 was searched. The syndrome patterns, characteristics of prescriptions and frequency of syndrome were summarized.Results There were 20 syndrome types of Behcet's disease in the 58 collected literature. Damp and hot accumulating poison, spleen and kidney Yang deficiency, liver and kidney Yin deficiency, and hot and damp were common syndromes, occupying 55.4%. Hot and damp occupied 41.3%;Yin deficiency occupied 18.1%;Yang deficiency occupied 12.9%. Liver, spleen and kidney were the main disease locations, occupying 94.4%. There were 197 Chinese herbs in total, and could be classified and further combined into 19 types, among which heat-clearing drugs, tonics, activate blood and removing blood stasis were commonly adopted, occupying 50.6%. There were 66 prescriptions collected, among which 31 were set prescriptions, and 35 were prescriptions formed by self-experience. Longdan Xiegan Decoction and Gancao Xiexin Decoction were commonly adopted.Conclusion Syndrome types and TCM prescription characteristics of Behcet's disease had certain regularity, which can provide reference for clinical diagnosis and treatment.
4.Early Goal Lactate Clearance Rate Therapy in Patients with Severe Sepsis or Septic Shock:A Meta Analysis

Objective To investigate the effect of early goal lactate clearance rate therapy in patients with severe sepsis or septic shock.Methods Articles were retrieved from PubMed,Embase,Cochrane Library,Wanfang and CNKI before March 5,2016.Inclusion criteria included the subjects concerning patients with severe sepsis or septic shock reported as randomized controlled trial (RCT),which endpoints were the mortality,the length of ICU stay and hospital stay.RevMan 5.3 software was used for Meta analysis.Results There were 5 RCTs meeting inclusive criteria including 860 patients.It was shown by Meta analysis that early goal lactate clearance rate therapy was associated with decrease in the 28-day mortality(RR =0.73,95% CI:0.60-0.88,P < 0.01),shorten the length of ICU stay (WMD =-2.41,95% CI:-4.68--0.14,P < 0.05),but not associated with decrease of the length of hospital stay(WMD =-0.13,95 % CI:-4.58-4.31,P =0.95).Conclusion Early goal lactate clearance rate therapy was associated with significant improvement in 28-day mortality and the length of ICU stay but not with the length of hospital stay in patients with severe sepsis or septic shock.
5. Studies on the correlation of PON1 gene rs854572 single nucleotide polymorphism to clopidogrel resistance

Objective To investigate the correlation of the single nucleotide polymorphism (SNP) of PON1 (Paraoxonase-1) gene rs854572 to the occurrence of clopidogrel resistance (CR). Methods A case-control method was employed in present study. A total of 850 hospitalized patients with coronary artery diseases (CAD) in General Hospital of Shenyang Command were enrolled. The residual platelet aggregation rate (RPA) induced by 20μmol/L of adenosine diphosphate (ADP) was detected by optical nephelometry, and RPA≥70% was defined as CR. Accordingly, all the enrolled 850 patients were then divided into CR group (n=215) and non-CR (NCR) group (n=635). Polymerase chain reaction (PCR) and pyrophosphate sequencing were executed to determine the genotypes and the allele frequencies of PON1 gene rs854572. Results The genotype frequencies in rs854572 of PON1 gene conformed well to the Hardy-Weinberg equilibrium in both CR group and NCR group. Three frequencies of genotype CC, CG and GG were 23.7%, 49.3%, 27.0% in CR group, and 24.1%, 50.2%, 25.7% in NCR groups, respectively. No significant difference in genotype and allele frequency existed between CR group and NCR group (P=0.93 and 0.76, respectively). Logistic regression analysis revealed that no correlation between rs854572 SNP of PON1 gene and the formation of CR in patients with CAD after adjustment of correspondent factors including age, gender, body mess index, smoking, hypertension, diabetes mellitus and hyperlipidemia. Conclusions It is considered that no correlation exists between PON1 gene rs854572 polymorphism and clopidogrel resistance in patients with coronary heart disease.
6.Risk and Precautions against Single Dispensing in Emergency Pharmacy

OBJECTIVE:To reduce the risk of single dispensing,and to improve the quality of pharmaceutical care in emergen-cy pharmacy. METHODS:Single dispensing environmental factors,human factors and social factors of emergency pharmacy in our hospital were analyzed;the suggestions and measures were put forward,risk intervention mechanism was established and manage-ment effects were evaluated. RESULTS:Through improving the pharmacy hardware and software,carrying out the relevant sys-tem,enhancing training,improving the process of dispensing and window order,promoting the drug management and carrying out near error management. Compared with before management(Oct.-Dec. 2014),the times of near error and disputes significantly re-duced after management(Feb.-Apr. 2015)(P<0.05),and medication error reduced to zero. CONCLUSIONS:As single dispens-ing,risk management and the improvement of prescription audit ability can improve pharmaceutical care in emergency pharmacy.
7.Nicotine effects on the osseointegration of implants with different treatments

BACKGROUND:Studies have confirmed that nicotine affects the activity of osteoblasts, osteoclasts, fibroblasts and erythrocytes.
OBJECTIVE:To study the nicotine effects on osseointegration and the expression of osteoprotegerin and bone morphogenetic protein 2 after implantation of dental implants with surface treatment by sandblasting or acid etching.
METHODS:Twenty-four Sprague-Dawley rats were randomly assigned to two groups and received daily injections for 2 weeks as folows: Nicotine 2 mg/kg twice for experimental group, saline solution for control group. Then the titanium implants with surface sandblasting or acid etching were implanted into the tibiae folowed by continuous nicotine or normal saline injection. At weeks 2 and 4 after implantation, the implants and surrounding bone tissue were prepared for CT, X-ray and hematoxylin-eosin staining examinations to evaluate bone healing and expression levels of bone-related genes were measured by quantitative RT-PCR.
RESULTS AND CONCLUSION: Compared with the control groups, the degree of osseointegration and the expression of osteoprotegerin and bone morphogenetic protein 2 in the experimental groups were decreased significantly (P < 0.05), except that the expression of bone morphogenetic protein 2 in the experimental group with acid etching was not significantly reduced. In addition, the expression of bone morphogenetic protein 2 in the experimental group with acid etching was higher than that in the experimental group with sandblasting at 2 weeks after implantation (P < 0.05). The X-ray and CT show that the quantities of new generation bone and the degree of bone mineralization of the sandblasting group were significant lower than those of the acid etching group under the intervention of nicotine. Hematoxylin-eosin staining showed that the activity and quantity of osteoblasts around the implants down-regulated significantly, but acid etching-treated implants showed better outcomes than sandblasting-treated implants.
8.Advances on Genetic Research of Ossification of the Posterior Longitudinal Ligament

[Summary] The ossification of the posterior longitudinal ligament ( OPLL ) is caused by environmental , genetic and other factors.With the development of genomics research , researchers have found that mutations are important factors inducing OPLL . Despite the genomics research on OPLL has made important advances , no highly correlation has been found between OPLL and the exact single nucleotide polymorphisms (SNPs) loci.The pathogenesis of OPLL needs further exploration .

With data support from abundant research ,epithelial growth factor receptor ( EGFR) signaling pathway is associated with radiotherapy resistance .EGFR inhibition has become one of methods to increase radio-sensitivity.As the first EGFR monoclonal antibody developed by China and Cuba ,nimotuzumab demonstrates its distinct clinical characteristics and has been extensively explored in head and neck cancer , esophageal cancer , high grade glioma .This article mainly reviews the present situation and recent advances on the radiosensitivity re -search of nimotuzumab .
10.Meta Analysis of the Association of Polymorphism in IL-28B Gene and Virological Response to Interferon in HBeAg-positive Chronic Hepatitis B

Objective To explore the association between the polymorphism in IL-28B gene and the interferon virological response in HBeAg-posi-tive chronic hepatitis B patients. Methods A literature search was performed on PubMed,web of Science,China Bio Medicine(CBM)and wan-Fang database. The references was screened with the inclusion and exclusion criteria,and then data were extracted and methodological quality of the studies was judged. Statistical analysis was carried out with RevMan 5.3 and STATA 11.0. Results Totally 7 publications were recruited for the study,including 1 345 patients. The meta-analysis showed that there was no significant difference of interferon response between HBeAg-positive chronic hepatitis B patients with rs8099917TG/GG alleles and rs8099917TT alleles (P= 0.20). There was no statistical difference between Rs12979860CC and CT/TT allele for the sustained virological response(P=0.75). Conclusion Our current meta-analysis suggests that there was no correlation of rs8099917,rs12979860 polymorphism in IL-28B with the sustained virological response to interferon in HBeAg-positive chronic hepatitis b.
